Effect of Plant Growth Regulators and Seed Priming Treatment on the Germination and Early Growth of Snapdragon (Antirrhinum majus L.) (식물생장조절제 및 priming 처리가 금어초 종자의 발아와 초기생육에 미치는 영향)

  • In this study, we investigated the effect of growth regulators, seed priming, and light condition for the germination and early growth in Snapdragon (Antirrhinum majus L.). The optimum concentration of growth regulator for the promotion of germinability turned out to be 250 uM of $GA_3$. The germination enhancement in combination of $GA_3+BAP$ was not significant. The optimum priming condition for the maximum germinability was 2 day treatment of 200 mM of $KNO_3$. which increased germination by 14% and shortened the day to germination by 3.5 days. The optimum temperature for germination was $15{\sim}25^{\circ}C$. The germination frequency was decreased to lower than 20% at $30^{\circ}C$, which showed that seeds of Snapdragon germinates better at low temperature than high temperature. The germination frequency was different at light condition; it was low at dark condition, but was increased by $15{\sim}20%$ at red light condition. The combination of $GA_3$ and seed priming showed better germinability than the single treatment of $GA_3$ and seed priming.

  • The batch compositions and physical properties of slag-ceramics were studied with respect to their formability from the molten state and conditions of nucleation and crystal growth treatment. The selected batch compositions for nucleation and growth studies were slag, 56%; silica sand, 28%; $Na_2O+MgO$, 8% and $TiO_2+$chromite, 8%. The optimum nucleation condition was the temperature of 75$0^{\circ}C$ with 6 hrs. holding time and the optimum growth condition was the temperature 975$^{\circ}C$ with zero holding time. The slag-ceramics prepared under the above conditions showed the best developed microtexture. The grown crystals were identified as diopside with the average grain size of 5.7$\mu\textrm{m}$, and the amount of crystal grown were about 53% by weight. The prepared specimens of slag-ceramics showed the microhardness, 793kg/$\textrm{mm}^2$; MOR, 1,050 kg/$\textrm{cm}^2$ and thermal expansion coefficient, $85{\div}10^{-7}$cm/cm/$^{\circ}C$($25^{\circ}C$~$700^{\circ}C$).

Optimization of in Vitro Cultivation of Inonotus Obliquus

  • This study was performed to get the basic information concerned to the optimum culture condition of Inonotus obliquus. Several solid media, PDA, MEA and Czapek-Dox, and three liquid media were adopted for the in vitro cultivation. Some main features of the fungal morphological characteristics under cultivation conditions were observed and described. Preliminary results showed that appearance of the mycelial mat, hyphal size and substrate pigmentation differed according to the media. The PDA medium was the most favorable substrate for the growth on solid culture, followed by MEA and Czapek-Dox media. Concerned to the addition of amino acids, 5 amino acids, such as alanine, alginine, isoleucine, leucine and threonine, enhanced to the mycelial growth. Isoleucine was shown the best fungal growth. An important morphological hyphal structure for the fungus, the setae, was found in abundance and diverse its shape and size. In liquid culture, fresh potato broth was the best growth stimulant of the fungus, followed by Malt extract and potato broth. Addition of yeast extract to the liquid media had improved the biomass, but not laccase production.

The growth YMnO$_3$ single crystals using a floating zone method (부유대용융법에 의한 YMnO$_3$단결정 성장)

  • High quality crystals of $YMnO_3$, which is interested in non-volatile memory device application, were grown by the floating zone method. Optimum condition for powder synthesis was established to be $1200^{\circ}C$ for 10 hrs and optimum condition for sintering of $YMnO_3$feed-rod was established to be $1500^{\circ}C$ for 10hrs respectively. It was found from non-seeded growth experiment that $YMnO_3$crystal was grown preferentially to the [1010] orientation. The $YMnO_3$single crystal, which was grown to the direction of perpendicular to C-axis, was typically 5mm in diameter, 50 mm in length and showed dark-blue color.

