The Study on Pre-treatment Method of Filter for Analysing the Heavy Metals in Air Quality (대기중의 중금속 분석을 위한 여지의 전처리 방법에 관한 연구)

  • This study was carried out to establish the standardized analysis method in order to decide accurate concentrations of hazardous metals in the air. Acid decomposition method used usually was compared to microwave digestion method. Comparing results of tested background concentration in blank filter, we found that the magnitude of element concentration was Na, Ca, K, Mg, Al and Ba by order. The element concentrations of Glass fiber filter were higher than those of Quartz fiber filter, and the number of undetected components in Glass fiber filter was lower than that in Quartz fiber filter. Thus it is supposed that the concentration of background elements in Glass fiber filter was higher than those in Quartz fiber filter The extraction rate of microwave digestion method was superior to those of acid decomposition method during the test of blank filter as well as SRM of NIST. In case of the SRM of NIST, the average extraction efficiency of acid decomposition Method and microwave pretreatment is 53.8∼82.7%, 81.3∼97.1%, respectively. This result might be caused by the closed system of Microwave, which make outflow and loss of components less. Also microwave digestion method has other merits such as the minimization of time, reagents, and contamination. Furthermore, if the extraction condition, extraction time and used acids are optimized, the better results will be represented.

  • A low energy ion irradiation system based on the deuterium arc ion source with a high perveance of 1 µP for a single extraction aperture has been successfully developed for the investigation of ion irradiation on plasma-facing components including the first mirror of plasma optical diagnostics system. Under the optimum operating condition for mirror testing, the ion source has a beam energy of 200 eV and a current density of 3.7 mA/cm2. The ion source comprises a magnetic cusp-type plasma source, an extraction system, a target system with a Faraday cup, and a power supply control system to ensure stable long time operation. Operation parameters of plasma source such as pressure, filament current, and arc power with D2 discharge gas were optimized for beam extraction by measuring plasma parameters with a Langmuir probe. The diode electrode extraction system was designed by IGUN simulation to optimize for 1 µP perveance. It was successfully demonstrated that the ion beam current of ~4 mA can be extracted through the 10 mm aperture from the developed ion source. The target system with the Faraday cup is also developed to measure the beam current. With the assistance of the power control system, ion beams are extracted while maintaining a consistent arc power for more than 10 min of continuous operation.

Optimization Condition of Astaxanthin Extract from Shrimp Waste Using Response Surface Methodology (반응 표면 분석법을 사용한 새우껍질에서 astaxanthin 추출 조건의 최적화)

  • A 17-run Box-Behnken design (BBD) was used to optimize the extraction conditions of astaxanthin from shrimp waste. Three factors such as ratio of ethanol to raw material, extraction temperature ($^{\circ}C$) and extraction time (min) were investigated. The adjusted coefficient of determination ($R^2{_{adj}}$) for the model was 0.9218, and the probability value (p=0.0003) demonstrated a high significance for the regression model. The optimum extraction conditions were found to be: optimized ratio of ethanol to raw material 29.7, extraction temperature $49.5^{\circ}C$ and extraction time 59.9 min. Under these conditions, the mean extraction yield of astaxanthin was $17.80{\mu}g/g$, which was in good agreement with the predicted model value. Under these conditions, validation experiments were done and the mean extraction yield of astaxanthin was $17.77{\mu}g/g$, which is in good agreement with the predicted model value.

Salting-out extraction of ginsenosides from the enzymatic hydrolysates of Panax quinquefolium based on ethanol/sodium carbonate system

  • Background: Salting-out extraction (SOE) had been developed as a special branch of aqueous two-phase system recently. So far as we know, few reports involved in extracting ginsenosides with SOE because of the lower recovery caused by the unique solubility and surface activity of ginsenosides. A new SOE method for rapid pretreatment of ginsenosides from the enzymatic hydrolysates of Panax quinquefolium was established in this article. Methods: The SOE system comprising ethanol and sodium carbonate was selected to extract ginsenosides from the enzymatic hydrolysates of Panax quinquefolium, and HPLC was applied to analyze the ginsenosides. Results: The optimized extraction conditions were as follows: the aqueous two-phase extraction system comprising ethanol, sodium carbonate, ethanol concentration of 41.51%, and the mass percent of sodium carbonate of 7.9% in the extraction system under the experimental condition. Extraction time had minor influence on extraction efficiency of ginsenosides. The results also showed that the extraction efficiencies of three ginsenosides were all more than 90.0% only in a single step. Conclusion: The proposed method had been successfully applied to determine ginsenosides in enzymatic hydrolysate and demonstrated as a powerful technique for separating and purifying ginsenosides in complex samples.

  • In order to develop a high brightness ion source using plasma, the ion beam extraction system with an aperture of $100{\mu}m$ in diameter has been designed and constructed. It is observed that over 500nA of He ion beam current can be extracted. With such an optimized condition, $\~10^3\;A/cm^2sr$ beam brightness can be measured by emittance scanner, which is believed to be a promising result for developing next generation FIB.

  • This study examined the optimization of hot-water extraction conditions for maximizing the total polyphenol compounds (TPC) extracted from amaranth leaf. The effects of three independent variables, including extraction temperature, extraction time and ethanol concentration on TPC were investigated using central composite design (CCD). The concentration of TPC increased with increased levels of extraction temperature and time. The extraction temperature and the ethanol concentration showed the significant effect on TPC production (p<0.05). The predicted values at the optimized condition were acceptable when compared to the experimental values ($R^2=0.9566$). The optimum extraction conditions were as follows: temperature of $90.1^{\circ}C$, time of 50 min and ethanol concentration of 61.6% (v/v) for the maximum TPC of 12.6 mg GAE/g DM.

