$H_2O_2$로 유도된 WB-F344 세포의 GJIC 억제에 대한 색상별 파프리카 추출물의 보호 효과 (Protective Effects of Lipophilic Extracts from Different Colored Paprikas on Inhibition of $H_2O_2$-induced Gap Junctional Intercellular Communications)

  • This study analyzed phytochemicals, including various carotenoids, tocopherol and L-ascorbic acid, in green, yellow and orange paprikas (GP, YP and OP) and measured the preventive effects of lipophilic extracts from different colored paprikas on the blockage of gap junctional intercellular communication (GJIC), which is known as a cellular event associated with tumor promotion. Main carotenoids were lutein and ${\beta}$-carotene in GP, lutein, ${\beta}$-carotene, capsanthin, violaxanthin, ${\beta}$-carotene and capsorubin in YP, and lutein, ${\beta}$-carotene, cryptoxanthin and zeaxanthin in OP. Total carotenoid contents were $65.54{\pm}15.87$ mg/100 g dw in OP, $11.98{\pm}0.69$ mg/100 g dw in YP and $10.30{\pm}1.43$ mg/100 g dw in GP. Tocopherol contents were highest in GP compared with in YP and OP, whereas L-ascorbic acid contents were very high in all paprikas. We determined the non-cytotoxic levels of paprika extracts by MTT assay, which showed less formation of reactive oxygen species (ROS) induced by $500{\mu}M$ $H_2O_2$ for 1h. Finally, we showed that pretreatment of paprika extracts prevented inhibition of GJIC induced by $500{\mu}M$ $H_2O_2$ by the scrape-loading/dye-transfer technique. In conclusion, each colored paprika has unique phytochemicals and showed a protective effect on inhibition of GJIC.

모세관 기체 크로마토그래미에서 HETP에 영향을 미치는 인자 (Factors Affecting HETP in Capillary Gas Chromatography)

  • 모세관 기체 크로마토그래피에서 HETP에 영향을 미치는 인자를 고찰하기 위해서 이동상의 유량에 대한 체류분포곡선을 측정하였다. 상관도($r^2$)를 기준으로 측정된 분포곡선으로부터 1, 2차 모멘트와 HETP를 계산하여, 실험식의 매개변수를 추산하였다. 실험식은 이동상의 유속에 대한 함수로서 표시되었으며 이론적인 Golay 식도 고려하였다. 실험에 사용된 시료는 글 표피에 많이 포함된 항암제인 d-limonene와 POH이었다. 실험결과에 의하면, 유속이 증가함에 따라 d-limonene과 POH의 HETP가 증가하였다. d-limonene의 상관도는 0.8265에서 0.8465이었고, POH에서는 0.9353에서 0.9374범위를 나타내었다. $H_g$, $H_{l}$$H_{c}$를 각각 계산하여 전체 HETP에 미치는 영향을 고찰한 결과, d-limonene과 POH의 $H_{g}$가 HETP에 가장 큰 영향을 주었지만, $H_{l}$는 거의 영향을 미치지 않았다. HETP에 관하여 최적 이동상의 유속이 존재하였다.

Lycopene Content and Fruit Morphology of Watermelon (Citrullus lanatus) Germplasm Collections

  • High-quality and high-phytonutrient watermelon fruits have strong market opportunities besides their health related benefits. Hence, investigating quality and nutritional related traits of watermelon genetic resources could provide important baseline data in breeding for increased lycopene content thereby increasing the marketability of watermelon. To this end, we have examined some fruit morphological traits and lycopene content of 105 genetic resources. The morphological characters were recorded on the field and inside laboratory and lycopene was measured using spectrophotometric and HPLC methods. Watermelon fruits have shown a diverse morphological characters. Red and pink fleshed fruits dominated in the entire collections. Fruits with higher thickness of rind were found to exhibit less soluble solid content (SSC). Korean origin fruits were characterized by intermediate SSC while USA, RUS, TJK, TKM, TWN, and URY originated fruits had the highest SSC. The lycopene content varied between 41.37 and 182.82 ㎍/g, 2.81 and 163.72 ㎍/g, and 3.54 and 255.47 ㎍/g using HPLC, UV-Vis, and microplate reader instrumnets, respectively. Red- and pink-fleshed fruits had the highest levels of lycopene content compared to the yellow- and orange-fleshed. Lycopene content had a significant positive correlation with SSC, however, no correlations were detected between lycopene and other quantitative fruit morphological characters.

