Evaluation of the in vivo Antithrombotic, Anticoagulant and Fibrinolytic Activities of Lumbricus rubellus Earthworm Powder

  • A saline suspension of Lumbricus rubellus earthworm powder (EWP) was administered to rats (1 g/kg/day) orally for 15 days to evaluate an oral effectiveness for thrombotic disorders. Blood was drawn at 2-day interval after the administration. Several parameters for antithrombotic, anticoagulant and fibrinolytic activities were measured, including platelet aggregation, clotting time, plasmin activity and the levels of FDP (fibrin/fibrinogen degradation products), D-dimer, and t-PA antigen. It did not affect platelet aggregation induced by ADP and collagen but anticoagulant activity (aPTT and TT) was gradually increased to two-folds for the first 5 days of administration and back to normal. Fibrinolytic activity of euglobulin fraction was highest on the 11 th day after the administration. The level of FDP was elevated to be comparable to the positve control$ (5-10 {\mu}g/ml)$ after 9-day treatment. Oral administration of the EWP could also reduce the formation of venous thrombus induced with viper venom. Complete blood count (CBC) profiles were within normal ranges except for a slight increase in white blood cells after the oral administration for 15 days. These results suggested that the EWP may be valuable for the prevention and/or treatment of thrombotic diseases.

Aspartate aminotransferase activity in the pulp of teeth treated for 6 months with fixed orthodontic appliances

  • Objective: To measure aspartate aminotransferase (AST) activity in the pulp of teeth treated with fixed appliances for 6 months, and compare it with AST activity measured in untreated teeth. Methods: The study sample consisted of 16 healthy subjects (mean age $25.7{\pm}4.3$ years) who required the extraction of maxillary premolars for orthodontic reasons. Of these, 6 individuals had a total of 11 sound teeth extracted without any orthodontic treatment (the control group), and 10 individuals had a total of 20 sound teeth extracted after 6 months of orthodontic alignment (the experimental group). Dental pulp samples were extracted from all control and experimental teeth, and the AST activity exhibited by these samples was determined spectrophotometrically at $20^{\circ}C$. Results: Mean AST values were $25.29{\times}10^{-5}U/mg$ (standard deviation [SD] 9.95) in the control group and $27.54{\times}10^{-5}U/mg$ (SD 31.81) in the experimental group. The difference between these means was not statistically significantly (p = 0.778), and the distribution of the AST values was also similar in both groups. Conclusions: No statistically significant increase in AST activity in the pulp of mechanically loaded teeth was detected after 6 months of orthodontic alignment, as compared to that of teeth extracted from individuals who had not undergone orthodontic treatment. This suggests that time-related regenerative processes occur in the dental pulp.

Association between systemic disease activity restriction and oral health

  • 연구의 목적은 전신질환의 활동제한과 구강건강의 연관성을 알아보고자 국민건강영양조사 제7기 원시자료를 활용하여 19세 이상 남자 2,574명, 여자 3,250명전체성인 5,824명의 응답을 분석하였다. 구강건강의 씹기와 말하기 문제가 있을 때 성인의 전신질환 활동제한이 많은 것으로 나타났으며, 치주질환 치료, 발치 및 구강 내 소수술, 보철물 제작·수리의 치과치료를 받았을 때 전신질환의 활동제한이 유의미하게 높았다(p<.001). 전신질환으로 인한 활동제한에 영향력을 주는 요인으로는 연령(1.03배), 남자(0.84배), 교육수준(0.57, 0.45, 0.31배), 음주(0.32배), 씹기(1.86배)와 말하기(1.84배)문제, 치주질환 치료(1.27배), 부러진 치아치료(2.1배)를 받았을 때 전신질환의 활동제한이 발생할 확률이 높았다. 또한 씹기와 말하기 문제가 있을 때 삶의 질이 낮아지는 것이 통계적으로 유의미하게 나타났다.

Antibacterial Activity of Oleanolic Acid from Physalis angulata against Oral Pathogens

  • A methanol extract of Physalis angulata exhibited in vitro antibarterial activity against oral pathogens such including Streptococcus mutans and Porphyromonas gingivalis. The methanol extract of Physalis angulata was further fractionated with ethyl acetate, n-butanol and water, in which the ethyl acetate fraction exclusively showed antibacterial activity. An active antibacterial compound from the ethyl acetate fraction was purified to a single compound using silica gel column chromatography and identified as oleanolic acid by $^{13}$ C-NMR, $^1$H-NMR and EI-MS. MIC of oleanolic acid against S. mutants and p. gingivalis were determined to be 50 and 25 ug/mL, respectively. The Antibacterial activity of oleanolic acid from Physalis angulata suggested that it has potential as an anticarcinogenic and antiperiodontic ingredients in various foods and oral care products.

