As for Services of the Public Libraries toward the Youth (공공도서관의 청소년들에 대한 봉사에 대하여 - 학교도서관과의 협조적 측면에서)

  • The following results have been driven out from a survey of the actual conditions of the public libraries service in Korea, made through questionnaire in reference with the actual service for youth that are presently done by the foreign countries' libraries, in order to prepare a perfect and objective base in establishing a service activities of the public libraries in this country: (1) It was found that the public libraries' loarning hours during the week days which accords with the schooling hours of the schools are shown to 50 %. On Saturdays during Summer season seven libraries and during Winter season twelve libraries would close their loaning before 17 : 00 hours, and on Holidays and Sundays fifteen libraries are not doing their loarning. (2) The followings are the present status of the service activities for the youth by the public libraries: a. It was found that they do introduction activity (50 %) of newly published books as a service for inducing reading interestings of the youth, and except this it was found that others are on beginning. b. The service activities by the data for the visual and auditory sense are almost negelected. and the rate of doing such activity is shown to 9.74%. c. It has been found that little information service activity is for inducing the youth to the libraries. d. As a cooperative activity with the school's, a passive service activity to have pursuaded to visit the public libraries by each class, are mostly being done (27.27%). Some research meeting for solving all the matters between the school libraries are not held mostly.

Selection method of sports talents using physical activity promotion system (건강체력평가시스템을 활용한 스포츠영재 선발방법)

  • There are many problems to single sports talents out of test applicants. The physical activity promotion system has been performed to all elements school students of both 5 and 6 grades in order to evaluate overall health and physical activities since 2009. This system includes some variables which could measure the students' sports latent power, so that the system could be used to single out sports talents. In this work, we propose a primary screening method that element school teachers might evaluate sports talents based on the data of the physical activity promotion system. Two more sports talent indices which are sports flexibility index and sports endurance index are defined. The selection method of sports talents is developed by using sports latent indices including sports power and cardiorespiratory indices. This method is efficient from the view of time and cost aspects, since we do not need to remeasure all elements school students again.

  • The purpose of this study was to analyze the elementary teachers' conceptions on the inquiry activity about 'make a fossil model' focusing the 6th, 7th, 2007 revised, and 2009 revised elementary science textbook. This study is intended to find causes of teachers' conceptions and to derive educational implications. We have analyzed questionnaires which were collected from 77 elementary school teachers. It was carried out about comparisons actual fossil with fossil model in each step of the inquiry activity, the lesson goal, the thought for changes of the inquiry activity, the improvement of the inquiry and conception related fossil. Teachers have a general understanding about the contents of the inquiry activity but some of them have partially incorrect understanding. And they emphasize the understanding of the concept on lesson goal. It is necessary to improve inquiry activity reflecting teachers' thinking about the merit and weaknesses. There was not statistically difference in understanding of fossil related concepts according to teachers' career, teaching or training experience. Through this research, there is a need to improve a textbook, a guide book, a teachers' training. So, teachers who have professionalism about science textbook be able to teach students.

Analysis of Teaching Strategies, Types of Inquiry Activities and the Relationship between Inquiry Activities and Concepts Presented in Elementary School Science Textbooks - Focusing on Earth Science - (초등학교 과학교과서에 제시된 탐구활동의 교수전략, 유형, 개념과의 연관성 분석 - 지구과학 영역을 중심으로 -)

  • This study was to analysis teaching strategies, types of inquiry activities and the relationship between science concepts and inquiry activities presented in science textbooks. As a result of the study, first, the inquiry-based teaching strategies presented in science textbooks were experiment, simulation, demonstration, and field study. Second, there were 53 inquiry activities presented in 8 units related to the earth science area of science textbooks, and the types of inquiry activities were experimental observation (EO) 18, mock activity (SA) 20, investigation discussion and presentation (IP). It was analyzed as 12, data interpretation (ID) 2, and express (EX) 1 piece. Third, the relationship between inquiry activities and science concepts presented in science textbooks was analyzed. As a result of the analysis, out of a total of 42 inquiry activities, 21 inquiry activities corresponded to EA (explicit activities), in which the result of inquiry activities was directly and explicitly linked to science concepts. And IA (implicit activities), which is an implicit inquiry activity in which science concepts must be inferred using the results of inquiry activities, were analyzed as 21. In particular, IA (implicit activities), which is an implicit inquiry activity, can be said to be the result of reflecting the characteristics of earth science areas where many simulated activities (SA) are used. This is considered to be a matter to be considered in the process of developing various elementary science textbooks in the future.

