Comparisons between Micro-Kjeldahl and Near Infrared Reflectance Spectroscopy for Protein Content Analysis of Malting Barley Grain (근적외분광분석법과 Micro-Kjeldahl 법 간의 맥주보리 종실의 단백질함량 분석 비교)

  • Near Infrared Reflectance Spectroscopy(NIRS) has been used as a tool for the rapid, accurate, protein assay of malting barley. NIRS used in this study was filter type instruments, Neotec 102. The objective of this study was to obtain the best calibration equation, for the rapid, ease and accurate protein content analysis of malting barley using NIRS system. The optimum wavelength for protein content analysis used NIRS were 2095nm, 2095/1941nm, 2095/1941/2282nm, 2905/1941/2282/2086nm, respectively. Mean protein content with this calibration equation in NIRS analysis was 10.59%, while 10.60% in Micro-Kjeldahl one. The range of protein content in Micro-Kjeldahl was 8.66~12.66% and that in NIRS was 8.80~12.35%. When 18 other varieties produced in 1992 were analysed with 2095nm, 2095/1941nm, 2095/1941/2282nm, 2095/1941/2282/2086nm equation, standard deviation of difference (SDD)and standard error of performence(SEP) and $R^2$ values were 0.47, 0.43, 0.95, respectively. Both the mean protein content by Micro-Kjeldahl and by NIRS was 10.25%. With this equation, analysied 31 varities produced in 1993, SDD and SEP and r values were 0.69, 0.67, 0.91, respectively, and that bias value was 0.65. In this analysis, mean protein content by Micro-Kjeldahl was 10.17% and by NIRS was 10.81%. The range of protein content in Micro-Kjeldahl was 7.58~14.29%, What that in NIRS was 8.63~13.93%. After adjusted bias in the best calibration equation, mean protein content of Micro-Kjeldahl was 10.17% and that of NIRS was 10.09%, without variance of SDD, SEP and r values.

Real-time controlled deposition of anti-reflection and high-reflection coatings for semiconductor laser (반도체 레이저 단면의 실시간 무반사 및 고반사 코팅)

  • We have obtained the optimum thickness of anti-reflection(AR) coating on one of facets of a $\1.55mu\textrm{m}$ InGaAsP MQW FP semiconductor laser by in-site monitoring of the light emitted from the rear facet during the film deposition on the fore facet. The optimum thickness of $SiO_x$ thin film whose refractive index is 1.85 was found to be 188 nm. The reflectivity of the coated facet was calculated by the threshold current ratio of before and after AR coating, which was obtained from exprimental data, and it was about 2$\times$ $10^{-4}$. The results show that the output power is increased by 87% at bias current 60 mA, the slope efficiency is increased by 3.4 times, and the threshold current is increased by 2.64 times. By in-situ depositing of the $Si/SiO_2$ thin film HR coating on the rear facet, the output power was increased by 160% than before the AR and HR coatings, the slope efficiency was increased by 3.8 times, the threshold current was increased by 1.07 times, which is similar to the value of before AR coating. Due to the AR and HR coatings the output light power characteristics were enhanced.

Optimum Condition and Color Mechanism for Gold Color Glaze in Diopside Crystallization (Diopside 금색 결정 유약의 발색 기구)

  • Generally, the color gold has had a biased conception due to its traditional use. Thus, this bias has resulted in a lack of usage of golden glaze on ceramics and also a lack of extensive studies of such glazes. In this paper, optimum conditions and mechanism of formation of gold color crystallization glaze containing $Fe_2O_3$(hematite), which is developed for gold colors of ceramic glazes, were studied. Experimental result showed that there are pyroxene based on diopside and $TiO_2$ phase in the base of a crystallization glaze with a value of $TiO_2$ of 6 wt% confirmed by XRD and Raman Spectroscopy. When $Fe_2O_3$ was used as a colorant for the gold color, the $TiO_2$ peak became extinct and the intensity of the diopside peak was sharper. Feldspar of 60 wt%, talc of 20 wt% and limestone of 20 wt% were used as the starting materials and these were tested using a three component system. The best result of test was selected and extended to its vicinity as an experiment to determine $TiO_2$ and $Fe_2O_3$ contents. The glaze with $TiO_2$ of 6 wt% and $Fe_2O_3$ of 12 wt% addition showed stable pyroxene based diopside crystals and the development of gold color. This gold color was obtained with CIE-$L^*a^*b^*$ values of 51.27, 4.46, 16.15 (a grayish yellow brown color), which was gained using the following firing conditions: temperature increasing speed $5^{\circ}C$/min, holding for 1 h at $1280^{\circ}C$, annealing speed $3^{\circ}C$/min till $1100{\circ}C$, holding for 2 h at $1100{\circ}C$, and finally natural annealing.

