Studies on the Culture of Haploid Tobacco Leaf (담배 반수성의 유엽배양에 관한 연구)

  • Young haploid leaf derived from the anthers of tobacco plant was cultuerd and plantlets of various ploidies were obtained. When the leaf was put on the medium supplemented with kinetin as growth regulator, plantlets developed directly from the leaf, and the plants coming out in early stage of culture were all haploid. Plants developing in later stage were mostly haploids with some exception of diploid and aneuploid. Leaves were also cultured on the callus-inducing media supplemented with 2,4-D and kinetiion, and the calluses were sub-cultured for six months. Plants developed from these calluses were mostly aneuploids of various chromosome numbers. In view of the fact that the plants directly developed from the leaf were all haploid, the tissue of the original leaf explant was assumed to be uniform as far as chromosome number was concerned. On the other hand, it seemed that the occurrence of various ploidies in the plants derived from the calluses of same origin was the result of the influence of in vitro culture. Apical meristem tissues and various multicellular bodies were formed in the epidermal and inner mesophyll tissues as well as in the sub-epidermal cells.

PRODUCTION OF GINSENOSIDES THROUGH IN VITRO CULTURE OF GINSENG(Panax ginseng C.A. MEYER)

  • Ginseng root explants and calli induced from selected cell lines were cultured on modified Murashige and Skoog's media supplemented with different concentrations of organic or inorganic compounds and plant growth requlators to clarify the effects of chemical composition and plant growth regulators in the medium on the growth of ginseng calli and the production of ginseng saponin. For optimum growth of calli, the concentrations of 2, 4-D and sucrose were the range of 1 to 3 mg/${\ell}$l and 1 to $3\%,$ respectively. And it was clarified that sucrose, nitrogen, phosphate, calcium, magmesian plant growth regulators and their concentrations influcenced the relative biosynthesis of saponin in tissue cultures of Panax ginseng. The patterns of ginsenosides, pharmacologically useful component, were different among the cell lines and contents of ginsenosides were much higher in selected cell lines than in original cell line.

First Report of Anthracnose Disease Caused by Colletotrichum gloeosporioides in Stonecrop

  • In year of 2013, anthracnose disease symptoms were observed on stem of stonecrop in a plant resource field, Southern Forest Resource Research Center, Jinju, South Korea. The disease symptoms were appeared 2-5 cm smudged irregular black spot color on the plant stems. Suspected pathogen was isolated from the lesion with surface sterilization method and identified it as Colletotrichum gloeosporioides by morphological characteristics and phylogenetic analyses. Artificially inoculated the isolated pathogen led to the similar disease symptom as originally observed in the field. The fungal pathogen was re-isolated from the inoculated lesion and it exhibited the same characteristics compared to the original isolate. This is the first report of anthracnose of stonecrop grown in open fields, caused by Colletotrichum gloeosporioides in Korea.

PLANT CELL WALL WITH FUNGAL SIGNALS MAY DETERMINE HOST-PARASITE SPECIFICITY

  • For improvement of plants in disease resistance, it is most important to elucidate the mechanism to perceive and respond to the signal molecules of invaders. A model system with pea and its pathogen, Mycosphaerella pinodes, showed that the fungal elicitor induced defense responses in all plant species tested but that the suppressor of the fungus blocked or delayed the expression of defense responses and induced accessibility only in the host plant. In the world, many researchers believe that the pathogens` signals are recognized only on the receptors in the plasma membranes. Though we found that the ATPase and polyphosphoinositide metabolism in isolated plasma membranes responded to these fungal signals, we failed to detect specific actions of the suppressor in vitro on these plasma membrane functions. Recently, we found that ATPase (NTPases) and superoxide generating system in isolated cell wall were regulated by these fungal signals even in vitro, especially, by the suppressor in a strictly species-specific manner and also that the cell wall alone prepared an original defense system. The effects of both fungal signals on the isolated cell wall functions in vitro coincide perfectly with those on defense responses in vivo. In this treatise, we discuss the key role of the cell wall, which is plant-specific and the most exterior organelle, in determining host-parasite specificity and molecular target for improvement of plants.

A Study on the Determination of Starting Head by Comparing The Generating Power in Single Action Tidal Power Plant (발전량 비교를 통한 창조식 조력발전의 기동낙차 결정에 관한 연구)

  • Because of its predictability of the energy cycle and huge scale power output, the ocean energy from tidal power utilization has always received attention as a great energy source, even though its development cost, including the embankment construction, is so much higher than that of any other energy source. Nevertheless, nowadays many projects are being planned on account of institutional support from the government and the recent advance of construction technology. In Korea, the new industry field operating and managing the tidal power plant has already opened. But we are facing a number of problems for optimal operation of tidal power plant that are a lack of operation experience and a skill of professional management and others. This paper suggests a novel way to determine the starting head of power generation by generating power comparison method For this new method, the paper discusses many factors including changing the volume of the basin, the number of operating turbines and gates and forecasting the tidal amplitude and the characteristic curve of turbine and gate. Finally we verified that it can increase about 2% an annual power generation compared with the conventional method using the original operational function made in the plant design process.

