The role of cytogenetic tools in orchid breeding

  • Orchidaceae species account for one-tenth of all angiosperms including more than 30,000 species having significant ecological, evolutionary, and economic importance. Despite Orchidaceae being one of the largest families among flowering plants, crucial cytogenetic information for studying species diversification, inferring phylogenetic relationships, and designing efficient breeding strategies is lacking, except for 10% or less of orchid species cases involving mostly chromosome number or karyotype analysis. Also, only approximately 1.5% of the identified orchid species from less than a hundred genera have genome size data that provide crucial information for breeders and molecular geneticists. Various molecular cytogenetic techniques, such as fluorescence in situ hybridization (FISH) and genomic in situ hybridization (GISH), have been developed for determining ploidy levels, analyzing karyotypes, and evaluating hybridity, in several ornamental crops including orchids. The estimation of genome size and the determination of nuclear DNA content using flow cytometry have also been employed in some Orchidaceae subfamilies. These different techniques have played an important role in supplementing beneficial knowledge for effective plant breeding programs and other related plant research. This review focused on orchid breeding summarizes the status of current cytogenetic tools in terms of background, advancements, different techniques, significant findings, and research challenges. Principal roles and applications of cytogenetics in orchid breeding as well as different ploidy level determination methods crucial for breeding are also discussed.

Lecanorchis japonica var. insularis (Orchidaceae: Vanilloideae), a new variety from Jejudo Island, Korea

  • A new variety, Lecanorchis japonica var. insularis (Orchidaceae: Vanilloideae), a mycoheterotrophic orchid from Jejudo Island, Korea, is described and illustrated. It usually grows with L. japonica var. japonica, L. japonica var. hokurikuensis and L. kiusiana in evergreen forests. The newly described variety is distinguished from L. japonica vars. japonica, hokurikuensis, and kiiensis by having fully opened, larger flowers and an obtuse labellum apex.

A Review of Orchid Mycorrhizae in Korea

  • Orchids are evolutionally known to be the most advanced plants in the order Liliales, and comprise approximately 1,000 genera and 35,000 species world-wide. In Korea, more than 110 species of Orchidaceae have been reported to be cultivated or to be collected in the wild. Orchids aye mostly dependant on orchid mycorrhizae(OM) throughout or in part of their life cycle. The OM endomycorrhizae belonging to basidiomycetes or rarley ascomycetes are needed for orchid seed germination. Various fungi, including plant pathogenic, antagonistic and symbiotic fungi, were isolated from the roots of orchid native to Korea. The OM fungi collected from the roots of Cymbidium goeringii were three species of Rhizoctonia namely, R. repens (anamorph state of Tulsanella repens), R. endophytica (Ceratobasidium cornigerum), and an unidentified species (possibly an anamorph of T. calospora). These symbiotic fungi induced peloton in the cortical cells of orchid roots, and differed biologically and in 18s rDNA sequences from plant pathogenic Rhizoctonia species. Also, the mycorrhyzal fungi enhanced the orchid root absorption of nitrogen sources and minerals from the soil. The activity of mycorrhizal fungal hyphae in the roots caused prevention from pathogenic fungi. In nature, the peloton is observed in the cortical cells of Cymbidium goeriingii roots, indicating mycorrhizal colonization in the native orchid roots. On the other hand, pathogenic fungi such as Fusarium and/or Rhizoctonia species are mostly isolated from commercial orchid plants. These suggest that application of symbiotic mycorrhizal fungi should be needed for orchid cultivation in nurseries and at the time of transplanting.

한국 해오라비난초속(난초과)의 미기록 식물: 큰해오라비난초 (A new record of Habenaria (Orchidaceae) to Korean flora: H. dentata (Sw.) Schltr.)

  • 국내 미기록 식물인 큰해오라비난초[Habenaria dentata (Sw.) Schltr.]를 경상남도 합천군 쌍책면과 가회면에서 확인하였다. 큰해오라비난초는 해오라비난초와 형태적으로 가장 유사한 종으로, 꽃받침이 꽃잎보다 크고 흰색이며, 입술꽃잎 곁갈래 가장자리에 얕게 갈라진 톱니가 있는 특징으로 구분된다. 새로운 국명은 해오라비난초에 비해 개체가 크며, 많은 꽃이 달리는 특징에 따라 '큰해오라비난초'로 하였고, 이에 대한 기재, 도해, 사진 및 유사분류군과의 검색표를 제시하였다.

Pollination of Cleisostoma scolopendrifolium (Orchidaceae) by megachilid bees and determinants of fruit set in southern South Korea

  • We investigated the pollinators of Centipede's foot orchid (Cleisostoma scolopendrifolium) at five locations in southern Korea. Only one species of megachilid bee (Megachile yasumatsui) was observed at three of the sites (Mokpo, Haenam, and Wando). We assumed that the megachilid bee worked as a sole pollinator of C. scolopendrifolium based on the visiting behavior of the bees the attached pollinia. Fruits were observed at the three locations that bees visited: 74% fruit set at Mokpo, 59% at Wando, and 31% at Haenam. By contrast, at two of the locations where megachilid bees did not visit the plants (Naju and Jindo), there were no fruits set. The differences in fruit set rates at the three orchid populations where bees were observed appear to be related to bee abundance and flower visitation rate rather than to differences in flower abundance. The pollination interaction between C. scolopendrifolium and megachilid bees appears to involve sexual deception since only male bees were attracted to the flowers. The underlying mechanism involved in this interaction needs investigation.

