The effect of a new hepatoprotective agent, YH-439, on the hepatobiliary transport of a model organic cation (OC), TBuMA (tributylmethylammonium), was investigated. The area under the plasma concentration-time curve (AUC) from time zero to 4 h following iv administration of TBuMA (6.6 $\mu$mol/kg) was increased significantly when YH-439 in corn oil (300 mg/kg) was orally administered to rats 24 h prior to the experiment. Nevertheless, the cumulative biliary excretion of TBuMA remained unchanged. As a consequence, the apparent biliary clearance ($CL_b$) of TBuMA was decreased significantly as a result of YH-439 pretreatment, consistent with the fact that the in vivo excretion clearance of TBuMA across the canalicular membrane ($CL_{exc}$) was not changed by the pretreatment. The in vitro uptake of TBuMA into isolated hepatocytes was decreased by one half by the pretreatment, owing to a decrease in the apparent V$_{max}$ and $CL_{linear}$, but the $K_m$ for the process remained constant. Most interestingly, however, the sinusoidal uptake of glucose, a nutrient, into hepatocytes was not influenced by the pretreatment, suggesting the YH-439 pretreatment specifically impaired the sinusoidal uptake of OCs. Thus, the OC-specific inhibition of hepatic uptake, without influencing the uptake of glucose, a nutrient, appeared to be associated with the hepatoprotective activity of YH-439.
Effects of cadmium (Cd) intoxication on renal endosomal accumulation of organic cations $(OC^+)$ were studied in rats using $^{14}C-tetraethylammnium$ (TEA) as a substrate. Cd intoxication was induced by s.c. injections of 2 mg Cd/kg/day for $2{\sim}3$ weeks. Renal cortical endosomes were isolated and the endosomal acidification (acridine orange fluorescence change) and TEA uptake (Millipore filtration technique) were assessed. The TEA uptake was an uphill transport mediated by $H^+/OC^+$ antiporter driven by the pH gradient established by $H^+-ATPase.$ In endosomes of Cd-intoxicated rats, the ATP-dependent TEA uptake was markedly attenuated due to inhibition of endosomal acidification as well as $H^+/TEA$ antiport. In kinetic analysis of $H^+/TEA$ antiport, Vmax was reduced and Km was increased in the Cd group. Inhibition of $H^+/TEA$ antiport was also observed in normal endosomes directly exposed to free Cd (but not Cd-metallothionein complex, CdMt) in vitro. These data suggest that during chronic Cd exposure, free Cd ions liberated by lysosomal degradation of CdMt in proximal tubule cells may impair the endosomal accumulation of $OC^+$ by directly inhibiting the $H^+/OC^+$ antiporter activity and indirectly by reducing the intravesicular acidification, the driving force for $H^+/OC^+$ exchange.
This study was conducted to evaluate the effects on soil chemical properties and plant growth after applying soldier fly compost (SFC). Treatments were as follows. No fertilizer (NF), control, SFC1 (SFC 250 kg/10a), SFC2 (SFC 500 kg/10a) and SFC3 (SFC 1,000 kg/10a). As compared to control in the pot test, organic matter (OM) and exchangeable sodium (Ex-Na) of SFC3 treatment were increased, and growth and nutrient uptake of young radish were not significantly different. Correlation coefficient between soil chemical factors like total nitrogen (T-N), OM, and CEC and uptake of nitrogen (N) and phosphorus (P) was significantly positive (p≤0.05). Compared to control in the field test, electrical conductivity, T-N, OM, Av.-P2O5, and CEC was increased, and lettuce growth was not significantly different. Correlation coefficient between application amount of SFC and T-N, OM, and Av.-P2O5 was significant positively (p≤0.05). These results indicated that the application of SFC improved nutrient availability of soil by increasing OM and CEC.
