정상 성인의 혈청 인슐린 농도에 따른 비만지수, 혈청 지질, 혈압 및 영양소 섭취상태 비교 (Comparisons of BMI, Blood Lipid Profile, Blood Pressure and Nutrient Intakes by Serum Insulin Concentrations)

  • This study was conducted to figure out the differences of the BMI, blood lipids, blood pressure and nutrient intakes by serum insulin concentration among adults in Korea. Ninety-three subjects(male : n=37, female : n=56) participated in this study. The subjects were grouped by serum insulin concentrations into two groups-hyperinsulinemia group(n=17) and normoinsulinemia group(n=76). Anthropometric measurements, blood lipid profiles, blood pressure, oral glucose tolerance test, and daily nutrient intakes were analyzed. And serum glucose and insulin secretion pattern as shown through and oral glucose tolerance test were performed. BMI (p<0.05) and WHR(p<0.01) were significantly higher in hyperinsulinemic group than in normoinsulinemia group. There was no differences in fasting blood glucose level between groups, but total glucose area (p<0.001) and insulin glucose resistance(p<0.001) were significantly higher in hyperinsulinemia group than in normoinsulinemia higher in hyperinsulinemia group, but HDL-cholesterol(p<0.05), and systolic blood pressure(p<0.05) were significantly higher in hyperinsulinemia group, but HDL-cholesterol(p<0.001) was significantly lower in hyperinsulinemia group. They showed significant differences in energy, carbohydrate, potassium, riboflavin, niacin and dietary fiber intakes(p<0.05), the intake of those nutrients were low in normoinsulinemia group. Further investigation is necessary to determine the effects of amounts and types of carbohydrate and dietary fiber on serum insulin concentrations.

Soybean isoflavone extract improves glucose tolerance and raises the survival rate in streptozotocin-induced diabetic rats

  • The present study evaluated the effect of various dosages of soybean isoflavone extract on body weight changes, glucose tolerance and liver function in streptozotocin-induced diabetic rats. One group of normal rats (normal control) was fed an AIN-76-based experimental diet and four groups of diabetic rats were fed the same diet supplemented with four different levels of soybean isoflavone extract for seven weeks. The daily dosages of pure isoflavone for four diabetic groups were set to be 0 mg (diabetic control), 0.5 mg (ISO-I), 3.0 mg (ISO-II) and 30.0 mg (ISO-III) per kilogram of body weight, respectively. The daily consumption of isoflavone at the level of 3.0mg per kilogram of body weight resulted in the suppression of body weight loss and increased the survival rate of diabetic animals one and half times compared to that of the diabetic control group. Blood glucose levels in a fasting state and after the oral administration of glucose were significantly lower in the ISO-II group during the oral glucose tolerance test. The ISO-II group showed a tendency to elongate the gastrointestinal transit time. The activity of serum aminotransferases, indicator of liver function, was not negatively affected by any intake level of isoflavone. The present study demonstrated that the soybean isoflavone extract may be beneficial to diabetic animals by improving their glucose tolerance and suppressing weight loss without incurring hepatotoxicity at the daily dosage of 3.0 mg per kg of body weight.

  • The blood glucose and plasma immunoreactive insulin(IRI) levels were measured glucose tolerance test in 7 healthy subjects and 6 patients with chronic liver diseases. The glucose tolerance was impaired in 5 of the 6 patients and normal in 1. Plasma IRI responses were markedly increased and delayed in all patients, suggesting insulin resistance. Patients with more glucose intolerance showed less increase in plasma IRI than the group with less intolerance. It is suggested that some insulin antagonists may decrease the peripheral insulin sensitivity and stimulate compensatory hyperactivity of pancreatic islets. If the compensatory hyperactivity is inadequate due to genetic predisposition to diabetes mellitus or exhaustion of ${\beta}$-cells of pancreatic islsts, the glucose intolerance and overt diabetes mellitus may ensue.

