Organic Solvent-Tolerant Esterase from Sphingomonas glacialis Based on Amino Acid Composition Analysis: Cloning and Characterization of EstSP2

  • Organic solvent-tolerant (OST) enzymes are widely applied in various industries for their activity and stability in organic solvents, for their higher substrate solubility, and for their greater stero-selectivity. However, the criteria for identifying OST enzymes largely remain undefined. In this study, we compared the amino acid composition of 19 OST esterases with that of 19 non OST esterases. OST esterases have increased the ratio of Ala and Arg residues and decreased the ratio of Asn, Ile, Tyr, Lys, and Phe residues. Based on our amino acid composition analysis, we cloned a carboxylesterase (EstSP2) from a psychrophilic bacterium, Sphingomonas glacialis PAMC 26605, and characterized its recombinant protein. EstSP2 is a substrate specific to p-nitrophenyl acetate and hydrolyzed aspirin, with optimal activity at $40^{\circ}C$; at $4^{\circ}C$, the activity is approximately 50% of its maximum. As expected, EstSP2 showed tolerance in up to 40% concentration of polar organic solvents, including dimethyl sulfoxide, methanol, and ethanol. The results of this study suggest that selecting OST esterases based on their amino acid composition could be a novel approach to identifying OST esterases produced from bacterial genomes.

Organic Solvent Stable Lipase from Pseudomonas sp. BCNU 171 (Pseudomonas sp. BCNU 171이 생산하는 유기용매 내성 리파아제)

  • An organic solvent stable lipase from solvent-tolerant Pseudomonas sp. BCNU 171 had an optimal pH of 8 and an optimal temperature of 37℃. This crude extracellular lipase from BCNU 171 exhibited increased stability in the presence of various types of solvents at high concentrations (25%, v/v). The lipase stability was found to be highest in the presence of xylene (137%), followed by toluene (131%), octane (130%), and butanol (104%). Overall, BCNU 171 lipase tended to be more stable than immobilized commercial lipase (Novozyme435) in the presence of organic solvents. Furthermore, BCNU 171 lipase maintained about 90% of its enzyme original activity in the presence of NH4+, Na+, Ba2+, Hg2+, Ni2+, Cu2+, and Ca2+ion and significantly increased its enzyme activity in the presence of various emulsifying agents. Thus, the organic solvent stable lipase from Pseudomonas sp. BCNU 171 could be usable as a potential whole cell biocatalyst and for synthetic applications of enzymes for industrial chemical processes in organic solvents without using immobilization.

  • This research was carried to investigate the effects of several organic solvents on the enzymatic transphosphatidylation in emulsion and two-phase solvent systems. The solvents having a similar dielectric constant with diethylether were effective for the enzyme activity. Diethylether and butylacetate were the most effective solvents, when added 12-15%(v/v) and 10-40%(v/v), respectively, for the synthesis of phosphatidylglycerol, phosphatidylethyleneglycol and phosphatidylpropyleneglycol. In the emulsion system, the size of ovolecithin liposome was increased and the clearness of the phospholipid bilayer was reduced as increasing the diethylether concentration. In the twophase solvent system, the rapidest reaction was obtained when water-organic solvent ratio was close to 1. The ratio of aqueous phase. however, should be lowered to 37% to gain the sole product of transphosphatidy1ation, without phosphatidohydrolysis.

Deactivation kinetics of C. rugosa lipase

  • To extend the spectrum of enzyme utilization in the organic solvent system, C. rugosal lipase was selected as a model enzyme because its substrate is soluble to organic solvent. One of the serious disadvantages in this system was the deactivation of the lipase. The pattern of lipase deactivation was the biphasic model. The activation energies for the deactivation were 14.05${\times}$10$^4$ KJ/Kg mole in the first phase and 3.59 ${\times}$ 10$^4$ KJ/mole in the second phase. The several factors were studied for their influences on the pattern of deactivation. Iso-octane as organic solvent influenced more on the first phase than the second phase. Urea as the reagent affecting boty hydrophobic interaction and hydrogen bond of enzyme also influencea more on the first phase. And the optimum pH for the activity was not correlated to that of the stability.

  • A flocculent aggregate produced by Lactococcus lactis subsp. lactis in broths containing Tween 80, including MRS broth, had a microscopic structure of intertwined thread-like filaments. The filamentous structure was not elongated bacterial cells, but consisted of an organic solvent-soluble portion and an insoluble solid. L. lactis subsp. lactis grown at $25^{\circ}C$ for 15 days in tryptic soy broth with 0.1% Tween 80 and 1.0% malt extract produced 13 mg/l of flocculent aggregate, which contained 0.84 g/g of organic solvent-soluble component. The organic solvent-soluble part was identified as 10-hydroxyoctadecanoic acid.

  • Radio iodination of various aromatic derivatives (aniline, toluene, iodobenzene, acetanilide, benzene, benzoic acid) were achieved at low temperature by a chloroamine-T procedures in presence of polar solvent(dioxane). Organic base (piperidine) was used as the catalyst. Iodine replacement reaction had occured on the aromatic or alicyclic ring by this reaction, and the kind and ratio of iodinated products were proved to be different from those of usual iodide reaction in organic solvent at low temperature. The reactivity of various aromatic or alicyclic compounds towards the present iodination system was evaluated and the scope and limitation of the present procedures in the preparation of radio pharmaceuticals were discussed.

Preparation of Silver Nanocolloids Using Silver Alkylcarbamate Complex in Organic Medium with PVP Stabilizer

  • We present a method for chemically reducing silver alkylcarbamate complex with hydrazine to synthesize silver nanocolloids in an organic solvent using polyvinylpyrrolidone (PVP) as the stabilizer. To determine the optimal conditions for preparing stable silver colloids of controlled size and shape, the silver 2-ethylhexylcarbamate (Ag-EHCB) complex, PVP, hydrazine, and 2-propanol solvent concentrations in the reaction mixture were varied. The initial colloid has a mean particle diameter of 5-80 nm, and it exhibits an absorption band with various shapes in the UV region with a maximum near 420 nm. UV-vis spectroscopy, TEM, and X-ray diffraction techniques were used to investigate the formation and growth process of the metallic silver nanocolloids.

  • Using various surfactants, multi-walled carbon nanotube (MWNT) was dispersed in organic solvent, isopropylalcohol(IPA). To refine the MWNT and give the functional group, MWNT was treated with sulfuric acid/nitric acid(v/v=3/1). The cationic, nonionic and anionic surfactants were used as MWNT dispersion agents in the organic solvent. Dispersion effect of various surfactants was observed by optical microscope and HR-TEM. Surface resistivities of MWNT dispersions were measured after coating on PET film. MWNT was dispersed well by poly vinyl pyrrolidone(PVP), nonionic surfactant.

  • Improvement of the photovoltaic efficiency via exposure of organic solar cells to solvent-vapor at room temperature is reported. Carbon disulfide ($CS_2$) vapor treatment can induce Poly(3-hexylthiophene) (P3HT) self-organization into ordered structure leading to enhanced hole transport and light absorption. The power conversion efficiency (PCE) of the organic solar cells can be increased from 0.89 to 1.67% by solvent-vapor treatment.