Biosynthesis of (R)-(-)-1-Octen-3-ol in Recombinant Saccharomyces cerevisiae with Lipoxygenase-1 and Hydroperoxide Lyase Genes from Tricholoma matsutake

  • Tricholoma matsutake is an ectomycorrhizal fungus, related with the host of Pinus densiflora. Most of studies on T. matsutake have focused on mycelial growth, genes and genomics, phylogenetics, symbiosis, and immune activity of this strain. T. matsutake is known for its unique fragrance in Eastern Asia. The most major component of its scent is (R)-(-)-1-octen-3-ol and is biosynthesized from the substrate linoleic acid by the sequential reaction of lipoxygenase and peroxide lyase. Here, we report for the first time the biosynthesis of (R)-(-)-1-octen-3-ol of T. matsutake using the yeast Saccharomyces cerevisiae as a host. In this study, cDNA genes correlated with these reactions were cloned from T. matsutake, and expression studies of theses genes were carried out in the yeast Saccharomyces cerevisiae. The product of these genes expression study was carried out with Western blotting. The biosynthesis of (R)-(-)-1-octen-3-ol of T. matsutake in recombinant Saccharomyces cerevisiae was subsequently identified with GC-MS chromatography analysis. The biosynthesis of (R)-(-)-1-octen-3-ol with S. cerevisiae represents a significant step forward.

Antimicrobial and Anticancer Activity of Korean Traditional Soy Sauce and Paste with Chopi (초피첨가 전통장류의 항균 및 항암활성)

  • The fruits of Zanthoxylum piperitum are known as having various physiology vitality, and the abstraction ingredient of the pericarp is also known as having strong antibiotic activities against various bacteria. Therefore, this study was carried out to estimate the effect of physiology vitality when the abstraction ingredient of Z. piperitum was added in soy sauce(Chopi-kanjang) and soybean paste(Chopi-doenjang). For the antibiotic activity against the pathogens of sitotoxism such as Staphylococcus aureus, Salmonella typhimurium, Vibrio parahemolyticus, Escherichia coli 0157:H7, the extracts of the Chopi-kanjang was added 1%, 2%, 4% pericarp of Z. piperitum in the manufacturing process of soy source. According to the results, the growth of E. coli 0157:H7 and V. parahemolyticus were respectively inhibited as 70% and 50% by the Chopi-kanjang added 2% of the ingredient. For the antibiotic effects of the aforementioned Chopi-kanjang against Sal. typhimurium and Sta. aureus, the growth of those pathogens was also inhibited between 40% and 60% according to the manufacturing period of Chopi-kanjang. It was confirmed that the antibiotic activity using the mixture of the abstraction ingredient and Chopi-doenjang was lower than those of Chopi-kanjang. In order to estimate the anticancer activity using by caspase-3 activity, the mixture of the abstraction ingredient of the pericarp of Z. piperitum and Chopi-kanjang was treated to leukemia cells. According to the results, the activities of caspase-3 using the mixture added 1%, 2% and 4% of the abstraction ingredient were respectively increased as much as 4, 12, 15 times comparing with the control which was treated with the soy source only. It could be that the mixture of the abstraction ingredient of the pericarp of Z. piperitum and soy source induced apoptosis, and the mixture of the abstraction ingredient and soybean paste had no effect on the activity of caspase-3. In order to find out the death of the aforementioned cells caused by necrosis or apoptosis, DNA fragmentation in the cell was examined. U-937 cells showed apoptotic DNA fragmentation in the incubation with Chopi-kanjang extract.