The issue of whether or not the presence NOx (NO and oxidized metabolites) in the hepatocytes at pathological levels affects the functional activity of transport systems within the sinusoidal membrane was investigated. For this purpose, the effect of the pretreatment of isolated hepatocytes with sodium nitroprusside (SNP), a spontaneous NO donor, on the sinusoidal uptake of tributylmethylammonium (TBuMA) and triethylmethyl ammonium (TEMA), representative substrates of the organic cation transporter (OCT), and taurocholate, a representative substrate of the $Na^+$/taurocholate cotransporting polypeptide (NTCP), was measured. The uptake of TBuMA and TEMA was not affected by the pretreatment, as demonstrated by the nearly identical kinetic parameters for the uptake ($i.e., V_{max}, K_{m} and CL_{linear}$). The uptake of mannitol into hepatocytes was not affected, demonstrating that the membrane integrity remained constant, irregardless of the SNP prutreatment. On the contrary, the uptake of taurocholate was significantly inhibited by the pretreatment, resulting in a significant decrease in V_{max}$, thus providing a clear demonstration that NOx preferentially affects the function of NTCP rather than OCT on the sinusoidal membrane. A direct interaction between NOx and NTCP or a decrease in $Na^+/K^+$ ATPase activity as the result of SNP pretreatment might be responsible for this selective effect of NOx.
Paeonol has neuroprotective function, which could be useful for improving central nervous system disorder. The purpose of this study was to characterize the functional mechanism involved in brain transport of paeonol through blood-brain barrier (BBB). Brain transport of paeonol was characterized by internal carotid artery perfusion (ICAP), carotid artery single injection technique (brain uptake index, BUI) and intravenous (IV) injection technique in vivo. The transport mechanism of paeonol was examined using conditionally immortalized rat brain capillary endothelial cell line (TR-BBB) as an in vitro model of BBB. Brain volume of distribution (VD) of [$^3H$]paeonol in rat brain was about 6-fold higher than that of [$^{14}C$]sucrose, the vascular space marker of BBB. The uptake of [$^3H$]paeonol was concentration-dependent. Brain volume of distribution of paeonol and BUI as in vivo and inhibition of analog as in vitro studies presented significant reduction effect in the presence of unlabeled lipophilic compounds such as paeonol, imperatorin, diphenhydramine, pyrilamine, tramadol and ALC during the uptake of [$^3H$]paeonol. In addition, the uptake significantly decreased and increased at the acidic and alkaline pH in both extracellular and intracellular study, respectively. In the presence of metabolic inhibitor, the uptake reduced significantly but not affected by sodium free or membrane potential disruption. Similarly, paeonol uptake was not affected on OCTN2 or rPMAT siRNA transfection BBB cells. Interestingly. Paeonol is actively transported from the blood to brain across the BBB by a carrier mediated transporter system.
We have investigated the transport characteristics of synthetic antioxidant and free radical scavenger, $\alpha$-phenyl-n-tert-butyl nitrone (PBN) at the blood-brain barrier (BBB) by in vitro uptake study in conditionally immortalized rat brain capillary endothelial cell line (TR-BBB). Also, the efflux of PBN from brain to blood is estimated using the brain efflux index (BEI) method. Choline is a charged organic cation, including nitrogen-methyl group and shows the carrier-mediated distribution to the brain. [$^3$H]Choline uptake by TR-BBB cells was significantly inhibited by PBN with $IC_{50}$/ of 1.2 mM, which appears to be due to similar structures between choline and PBN. And, PBN was microinjected into Par2 of the rat brain by BEI method, and was eliminated from the brain with an apparent elimination half-life of about 2 min. Also, [$^3$H]choline efflux was significantly inhibited by PBN using BEI method. In conclusion, the efflux transport of PBN takes place across the BBB and PBN may be transported into the brain and eliminated from the brain by BBB choline transporter.
Sulfate adsorption in forest soils is a process of sulfur dynamics playing an important role in plant uptake, cation movement, acid neutralization capacity and so on. The relationship between sulfate adsorption and some physicochemical properties of four forest soils was investigated. Extractable sulfate contents and sulfate adsorption capacity (SAC) in the forest soils varied much among study sites. Extractable sulfate contents were more in sub-surface soils with lower organic matter and greater Al and Fe oxides than in surface soils. The average contents of $Al_d$ and $Fe_d$ in the sub-surface soils were 8.49 and $12.45g\;kg^{-1}$, respectively. Soil pH, cation exchange capacity and clay content were positively correlated with the extractable sulfate contents and SAC. Organic carbon content, however, was negatively correlated with the extractable sulfate contents, implying the competitive adsorption of sulfate with soil organic matter. Considerably significant correlation was found between inorganic + amorphous Al and Fe oxides and the sulfate adsorption, but crystalline Al and other fractions of Fe oxide showed no correlation. Relatively close relationship between the adsorbed sulfates and soil pH, cation exchange capacity, or amorphous Al oxides indicates that the accelerated soil acidification may substantially reduce the potential for sulfate adsorption contributing to sulfur flux in forest ecosystems.