Anti-hyperglycemic effect and single oral toxicity of SPP003

  • The SPP003 is a mixture of water extract from Schizandrae Fructus, Poligoni multiflori Radix, Ginseng Radix Akba and Hoelen. The aim of this study was to investigate the anti-hyperglycemic effect of SPP003 in normal and streptozotocin (STZ)-induced diabetic rats, and to monitor the toxicity of SPP003. Oral glucose tolerance test (OGTT) was performed after oral administration of SPP003 100, 300, 600 and 900 mg/kg in normal rats. Blood glucese concentration was measured at -30 min (vehicle, SPP003 or tolbutamide 60 mg/kg, 0 min (glucose treatment), 60, 120 and 180 min. Rats were administerd STZ 65mg/kg (0.1M citrate buffer, pH 4.5) intraperitoneally to induce diabetes and administered vehicle, Spp003 (100, 300 and 600 mg/kg) or tolbutamide (60 mg/kg) orally once a day for 4 weeks. Blood glucose level was measured a 0, 4, 7, 14, 21 and 29 day after initial drug administration. A single oral toxicity of SPP003 was studied in Sprague-Dawley rats of both sexes. In this study, rats were administered with doses of 1, 2 and 5 g/kg of SPP003. In glucose tolerance test, SPP003 900 mg/kg markedly decreased glucose concentration at 1 hr after glucose treatment. Blood glucose levels were much higher in STZ-diabetic rats. These increases were significantly attenuated by SPP003 600 mg/kg. SPP003 did not show any signigicant toxicity. These findings suggest that SPP003 has hypoglycemic properties in STZ-diabetic rats.

고지방식유래의 뇌졸중 유발 고혈압 흰쥐(SHRSP)에 있어서 수용성 식이 섬유의 급여가 근섬유 형태에 따른 GLUT4 발현에 미치는 영향 (Effects of Soluble Dietary Fiber on Skeletal Muscle GLUT4 Protein Contents in SHRSP Fed a High-Fat Diet)

  • The Purpose of this study is to investigate the effect of soluble dietary fiber psyllium on insulin sensitivity and skeletal muscle glucose transporter 4(GLUT4) protein expression in stroke-prone hypertensive rats(SHRSP) fed a high-fat diet containing 5% of psyllium or cellulose from five to nine weeks of age. Obtained results were as follows : (1) In the psyllium diet group fasting plasma glucose level was significantly reduced and glucose levels upon oral glucose tolerance test were significantly lower than cellulose diet group at 30 min(p<0.05) and 60 min(p<0.01) (2) Skeletal muscle GLUT4 contents were significantly increased in the soleus(slow twitch) and extensor digitorum longus(fast twitch) muscle of psyllium diet group. (3) However there was no difference in insulin levels in the fasting and oral glucose tolerance test. These results indicated that psyllium diet improves peripheral insulin sensitivity but not insulin secretion. In conclusion our present finding suggest that soluble fiber diet is effective to increase insulin sensitivity in SHRSP. From these results it was suggested that soluble dietary fiber supplementation effectively prevents insulin resistance.

Genetic Association Analysis of Fasting and 1- and 2-Hour Glucose Tolerance Test Data Using a Generalized Index of Dissimilarity Measure for the Korean Population

  • Glucose tolerance tests have been devised to determine the speed of blood glucose clearance. Diabetes is often tested with the standard oral glucose tolerance test (OGTT), along with fasting glucose level. However, no single test may be sufficient for the diagnosis, and the World Health Organization (WHO)/International Diabetes Federation (IDF) has suggested composite criteria. Accordingly, a single multi-class trait was constructed with three of the fasting phenotypes and 1- and 2-hour OGTT phenotypes from the Korean Association Resource (KARE) project, and the genetic association was investigated. All of the 18 possible combinations made out of the 3 sets of classification for the individual phenotypes were taken into our analysis. These were possible due to a method that was recently developed by us for estimating genomic associations using a generalized index of dissimilarity. Eight single-nucleotide polymorphisms (SNPs) that were found to have the strongest main effect are reported with the corresponding genes. Four of them conform to previous reports, located in the CDKAL1 gene, while the other 4 SNPs are new findings. Two-order interacting SNP pairs of are also presented. One pair (rs2328549 and rs6486740) has a prominent association, where the two single-nucleotide polymorphism locations are CDKAL1 and GLT1D1. The latter has not been found to have a strong main effect. New findings may result from the proper construction and analysis of a composite trait.