Molecular Identification of Mycorrhizae of Cymbidium kanran (Orchidaceae) on Jeju Island, Korea

  • A fungal internal transcribed spacer region was used to identify the mycorrhizae of Cymbidium kanran. The family Russulaceae was found to be the most frequently occurring group in both root and soil samples. In phylogenetic analyses, the majority of the Russulaceae clones were clustered with Russula brevipes and R. cyanoxantha. Therefore, C. kanran may form symbiotic relationships with the genus Russula.

Hydrocarbon patterns in Cleisostoma scolopendrifolium (Orchidaceae) as a key mechanism for pollination

  • Cleisostoma scolopendrifolium is an orchid species solely pollinated by the male bee Megachile yasumatsui. Although C. scolopendrifolium is an endangered species in Korea, little is known about its pollination mechanisms or the profiles of its chemical attractants. This study provides evidence that the Cleisostoma orchid attracts male bees as pollinators by mimicking female mating signals. We found 13 hydrocarbons in the Cleisostoma orchid flower presumed to be involved in sex pheromone mimicry: five alkanes (tricosane, pentacosane, heptacosane, nonacosane, and hentriacontane), compounds of cuticular hydrocarbons which function as chemical cues for the recognition of mates and species in social insects; and eight alkenes ((z)-9-tricosene, (z)-9-pentacosene, (z)-11-pentacosene, (z)-9-heptacosene, (z)-11-heptacosene, (z)-9-nonacosene, (z)-11-nonacosene, and (z)-11-hentriacontene) which serve as sex pheromones in several insects. We suggest that these hydrocarbons play a key role in the pollination mechanism between Cleisostoma orchids and Megachile bees.

Two new generic records in the orchid flora of Myanmar

  • The species diversity of Orchidaceae is believed to be extremely high in Myanmar. Like in other plant groups, little basic inventory work has been done thus far. During floristic surveys conducted in Popa Mountain Park (Mandalay Region), Natma Taung National Park (Chin State), and in the Ywangan area (southwestern Shan State), we encountered two interesting orchid species which were determined to be new generic records in the flora of Myanmar. Thuniopsis cleistogama was previously considered endemic to Yunnan Province (China) and was only known from the type locality. However, here the species is newly reported from two localities in central and western Myanmar. Disperis neilgherrensis is known from several widely scattered localities in tropical and subtropical Asia. This tiny orchid species was discovered in the Ywangan area in the southwestern area of the Shan State of Myanmar. These two species will be valuable additions to the updated checklist of the orchids of Myanmar.

한국남부 자생 닭의난초 (난초과)의 시 공간에 따른 결실률 변이 (Spatial and temporal variation on fruit set in Epipactis thunbergii (Orchidaceae) from southern Korea)

  • 난초 열매의 결실의 시 공간적 변화가 장기적으로 개체군의 생존에 영향을 끼칠 수 있으며 수 세대에 걸쳐 유전적 다양성에 영향을 미칠 것이다. 본 연구의 목적은 화밀을 분비하는 육상 난초인 닭의난초를 대상으로 자연 조건에서 시 공간의 결실 수준을 조사하고 교배계를 파악하는 데 있다. 본 연구자들은 전라남도 해남군 징의리 해안 1.5 km 걸쳐 위치한 4곳 집단에서 2년간의 조사 기간 동안 수분실험을 행하였다. 2년 동안 결실률이 집단 내 작은 슈트의 모임인 패치 내에서 유사했다. 대조적으로, 패치 간에는 결실룰이 유의 한 수준에서 차이를 보였다. 또한 큰 화서를 지닌 식물이 작은 화서를 지니는 식물보다 훨씬 더 많은 열매 생성을 보였다. 닭의난초의 결실률은 2 년의 조사 기간 동안 비슷하였으나(14.1%) 연구된 보상을 주는(rewarding) 난초84종의 평균 결실률(37.1%) 보다 낮았다. 인위적인 자가 수분(90.5-95.2%), 인위적인 인화(隣花)수분(geitonogamy: 94.7-95.0%), 그리고 인위적인 타가 수분(91.3-91.4%)등에 의해서는 결실률이 상당한 수준으로 증가되었다. 꽃피기 전 수술을 제거 한 경우에 열매가 전혀 맺히지 않았으며 꽃 스스로 자동으로 수분이 일어나지 않았다. 본 연구 결과에 의하면 닭의난초는 자가화합성이고, 열매를 생성하기 위해서는 수분매개체인 곤충류가 필요하며, 2년간 조사 기간 중 징의리 집단들의 환경이 비슷하였음을 암시하고 있다. 본 연구 결과는 결실률이 시간보다 공간적으로 더욱 뚜렷하다는 점을 강조하고 있다.

Genetic diversity and population structure of endangered Neofinetia falcata (Orchidaceae) in South Korea based on microsatellite analysis

  • Population genetic assessment is essential for the conservation and management of endangered and rare plants. Neofinetia falcata is endangered epiphyte orchid and protected by law in Korea. In Korea, this species is only found on islands in the South Sea of Korea (including Jeju-do) and the southern coast of the Korean Peninsula. We developed nine microsatellite makers to assess the genetic diversity and population genetic structure of three populations of N. falcata. The genetic diversity at the species level was low, which can be attributed to inbreeding or fragmentation into small, isolated populations. A recent bottleneck was detected in one population, likely due to overcollection. N. falcata exhibited moderated levels of differentiation among populations, with the three populations were divided into two clusters based on genetic structure. The genetic diversity and structure of N. falcata are affected by restricted gene flow by pollen or seeds due to isolation and geographic distance. Strategies for in situ and ex situ conservation of this species are been proposed based on the results of our study.