Effect of deep eutectic solvent (DES) on the extraction of asiaticoside and madecassoside from Centella asiatica (병풀(Centella asiatica)로부터의 asiaticoside와 madecassoside의 추출효율에 미치는 DES의 영향)

  • Centella asiatica (C. asiatica) extracts, including asiaticoside and madecassoside, are used in ointments to treat the wound and atopic dermatitis due to their antibacterial and skin-regenerating effects in Asia. Therefore, research on the cultivation and extraction efficiency of C. asiatica is being actively conducted to increase commercialization efficiency. In this study, various deep eutectic solvents (DESs) were prepared and used as the extraction solvents according to the mole ratio between the hydrogen bond acceptor (HBA) and hydrogen bond donor (HBD). And then, the extraction yields in distilled water (DW) and methanol (MeOH), commonly used extraction solvents for C. asiatica, were compared and analyzed by HPLC in the optimized operating condition. As a result, a mixture of DW and DES at a ratio of 3:7 showed about 1.4 times higher extraction efficiency than MeOH only. Conversely, the extraction efficiency in a mixture of MeOH and DES at a ratio of 3:7 was about 6 % lower than that in MeOH only.

Beta glucan: Heath benefits of optimal ingredients via the mixture ratio of rice bran and Sarcodon aspratus

  • Beta-glucan is a polymerized polysaccharide on beta-1,3 chemical bonds. It has the main pharmacological action of various anticancer mushrooms and is colorless and odorless. In addition, it enhances phagocytosis of macrophages against mycobacterium tuberculosism and increases resistance of host against food poisoning bacteria. Owing to these advantages, beta-glucan was used in various health supplement foods such as immune boosters, hypotensive agent and hypoglycemic agent. Our study was aimed to investigate the effective components of beta-glucan, which was optimized via the contents of Sarcodon aspratus and rice bran. First, the mixture ratio condition of Sarcodon aspratus and rice bran were respectively 10:0, 7:3, 5:5, 3:7, and 0:10. The raw materials were fabricated using hot water extraction method. Distilled water of 100 mL was added to the raw material and extracted. After the extraction processed, the content of effective components was analyzed. The absorbance of beta-glucan was measured using a beta-glucan kit (Megazyme, (1-3) (1-4) BETA-D-GLUCAN ASSAY KIT). The absorbance analysis was repeated three times for accurate analysis. After the extracts were lyophilized, the yields of extracted raw materials according to the mixture ratio conditions of Sarcodon aspratus and rice bran (10:0, 7:3, 5:5, 3:7, and 0:10) were respectively 33.1%, 34.5%, 35.3%, 30.7%, and 24.3%. The absorbance was 1.52, 1.47, 1.50, 1.79, and 1.56, respectively. As a result, the optimum ratio of beta-glucan is 3:7 at Sarcodon aspratus and rice bran. This study suggested that the optimal amount of beta-glucan could be used as a health supplement foods and food additive such as immune boosters, hypotensive agent and hypoglycemic agent. However, the new condition (temperature, time) of hot water extraction to maximize the content of beta-glucan could be considered, could be necessary to compare with the existing extracts.

Optimization of Beverage Preparation from Schizandra chinensis Baillon by Response Surface Methodlogy (반응표면 방법에 의한 오미자 음료 제조의 최적화)

  • To develop the beverage from Schizandra chinensis Baillon, extraction condition was optimized, using the fractional factorial design with 3 variables and 3 levels, by a RSM computer program. The optimum extraction time, extraction temperature and alcohol concentration levels were 3 hrs, $80{\sim}85^{\circ}C$ and 0%(water) for a desired yield, color and non-volatile organic acid. And to obtain the optimum recipe, amounts of extract, sugar and citric acid were optimized, using the fractional factorial design with 3 variables and 3 levels, by a RSM program. The optimum extract, sugar and citric acid levels were 4%, 9.6%n and 0.06%, respectively, for a disired flavor and overall acceptance.

Optimizing the Hot-water Extraction Conditions for Acanthopanacis cortex Using Response Surface Methodology (반응표면분석법을 이용한 오가피의 열수 추출조건 최적화)

  • In order to examine the effective components and antioxidative characteristics of Acanthopanacis cortex, response surface methodology was used to optimize the hot water extraction process by analyzing and monitoring the extraction condition characteristics. The total extraction yield was optimized using the extraction temperature of $84.28^{\circ}C$, extraction time of 3.59 hr and 18.37 mL/g as the amount of solvent per sample. For the effective component of eleutheroside E, the optimal extraction temperature, time and amount of solvent per sample were $81.16^{\circ}C$, 3.56 hr, and 19.06 mL/g, respectively; also, for total phenolics, the optimal conditions were $88.45^{\circ}C$, 4.33 hr, and 21.12 mL/g, respectively. Regression equations were deduced for each variable and then eleutheroside E, chlorogenic acid, and antioxidative characteristics were superimposed with the optimal conditions to obtain values for each extraction process factor. The predicted results were $81{\sim}88^{\circ}C$, $3.5{\sim}4.3hr$, and $18{\sim}21mL/g$, respectively. The estimated values for the optimal extraction conditions of Acanthopanacis cortex's effective components and antioxidative characteristics were similar to the experimental results.