Efficacy and safety of Panax ginseng berry extract on glycemic control: A 12-wk randomized, double-blind, and placebo-controlled clinical trial

  • Background: Antihyperglycemic effects of Panax ginseng berry have never been explored in humans. The aims of this study were to assess the efficacy and safety of a 12-wk treatment with ginseng berry extract in participants with a fasting glucose level between 100 mg/dL and 140 mg/dL. Methods: This study was a 12-wk, randomized, double-blind, placebo-controlled clinical trial. A total of 72 participants were randomly allocated to two groups of either ginseng berry extract or placebo, and 63 participants completed the study. The parameters related to glucose metabolism were assessed. Results: Although the present study failed to show significant antihyperglycemic effects of ginseng berry extract on the parameters related to blood glucose and lipid metabolism in the total study population, it demonstrated that ginseng berry extract could significantly decrease serum concentration of fasting glucose by 3.7% (p = 0.035), postprandial glucose at 60 min during 75 g oral glucose tolerance test by 10.7% (p = 0.006), and the area under the curve for glucose by 7.7% (p = 0.024) in those with fasting glucose level of 110 mg/dL or higher, while the placebo group did not exhibit a statistically significant decrease. Safety profiles were not different between the two groups. Conclusion: The present study suggests that ginseng berry extract has the potential to improve glucose metabolism in human, especially in those with fasting glucose level of 110 mg/dL or higher. For a more meaningful benefit, further research in people with higher blood glucose levels is required.

비만형 동물모델에서 복합 한약 추출물 CAPA의 비만 및 당대사 개선 효능 (The Mixed Herbal Extract, CAPA, Prevents Obesity and Glucose Intolerance in Obese Mice)

  • Objectives: This study investigated the effects of the mixed herbal extract from Cinnamomum cassia Blume, Atractylodes macrocephala Koidzumi, Pueraria lobata Benth, and Aconitum carmichaeli Debx (CAPA) on obesity and glucose tolerance in obese mice. Methods: Animals were divided in 6 groups, normal diet, high fat diet (HFD), HFD with CAPA 100 mg/kg (CAPA 100), CAPA 300 mg/kg (CAPA 300), and metformin 200 mg/kg or lorcaserin 10 mg/kg as positive controls, and treated for 16 weeks. Body weight, food intake, fasting blood glucose, and body temperature were checked every week and then organs, blood serums were collected after treatment. The oral glucose tolerance test was also carried out after treatment. Results: Compared to HFD, CAPA extract treated mice showed significant decreases in body weight, adipose tissue weight, lipid accumulations in liver and serum lipid levels without a reduction of food intake. And fasting glucose and glucose tolerance were all improved in the CAPA treated mice. Conclusions: Our results suggest that CAPA extract can prevent diet induced obesity and glucose intolerance without a reduction of energy intake in obese mice.

Pharmacognostic Profile of Trigonella Seed and Its Hypoglycaemic Activity

  • Pharmacognostic study was carried out on the seeds of Trigonella foenumgraecum L. (fenugreek) in order to establish its pharmacognostic characteristics. In view of its renewed interest as a dietary supplement among the local inhabitants afflicted with diabetes, its hypoglycaemic activity in normal as well as diabetic rats was also investigated. Oral glucose tolerance test showed that a suspension of the powdered trigonella seed inhibited the increase in blood glucose levels in normal rats that had been treated with an oral glucose load prior to the test. A reduction in the blood glucose levels was also observed when a suspension of the powdered seed was given by gastric intubation to the streptozotocin-induced diabetic rats. These findings suggested that trigonella seeds possessed some hypoglycaemic activities that might be useful to the diabetics.

고지방 식이로 비만이 유도된 C57BL/6 마우스에서 식이 기간에 따른 비만 관련 지표 변화에 대한 연구 (The Study of the Changes of Obesity-Relating Biomarkers in High Fat Fed-Induced C57BL/6 Mice)

  • Objectives: The prevalence of obesity continues rise and obesity and metabolic syndrome is a major problem in global health care. Animal models are used in the drug discovery of novel treatment for obesity. One of common models of obesity is a high fat diet induced obesity in a C5BL/6 mouse, and the development of obesity and glucose tolerance in mouse model is different according to period of diet. Therefore, this study was performed to observe the development of obesity and glucose tolerance during a high fat diet (HFD). Methods: Male C57BL/6 mice, 5 weeks of age, were fed on a standard chow diet as a normal diet (18 kcal% fat) or a HFD (60 kcal% fat) for up to 16 weeks. The various factors related with obesity and insulin resistance were measured at 8, 12, and 16 weeks. Results: The weights of body and epididymal fat were gradually increased for 8~16 weeks, however the change of hyperglycaemia and glucose tolerance have shown different with that of body weight. Blood glucose, oral glucose tolerance and insulin tolerance were increased more clearly at week 12 and 16 than week 8. Lipid accumulation of liver and body temperature were also significantly increased at week 16, compared with normal group. Conclusions: The developments of obesity and related factors were different by a HFD period in a C57BL/6 obese mice. This result suggests that the development of obesity with glucose tolerance and liver lipid may induce clearly by a HFD for 16 weeks.