This experiment was carried out to investigate the effects of amount of nitrogen application with the barley straw application on the changes in soil physical and chemical properties. nutrient uptake and percentage recovery of chemical fertilizer N in the rice plant from 1997 to 1998. The soil physical properties, such as bulk density, hardness, porosity and gaseous phase were improved by barley straw application. There was also improvement or increment in the soil chemical properties, such as pH, organic matter, T-C. T-N, available $SiO_2$, exchangeable K and cation exchange capacity, but decrease in available $P_2O_5$. The $Fe^{+{+}}$ content in soil after barley straw application was high from tillering stage to panicle forming stage, but becoming lowered toward the heading stage, while $Mn^{+{+}}$ content was increased. N uptake with barley straw application was increased in the N $126kg\;ha^{-1}$ plot, but decreased in the N $141kg\;ha^{-1}$ plot. The uptake of fertilized N was continued longer in barley straw application than none-application plot. Percentage recovery of chemical fertilizer N in rice straw was around 1% at tillering stage, but was highly increasing till maximum tillering stage, while the recovery was generally low in barley straw application. Meanwhile, fertilizer P uptake in barley straw application was high, but potassium uptake was low at all different levels of N application.
This study was carried out to investigate the effects of fertilizer and organic resource annual dressing for 30 years of Jeonbug series (silt loam) on soil properties and rice N uptake in paddy field soil. In the study field, treatments including control (NPK), NPK+rice straw, NPK+rice straw compost and nitrogen fertilization levels at 0, 100, 150, 200, 250 kg $ha^{-1}$ have been imposed for 30 years. Soil hardness and bulk density decreased from 15.7 mm and 1.381 Mg $m^{-3}$ in the control to 12.5 mm and 1.244 Mg $m^{-3}$ in NPK+rice straw compost treatment, respectively, indicating improvement of soil physical conditions such as porosity. Co-application of straw compost with NPK also result in a better chemical properties than NPK alone as it increased available phosphate (from 96 to 133 mg $kg^{-1}$), available silicate (from 81 to 116 mg $kg^{-1}$), and cation exchange capacity (from 9.8 to 11.4 $cmol_c\;kg^{-1}$). Soil organic matter concentration of top soil (0 to 7.5 cm in depth) was higher in NPK+rice straw and NPK+rice straw compost than in control. Fertilizer N uptake amount was much higher in NPK+rice straw (nitrogen fertilization level; 250 kg $ha^{-1}$) and NPK+rice straw compost (nitrogen fertilization levels; 200, 250 kg $ha^{-1}$) plots compared to the control (nitrogen fertilization level; 100 kg $ha^{-1}$) plot. Nitrogen use efficiency was showed significantly high in the NPK+rice straw compost (nitrogen fertilization levels; 100, 150 kg $ha^{-1}$) plot compared to the control (nitrogen fertilization level; 100 kg $ha^{-1}$) plot. Therefore, it was suggested that application of organic inputs is helpful in improving soil fertility and physical conditions and thus in N uptake.
The chemical properties of oak tree wood vinegar and the effect of wood vinegar on the tomato seedling were investigated to apply wood vinegar efficiently to the organic - and natural farming system. On the basis of the results from chemical properties of the oak tree wood vinegar, mineral nutrient contents of wood vinegar was low. Therefore, wood vinegar could not be a suitable nutrient source for the plant growth at 500∼1000 times dilution level. which commonly used in the farming, if only wood vinegar is supplied for the nutrient source for the plant growth. The application of wood vinegar increased root growth up the 500 times dilution level while decreased shoot growth. Furthermore. the anion concentrations such as nitrate and phosphate of the plant were decreased by the application of wood vinegar while cation concentrations such as K. Ca. and Mg were increased. Phenolic compounds of wood vinegar such as chlorogenic acid and ferulic acid enhanced the root growth. Interestingly the application of ferulic acid increased both root and shoot growth at the level of 10$^{-4}$ M concentration. It indicated that the effect of wood vinegar on the production of healthy plant seedling may be due to the beneficial root growth by phenolic compounds such as chlorogenic acid and/or ferulic acid of the wood vinegar. However. the effect of the wood vinegar on the plant growth could be influenced by synergism or antagonism of different phenolic compounds in wood vinegar used. In addition. drench in the soil of wood vinegar may be more beneficial compared to foliar application for the improvement of root activity and plant growth.
