Studies on the Control of Fusarium Wilt of tile Cucurbitaceous Plants(1) Investigation on the Pathogenicity of Fusarium Isolates from tile Wilted Cucurbitaceous Plants (오이류 덩굴 쪼김병 (만할병) 방제에 관한 연구 (1) 오이류 덩굴쪼김병균(만할병균)의 기생성에 관한 조사)

  • These studies aimed to investigating the pathogenicity of Fusarium isolates from the wilted cucur bitaceous plants and the pathogenicity of 24 isolates to cucumber, oriental cantaloup, oriental pickling melon, sponge gourd. muskmelon and watermelon were as follows. 1) The isolates from wilted watermelon and melon severely and one of the isolates was slightly pathogenicon oriental pickling melon or oriental cantaloup. 2) The isolates from wilted cucumber were four types. some of the isolates were severely pathogenic on cucumber and infected oriental cantaloup. oriental pickling melon and melon. Some of the isolates were not pathogenic or slightly pathogenic on cucumber. but these were severely pathogenic on oriental pickling melon. One of the isolates was severely pathogenic on oriental cantaloup oriental pickling melon and melon, but no pathogenic on cucumber and one of the isolates was also severely pathogenic on oriental cantaloup. oriental pickling melon and melon and moderatly pathogenic' on water melon. 3) The isolates from wilted oriental cantaloup were four types some of the isolates infected oriental cantaloup, oriental pickling melon and melon severely and some of the isolates were pathogenic on oriental cantaloup. oriental pickling melon. melon and watermelon. One of the isolates, was slightly pathogenic on oriental pickling melon and melon but one of the isolates was highly pathogenic on melon and infected watermelon slightly. 4) Wilt Fusarium of the cucurbitaceous plants could divide into the group of wilt Fusarium of watermelon, cucumber and muskmelon according to the pathogenicity but it will have to rearrange to one form species from several form species of wilt Fusarium of the cucurbitaceous plants and ought to divide to races according to pathogenicity on severely fixed differential varieties which selected from the cucurbitaceous plants. because of the degree of pathognicity and host range are not surely fixed between isolates of wilt Fusarium of the cucurbitaceous plants tested.

Monitoring of Organoleptic and Physical Properties on Preparation of Oriental Melon Jelly (참외젤리 제조에 따른 관능적 및 물리적 특성 모니터링)

  • To prepare oriental melon jelly having highly favorite, oranoleptic and physical properties of jelly with special to the content of glucomannan, oriental melon concentration juice and vinegar was monitored by four-dimensional response surface methodology. Mouth-feel showed maximum sensory score in 0.09% glucomannan, 3.17% oriental melon concentration juice and 1.25% oriental melon vinegar. Organoleptic taste showed maximum sensory score in 0.12% glucomannan, 3.64% oriental melon concentration juice and 0.91% oriental melon vinegar. Overall palatability showed also maximum sensory score in 0.10% glucomannan, 3.07% oriental melon concentration juice and 1.34% oriental melon vinegar. The sensory socore of overall palatability was more than 7.0 in the texture ranges of 0.20~0.31 cm/g (softness) and 8.0~12.5 gㆍcm (jelly strength).

Effect of Double-Cropping Systems on Nematode Population in Plastic Film House Soils of Oriental Melon Cultivation (이모작에 따른 참외 재배 비닐하우스 토양의 선충밀도 변화)

  • BACKGROUND: Crop rotation is often used as a solution to eradicate nematodes in soils used in plastic film houses for long-term cultivation of oriental melon. However, it is not clear if the double-cropping is effective in reducing nematode populations in soils. METHODS AND RESULTS: Nematode population in plastic film house soil was measured during oriental melon cultivation from April to July in short term crop rotation systems of oriental melon. Double-cropping of chinese cabbage in open-field for 3-4 months following oriental melon in plastic film houses could not prevent the build-up of high population density of nematodes. However, double-cropping of dropwort in flooded soil for 3-4 months following oriental melon in plastic film houses could effectively reduce the nematode population during the successive year of oriental melon cultivation. The reduced nematode population in soils of oriental melon-dropwort double-cropping system was continued until the mid season of progressive year oriental melon cultivation. Application of nematicide to soil before growing oriental melon in the oriental melon-dropwort double-cropping was very effective in preventing the build-up of high population density of nematode in plastic film house soils. CONCLUSION: Short-term introduction of crop rotation was not effective in suppression of high population density of nematodes in plastic film house soils of long-term year-to-year production of oriental melon. For securing the soil productivity and sustainability of plastic film house, various physical, chemical, and agronomic practices should be properly combined together.

