NAG Activity and Urinary Excretion of Hippuric Acid Among Workers Exposed to Aromatic Organic Solvents (방향족 유기용제 폭로근로자들의 요중 N-Acetyl-β-Glucosaminidase Activity와 마뇨산농도)

  • In order to investingate the renal dysfunction in workers exposed to aromatic organic solvents, 105 male exposed workers and 109 controls were participated in this study. This study was conducted to examine the correlation among the concentration of aromatic organic solvents in air, worker's urinary NAG activity and urinary hippuric acid. The results were as follows : 1. The exposure level of aromatic organic solvent mixture caculated by the equation proposed by ACGIH was ranged from 0.27 to 0.54 and toluene concentration was ranged from 18.3ppm to 48.3ppm. 2. NAG activity in the urine of control and exposed group were $36.1{\pm}14.2nmolMU/mg$ creatinine and $52.4{\pm}26.1nmolMU/mg$ creatinine, respectively. Hippuric acid concentration in the urine of control and exposed group were $191.1{\pm}194.2mg/g$ creatinine and $789.3{\pm}784.1mg/g$ creatinine, respectively. 3. Correlation coefficent between the exposure level of aromatic organic solvent mixture was significantly related to urinary NAG activity(r=0.542) and urinary hippuric acid(r=0.752). 4. Correlation coefficient between NAG activity and hippuric acid concentration was 0.357. This study suggested that urinary NAG activity was a good renal function index for aromatic organic solvents exposure and urinary hippuric acid was an index for the biological monitoring of toluene exposure.

A Study on the Efficacy of Dietary Supplementation of Organic Acid Mixture in Broiler Chicks (육계에 대한 복합 유기산제의 첨가급여 효과에 관한 연구)

  • This experiment was conducted to investigate the effects of dietary supplementation of organic acid mixture on growth performance, cecal microflora, blood characteristics and immune response in broiler chicks and to prove the possibility of organic acid mixture as an alternative to antibiotics growth promotor. A total of four hundred eighty, 1-day-old male broiler chicks (Ross$\times$Ross 308) were randomly divided into 4 groups with 4 replicates of 30 birds each. The treatments were NC (free antibiotics), PC (basal diet with virginiamycin 10 ppm and salinomycin 60 ppm), 0.3% organic acid, and 0.5% organic acid. The final body weight and body weight gain were significantly higher in organic acid 0.5% than NC (P<0.05). The feed conversion ratio in all treated groups were significantly improved as compared to that of NC (P<0.05). The carcass rate and relative organs weight were not significantly difference among the groups. The relative weight and length of small intestine in PC were significantly decreased than the other groups. The numbers of cecal coliform bacteria and Salmonella in all treated groups were significantly lower than NC (P<0.05). The number of cecal lactic acid bacteria was not different among the groups. No significant differences among the groups were observed in the contents of total cholesterol, triglyceride, blood urea nitrogen (BUN), albumin, aspartate aminotransferase (AST), and alanine aminotransferase (ALT) in blood serum. The contents of total protein and globulin in blood serum of PC and organic acid treated groups were significantly increased as compared to those of NC (P<0.05). Therefore, albumin:globulin ratio of PC and organic acid treated groups was significantly lower than NC (P<0.05). The total white blood cell (WBC), heterophil, lymphocyte, and stress indicator (heterophil:lymphocyte ratio) were not significantly different among the groups. No significant difference was observed on the expression rate of splenic cytokines mRNA in organic acid treated groups compared to the control. Consequently, supplemental organic acid mixture improved the growth performance, and influenced positive effects on the intestinal microflora by inhibiting the growth of harmful bacteria without any adverse effects on relative weights of organs and blood biochemical parameters in broiler chicks.

