Effects of galactagogue herbal tea containing Chrysanthemum indicum as the main component on milk production in postpartum rats (감국을 주성분으로 하는 허브차의 투여가 출산 후 쥐의 유즙 생성 및 관련 호르몬 분비에 미치는 영향)

  • Purpose: Breastfeeding is the optimal method for feeding a newborn. But insufficient breast milk is the major reason why mothers give up breastfeeding. Herbal galactagogues have been used increasingly to treat postpartum hypogalactia. This study examined the effect of an herbal tea containing Chrysanthemum indicum, as the main ingredient, on milk production in lactating rats. Methods: The herbal tea contained C. indicum (27%), Foeniculum vulgare Mill (21%), Pimpinella anisum (18.2%), Carum carvi (16.1%), Urtica dioica (14.7%), and Gardenia jasminoides (3%). Sixteen lactating Sprague-Dawley (SD) rats were divided randomly into 2 groups, the normal control group (NC; n = 8), and the galactagogue herbal tea group (GHT; n = 8) for 7 days. Lactating rats were administered the decoction of an herbal galactagogue mixture by oral gavage or the same amount of distilled water and milk production was assessed by measuring the pups' weights during the suckling period. The blood concentrations of prolactin, cortisol, oxytocin levels and mammary gland tissues were examined to assess the effects of the galactagogue. Results: Milk production was 9.2% higher in the GHT group given the herbal tea than in the NC group and the difference was statistically significant. The cortisol level in the GHT group was 17.2% higher than the NC group. The herbal tea containing C. indicum increased the size of the alveoli epithelium cells and the mammary lobe. Conclusion: The present study revealed the potential of herbal tea containing C. indicum to enhance milk production in postpartum SD rats.

Clinical Efficacy of Pimobendan on Dogs with Chronic Mitral Valvular Diseases (만성 이첨판 폐쇄부전증 이환된 견에서 피모벤단의 임상적 효과)

  • Pimobendan is a recently developed cardiac drug which is useful to control moderate to severe congestive heart failure (CHF) from chronic mitral valvular diseases (CMVI). Because of controversy related to the efficacy and safety of pimobendan in dogs, the optimal efficacy and safety of pimobendan was assessed in 20 dogs with CMVI in this study. Scores for quality of life, respiratory failure, circulatory failure and heart failure were evaluated along with radiographical and echocardiographical assessments for about 2 months period after the addition of pimobendan into the regular cardiac medications. This study proved clear evidence that pimobendan had beneficial therapeutic effects in dogs with advanced CMVI, without particular adverse effects. However, further studies are warranted to address the drug interaction with other cardiac therapeutics and to assess therapeutic effects in CHF from other type of heart diseases in dogs and other animals.

Adaptive Beacon Scheduling Algorithm to Reduce End-to-End Delay in Cluster-tree based LR-WPAN (클러스터-트리 기반 LR-WPAN에서 End-to-End 지연시간을 줄이기 위한 적응적 Beacon 스케줄링 알고리즘)

  • In this paper, we propose an adaptive beacon scheduling algorithm to control a reception period of actual data according to variation of amount of traffic in IEEE 802.15.4 LR-WPAN(Low Rate-Wireless Personal Area Network) with the cluster-tree topology. If a beacon interval is shortened, the amount of the traffic a node receives can be increased while consumption of the energy can be also increased. In this sense, we can achieve optimal on orgy consumption by controlling the beacon interval when the amount of data to be received is being decreased. The result of simulation using NS-2 shows that the proposed algorithm improves performances in terms of packet loss rate and end-to-end delay compared with algorithm using a fixed beacon interval. For a design of cluster-tree based LR-WPAN managing delay-sensitive services, the proposed algorithm and the associated results can be applied usefully.

