Efficacy of Lactobacillus fermentum Isolated from the Vagina of a Healthy Woman against Carbapenem-Resistant Klebsiella Infections In Vivo

  • Carbapenem-resistant Enterobacteriaceae (CRE) that produce Klebsiella pneumoniae carbapenemase are increasingly reported worldwide and have become more and more resistant to nearly all antibiotics during the past decade. The emergence of K. pneumoniae strains with decreased susceptibility to carbapenems, which are used as a last resort treatment option, is a significant threat to hospitalized patients worldwide as K. pneumoniae infection is responsible for a high mortality rate in the elderly and immunodeficient individuals. This study used Lactobacillus fermentum as a candidate probiotic for treating CRE-related infections and investigated its effectiveness. We treated mice with L. fermentum originating from the vaginal fluid of a healthy Korean woman and evaluated the Lactobacilli's efficacy in preventive, treatment, nonestablishment, and colonization mouse model experiments. Compared to the control, pre-treatment with L. fermentum significantly reduced body weight loss in the mouse models, and all mice survived until the end of the study. The oral administration of L. fermentum after carbapenem-resistant Klebsiella (CRK) infection decreased mortality and illness severity during a 2-week observation period and showed that it affects other strains of CRK bacteria. Also, the number of Klebsiella bacteria was decreased to below 5.5 log10 CFU/ml following oral administration of L. fermentum in the colonization model. These findings demonstrate L. fermentum's antibacterial activity and its potential to treat CRE infection in the future.

비경구 투여한 Streptococcus mutans 균체 및 Glucosyltransferase에 대한 마우스의 면역항체반응 (ANTIBODY PRODUCTION BY PARENTERAL ADMINISTRATION OF STREPTOCOCCUS MUTANS AND GLUCOSYLTRANSFERASE IN MICE)

  • 치아우식증을 예방하기 위한 백신연구를 위하여 주원인균인 Sterptococcus mutans 균체항원과 이 균에서 분리한 glucosyltransferase (GTF)를 항원으로 하고 이에 면역조절기능이 있는 retinoic acid (RA)를 첨가하여 투여경로와 백신조성이 이들 항원에 대한 면역반응에 미치는 영향을 실험하였다. 균체항원(Ingbritt strain)을 마우스의 피하에 Complete Freund's Adjuvant와 함께 투여하여 생산되는 혈청내 응집항체가는 serotype e (LM-7)와는 강한 교차반응을 보였으나 serotype f (OMZ-175)와는 거의 교차반응을 일으키지 않았다. 면역혈청내 항-GTF 및 항-Ag I/II 항체중 항-GTF IgA는 피하로 투여시 전혀 검출되지 않았으나 이에 RA를 첨가하면 다량의 항체 생산을 관찰하였고 그 정도는 경구투여시의 생산량을 능가하였다. GTF를 alum과 함께 투여하여 생산되는 혈청내 항-GTF 항체중 IgM은 피하로 투여시 상당량이 검출되었고 RA를 첨가하면 그 생산이 증가되었으며 경구로 투여시 대조군에 비하여 약간증가를 보였으나 피하투여시의 그것에는 미치지 못하였다. GTF-특이 IgG는 경구투여시는 전혀 검출되지 아니하였고, 피하투여시에만 현저한 증가를 보였으며, RA첨가는 이에 영향을 미치지 못하였다. 항-GTF IgA는 피하로 투여시 전혀 검출되지 아니하였으나 이에 RA를 첨가하면 증가된 항체생산을 관찰하였고 그 정도는 경구투여시의 생산량을 능가하였다. 이상의 실험성적은 GTF에 대한 항체생산은 투여경로와 항원의 종류에 따라 다양한 반응을 나타내며 RA는 이를 백신에 첨가하면 피하경로를 이용하여 면역하더라도 경구투여와 유사한 IgA-매개 면역반응으로 조절시킬 수 있는 가능성을 나타내었다.

경기지역 일부 고등학생들의 구강건강 행태 분석 (A analysis on oral health behaviors of High School students in Gyeonggi)

  • Objectives : The purpose of this study was to examine the oral health behavior and examine closely the connection factor and plans oral health promotion. Methods : This survey was conducted on 370 high school students in Kyunggi-do Suwon from November 21 to 23, 2011. 352 questionnaires were collected and analyzed. The collected data was analyzed using the statistical package SPSS WIN 15.0 using frequency, mean and standard deviation analysis, T-test, one-way ANOVA and Duncan's test correlation analysis. Results : The results state that 73.3% of students believe brushing teeth is very important and 26.7% of the students method of tooth brushing is up and down. for correct teeth brushing, they think choosing a brush is important(70.7%) also their brush shapes were uneven form(44.3%) and flat form(43.5%). 31.4% of students are using an oral hygiene device and 31.4% of students had received oral health education. 42.7% of female students get information about oral health from their dental clinics. 24.8% of male students receive information on oral health from parents and relatives. They think the best educator for oral health education will be a Dental hygienist(46.5%), or Dentist(48.0%). we found that a correlation between oral health interest, importance, knowledge and condition effect each other(p<.001). It means that if students have high oral health interest, oral care importance and oral health knowledge which can improve their oral health condition. Conclusions : The results of this study state that we need continuous oral health education systems and prevention programs to raise awareness among students and for the improvement of their oral health.