  • Consumer preference, marketing environment and various agricultural policy were changed recently. So it is necessary to analyze the consumption behavior for selection the target market. In this paper, we analyzed the consumption behavior and the brand positioning using oriental melon. From the results of analysis, firstly, consumers' age, job, education and the number of member of household were effected to consumption behavior of oriental melon. Secondly, the marketing factor was the most important when purchasing oriental melon. Thirdly, the brand of Sung-Ju Oriental Melon was evaluated to the best brand on brand name, brand image and brand message.

  • In order to utilize non-commercial melon as alcoholic beverage material, in this study the optimum fermentation method and manufacture conditions were investigated and thereafter the properties of final product were determined. As for melon wine made with only melon juice, sensorial properties, particularly flavor, were not acceptable as alcoholic beverage. To improve the lack of melon wine, we made mash with cooked rice and Nuruk, and then added melon juice during fermentation. Acceptable sensorial properties were obtained in melon wine when 30% extract juice of oriental melon was added after 4 day of fermentation. The alcoholic beverage added melon juice showed much higher yellowness value than that without melon juice indicating that a clear yellow color similar to melon was actually observed in final product. Adding melon juice led little changes in amount and composition of free sugars, organic acids and amino acids and could improve sensorial properties.

Potential Reasons for Prevalence of Fusarium Wilt in Oriental Melon in Korea

  • This study aims to examine the potential reasons for the current prevalence of the fusarium wilt in the oriental melon. Twenty-seven Fusarium isolates obtained from oriental melon greenhouses in 2010-2011 were identified morphologically and by analysis of elongation factor-1 alpha gene (EF-$1{\alpha}$) and internal transcribed spacer (ITS) rDNA sequences as 6 Fusarium species (8 isolates of F. oxysporum, 8 F. commune, 5 F. proliferatum, 3 F. equiseti, 2 F. delphinoides, and 1 F. andiyazi), which were classified as same into 6 EF-$1{\alpha}$ sequence-based phylogenetic clades. Pathogenicity of the Fusarium isolates on the oriental melon was highest in F. proliferatum, next in F. oxysporum and F. andiyazi, and lowest in the other Fusarium species tested, suggesting F. proliferatum and F. oxysporum were major pathogens of the oriental melon, inducing stem rots and vascular wilts, respectively. Oriental melon and watermelon were more susceptible to F. oxysporum than shintosa and cucumber; and cucumber was most, oriental melon and watermelon, medially, and shintosa was least susceptible to F. proliferatum, whose virulence varied among and within their phylogenetic subclades. Severe root-knot galls were formed on all the crops infected with Meloidogyne incognita; however, little indication of vascular wilts or stem and/or root rots was shown by the nematode infection. These results suggest the current fungal disease in the oriental melon may be rarely due to virulence changes of the fusarium wilt pathogen and the direct cause of the severe root-knot nematode infection, but may be potentially from other Fusarium pathogen infection that produces seemingly wilting caused by severe stem rotting.

Whole genome re-sequencing and development of SSR markers in oriental melon (참외 전장유전체 염기서열 분석 및 SSR 마커 개발)

  • The objective of this study was to use 'Danta PR', NGS (Next Generation Sequencing) technology for genome resequencing to develop polymorphic makers between Chinese oriental melon, 'Hyangseo 1' and Korean oriental melon. From the resequencing data that covered about 81 times of the genome size, 104,357 of SSR motifs and Indel, and 1,092,436 of SNPs were identified. 299 SSR and 307 Indel markers were chosen to cover each chromosome with 25 markers. These markers were subsequently used to identify genotypes of 'Danta PR' BC1 (F1 x 'Danta PR') population and a genetic linkage map was constructed. SSR, Indel, and SNPs identified in this study would be useful as a breeding tool to develop new oriental melon varieties.