Geochemical Indicators for the Recovery of Sediment Quality after the Abandonment of Oyster Crassostrea gigas Farming in South Korea (굴(Crassostrea gigas)양식 중단 이후 퇴적물 질 회복에 관한 지화학적 지표 탐색)

  • In order to evaluate the recovery of fishing ground environment after the cessation of aquaculture farming, we examined the variation in sediment quality over time using different geochemical factors and investigated whether these factors are indicators of sediment quality recovery. The study area was an oyster Crassostrea gigas, farm in Tongyeong, Geyongsangnam-do, South Korea, where aquaculture activities had been carried out for 20 years, and the changes in water and sediment environment were monitored for 14 months after the abandonment of the farm. The mean water depth was 14 m, with a tidal range of 3 m, and seawater current velocity ranged from 4.7 to 7.0 cm/s. After the abandonment of the farm, total organic carbon and total nitrogen concentrations in the surface sediments decreased significantly over time compared to those in the control site; in particular, immediately after farm abandonment, acid-volatile sulfide concentrations decreased relatively rapidly. Carbohydrate and protein concentrations in the sediments showed no significant changes; however, lipid concentrations markedly decreased. Further studies on other aquaculture taxa and various aquaculture areas are needed to establish new policies for environmental management of fishing grounds, such as relocation of fishing grounds and determination of optimal fallowing periods.

Prognostic Factors and Therapeutic Outcomes in 22 Patients with Pleomorphic Xanthoastrocytoma

  • Objective : Pleomorphic xanthoastrocytoma (PXA) is a rare primary low-grade astrocytic tumor classified as WHO II. It is generally benign, but disease progression and malignant transformation have been reported. Prognostic factors for PXA and optimal therapies are not well known. Methods : The study period was January 2000 to March 2012. Data on MR findings, histology, surgical extents and adjuvant therapies were reviewed in twenty-two patients diagnosed with PXA. Results : The frequent symptoms of PXA included seizures, headaches and neurologic deficits. Tumors were most common in the temporal lobe followed by frontal, parietal and occipital lobes. One patient who died from immediate post-operative complications was excluded from the statistical analysis. Of the remaining 21 patients, 3 (14%) died and 7 (33%) showed disease progression. Atypical tumor location (p<0.001), peritumoral edema (p=0.022) and large tumor size (p=0.048) were correlated with disease progression, however, Ki-67 index and necrosis were not statistically significant. Disease progression occurred in three (21%) of 14 patients who underwent GTR, compared with 4 (57%) of 7 patients who did not undergo GTR, however, it was not statistically significant. Ten patients received adjuvant radiotherapy and the tumors were controlled in 5 of these patients. Conclusion : The prognosis for PXA is good; in our patients overall survival was 84%, and event-free survival was 59% at 3 years. Atypical tumor location, peritumoral edema and large tumor size are significantly correlated with disease progression. GTR may provide prolonged disease control, and adjuvant radiotherapy may be beneficial, but further study is needed.

A Study on the Methodology for a Clinical Trial of an Antidepressive Korean Herb by Comparison with St. John's Wort Trials (성요한초제제 임상시험 고찰을 통한 한약기원 항우울제 임상시험 방법론 연구)

  • Objectives : This study was undertaken to learn what should be considered in a good clinical trial investigating a herbal medicine as an antidepressant. Methods : Five well-designed clinical trials published from 2000 to 2006 investigating SJWE in depressive disorder were selected. The trials were reviewed and compared in terms of methodology such as trial design, patient selection, efficacy & safety evaluation, and so on. On the basis of this review of the trials and the regulations and guidelines of KFDA, we suggest some points to be considered for a good clinical trial of a herb for depression. Results : Although every trial had its own unique design, procedure, objectives and so on, all trials used randomizing and double blinding methods. If there is no ethical problem, a placebo-controlled design should be considered in a herbal antidepressant clinical trial for depression. Conclusions : Some points to be considered in an optimal & good clinical trial for an antidepressive herbal medicine were suggested as follows: 1) randomizing and double blinding manner is essential, 2) if there is no ethical problem, placebo control design should be considered, 3) the trial period should be 6 weeks, 4) out-patients will be recruited as subjects, 5) investigators will be well-trained psychiatrists or medical doctors, 6) the number of subjects should be calculated by statistical methods, 7) subjects should be diagnosed by DSM-IV criteria, 8) subjects who have current risk of committing suicide should be excluded, etc.

Cultural characteristics and formation of fruiting body in Lentinula edodes (표고버섯의 배양적 특성 및 자실체 형성에 관한 연구)

  • Lentinula edodes is known by oak mushroom. It has been favored as delicious and nutritious food and the low-calorie food with a high nutritional value. It is also functional food since it contains a material well-known for its medicinal benefits. Since the growth and quality of oak mushrooms are sensitively affected by environmental conditions, an adequate environmental control is very essential to improve the yield and quality under protected cultivation. The main objectives of the study were to investigate cultural characteristics of mycelial growth and in vitro fruiting of Lentinula edodes. The optimum culture media for mycelial growth of L. edodes were PDA and MYA. Similarly, optimum temperature was $25^{\circ}C$. Malt extract(2%) and yeast extract(0.2%) were optimum carbon and nitrogen sources. Optimal culture period was 110~120 days in sawdust medium. Mycelial growth in medium(61 mm/7 days) Quercus mongolica extract the most good. Among different five log types, highest mycelial growth and fruiting productivity were observed in Quercus variabilis sawdust(20.9%).