The Essential Oil of Artemisia iwayomogi Kitamura Induces Apoptosis on Human Oral Epidermoid Carcinoma Cells

  • The aerial part of Artemisia iwayomogi Kitamura has traditionally been used for inflammation, infectious disease, cancer, pyretic, diuretic, liver protective effect, and choleretic purposes in Korea. We investigated that the essential oil induces apoptosis in KB cell as evidenced by Hoechst-33258 dye staining, flow cytometry (cell cycles), and DNA fragmentation for nuclear condensation and Western blotting for activation of caspases-3, -8, -9, Bax, Bcl-2, cytochrome c, and poly (ADP-ribose) polymerase (PARP) cleavage. In the present study, we found that the essential oil could induce apoptosis in KB cells, as characterized by DNA fragmentation, activation of caspase-3, -8, and -9, and PARP cleavage. The efficacious induction of apoptosis was observed as a dose-dependent. The essential oil-induced apoptotic cell death was accompanied by up-regulation of Bax and down-regulation of Bcl-2. The essential oil also caused the loss of mitochondrial membrane potential and cytochrome c release from mitochondria to cytosol. These findings indicate that mitochondrial pathways might be involved in the essential oil-induced apoptosis and enhance our understanding of the anticancer function of the essential oil in herbal medicine.

유전자재조합 인간 골형성단백2 및 생흡수성고분자를 이용한 골형성유도체의 개발 (DEVELOPMENT OF BONE REGENERATING MATERIAL USING BONE MORPHOGENETIC PROTEIN(rhBMP-2) AND BIORESORBABLE POLYMER)

  • We tested the bone regenerating capacity and histologic response of bioresorbable matrix-type implant, which was made with Poly(lactide-co-glycolide)(PLGA) and bone apatite for the carrier of bone morphogenetic protein(BMP). The critical size defect of 8mm in diameter was created at the calvaria of SD rats(n=18), and repaired with polymer implant with $15{\mu}g$ of rhBMP-2(n=9) or without it(n=9). At 2 weeks, 1 months after implantation, the animals were sacrificed(3 animals at every interval and group) and histologically evaluated. The calvarial defect which was repaired with polymer with BMP healed with newly formed bone about 70% of total defect. But that without BMP showed only 0 to under 30% bony healing. Inflammatory response was absent in both group through the experimental period, but there's marked foreign body giant response though it was a little less significant in polymer with BMP group. As the polymer was resorbed, the space was infiltrated and replaced by fibrovascular tissue, not by bone. In conclusion, our formulation of bioresorbable matrix implant loaded with bone morphogenetic protein works good as a bone regenerating material. However, it is mandatory to devise our system to have better osteoinductive and osteoconductive property, and less multinucleated giant cell response.

Oral Delivery of Probiotics Using pH-Sensitive Phthalyl Inulin Tablets

  • Probiotics show low cell viability after oral administration because they have difficulty surviving in the stomach due to low pH and enzymes. For the oral delivery of probiotics, developing a formula that protects the probiotic bacteria from gastric acidity while providing living cells is mandatory. In this study, we developed tablets using a new pH-sensitive phthalyl inulin (PI) to protect probiotics from gastric conditions and investigated the effects of different compression forces on cell survival. We made three different tablets under different compression forces and measured survivability, disintegration time, and kinetics in simulated gastric-intestinal fluid. During tableting, there were no significant differences in probiotic viability among the different compression forces although disintegration time was affected by the compression force. A higher compression force resulted in higher viability in simulated gastric fluid. The swelling degree of the PI tablets in simulated intestinal fluid was higher than that of the tablets in simulated gastric fluid due to the pH sensitivity of the PI. The probiotic viability formulated in the tablets was also higher in acidic gastric conditions than that for probiotics in solution. Rapid release of the probiotics from the tablet occurred in the simulated intestinal fluid due to the pH sensitivity. After 6 months of refrigeration, the viability of the PI probiotics was kept. Overall, this is the first study to show the pH-sensitive properties of PI and one that may be useful for oral delivery of the probiotics.

Identification and Detection of Streptococcus anginosus Using Species-Specific 16S rDNA Primers

  • This study was undertaken to develop PCR primers for the identification and detection of Streptococcus anginosus using species-specific forward and reverse primers. These primers targeted the variable regions of the 16S ribosomal RNA coding gene(rDNA). The primer specificity was tested against 12 S. anginosus strains and 6 different species(10 strains) of oral bacteria. The primer sensitivity was determined by testing serial dilutions of the purified genomic DNA of S. anginosus ATCC $33397^T$. The data showed that species-specific amplicons were obtained from all the S. anginosus strains tested, but not in the six other species. The PCR could detect as little as 0.4pg of the chromosomal DNA from S. anginosus. This suggests that the PCR primers are highly sensitive and applicable to the detection and identification of S. anginosus.

The Ethanol Extract of Croton Seed Inhibits the Oral Pathogen, Streptococcus mutans

  • It is noted that Streptococcus mutans (S. mutans) triggers dental caries establishment by two major factors: the synthesis of organic acids, which demineralize dental enamel, and the synthesis of glucans, which mediate the attachment of bacteria to the tooth surface. Therefore, it is noted that the development of a more effective, substantial and safe preventive agent that works against dental caries and periodontal disease is required at this time. For this reason, the present study was designed to investigate the effect of croton seed ethanol extracts on the growth, acid production, adhesion, and insoluble glucan synthesis of S. mutans. In this case, the ethanol extract of croton seed showed concentration dependent inhibitory activity against the growth, acid production and adhesion of S. mutans. Especially, it is important to note that it has produced significant inhibition at the concentration of 0.1 and 0.2 mg/ml as compared to the control group. Moreover, these results suggest that the application of croton seed extract may be considered to be a useful method for the prevention of dental caries.