  • Amorphous silicon thin-film transistors (a-Si TFTs) were incorporated into Mo-tip-based triode-type field emitters and diode-type ones of carbon nanotubes for an active-matrix cathode (AMC) plate of field emission displays. Also, we developed a novel surface-treatment process for the Mo-tip fabrication, which gleatly enhanced in the stability of field emission. The field emission currents of AMC plates on glass substrate were well controlled by the gate bias of a-Si TFTs. Active-matrix field emission displays (AMFEDs) with these AMC plates were demonstrated in a vacuum chamber, showing low-voltage matrix addressing, good stability and reliability of field emission, and highly uniform light emissions from the anode plate with phosphors. The optimum design of AMFEDs including a-Si TFTs and a new light shield/focusing grid is discussed.

  • Multi-dimensional hydrodynamic and water quality models are widely used to simulate the physical and biogeochemical processes in the surface water systems such as reservoirs and estuaries. Most of the models have adopted the Eulerian grid modeling framework, mainly because it can reasonably simulate physical dynamics and chemical species concentrations throughout the entire model domain. Determining the optimum grid cell size is important when using the Eulerian grid-based three-dimensional water quality models because the characteristics of species are assumed uniform in each of the grid cells and chemical species are represented by concentration (mass per volume). The objective of this study was to examine the effect of grid-size of a three dimensional hydrodynamic and water quality model (EFDC) on hydrodynamics and mass transport in the Saemangeum Reservoir. Three grid resolutions, respectively representing coarse (CG), medium (MG), and fine (FG) grid cell sizes, were used for a sensitivity analysis. The simulation results of numerical tracer showed that the grid resolution affects on the flow path, mass transport, and mixing zone of upstream inflow, and results in a bias of temporal and spatial distribution of the tracer. With the CG, in particular, the model overestimates diffusion in the mixing zone, and fails to identify the gradient of concentrations between the inflow and the ambient water.

Wide-bandwidth SQUID Current Amplifier and Control Electronics for X-ray Microcalorimeter (X-선 미소열량계 신호 검출을 위한 광대역 SQUID 전류증폭기와 조절 회로)

  • Wide-bandwidth SQUID current amplifier and its control electronics have been constructed for detecting pulse outputs of a superconducting microcalorimeter. The current amplifier made of a double relaxation oscillation SQUID (DROS) has a bandwidth of 1.2 MHz and typical white noise level of about 6 pA/(equation omitted) Hz. To increase the dynamic range of the current amplifier, the flux-locked loop (FLL) has additional circuits to reset the integrator and to count reset numbers which present the number of passed flux quanta. In this system, dynamic range covers from -65 mA to +65 mA. SQUID electronics are controlled by software to get the optimum FLL condition, and to control the current to bias the transition edge sensor (TES). The electronics are shielded from the outside electromagnetic noises by using an aluminum case of 66 mm ${\times}$ 25 mm ${\times}$ 100 mm, and consist of 2 separate printed-circuit-boards for the current amplifier and the control electronics, respectively. The SQUID current amplifier and its control electronics will be used in TESs for detecting photons such as UV and X-ray with high energy resolution.

Electrical Properties of DC Sputtered Titanium Nitride Films with Different Processing Conditions and Substrates

  • Deposition of TiN$_{x}$ film was conducted with a DC sputtering technique. The effect of the processing parameters such as substrate temperature, deposition time, working pressure, bias power, and volumetric flowing rate ratio of Ar to N$_{2}$ gas on the resistivity of TiN$_{x}$ film was systematically investigated. Three kinds of substrates, soda-lime glass, (100) Si wafer, and 111m thermally grown (111) SiO$_{2}$ wafer were used to explore the effect of substrate. The phase of TiN$_{x}$ film was analyzed by XRD peak pattern and deposition rate was determined by measuring the thickness of TiNx film through SEM cross-sectional view. Resistance was obtained by 4 point probe method as a function of processing parameters and types of substrates. Finally, optimum condition for synthesizing TiN$_{x}$ film having lowest resistivity was discussed.