Effect of Oriental Melon Vinegar Treatment on Growth and Disease Control of Oriental Melon (참외식초 처리가 참외 생육 및 병충해 방제에 미치는 영향)

  • Oriental melon vinegar was prepared by two stage fermentations of alcohol and acetic acid. In the alcohol fermentation using oriental melon residual products, alcohol content showed 7.43% in 17$^{\circ}$ brix of initial sugar concentration and 80 h of fermentation time. In the acetic acid fermentation using oriental melon alcohol, acidity showed 5.25% in 250rpm of agitation rate and 200 h of fermentation time. The cultivation esults of oriental melons using its vinegar are as follows. Quantity and quality of samples treated with 500, 1,000 and 2,000 times of oriental melon vinegar were higher than that of control : weight, quality, sugar content and goods production rate were higher to degree of 33∼42 g/piece, 370 ∼460kg/10a, 0.6∼.9$^{\circ}$ Brix, 2∼5%, respectively. Coods production yield of samples treated with 500, 1,000 and 2,000 times of oriental melon vinegar was higher (400∼610 kg/10a) than that of control. The results of control of powdery mildew on oriental melons using oriental melon vinegar as the diluted solution with 500 and 1000 times were identical for control value that used by agrochemical. Powdery mildew were exterminated by 2nd treatment of the diluted solution. In case of aphids, the diluted solution with 500, 1,000 and 2,000 times of oriental melon vinegar exterminated thoroughly by 2nd treatment.

Hypoglycemic Activity of the Hexane Extract of Oriental Melon (Cucumis melo L. var. makuwa Makino) Seeds and Its Active Compounds

  • The aim of this work is to evaluate the potential of oriental melon (Cucumis melo L. var. makuwa Makino) seeds for the management of type 2 diabetes by controlling glucose absorption. The ${\alpha}$-glucosidase and ${\alpha}$-amylase inhibitory effects of the hexane extracts from oriental melon seeds were investigated. A bioassay-guided fractionation technique was used to elucidate the principal active components. The results show that the hexane extract from oriental melon seeds exhibited high inhibitory activities against ${\alpha}$-glucosidase and ${\alpha}$-amylase. The hexane extract was further fractionated into four sub-fractions. Among them, the sub-fraction F-1 exhibited the most potent anti-diabetic effect. The active components were isolated and identified by gas chromatography/mass spectroscopy (GC-MS). Free fatty acids showed significant hypoglycemic activity (p < 0.001) and fatty acid composition influenced enzyme inhibitory activities. These results suggest that oriental melon seeds could be used to prevent type 2 diabetes.

Effect of Silicate Fertilizer on Oriental Melon in Plastic Film House (시설재배 참외에 대한 규산 비료 시용 효과)

  • Although the requirement and optimum soil level of Si for oriental melon are still not well understood, silicate fertilizer is commonly applied to the oriental melon in plastic film houses where soil silicate level is relatively high. In this research the effects of silicate fertilizer on growth, fruit yield and fruit quality of oriental melon, and soil properties were investigated in plastic film house where the soil available silicate was $212mg\;SiO_2\;kg^{-1}$. Silicate fertilizer was applied in the rates of 100, 200, and $300kg\;10a^{-1}$. The application of silicate fertilizer could not increase the early growth of oriental melon, and also the fruit yield and quality were not different among the treatments. Available Si and P contents in soils and also Si and P contents in leaf of oriental melon of the different treatments were not significantly different. In the relationship between total Si in oriental melon leaf and soil silicate extracted by 1 N sodium acetate, optimum soil available silicate level for oriental melon was found to be around $100mg\;SiO_2\;kg^{-1}$. These results indicate that the additional silicate fertilization in soils of available silicate higher than $100mg\;SiO_2\;kg^{-1}$ is unnecessary, and such application of silicate can not have any beneficial effect on the growth and fruit yield of oriental melon.