  • With the recent proliferation of Hallyu content, national interest in Korea and the demand for designs with traditional Korean patterns as a motif is on the rise. In addition, as customers' design requirements tend to value more and more detailed and differentiated handcrafted designs, research on the motifs used in nail experts' original nail design development and nail design is continuously needed. Therefore, the purpose of this study is to expand the scope of nail design motifs with high practical value by producing nail design works applying plant patterns of Wadang, used during the Unified Silla Period, and to diversify nail art expression techniques. The research reviewed domestic and foreign literature related to Wadang during the Unified Silla Period, analyzed the collection at the National Museum of Korea, and categorized the motifs based on the characteristics of plant patterns and Wadang during the Unified Silla Period. The tangible motifs are intended to be presented as a fusion of nail art design works that utilizes both flat and stereoscopic art techniques. Through this study, it was confirmed that the plant pattern of Wadang from the Unified Silla Period is an attractive motif that can be expressed in various nail designs expressing Korean emotions and traditional beauty, and furthermore, it can be used as basic data for the idea of various beauty design areas.

Classification of Herbs in Vegetable Part, Pen-tsao-kang-mu(Bon-cho-kang-mok) (본초강목(本草綱目) 채부(菜部)에 수재된 본초(本草)의 분류(分類))

  • Pen-tsao-kang-mu(Bon-cho-kang-mok), chinese medicinal plant book, was written by Lee sijin(1578), China. The subject of this study were 158 articles of vegetable part in Pen-tsao-kang-mu. Among them only 139 articles were able to be identified by authority of several references. By Engler's system they were classified into 8 divisions, 10 classes, 6 subclasses, 31 orders, 22 suborders, 52 families, 85 genera, 99 species, 12 varieties and 2 forma, and were confirmed 113 kinds of original plants. Among the divisions, Angiospermae was the most numerous division with 92 kinds(80.70%) and the second division was Fungi with 12 kinds(10.53%). The next was Rhodophyta with 4(3.51%) kinds. Other 19 articles were unable to be classified because of their ambiguous name.

  • Pen-tsao-kang-mu(Bon-cho-kang-mok), chinese medicinal plant book, was written by Si-jin Lee, 1578, China. The subject of this study were 210 articles of grain part in Pen-tsao-kang-mu. Among them only 193 articles were able to be identified by authority of several references. By Engler's system they were classified into 4 divisions, 5 classes, 3 subclasses, 28 orders, 17 suborders, 46 families, 95 genera, 100 species, 11 varieties and 1 form, and were confirmed 112 kinds of original plants. Among the divisions, angiospermae was the most numerous division with 107 kinds(91.98%) and the second division was gymnospermae with 3 kinds(2.70%). The next were phaeophyta and fungi with 1 kind(0.90%) on each. Other 17 articles were unable to be classified because of their ambiguous name.

Root Colonizing and Biocontrol Competency of Serratia plymuthica A21-4 against Phytophthora Blight of Pepper

  • The biocontrol agent Serratia plymuthica A21-4 readily colonized on the root of pepper plant and the bacterium moves to newly emerging roots continuously. The colonization of A21-4 on the pepper root was influenced by the presence ofPhytophthora capsici in the soil. When P. capsici was introduced in advance, the population density of A21-4 on the root of pepper plant was sustained more than $10^6$ cfu/g root until 3 weeks after transplanting. On the other hand, in the absence of P. capsici, the population density of A21-4 was reduced continuously and less than $10^5$ cfu/g root at 21 days after transplanting. S. plymuthica A21-4 inhibited successfully the P. capsici population in pepper root and rhizosphere soil. In the rhizosphere soil, the population density of P. capsici was not increased more than original inoculum density when A21-4 was treated, but it increased rapidly in non-treated control. Similarly, the population density of P. capsici sharply increased in the non-treated control, however the population of P. capsici in A21-4 treated plant was not increased in pepper roots. The incidence of Phytophthora blight on pepper treated with A21-4 was 12.6%, while that of non-treated pepper was 74.5% in GSNU experimental farm experiment. And in farmer's vinyl house experiment, the incidence of the disease treated with the fungicide was 27.3%, but treatment of A21-4 resulted in only 4.7% of the disease incidence, showing above 80% disease control efficacy.

Development of Molecular Detection of Three Species of Seed-Transmissible Viruses Useful for Plant Quarantine

  • Three pairs of specific primers were developed for rapid and precise RT-PCR detection of three seed-transmissible viruses, namely Peanut clump virus (PCV, Pecluvirus), White clover mosaic virus (WCIMV, Potexvirus) and Carrot red leaf virus (CaRLV, Luteovirus). Each primer set was found in conserved region through multiple sequence alignment in the DNAMAN. Total nucleic acids extracted from PCV-, WCMV-, and CaRLV-infected seeds and healthy plants were used for RT-PCR detection using each virus-specific primer, Sizes of PCV, WCIMV, and CaRLV PCR products were 617bp (PCV-uni5 and PCV-uni3 primers), 561bp (WCMV-CP5 and WCMV-CP3 primers), and 626bp (CL1-UP and CL2-DN primers); which corresponded to the target sizes. Nucleotides sequences of each amplified cDNA were confirmed which belonged to the original virus. This study suggests that these virus-specific primer sets can specifically amplify viral sequences in infected seeds. Thus, they can be used for specific detection of three viruses (PCV, WCMV and CaRLV) from imported seed samples for plant quarantine service.