Inhibitory Effects of Organic Acids against Pectinolytic Yeasts Isolated from Decayed Citrus (연부현상이 발생한 감귤로부터 분리한 효모에 대한 유기산의 생육 저해 효과)

  • Organic acids are known as natural sanitizers. We examined the sanitizing effects of five organic acids (acetic acid, propionic acid, citric acid, malic acid, and lactic acid) and their persistence on three pectinolytic yeast strains isolated from decayed citrus, and the persistence of their sanitizing effects was determined during storage at $4^{\circ}C$ and $16^{\circ}C$. The 7~8 log CFU/mL of the mixed three yeast mixture was exposed to various concentrations of each organic acid for 1 min. The yeast mixtures decreased under detection limit(1 log CFU/mL) in 1% of acetic acid, followed by in 3% of propionic acid with the reduction of 5 log CFU/mL. The citric acid, malic acid, and lactic acid decreased the number of yeasts under detection limit at 7.5%. When treated with deionized water and 1~5% of organic acids were treated on the surfaces of citrus contaminated by yeasts, total numbers of the yeasts decreased under detection limit(3 log CFU) at 5% of acetic acid and 4 log CFU/piece at 5% propionic acid compared with deionized water. When treated with acetic acid and propionic acid on the stem ends of the contaminated citrus, total numbers of the yeasts significantly decreased 0.5 log CFU/piece at 3% of both organic acids. During storage at $4^{\circ}C$ and $16^{\circ}C$ for 20 days, total number of yeasts significantly decreased at 2% acetic acid compared with deionized water. This study suggested that organic acids could be used to sanitize microbial contaminants from citrus for storage and transportation.

Effects of Dietary Supplementation of Organic Acid and Antibiotics Mixture on Growth Performances and Blood Metabolites in Growing Pigs (사료내 유기산제 및 항생제의 혼합첨가가 육성돈의 성장과 혈액성상에 미치는 영향)

  • This study was carried out to evaluate the effect of replacing antibiotics by organic acid mixture on growth performances and blood metabolites in growing pigs. Twenty-five crossbred pigs (Large White${\times}$Landrace) at the age of 79 days were fed five different diets by supplementing organic acid mixture and chlortetracycline. The experimental diets were consisted of diets without antibiotics supplementation (control), diets added 100mg/kg of chlortetracycline to control diet (T1), diets added 100mg/kg of chlortetracycline and 0.1% of $Acidomix^{(R)}$ (comprising formic acid 25%, sorbic acid 10%, fumaric acid 10%) to control diet (T2), diets added 0.1% of $Acidomix^{(R)}$ to control diet (T3), and diets added 0.3% of $Acidomix^{(R)}$ to control diet (T4). The changes in feed conversion ratio, average daily gain and blood metabolites were investigated. Twenty-five pigs were allotted to five treatments with five replications of each and the experiment was conducted on the basis of complete randomized design for 6 weeks. Average daily gain was significantly (p<0.05) different between T4 and control diets. All treatments including diets added $Acidomix^{(R)}$ and chlortetracycline were slightly higher than control diets. The feed intakes did not show a significant difference between the control and other treatments, and did not give change in feed intake by the addition of $Acidomix^{(R)}$. No differences on feed conversion ratio among treatments were observed but T4 and T3 treatments showed lower value than other treatments. The concentrations of blood urea nitrogen, total cholesterol and triglyceride were significantly (p<0.05) different among treatments, but their values were within the normal range representing that effects on blood profile by organic acid or antibiotics supplementation were not found. The results from this study indicated that adding chlortetracycline or organic acid mixtures to diets showed tendency to improve average daily gain and feed conversion ratio in growing pigs. These results showed that antibiotics could be replaced by organic acid mixture in growing pig diets, leading to stimulated growth and improved feed conversion ratio.

Study on the Development of Horticultural Media using Recyled Used-mushroom-media (버섯폐배지를 재활용한 원예용 상토 개발에 관한 연구)

  • As for chemical characteristics of cultivated media waste, the total content of nitrogen, phosphoric acid, and potassium was 6.93%; organic matter content was 67.78%; pH and EC were 7.20 and 1.46 ds/m, respectively. Organic acids identified during stabilization included citric acid, oxalic acid, fumaric acid, and acetic acid, which was highest. During the stabilization of cultivated media waste, temperature decreased and pH increased on the 15th day, and the germination index was 88 after the period, indicating that at least 15 days of stabilization is needed when cultivated media waste is used for growth material of crop. As for germination characteristics, when the mixture rate of cultivated media waste was 20% and under, germination of cucumber and oriental melon was favorable without difference compared to the control plots. For lettuce and Chinese cabbage, no statistically significant difference was found when the rate was 10% and under compared to the control plots. As for seedling quality after 20 days of sowing, cucumber, oriental melon, and Chinese cabbage showed favorable growth only when the mixture rate of cultivated media waste was 10% and under; lettuce showed lower growth characteristics regardless of mixture rate when compared to the control plots. Cultivated media waste may be significantly useful for cucumber and oriental melon, but not for Chinese cabbage and lettuce.