단고추 종류와 데침처리에 따른 영양성분 변화 및 잔존율 비교 (Changes of Nutrient Composition and Retention Rate of Sweet Pepper by Different Types and Blanching Method)

  • Purpose: To provide the nutritional information of sweet peppers, we have evaluated changes of nutrient content and retention rates by different sweet pepper types and blanching method. Methods: Four types of raw and blanched ($100^{\circ}C$, 30 sec) sweet peppers were prepared and their weight, nutrient content (3 water soluble vitamins, 10 minerals) and retention rates were analyzed. Results: The weight of blanched sweet peppers was decreased than that of raw sweet peppers. From the results of the two-way analysis, the type of sweet pepper had a greater influence than blanching method used on the thiamine, riboflavin, folate and vitamin C contents (but not niacin) (p<0.05). Red pimento contained the most thiamine (0.1 mg/100 g), riboflavin (0.16 mg/100 g), folate ($47.89{\mu}g/100g$) and total niacin (0.11 mg/100 g). Red paprika contained high levels of riboflavin (0.15 mg/100 g) and folate ($44.96{\mu}g/100g$). Orange paprika contained the highest vitamin C content (116.29 mg/100 g). In addition, the mineral content, with the exception of Se, was strongly influenced by the type of sweet pepper rather than the blanching method (p<0.001). The K content of sweet peppers was higher than other minerals. Overall, after blanching, high retention rates (over 70%) were observed in all types of sweet peppers. However, the retention rates of thiamin, vitamin C and Se showed lower levels (less than 70%). Conclusion: The vitamin and mineral contents depends more on the type of sweet pepper; although vitamin and mineral (except Se) contents showed some changes after blanching. Our results can be utilized to determine the effects of cooking processes on nutritional information.

Preparation and Characteristics of Calcium Lactate from Black Snail

  • Calcium lactate (CL) prepared from powdered black snail (PBS) or its ashed powder (ABS), was investigated for ideal manufacturing conditions to optimize color, solubility and sensory quality. Based on the amount of PBS and 100 mL lactic acid (LA), the yields of PBS-CL were 300% and 15 g in 10% LA and 260% and 20 g in 20% LA. Yields of ABS-CL based on the amount ABS and 100 mL LA were 400% and 60 g in 10% LA and 329% and 66 g in 20% LA. Both of the yields were decreased with an increase of the LA concentration on the basis of PBS and ABS amounts, but proportionally increased with the increment in the LA concentration on the basis of LA volume. Optimal preparation times of the dehydrated PBS-CL and ABS-CL were, respectively, 4 hr and 5 hr at 10$0^{\circ}C$, 3 hr and 4 hr at 12$0^{\circ}C$, and 1 hr and 2 hr at 15$0^{\circ}C$, which showed shorter time in preparing the dehydrated ABS-CL. PBS-CL and ABS-CL were confirmed to be Ca($CH_3$CHOH$CO_2$)$_2$ by the analysis results of IR and $^1$H-NMR. Calcium contents of the anhydrous PBS-CL and ABS-CL were individually 15.4% (w/w) and 17.3% (w/w) representing 84.2% and 94.5% or each theoretical value. Colors or PBS-CL and ABS-CL were light yellow and light-greenish white each. Solubilities of PBS- CL and ABS-CL in distilled water at pH 3~8 were 5.43 and 6.11 g/100 mL, respectively, which demonstrated higher mean solubilities rather than the 4.74 g/100 mL of standard CL. Solubilities of PBS-CL (3.14~5.03 g/100 mL) and ABS-CL (4.69~6.05 g/100 mL) against soup soy sauce, 3% brine, Soju (Korean distilled liquor), thick soy sauce, grape juice and orange juice were higher than those of standard CL (2.94~5.84 g/100 mL). ABS-CL was believed to have a wide use range due to its low sourness while different applications of PBS-CL in food are expected due to its mild astringent taste and strong savory taste despite its strong bitter taste as estimated by sensory evaluation.

