과실 및 채소중 유기인계 잔류 농약에 관한 연구 (Organophosphorus Insecticide Residues in Fruits and Vegetables)

  • Organophosphorus insecticide residues were investigated in six kinds of fruits and five kinds of vegetables.The materials used in this experiment were grape, musk melon, apple, peach, plum, apricot, lettuce, green pepper, cucumber, pumpkin and tomato which were collected from June to september 1986 in Seoul. Residual pesticides investigated were Diazinon, Parathion, MEP (Fenitrothion), Malathion, EPN, MPP (Fenthion), PAP (Phenthoate) and Dimethoate and all samples were analysed by gas chromatographic technique with NPD (Nitrogen Phosphorus Detector). No sample was found to approach proposed national maximum residue limits in Korea.

Development of an ELISA for the Organophosphorus Insecticide Chlorpyrifos

  • A selective enzyme-linked immunosorbent assay (ELISA) for the insecticide chlorpyrifos was developed. Four haptens for chlorpyrifos were synthesized and two of them were used as immunogens after coupling to keyhole limpet hemocyanin by two differe nt approaches. Rabbits were immunized with either of them and the sera were screened against 4 haptens coupled to ovalbumin (OVA). Using the sera of highest specificity, an antigencoated ELISA was developed, which shows an I50 of 160 ppb with a detection limit of 10 ppb. An antibody-coated ELISA was also developed, which shows an $I_{50}$ of 20 ppb with a detection limit of 0.1 ppb. The antibodies showed negligible cross-reactivity with other organophosphorus pesticides except for insecticides chlorpyrifos-methyl and bromophos-ethyl, which makes these assays suitable for the selective detection of chlorpyrifos.

  • A dipstick-type immunochemical biosensor for the detection of the organophosphorus insecticide fenthion was developed using a screen-printed electrode system as an amperometric transducer with polyclonal antibodies against fenthion as a bioreceptor. The assay of the biosensor involved competition between the pesticide in the sample and pesticide-glucose oxidase conjugate for binding to the antibody immobilized on the membrane. This was followed by measurement of the activity of the bound enzyme by the supply of the enzyme substrate (glucose) and amperometric determination of the enzyme reaction product ($H_2O_2$). The activity of the bound enzyme was inversely proportional to the concentration of pesticide. The optimized sensor system showed a linear response against the logarithm of the pesticide concentration ranging from $10^{-2}$ to $10^3\;{\mu}g/L$.

Using the sera of highest specificity, an antigen-coated ELISA was developed, which showed an I50 of 310 ng/mL with the detection limit of 20 ng/mL. The antibodies showed negligible cross-reactivities with other organophosphorus pesticides except for parathion-methyl, which makes the assay suitable for the selective detection of cyanophos.

  • Organophosphorus insecticides induce different clinical manifestations varying according to the different side groups attached to the phosphate, their rates of degradation, and their fat solubilities. In consequence of this variation, specific treatments are required for particular organophosphorus insecticides. We report a unusual case of intoxication with dimethyl organophosphorus insecticide in a 26-year-old woman. She manifested atypical and ultimately fatal symptoms including profound shock, refractory hyperglycemia, and hypothermia.

Development of an ELISA for the Organophosphorus Insecticide Isofenphos

  • A selective enzyme-linked immunosorbent assay (ELISA) for the insecticide isofenphos was developed. Three different analogues (haptens) of isofenphos were synthesized and were coupled to carrier proteins through the pesticide thiophosphate group t o use as immunogens or coating antigens. Rabbits were immunized with one of the haptens coupled to BSA for production of polyclonal antibodies and the sera were screened against each of the other two haptens coupled to ovalbumin (OVA). Using the sera of highest specificity, an antigen-coated ELISA was developed, which showed an I50 of 96 ng/mL with the detection limit of 2 ng/mL. The antibodies showed negligible cross-reactivity with other organophosphorus pesticides and the phenol metabolite of isofenphos, which makes the developed assay suitable for the selective detection of isofenphos. An antibody-coated ELISA was also developed, which showed an I50 of 580 ng/mL with a detection limit of 70 ng/mL.

  • A competitive enzyme-linked immunosorbent assay (ELISA) was developed for the quantitative detection of the organophosphorus insecticide bromophos. Three bromophos analogues (haptens) were synthesized and were coupled to carrier proteins to use as immunogens or coating antigens. Rabbits were immunized with either one of two haptens coupled to bovine serum albumin (BSA) for production of polyclonal antibodies, and the sera were screened against one of the haptens coupled to ovalbumin (OVA). Using the serum with highest specificity and an enzyme tracer, an antibody-coated ELISA was developed, which showed an $IC_{50}$ of 40 ng/mL with a detection limit of 7 ng/mL. The antibodies in this assay showed negligible cross-reactivity with other organophosphorus pesticides except with the insecticides chlorpyrifos and fenitrothion.

  • 본 연구에서는 동물세포에서 매우 높은 독성을 갖는 유기인 살충제 coumaphos를 생분해하기 위하여, 재조합 대장균으로부터 발현된 organophosphorus hydrolase (OPH)가 사용되었다. OPH 생산성을 향상시키고자, 균주의 배지에 비이온성계면활성제(Tween 20, PEG 1000)와 유기용매(glycerol, propanol, ethanol)에 대한 효과를 고찰하였다. 0.25% Tween 20과 0.5% glycerol를 배지에 첨가하였을 때, 최대의 OPH가 생산되었다. 초음파를 이용한 균주 파쇄 공정으로부터 생성된 OPH를 사용 하였을 때, 0.2, 0.5, 1.0, 2.0 mM coumaphos는 각각 100, 88, 84, 78%의 생분해효율을 나타내었다. 이 연구에서 얻어진 결과들은 coumaphos가 다양한 농도로 오염된 지역을 생물학적으로 처리할 수 있는 새로운 방법으로서 활용될 수 있을 것이다.

  • Recently, we reported an improved technology for the degradation of organophosphate nerve agents using whole cells of genetically engineered Escherichia coli that anchored and displayed the enzyme organophosphorus hydrolase on the cell surface. In this paper we report the immobilization of these cells on highly porous sintered glass beads and the subsequent application of the immobilized cell in a continuous-flow packed bed bioreactor for the biodetoxification of a widely used insecticide, coumaphos.
