β-Glucuronidase 저해 활성이 우수한 결명자를 첨가한 상황 균사체 배양액의 생리활성 (Biological Activities of Phellinus linteus Mycelium Culture with Cassiae Semen Extract on β-Glucuronidase Inhibitory Activity)

  • We examined the effects of biological activity Phellinus linteus mycelium culture with cassiae semen extract. Firstly, the optimal temperature, initial pH and culture period for mycelial growth in a liquid culture of P. linteus were determined, and they were $30^{\circ}C$, pH 5.0 and 8 days respectively. The five herbal materials were examined against several health functional efficacies, and, as a result, Cassiae semen was chosen, with its superior inhibitory effects in ${\beta}$-glucuronidase inhibitory activity, electron donating activity, ACE inhibitory, and ${\alpha}$-glucosidase inhibitory activities(95.3%, 80.9%, 96.1 and 24.2%, respectively). P. linteus fruit body was investigated on ${\beta}$-glucuronidase inhibitory activity, electron donating activity, ACE inhibitory, and ${\alpha}$-glucosidase inhibitory activities, and they were 54.7%, 81.9%, 30.0% and 20.1%, respectively. Accordingly, C. semen was used in the following experiment, to give an additive functional effect on the P. linteus. As the amount of C. semen in the cultural media increased, mycelial weight and ${\beta}$-glucan contents also increased, but final pH was not influenced. In addition, the ${\beta}$-glucuronidase inhibitory activity, electron donating activity, and ${\alpha}$-glucosidase inhibitory activity increased. P. linteus mycelium culture showed higher activities in the other three tests above, except for electron donating activity, when C. semen was added to the medium before cultivation.

동충하초 균사체 최적 성장을 위한 심부배양 조건에 따른 형태학적 변화 및 균사체 열수 추출물의 면역학적 특성 (Morphological Changes by Submerged Culture Conditions for the Mycelial Optimal Growth of Cordyceps sinensis and Immunological Properties of Hot Water Extract of Mycelium)

  • The mycelial dispersed growth of Cordyceps sinensis was optimized in submerged batch culture at initial pH of 5.0, 150 rpm, and $25^{\circ}C$. The morphological data showed much more dispersed growth of C. sinenesis at initial pH of 5.0. Also, projected area, main hyphal length and number of tips for the mycelial growth of initial pH 5.0 were higher than those of other initial pHs. The industrial medium for mycelial production of C. sinensis was determined to be molasses of 100 g and crushed brewery yeast of 10 g per liter as carbon and nitrogen sources, respectively. With these culture conditions, the maximum production of mycelia was approximately 30.0 g per liter by batch culture in 5-liter jar fermenter with no controlled pH. This result suggests that large-scale mycelia production of C. sinensis may be possible in submerged batch culture. The hot water extract of mycelia from C. sinensis was mainly composed of 83.0% carbohydrate, 11.8% protein, 1.9% lipid, and 2.4% ash and there were present glucose, mannose, galactose, and arabinose as molar ratio of 8.79 : 2.59 : 1.34 : 1.0 in the carbohydrate, respectively. In the experiment using spleen cell and macrophage, the extract showed potent mitogenic and immuno-stimulating activities and among various components, an important factor that contribute to the immunological activities was turned out to be carbohydrate moiety.

액체종균(液體種菌) 접종(接種)에 의한 표고톱밥재배(栽培) 효과(效果) (Effect of Sawdust Culture on Oak Mushroom, Lentinula edodes (Berk.) Pegler by Inoculation of the Liquid Spawn)