Effect of Reflux Conditions on Extraction Properties and Antioxidant Activity of Freeze Dried-Schisandra chinensis (열수추출조건이 동결건조 오미자의 추출 및 항산화 특성에 미치는 영향)

  • Reflux extraction properties of Schisandra chinensis were investigated with different extraction conditions of ethanol concentration (0-99%), extraction time (2-8 h), and extraction temperature ($40-100^{\circ}C$). Different chemical properties, such as reducing sugars (RS), titratable acidity (TA), Hunter's color values, total phenolic compounds (TPC), and antioxidant activity (DPPH and ABTS assays) were analyzed for the corresponding extracts. The results showed that RS and TA increased as the extraction temperature increased. For each parameter, the maximum value was achieved, when extraction was carried out with 50% ethanol for 8 h at $100^{\circ}C$. Redness ($a^*$) of the extract decreased as all 3 extraction parameters were increased. TPC increased significantly as the extraction time and temperature increased; further, the highest TPC was achieved, when extraction was carried out with 50% ethanol. The same tendency was observed for DPPH and ABTS radical scavenging activities. The highest TPC and antioxidant activity were obtained, when extraction was carried out with 50% ethanol for 4-6 h at $60-80^{\circ}C$, respectively.

Hepatoprotective Effects of Waxy Brown Rice Fermented with Agrocybe cylindracea

  • The methanol extract of waxy brown rice fermented with Agrocybe cylindracea was prepared. The extract was then freeze dried and fed to rats at the level of 0.5, 1.0, 2.0 g/kg body weight for 14 days, followed by the treatment with carbon tetrachloride for three consecutive days to induce hepatotoxicity. After sacrificing the rats, the enzyme activity of aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), lactate dehydrogenase (LDH), and ${\gamma}$-glutamyl transpeptidase (${\gamma}$-GTP) in serum was determined. Biochemical analysis on serum for albumin, total protein, triglyceride, and total as well as HDL-cholesterol were carried out along with a histopathological study of liver tissues. Based on these data, we suggest that the waxy brown rice cultured with A. cylindracea may exert hepatoprotective activity against hepatotoxicity caused by chemicals such as carbon tetrachloride.

Photocatalytic Systems of Pt Nanoparticles and Molecular Co Complexes for NADH Regeneration and Enzyme-coupled CO2 Conversion

  • Natural photosynthesis utilizes solar energy to convert carbon dioxide and water to energy-rich carbohydrates. Substantial use of sunlight to meet world energy demands requires energy storage in useful fuels via chemical bonds because sunlight is intermittent. Artificial photosynthesis research focuses the fundamental natural process to design solar energy conversion systems. Nicotinamide adenine dinucleotide ($NAD^+$) and $NADP^+$ are ubiquitous as electron transporters in biological systems. Enzymatic, chemical, and electrochemical methods have been reported for NADH regeneration. As photochemical systems, visible light-driven catalytic activity of NADH regeneration was carried out using platinum nanoparticles, molecular rhodium and cobalt complexes in the presence of triethanolamine as a sacrificial electron donor. Pt nanoparticles showed photochemical NADH regeneration activity without additional visible light collector molecules, demonstrating that both photoactivating and catalytic activities exist together in Pt nanoparticles. The NADH regeneration of the Pt nanoparticle system was not interfered with the reduction of $O_2$. Molecular cobalt complexes containing dimethylglyoxime ligands also transfer their hydrides to $NAD^+$ with photoactivation of eosin Y in the presence of TEOA. In this photocatalytic reaction, the $NAD^+$ reduction process competed with a proton reduction.

A Comparative Study on the Prevalence of Refractive Errors Between Urban and Rural Primary School Children (초등학생의 지역간 시력차이에 관한 연구 -대구 및 영천지역을 중심으로-)

  • The study was carried out to estimate the prevalence and possible causes of refractive errors in primary school children. At one primary school in urban Daegu, one in the medium-sized city of Yongcheon and 2 in rural areas, children wearing glasses and children with visual acuity of less than 0.7 were given our questions. 354 out of 378 children responded to our questions. The research was done from March 2000 to February 2001. The data were analyzed by percentage, x2 test, t-test, ANOVA and Scheff method. Children with eyesight problems were more prevalent in urban areas than rural areas. The lack of luminosity during reading (p=0.015), length of reading time (p=0.08) and posture which watching TV (p=0.023) appeared leading causes of progressing myopias. The visual impairment caused by refractive errors may affect the mental and physical activity of primary school children and lead to social isolation, loneliness and depression. However, little attention may be paid by professionals engaged in providing health care to the children. Once we have recognized the impact of visual impairment, we may be in a better position to prevent progressing refractive errors.