  • Auricularia auricula-judae is an edible mushroom, which is known as wood ear, free ear, black ear mushroom, and free jelly fish. This study was carried out to obtain the basic information for mycelial culture conditions of Auricularia auriculajudae. According to colony diameter and mycelial density, the media for suitable mycelial growth were PDA and MCM. The optimum temperature for mycelial growth was $25{\sim}30^{\circ}C$. Carbon and nitrogen sources were mannose and malt extract, respectively. The optimum C/N ratio was in the range of 10 to 1 with 2% glucose. Other minor components for the optimal growth were thiamine-HCl and biotin as vitamins, succinic acid and lactic acid as organic acids, and $MgSO_4{\cdot}7H_2O$ and $KH_2PO_4$ as mineral salts.

Optimization of Heating Conditions for Bacteria Associated with Food Hygiene (식품위생관련 세균에 대한 가열처리 조건의 최적화)

  • Response surface methodology(RSM) was applied to determine the optimum heating conditions(tem-perature and time) for the inactivation of bacteria associated with food hygiene. Coefficients of determinations(R2) for the cell growth were 0.9155(P<0.05) in Pseudomonas aeruginosa 0.8997(P<0.05) in Escherichia coli 0.9690(P<0.01) in Salmonella typhimurium 0.9393(P<0.05) in Enterobacter aerogenes and 0.7905 in Vibrio parahaemolyticus. The growth of strains was significantly affected by the temperature and time and reaction to temperature was most closely correlated to the inhibition of growth. On the basis of superimposed contour maps of cell growth the optimum range of heating conditions were 55.62-6$0^{\circ}C$ and 19.25-30 min.

  • Since the condition of immobilization must be optimized, it is very important to know whether and on how conditions bacterial cells retain their metabolic activity during immobilization process. A bioluminescence intensity had the maximum value when cell concentrations were between 1.0 and 1.2 measured at O.D660. The strontium alginate was used as an immobilization matrix and two independent factors for immobilization of Photobacterium phosphoreum with strontium alginate were optimized with the response surface methodology(RSM) considering degree of bioluminescence. As a result, the optimum concentration for immobilization was found to be 2.4%(w/w) for sodium alginate and 0.31M for strontium chloride, respectively. A dilution was carried out with 2.5%(w/v) NaCl solution that is an optimum environmental condition for growth of P. phosphoreum. Under the such condition of immobilization, hardness could be predicted as 4.66$\times$104N/$m^2$ and it took different time according to the volume of matrix to be immobilized completely.

  • For artificial cultivation of Phellinus pini (Thore. Fr.) Ames, we conducted some study on mycelium growth and optimum condition for fruitbody formation. The optimum condition for mycelial growth was $25{\sim}30^{\circ}C$ at pH $5.0{\sim}6.0$. Optimum sawdust media were oak sawdust+willow sawdust+rice bran (4.5:4.5:1, V/V) and oak sawdust+pine sawdust+rice bran (4.5:4.5:1, V/V) and the optimum spawn incubation period was about $33{\sim}34$ days. Mycelial growth in the inner portion of oak log was 40 mm after 60 days and duration for first fruitbody primordia formation was about 110 days after inoculation.

  • Ganoderma applanatum is one of the most popular medicinal mushrooms due to the various biologically active components it produces. This study was conducted to obtain basic information regarding the mycelial culture conditions of Ganoderma applanatum. Based on the colony diameter and mycelial density, PDA, YMA and MCM media were suitable for the mycelial growth of the mushroom. The optimum temperature for mycelial growth was found to be $25{\sim}30^{\circ}C$. The optimum carbon and nitrogen sources were mannose and dextrin, respectively, and the optimum C/N ratio was 2 to 10 when 2% glucose was used. Other minor components required for the optimal growth included thiamine-HCl and biotin as vitamins, succinic acid and lactic acid as organic acids, and $MgSO_4$ $7H_2C$, $KH_2PO_4$ and NaCl as mineral salts.

Optimal Condition of Mycelial Growth of Grifola frondosa (잎새버섯 균주의 균사체 생육 최적조건)

  • This study was carried out to obtain basic data on mycelial growth characteristics for an artificial cultivation of Grifola frondosa. Ten strains of G. frondosa were collected from Korea, China and Japan and investigated its optimal culture condition. Among four kinds of mushroom culture media, PDA medium was selected as the suitable culture medium. The optimal conditions for the mycelial growth of G. frondosa in PDA medium were $25^{\circ}C$ and $4{\sim}5$ of pH, respectively. The carbon and nitrogen sources for the optimum mycelial growth were fructose and peptone, respectively, and the highest mycelial growth was observed when the C/N ratio was $10{\sim}20$.
