태음조위탕(太陰調胃湯)의 항산화(抗酸化) 효능(效能)에 의한 간세포(肝細胞) 보호(保護) 효과(效果)에 관한 연구(硏究) (Effects of Taeumjowetang on Lipid Peroxidation by Free Radicals and Oxidative Damage of Hepatocytes by tert-Butyl Hydroperoxide)

  • Effects of Taeumjowetang on Lipid Peroxidation by Free Radicals and Oxidative Damage of Hepatocytes by tert-Butyl Hydroperoxide. 1. Purpose The present study was carried out to evaluate the antioxidant effects of Taeumjowetang in vitro. 2. Methods In this study, antioxidant effects of TJT on lipid peroxidation were determined according to the method of TBA. (Abbreviation) TJT : Taeumjowetang, TBA : 2-thiobarbituric acid. 3. Results : 1) TJT inhibited markedly peroxidation of linoleic acid during the autoxidation. 2) TJT inhibited lipid peroxidation induced by hydroxyl radical derived from H2O2-Fe2+ in rat liver homogenate. 3) TJT showed 66% scavenging effect on DPPH radical. 4) TJT exhibited a 25% inhibitory effect on superoxide generation from xanthine-xan thine oxidase system. 5) To investigate the antioxidative effects of TJT on the hepatocytes, cultured normal rat liver cells(Ac2F) were prepared and incubated with or without TJT. After 16~18hr, cells placed in DMEM medium without serum, and then incubated with 1mM t-BHP for 2hr. Viable cells were detected by MTT assay. In this test, TJT protected the cell death induced by t-BHP and significantly increased cell viability in the normal rat liver cell. Conclusion These results suggested that TJT might play a protective role in lipid peroxidation by free radicals.

목단피지황탕(牧丹皮地黃湯)이 위점막손상(胃粘膜損傷)에 미치는 영향(影響) (Effects of Mokdanpijihwang-tang on gastric mucosal damage in mice)

  • 1. Back ground and purpose An experimental study has done to examine the effect of defense and cure gastric mucasal damage of Mokdan-pijihwang-tang. 2. Methods Mice had intragastric injected with MJ extract before indomethacin treatment which induces homorrhage infarct and erosion artificially. Degree of lipid peroxidation, general morphology, change of mucous cell, the distribution of PNA, ICAM and distribution of apoptotic cell were objected. (Abbreviation) MJ : Mokdanpijihwang-tang, PNA : Peanut Agglutinin, ICAM : Intercellular Adhesion Molecule 3 Results 1) The degree of lipid peroxidation in INDO-group had increased conspicuously than control group. But the degree of lipid peroxidation in MJ-group had decreased than INDO-group and these decline had probability. 2) After indomethacin treatment, hemorrhage infarct and erosion had increased in stomach body. But in MJ-group, the configuration is normal, except the group intragastric injected with MJ extract at hour 24 before indomethacin treatment. 3) Surface mucous cell and neck mucous had disappeared in INDO-group. But in MJ-group tormal distribution had shown like control group except the group intragastric injected with MJ extract at hour 24 before indomethacin treatment. 4) PNA positive reaction had not shown in INDO-group. But medium PNA positive reaction had shown In Mj-group. 5) ICAM positive reacted cell had shown in INDO-group. The decrease of ICAM positive cell were shown than INDO-group. 6) A number of apoptotic cell was distributed in hemorrhagic erosion. A few number of apoptotic cell was distributed in MJ-group except some surface mucous. 4. Conclusion These results suggest that MJ has an effect on cure of gastric mucosal damage.

Streptozotocin 유발 당뇨 쥐의 혈당 및 혈청 지질에 미치는 한방추출물의 효과와 최적 농도에 관한 연구 (Study of Effects and Optimum Concentrations of Medical Herb Extracts on Blood Glucose and Serum Lipid in Streptozotocin-Induced Diabetic Rats)

  • Objectives : The purpose of this study was to investigate effects of extract from mixed culture with Tricholoma matsutake mycelium and Cordyceps militaris mycelium in oriental medicine and cereal midium (OCM) considered effective for the treatment of diabetes. Methods : We examined antioxidant activity and $\alpha$-glucosidase inhibit activity of OCM and orally administered OCM extracts to Streptozotocin-induced diabetic ratsat the dose of 100mg/kg, 200mg/kg and 300mg/kg body weight. Results : Total polyphenol content of extracts of OCM was 26.41 mg/g, DPPH radical scavenging activity was 32.24% and SOD-like activity was 31.43%. $\alpha$-glucosidase inhibit activity showed 85.3% of acarbose. Blood glucose levels of the OCM extracts-treated groups were lower than those of the diabetic control group (DC), total cholesterol and triglyceride contents were similar to the acarbose-treated group (PC) and ALT, AST were lower than the DC and PC groups. The 300 mg/kg body weight administered group (UM-300) were superior to other OCM-treated groups (UM-100, UM-200). Conclusions : These results showed extract from mixed culture with Tricholoma matsutake mycelium and Cordyceps militaris mycelium in oriental medicine and cereal midium has potentiality to beused in pharmaceutical products and medicinal food for preventive and treatment diabetes mellitus.