  • For cultivation on sawdust-bed of oak-mushroom until present time, inoculation of spawn on sawdust bed has been performed by sawdust spawn. But, liquid spawn may have advantages for rapid mass production of spawn, and now, sawdust-cultivation by liquid spawn inoculation should be applied instead of sawdust spawn. Therefore, investigations were performed to evaluate the effect of sawdust-cultivation by liquid spawn inoculation. The results were as follows: 1. When 11 kinds of liquid media were applied, the oak-mushroom culture medium was the most excellent in growth. Most suitable temperature at PDA was $25^{\circ}C$, and $22.5\sim27.5^{\circ}C$ in range were optimal for liquid culture. In liquid culture, amount of mycelial growth increases rapidly up to 40 days of cultivation. Incubation at fermentor brought yield of 106mg dry mycelia per 40ml media after 17 days. 2. In 1l-spawn bottle, growth of mycelium by inoculation of 20ml-liquid spawns were faster than 6g-sawdust spawn in spread of mycelia. On 2kg-bag culture, inoculations of 10ml-, 20ml- and 30ml-liquid spawns were all slower than 20g-sawdust spawn in mycelial spread. So, amount increasement in ampunt of liquid spawn should be discussed. Yields of mushrooms until third sproutings of 2kg-bag culture were 580g in 30ml-liquid spawn inoculation, but 510g, 486g and 470g from 20g-sawdust spawn, 20ml-liquid spawn and 10ml-liquid spawn, respectively. Thus, 30ml-liquid spawn inoculation was highest in yield.

Production and Transfer of In Vitro fertilized Hanwoo Embryos with Serum-free Media

  • As a simple and economical method for in vitro produced embryos, we have used BSA instead of serum for the production and embryo transfer of Hanwoo in vitro fertilized (IVF) embryos and obtained the following results: 1) When using serum (FBS; fetal bovine serum) or BSA-containing culture media as the initial culture media for immature oocytes, it is regarded as inappropriate to add only BSA to the culture solutions from maturation of the immature oocytes to development stage culture, but serum still needs be added though there is no significant difference in the concentration, with a change from 5% to 10%. 2) The results of culturing IVF embryos after development (4 cell stage) in the Medium199 solutions containing BSA instead of serum (FBS) showed that 0.3% BSA concentration is not optimal and 0.5% or higher BSA concentration has no significant difference among 0.5%, 0.7%, 1% and 2% (p > 0.05). 3) The post-freezing survival ratio after development in 5% FBS-Medium199 showed that 1% BSA concentration of the culture solution is the most suitable in the BSA concentrations of 0.3% (51%), 0.5% (67%), 0.7% (69%), 1% (77%) and 2% (75%). 4) The pregnancy rates of the transplanted fresh(not frozen) blastocyst had no significant concentration dependency (p > 0.5), and the average pregnancy rate was 63.8%. 14% of overweight calves were found among the calves given birth to by the transfer of IVF blastocysts cultured in the serum-added culture solution, but none was found in the experimental groups in which BSA was added instead of serum.

단세포단백질 생산을 위한 혼합배양의 생육조건 (Growth conditions of symbiosis for production of single cell protein)

  • Mouse의 대장에서 분리된 E. coli LI-10 균주를 보조균주로 하여 Cellulomonas sp. KL-6과 혼합배양한 결과, 균체증식은 주균주와 보조균주를 1 : 1(v/v)의 비로 혼합하였을 때 가장 좋았다. 혼합배양은 주균주 단독배양 보다 균체증식을 63% 정도로 증가시켰으며 두 균주의 분포도는 10 : 1 비율로 KL-6 균주가 주로 분포되어 있었다. 0.1%의 $CaCO_3$의 첨가는 무첨가에 비해 각 배양기간별로 pH를 상승시켜 어느정도의 균체증식을 가져왔다. Filter paper 배지에서 혼합배양시, 본 균주들은 cellobiose를 월등하게 많이 생산하였으나 glucose는 검출되지 않았다. 균체증식 최적배지에서 4일간 혼합배양하였을 때, $1.0\;g/{\ell}$의 균체량을 생산하여 기본배지인 CMC 배지에서 생산한 균체량보다 53% 정도가 증가되었다.

고추 소포자를 이용한 반수체 및 배가반수체 생산 (Production of Haploid and Doubled Haploid Plants from Isolated Microspore Culture of Hot Pepper (Capsicum annuum L.))