천궁 엑스 및 분획의 소염.진통작용 (Anti-inflammatory and Analgesic Activities of the Extracts and Fractions of Cnidii Rhizoma)

  • Cnidii rhizoma is one of the most important crude drugs used particularly for the treatment of female genital inflammatory diseases in traditional oriental medicine. In this study, its anti-inflammatory and analgesic activities were examined employing animal models. It was found that $H_2O$ extract and n-BuOH fraction inhibited significantly the edema formation after the subplantar injection of carrageenin at oral doses of 100 and 200 mg/kg in a dose-dependent fashion, where MeOH extract showed significant anti-inflammatory activity at a oral dose of 200 mg/kg. In Freund's complete adjuvant-induced arthritis, H2O extract and n-BuOH fraction exerted their significant inhibitory activity on the edema formation at oral doses of 100 and 200 mg/kg. The $H_2O$ extract and n-BuOH fraction also showed significant analgesic activity in a dose-dependent manner in acetic acid-induced writhing test.

  • The effects of application of mustard oil (MO), a small-fiber excitant and inflammatory irritant into the rat maxillary molar tooth pulp on arterial blood pressure and heat race, and electromyographic (EMG) activity of the jaw muscles were assessed in the anesthetized rats. In addition, Evans blue extravasation following pulpal MO application was measured. Application of MO into the tooth pulp produced a significant increase in mean arterial pressure and heat rate which gradually returned to baseline level. Application of MO into the tooth pulp induced a significant and short-lasting increase in EMG activity of digastric masseter and tongue muscle. Application of MO into the tooth pulp significantly increased the plasma extravasation of Evans blue dye. However, Application of mineral oil into the tooth pulp did not produce any significant changes in EMG activity of the digastric, masseter and tongue muscles, mean arterial pressure and heart rate, and plasma extravasation of Evans blue dye. These findings indicate that changes in arterial blood pressure, heart rate, jaw muscle activity and plasma extravasation accompanying noxious tooth pulp stimulation call be utilized as indirect measure of orofacial pain and inflammation.

Luteolin Induces the Differentiation of Osteoblasts

  • Luteolin is a flavonoid that exists in a glycosylated form in celery and green pepper. Flavonoids possess antioxidant and anti-inflammatory properties and can reduce the expression of key inflammatory molecules in macrophages and monocytes. It has been reported also that some flavonoids have effects on bone metabolism. The effects of luteolin on the function of osteoblasts were investigated by measuring cell viability, alkaline phosphatase activity, type I collagen production, osteoprotegerin secretion, Wnt promoter activity, BMP-2 and Runx2 expression and calcified nodule formation. Luteolin has no effects upon osteoblast viability but induced an increase in alkaline phosphatase activity, type I collagen production and a decrease in osteoprotegerin secretion in these cells. Luteolin treatment also upregulated BMP-2 mRNA expression. These results suggest that luteolin may be a regulatory molecule that facilitates the differentiation of osteoblasts.

구강 Streptococci가 가진 Sucrose 대사 효소의 활성도의 비교 (Relative Level of Sucrose Metabilizing Enzymes in Oral Streptococci)

  • Occurrence and distribution of sucrose metabolizing enzymes in oral streptococci had been studied. In these studies, the carbohydrate component of the culture medium had been glucose. I have extended these studies by analyzing bacterial culture supernatants for the relative content of hexosyltransferases, namely glucosyl and fructosyltransferase. As a carbohydrate, fructose was used. The growth measured for nine oral streptococci (Strptococcus mutans strains BHT, ING, AHT, 6715, LM-7, and SL-1 ; Streptococcus sanguis 903, 9811, and M-5) varied. The level of glucosyltansferase activity also varied among S. mutans strains, and its level in S. sanguis was relatively low. Fructosyltansferase activity of the various strains fluctuated more than of glucosyltransferase. S.mutans strain LM-7 had significantly higher level of both enzymes. As a whole, fructose-grown cultures had generally an agreeable trend of enzyme activity to those from glucose-grown cultures.

Anti-inflammatory Activity of Propolis

  • Propolid (bee-glue), known as a folk medicine, is a lipo;hilic material found in honeybee hives. In the present study on the anti-inflammatory effect of Korean propolis, it was extracted with ethanol, and used as a test material. The $LD_{50}$ value with the oral administration of ethanolic extract of Korean propolis (EEKP) was higher than 2g/kg in mice. The oral administration of the propolis extract (100mg/kg) significantly inhibited the development of hind paw edema induced by carrageenin in rats. the oral pretreatment of the propolis extract markedly inhibited the increase in vascular permeability and the number of writhing induced by acetic acetic acid in mice. Propolis extract, 50 and 100 mg/kg p.o. per day for 7 days, produced a significant inhibitory effect on granuloma and exudate formation in rats. This inhibitory effect was enhanced with the concomitant use of prednisolone (2.5 mg/kg). These results suggest that Korean propolis apparently has a strong anti-inflammatory activity.

  • Streptococcus vestibularis is a urease-producing oral bacterium. frequently isolated from vestibular mucosa of human oral cavity. Ureolysis by S. vestibularis and other ureolytic oral bacteria is believed to be crucially involved in oral microbial ecology and oral health. Genomic library of the S. vestibularis ATCC49124 was constructed in an E. coli plasmid vector and the urease-positive transformants harboring the urease gene cluster were isolated on Christensen-urea agar plates. The minimal DNA region required for the urease activity was located on a 5.6 kb DNA fragment. (omitted)