Design and Fabrication of InP/InGaAs PIN Photodiode for Horizontally Integrated OEIC's (수평집적형 광전자집적회로를 위한 InP/InGaAs PIN 광다이오드의 설계 및 제작)

  • OEIC(Optoelectronic Integrated Circuit)'s can be integrated horizontally or vertically. Horizontal integration approach is, however, more immune to parasitic and more universally applicable. In this paper, a structural modeling, fabrication and characterization of PIN photodiodes which can be used in the horizontal integration are performed. For device modeling, we build a transmission line model from 2-D device simulation, from which lumped model parameters are extracted. The speed limits of the PIN photodiodes can also be calculated under various structural conditions from the model. Thus optimum design of horizontally integrated PIN photodiodes for high speed operation are possible. Such InGaAs/InP PIN photodiodes for long-wavelength communications are fabricated using pit etch, epi growth, planarization, diffusion and metallization processes. Planarization process using both RIE and wet etching and diffusion process using evaporated Zn$_{3}P_{2}$ film are developed. Characterization of the fabricated devices is performed through C-V and I-V measurements. At a reserve bias of 10V, the dark current is less than 5nA and capacitance is about 0.4pF. The calculated bandwidth using the measured series resistance and capacitance is about 4.23GHz.

Novel $\alpha$-Glucosidase from Extreme Thermophile Thermus caldophilus GK24

  • $\alpha$-Glucosidase of an extreme thermophile, Thermus caldophilus GK24 (TcaAG), was purified 80-fold from cells to a homogeneous state and characterized. The enzyme exhibited optimum activity at pH 6.5 and $90^{\circ}C$, and was stable from pH 6.0 to 85 and up to $90^{\circ}C$. The enzyme had a half-life of 85 minutes at $90^{\circ}C$. An analysis of the substrate specificity showed that the enzyme hydrolyzed the non-reducing terminal unit of $\alpha$-1,6-glucosidic linkages of isomaltosaccharides and panose, $\alpha$-1,3-glycosidic bond of nigerose and turanose, and $\alpha$-1,2-glycosidic bond of sucrose. The gene encoding the TcaAG was cloned, sequenced, and sequenced in E. coli. The nucleotide sequence of the gene encoded a 530 amino acid polypeptide and had a G+C content of 68.4% with a strong bias for G or C in the third position of the codons (93.6%). A sequence analysis revealed that TcaAG belonged to the $\alpha$-amylase family. We suggest that this monomeric, thermostable, and broad-acting $\alpha$-glucosidase is a departure from previously exhibited specificities. It is, therefore, a novel $\alpha$-glucosidase.

A Research on DLC Thin Film Coating of a SiC Core for Aspheric Glass Lens Molding (비구면 유리렌즈 성형용 SiC 코어의 DLC 코팅에 관한 연구)

  • Technical demands for aspheric glass lens formed in market increases its application from simple camera lens module to fiber optics connection module in optical engineering. WC is often used as a metal core of the aspheric glass lens, but the long life time is issued because it fabricated in high temperature and high pressure environment. High hard thin film coating of lens core increases the core life time critically. Diamond Like Carbon(DLC) thin film coating shows very high hardness and low surface roughness, i.e. low friction between a glass lens and a metal core, and thus draw interests from an optical manufacturing industry. In addition, DLC thin film coating can removed by etching process and deposit the film again, which makes the core renewable. In this study, DLC films were deposited on the SiC ceramic core. The process variable in FVA(Filtered Vacuum Arc) method was the substrate bias-voltage. Deposited thin film was evaluated by raman spectroscopy, AFM and nano indenter and measured its crystal structure, surface roughness, and hardness. After applying optimum thin film condition, the life time and crystal structure transition of DLC thin film was monitored.