  • Haploid/double haploid plants developed from isolated microspores can significantly accelerate plant breeding. Haploid plants can naturally double their chromosomes to create a pure homozygous line of diploid plants. We present a method for producing embryos from isolated microspores of hot peppers (Capsicum annuumL.). We analyzed the polyploidization levels of the regenerated plants. The donor plants produced the optimal stage of microspores following short-term growth under low-intensity light, which resulted in high rates of embryogenesis and cotyledonary embryogenesis. To find an efficient culture method, liquid, doubled-layer, and 2-step cultures were tested. Liquid culture yielded the highest number of embryos, whereas the highest efficiency for cotyledonary embryogenesis was afforded by the doubled-layer culture. When normal cotyledonary embryos were transplanted onto a regeneration medium, they developed into complete plants. From these, 208 plants were tested via flow cytometric analysis, and 35.6% and 72.7% of the chromosomes from the Milyang-jare and LV2319 genotypes, respectively, were found to be spontaneous double haploids. These results are the same as those obtained on analyzing horticultural characteristics, including the size of leaves and the size and shape of fruits. The present study provides information on the practical application of isolated microspore culture of hot peppers, factors that affect embryogenesis, and methods for polyploidy testing.

켄터키 블루그래스에 있어서 캘러스 배양 및 식물체 재분화에 미치는 요인의 영향 (Factors Affecting Callus Culture and Plant Regeneration in Kentucky Bluegrass)

  • 켄터키 블루그래스의 최적 조직배양조건을 확립하기 위하여 성숙종자로부터 배발생 캘러스 유도 및 효율적인 식물체 재분화를 위한 품종별 배양효과, 배지종류 및 배지첨가물 등의 최적조건을 조사하였다. 배발생 캘러스 유도시 첨가되는 auxin으로는 2,4-D가 가장 효율적이었으며, 3mg/L 2,4-D와 0.1mg/L BA가 첨가된 MS배지에서 배발생 캘러스가 가장 높은 빈도로 유도되었다. 식물체 재분화는 배발생 캘러스를 1mg/L 2,4-D와 3mg/L BA가 첨가된 N6 배지에서 배양했을 때 57.7%의 높은 재분화율을 나타내었다. 기본배지의 종류에 따른 배양효율의 차이는 캘러스 유도에는 MS 배지가, 식물체 재분화에는 N6 배지가 효과적이었다. 켄터키 블루그래스의 품종에 따른 배발생 캘러스의 유도율과 재분화 능력은 큰 차이를 나타내어 Newport가 가장 높은 배양효율을 나타내었다. 본 연구를 통하여 확립된 단기간의 효율적인 재분화시스템은 분자육종을 통한 신품종 켄터키 블루그래스 개발에 유용하게 응용되어질 수 있을 것이다.

증숙 처리에 의한 산삼 부정 배양근의 저분자 진세노사이드 추출 (Extraction of Low Molecular Weight Ginsenosides from Adventitious Roots Culture of Wild Mountain Ginseng by Steam Processing)

  • Background: Hot steaming is known to be effective in improving the biological activities of plant extracts by breaking down useful compounds to low molecular weight ones. Methods and Results: This study aimed to develop an optimal extraction and steam processing method for enhancing the low molecular ginsenoside contents of the adventitious roots culture of wild mountain ginseng. The total ginsenoside was optimally extracted when 70% EtOH was used at $50^{\circ}C$, whereas low molecule ginsenoside such as Rg2, Rh1, Rh4 and Rk1 could be extracted using 70% EtOH at $70^{\circ}C$. The adventitious roots culture of wild mountain ginseng is known to contain four major ginsenosides, i.e., Rb2, Rb1, Rg1 and Rd, however new ginsenosides Rg6, Rh4, Rg3, Rk1 and Rg5 were new abundantly obtaind after steam processing method was applied. The contents of total ginsenosides were the highest when thermal steam processing was conducted at $120^{\circ}C$ for 120 min. Unlike ginsenosides such as Rg1, Re, Rb1, Rc, Rb2, and Rh1, which decreased after steam processing, Rg3, Rk1, and Rg5 increased after thermal processing. Steam processing significanltly reduced the content of Rb1, increased that of Rg6 by about ten times than that in the adventitious roots culture of wild mountain ginseng. Conclusions: Our study showed that the optimal extraction and steam processing method increased the content of total ginsenosides and allowed the extraction of minor ginsenosides from major ones.