식품 중 비페닐 분석법 개발 및 유효성 검증 (Development and Validation of Analytical Method for Determination of Biphenyl Analysis in Foods)

  • 본 연구에서는 국내 미설정 보존료인 biphenyl의 분석법 확립을 위해 국내외 다양한 분석법을 조사하고 비교 검토하였다. 식품용 기구 및 용기 포장공전 목재류 시험법의 경우 범용적 장비와 비교적 간단한 시험 전처리 방법이지만 회수율이 10% 이하로 측정되었으며 방해 물질 제거가 어려워 효과적인 시료 전처리와 분석방법이 될 수 없음을 확인하였다. 식품공전의 잔류농약 분석법의 경우 시료 전처리에 사용된 추출 용매는 아세토니트릴이고 정제는 아미노프로필 카트리지를 이용하였으며, 정성 및 정량분석이 가능하고 효과적으로 matrix를 제거하는 장점이 있으나, 30% 이하의 회수율과 전처리 시간이 많이 소요되는 것을 확인하였다. AOAC official method 분석법의 경우 n-heptane을 이용하여 추출 후 무수황산나트륨 칼럼을 이용하여 정제한 뒤 TLC와 spectrophotometry를 이용하여 확인 시험 결과 시료 전처리에 시간 소요가 많고 재현성과 숙련도가 요구되는 단점이 확인되었다. QuEchERs 방법은 시료를 아세토니트릴로 추출 후 dispersive SPE kit을 이용하여 전처리 후 GC-MS와 HPLC를 이용하여 확인 결과 시료 전처리 시간이 비교적 짧고 좋은 재현성이 있는 장점이 있으나, 소모품 사용 시 경제적 부담이 큰 단점을 가지고 있다. Anklam E 등의 시험법은 GC-FID이며 디클로로메탄, 메탄올, 헥산을 이용한 단순 추출 후 2시간 방치한 다음 GC-MS와 GC-FID로 분석하는데, 일부 시료에서 효과적인 matrix 제거가 어려운 것으로 확인되었다. 따라서 국내외 여러 시험방법을 검토 및 응용하여 간단한 시료 전처리와 분석법을 확립하였다. 시료 중의 biphenyl 분석에 사용한 칼럼은 Capcellpak UG 120 $C_{18}$($4.6mm{\times}250mm$, $5{\mu}m$), 이동상은 3차 증류수에 녹인 25 mM phosphoric acid(solvent A)와 아세토니트닐에 녹인 25 mM phosphoric acid(solvent B)를 이용하여 gradient로 분석하였다. UV 흡수 파장은 230 nm, 오븐 온도는 $40^{\circ}C$, 유속은 1.0 mL/min을 이용하였다. 직선성은 0.999($R^2$) 이상의 높은 직선성을 보였으며 직선성 각각의 농도에 대한 정밀도는 6.7% 이하, 정확도는 86.7~102.9%, 검량선 내 서로 다른 농도 3가지에 대한 일내 정밀도는 0.7~1.9%, 정확도는 95.8~104.0%로 확인되었다. 일간은 정밀도가 1.6~2.4%, 정확도는 96.7~102.4%로 확인되었다. 검출한계 $0.04{\mu}g/mL$, 정량한계는 $0.13{\mu}g/mL$로 측정되었다. 회수율 측정 결과 3가지 농도에서 정밀도는 1.8% 이하이고, 회수율은 92.7~99.4%로 측정되었다. 확립된 biphenyl의 분석법 확립의 적용성 검토를 위해 시료는 총 111건을 수집하였으며, 일본 현지에서 구입한 시료는 30건, 서울 경기 대형마트 및 중소형 식품 도매점에서 81건을 수집하여 총 111건에 대해 분석한 결과 모든 시료에서 불검출이 확인되었다. 향후 국내 미설정 보존료인 biphenyl을 사용한 식품에 대해 정량한계($0.13{\mu}g/mL$)까지 분석할 수 있는 분석법 확립과 국내에 비의도적으로 혼용될 수 있는 수입식품의 사전사후 관리에 기여될 수 있을 것으로 판단된다.