  • A separation/purification process for enzymatic sugar ester production was investigated The crude reaction mixture contained sugar ester and unreacted fatty acid in acetone. The reaction mixture was mixed with KOH/phosphate buffer. Hexane was then added to enhance phase separation. Three phases formed: a lower aqueous phase containing nothing of interest, a polar organic solvent middle phase that contained mostly fatty acid soaps, and a hexane-rich upper phase that contained mostly sugar ester. Distribution coefficient of each component was measured.

  • We have investigated to observe the photoisomerization using the mixture solutions in chloroform and LB monolayers mixed with DPPA and BASH containing azobenzene group which has reversible to cis-trans by light irradiation.. Spreading solutions for the LB films were Prepared in chloroform($5.0{\times}10^{-5}mol/L$). We investigated the photoisomerization and property of the organic ultra thin film of fatty acid containing azobenzene was Prepared on the glass plate by LB method. As a result, the absorption spectra of 8A5H and DPPA mixture of LB films was induced to photoisomerization by alternating irradiation of ultraviolet and visible light, because the condensation of pure azobenzene monolayers was loosened by the introduction of phospholipid into the monolayers, and the molecular high aggregation in pure azobenzene monolayers is also weakened by the introduction of phospholipid. We found that it was reversibly induced to cis-trans photoisomerization in a various mixture molar ratio.

Low Molecular Weight Organic Acids in Brassica pekinensis Rupr. and Growing soil Influenced by Simulated Nitrate Deposition

  • We investigated whether carboxylate exudation of Brassica pekinensis Rupr. was affected by nitrate deposition from simulated acid rain. A gas chromatographic (GC) analysis was employed for the determination of low molecular weight organic acids (LOA) in rhizosphere soils, bulk soil, roots and leaves of Brassica pekinensis Rupr.. Rhizosphere soils were collected after 8 weeks of plant growth by first removing the bulk soil from the root system and then by mechanical move off the rhizosphere soil that adhered to the root surface with soft brush. Soil and plant materials were simultaneously extracted with the mixture of methanol and sulfuric acid (100:7, v/v). Seven organic acids, oxalic, malonic, fumaric, succinic, maleic, L-malic and citric acid were identified and quantified by GC equipped with FID. Oxalic, L-malic, and citric acids were found in both the bulk and rhizosphere soils, while most LOAs were not detected in the control treatment. On the contrary, except maleic acid, all other organic acids were detected in the leaves and roots of cabbages treated with nitrate deposition.

  • A unique porous material with controlled pore characteristics can be fabricated by the freeze-drying process, which uses the slurry of organic material as the sublimable vehicle mixed with powders. The essential feature in this process is that during the solidification of the slurry, the dendrites of the organic material should repel the dispersed particles into the interdendritic region. In the present work, a model experiment is attempted using some transparent organic materials mixed with glass powders, which enable in-situ observation. The organic materials used are camphor-naphthalene mixture (hypo- and hypereutectic composition), salol, camphene, and pivalic acid. Among these materials, the constituent phases in camphor-naphthalene system, i.e. naphthalene plate, camphor dendrite, and camphor-naphthalene eutectic exclusively repel the glass powders. This result suggests that the control of organic material composition in the binary system is useful for producing a porous body with the required pore structure.

Growth Characteristics of Azotobacter vinelandii UWD Using Glucose and Organic Acids as Substrates (포도당 및 유기산을 이용한 Azotobacter vinelandii UWD 의 생장 특성)

  • Azotobacter vinelandii UWD synthesizes poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV), one of the biodegradable polymers, when odd and even number carbon sources are simultaneously added to a medium. In this study, we investigated the specific growth rate of Azotobacter vinelandii UWD on propionic acid and valeric acid. The specific growth rates were $0.183 hr^{-1} and 0.137 hr^{-1}$ at 1.0∼1.5 g/L of propionic acid and 1.0 g/L of valeric acid, respectively. When a mixture of 0.75 g/L of propionic acid and 0.5 g/L of valeric acid was added to the medium, the specific growth rate was 0.196 hr(sup)-1, which was equal to or higher than those of the individual organic acids. Among 10∼50 g/L of glucose cell growth was best at 20 g/L.