동해안 토속종 개다시마(Kjelimaniella crassifolia)와 이식종 다시마(Laminaria japonica)의 실내배양 연구 (Comparative Laboratory Culture Studies of the Native Kelp Kjellmaniella crassifolia and the Introduced Kelp Laminaria japonica in East Coast of Korea)

  • 이 연구는 동해 연안에서 토속종인 개다시마(K. crassifolia) 와 이식종인 다시마(L. japonica)의 실내 배양에서 온도와 조도에 대한 특성을 비교하였다. 유주자 단계에서 수온 $15{\~}20^{\circ}C$도일 때 24시간 안에 $95\%$의 포자 발아가 이루어진 반면 개다시마는 48시간 안에 $90\%$를 보였다. 배우체의 성장은 $10^{\circ}C$$15^{\circ}C$도에서 생장이 좋았는데 이입종인 다시마는 $80{\~}l20{\mu}mol{\cdot}m^{-2}s^{-1}$ 고조도에서 최대를 보였고 토속종인 개다시마는 $40{\~}80{\mu}mol{\cdot}m^{-2}s^{-1}$ 낮은 조도에서 최적을 보였다. 아포체의 성장은 개다시마는 $10^{\circ}C$, $40{\mu}mol{\cdot}m^{-2}s^{-1}$ 조도에서 생장이 좋은 반면 다시마는 수온 $10{\~}20^{\circ}C$$40{\~}l20{\mu}mol{\cdot}m^{-2}s^{-1}$ 조도의 다양한 조건에서 생장이 좋게 나타났다. 개다시마의 배양 최적 조건은 이입종 다시마에 비해 수온과 조도의 범위로 볼 때 매우 협소하게 나타나 이는 개다시마가 깊은 곳에 서식하는 바다환경의 특성을 잘 반영해주고 있다.

쥐 하이브리도마 세포배양을 위한 무혈청 배지개발( I ) -최적 배지성분의 결정- (Development of Serum-free Media for the Culture of Mouse Hybridoma (I) ; Determination of Optimal Media Composition)

  • 최근 단일클론항체의 상업적 이응가치가 증대하면서 그 수요량 또한 급격히 증가, 이를 뒷받침할 수 있는 대량생산 기술이 요구되고 있다. 본 연구에서는 쥐 하이브리도마 Alps 25-3의 대량배양을 위해 우선적으로 무혈청 배지 (serum-free media)의 개발을 시도하였다. 동물세포 배양액 IMDM과 Ham's F-12를 1:1로 혼합한 배지를 기본으로하여 ITES 용액 (insulin 10$\mu\textrm{g}$/$m\ell$, transferrin 10$\mu\textrm{g}$/$m\ell$, ethanolamine 10$\mu$M and selenium 30nM)을 첨 가하여 일차적으로 결정한 EBM(enriched basal medium)에 steroid hormones, vltamins, lipid, mineral 및 여러성분들을 각각 첨가하여 혈청과 대치될 수 있는 성분들의 최적농도를 조사하였다. 각 성분들의 개별효과와 함께 그들을 혼합하였을 때의 상호효과를 통해 혈청배지와 거의 동일한 성장을 보이는 무혈청배지 KM3(EBM with BSA 100$\mu\textrm{g}$/$m\ell$, mineral solution and 0.05% PEG)를 결정할 수 있었으며 이 배지에서의 최고 항체 생성량은 혈청배지의 약 80% 로 나타났다. 또한 무혈청 배지에서 여러 cell line의 성장과 그들의 항체 생성을 조사함으로써 다른 쥐 하이브리도마 세포배양에도 적응시킬 수 있음을 확인할 수 있었다.