2008년 국민건강영양조사에 근거한 과채류 주스 음용으로부터 한국인의 일인당 하루 총페놀, 총플라보노이드 및 항산화능 섭취량 추정 (Estimation of Daily Per Capita Intake of Total Phenolics, Total Flavonoids, and Antioxidant Capacities from Fruit and Vegetable Juices in the Korean Diet Based on the Korea National Health and Nutrition Examination Survey 2008)

  • 본 연구에서는 2008년도 국민건강영양조사 자료를 분석하여 우리나라에서 가장 많이 소비되는 상위 7종류의 과채류 주스(사과, 포도, 감귤, 오렌지, 석류, 파인애플, 토마토)를 선정하고, 이 주스들의 총페놀, 총플라보노이드 및 항산화능 함량을 정량 분석하였다. 2008년에 실시한 국민건강영양조사의 자료 분석을 통해 오렌지 주스(6.31 g/capita/day), 포도 주스(2.28 g/capita/day), 토마토 주스(1.50 g/capita/day), 감귤 주스(1.23 g/capita/day), 사과 주스(1.05 g/capita/day), 석류 주스(0.17 g/capita/day), 파인애플 주스(0.05 g/capita/day) 등 7가지 과채류 주스의 하루 일인당 주스 소모량을 산출하였다. 총 7종류의 과채류 주스의 섭취를 통해서 얻어진 일인당 하루 섭취량의 경우, 총페놀 섭취량은 11.70 mg GAE/capita/day, 총플라보노이드 섭취량은 1.65 mg CE/capita/day, 그리고 항산화능은 10.42 mg VCE/capita/day(DPPH 분석법)와 13.21 mg VCE/capita/day(ABTS 분석법)였다. 일인당 하루 총페놀 섭취량, 총플라보노이드 섭취량 및 항산화능 섭취량 추정은 과채류 주스의 페놀성 화학물질 등의 생리활성성분의 농도뿐만 아니라 일인당 하루 과채류 주스 소모량에도 영향을 받는 것으로 나타났다.

섬유소 분해효소 및 니코틴 분해세균을 이용한 잎담배의 발효촉진효과 (Studies on the promoted Aging of flue-cured leaf tobacco by cellulolytic Enzyme and Nicotinophiles)