Influence of Artificial Acid Rain on the Growth of Soybean Irradiated with Low Dose Gamma Radiation (저선량 감마선이 조사된 대두작물의 생육에 미치는 산성강우 효과)

  • This study was carried out to investigate the influence of artificial acid rain on the growth of soybean irradiated with the low dose gamma radiation. The growth patterns of Hwangkumkong and Bangsakong were not dependent on the acid rain treatment and the period of seed storage but the radiation dose. In Hwangkumkong, the optimal irradiation dose for the growth promotion was lower in the acid rain treatment group than the control group. The symptom of acid rain damage appeared broadly around pH 2.20, were higher in Hwangkumkong than in Bangsakong, and were more effective in the seeds of 1997 than 1995. But the damage symptom of Bangsakong was not dependent on the acid rain treatment and the irradiation of gamma radiaiton.

Maximum standardized uptake value at pre-treatment PET in estimating lung cancer progression after stereotactic body radiotherapy

  • Purpose: This study aimed to identify the feasibility of the maximum standardized uptake value (SUVmax) on baseline 18F-fluorodeoxyglucose positron emission tomography-computed tomography (FDG PET/CT) as a predictive factor for prognosis in early stage primary lung cancer treated with stereotactic body radiotherapy (SBRT). Materials and Methods: Twenty-seven T1-3N0M0 primary lung cancer patients treated with curative SBRT between 2010 and 2018 were retrospectively evaluated. Four patients (14.8%) treated with SBRT to address residual tumor after wedge resection and one patient (3.7%) with local recurrence after resection were included. The SUVmax at baseline PET/CT was assessed to determine its relationship with prognosis after SBRT. Patients were divided into two groups based on maximum SUVmax on pre-treatment FDG PET/CT, estimated by receiver operating characteristic curve. Results: The median follow-up period was 17.7 months (range, 2.3 to 60.0 months). The actuarial 2-year local control, progression-free survival (PFS), and overall survival were 80.4%, 66.0%, and 78.2%, respectively. With regard to failure patterns, 5 patients exhibited local failure (in-field failure, 18.5%), 1 (3.7%) experienced regional nodal relapse, and other 2 (7.4%) developed distant failure. SUVmax was significantly correlated with progression (p = 0.08, optimal cut-off point SUVmax > 5.1). PFS was significantly influenced by pretreatment SUVmax (SUVmax > 5.1 vs. SUVmax ≤ 5.1; p = 0.012) and T stage (T1 vs. T2-3; p = 0.012). Conclusion: SUVmax at pre-treatment FDG PET/CT demonstrated a predictive value for PFS after SBRT for lung cancer.

Fed-batch Cultivation of Escherichia coli YK537 (pAET-8) for Production of phoA Promoter-controlled Human Epidermal Growth Factor

  • Secretion of the expressed heterologous proteins can reduce the stress to the host cells and is beneficial to their recovery and purification. In this study, fed-batch cultures of Escherichia coli YK537 (pAET-8) were conducted in a 5-L fermentor for the secretory production of human epidermal growth factor (hEGF) whose expression was under the control of alkaline phosphatase promoter. The effects of feeding of glucose and complex nitrogen sources on hEGF production were investigated. When the fed-batch culture was conducted in a chemically de-fined medium, the cell density was 9.68 g/L and the secreted hEGF was 44.7 mg/L in a period of 60 h. When a complex medium was used and glucose was added in pH-stat mode, the secreted hEGF was improved to 345 mg/L. When the culture was fed with glucose at a constant specific rate of $0.25\;gg^{-1}h^{-1}$, hEGF reached 514 mg/L. The effects of adding a solution containing yeast extract and tryptone were further studied. Different rate of the nitrogen source feeding resulted in different levels of phosphate and acetic acid formation, thus affected hEGF expression. At the optimal feeding rate, hEGF production achieved 686 mg/L.