  • For the quality enhancement of harvested-year leaf tobacco to the quality of 2-year naturally aged leaf tobacco, cellulose and nicotine degradative bacteria were isolated and identified. Effects of artificial fermentation treated cellulase and nicotine degradative bacteria on the quality of leaf tobacco were investigated from the chemical and sensory points of view. 1, Changes in chemical composition of leaf tobacco resulted from the addition of cellulase extracted from Cellulomonas sp. [3ml(${\mu}{\textrm}{m}$ D-glucose/ml. mil-1) of enzymes solution 11009 of leaf tobacco] and nicotine degradative bacteria, Pseudomonas sp. 2ml(IX109 cells$\div$ 100g of leaf tobacco), and subsequently fermented at 40${\mu}{\textrm}{m}$$^{\circ}C$, 65% R. H. for 40 days are as follows : 1) Content of crude fiber decreased 12% It took 9 min, 53 sec. to reach full combustion in control group but took only 7 min. 47 sec. in the treated group, taking almost equal time to 2-year naturally aged leaf tobacco(7 min. 35sec.). 2) Light intensity of control group was 60.96% with bright lemon color but that of treated leaf tobacco accounted for 47.69 with orange to dark brown color series, which was almost equal to the value, 45.69, of 2-year naturally aged leaf tobacco. 3) Linoleic acid, serving mild taste among organic acids, amounted to 1.llmg/g in control group but increased to 1.35m9/9 in the treated leaf tobacco, identical to the content(1.35mg/g) of 2-year naturally aged leaf tobacco. 4) Content of solanone, on of the typical leaf tobacco flavor compounds, accounted for 2.95% in control group but increased to 2.87% in treated group. 5) Methyl furan, useful flavor compound in smoke composition, accounted for 17.6$\mu\textrm{g}$/cig. in control group but increased to 25.9$\mu\textrm{g}$/cig. in treated group. However, acroleine decreased from 69.3$\mu\textrm{g}$/cig. in control group to 58.6$\mu\textrm{g}$/cig. in treated group 2. In sonsory test, mild taste evaluation of control group scored 5.47 and treated group 7.93 which was evaluted almost equal to the value(8.00) of 2-year naturally aged leaf tobacco. Aroma evaluation of control group scored 5.60, treated group 8.20, and 2-year naturally aged leaf tobacco 8.33. In addition, total harmony taste of control group showed 5.67, treated group 8.07 (p<0.01), and 2-year naturally aged leaf tobacco 8.00. From these results, it can be said that quality of treated leaf tobacco is not inferior to that 2-year naturally aged leaf tobacco.

Inhibition of Cell Growth and Induction of Apoptosis by Euonymus Alatus (Thunb.) Sieb in Human Leiomyomal Smooth Muscle Cells

  • Objective : Euonymus alatus (Thunb.) Sieb (EA) is a traditional Korean herbal medicine, commonly used to treat tumors in Korea and China for centuries. Several earlier studies have indicated that EA exhibits anti-tumor properties, but its mechanism remains to be elucidated. In this study, we evaluated the molecular mechanism of EA in a human uterine leiomyomal smooth muscle cell (ULSMC) line. Methods : This study was evaluated by: (a), morphological changes by using acridine orange/ethidium bromide staining; (b), DNA fragmentation by TdT-mediated dUTP nick end labeling (TUNEL); and (c), sub-G1 cell analysis. Results : This study observed that EA treatment caused apoptotic cell death and depletion of intracellular glutathione (GSH) and that reduction of mitochondrial membrane potential was found to be involved in the initiation of apoptosis by EA. Conclusion : This results show that EA exerted clear cytotoxic effects and strongly inhibited the proliferation of ULSMC.

Synthesis and Luminescent Properties of $RE^3+(Eu^3+\;and\;Tb^3+$) Ions Activated CaGd4O7 Novel Phosphors

  • Trivalent rare-earth ($RE^{3+}=Eu^{3+}\;and\;Tb^{3+}$) ions activated $CaGd_4O_7$ phosphors were synthesized by a sol-gel process. After annealing at $1,500^{\circ}C$, the XRD patterns of the phosphor confirmed their monoclinic structure. The photoluminescence excitation spectra of $Eu^{3+}$ and $Tb^{3+}$ doped $CaGd_4O_7$ phosphor shows the broad-band excitations in the shorter wavelength region due to charge transfer band of completely filled $O^{2-}$ to the partially filled $Eu^{3+}$ ions and f-d transitions of $Tb^{3+}$ ions, respectively. The photoluminescence spectra show that the reddish-orange ions and green emission for $Eu^{3+}$ and $Tb^{3+}$ ions, respectively. Owing to the importance of thermal quenching property in the technological parameters, the temperature-dependent luminescence properties of these phosphors were measured for examing the suitability of their applications in the development of light emitting diodes (LEDs). In addition to those measurements, the cathodoluminescence properties were examined by changing the acceleration voltage and filament current. The calculated chromaticity coordinates of these phosphors were close proximity to those of commercially available phosphors for LED and field emission display devices.
