The objective of this study was to determine the effect of different height of drainpipe and growing media soilless cultivated cut rose in container culture. Two experiment were to examine the effect of the drainpipe height and media composition on yield and quality of cut rose, Four different drainpipe height (0, 3, 6, 9 cm) were treated to determine of optimal container type. Yield was the highest at 3 cm drainpipe height, but quality was not significantly affected by drainpipe height. Survival rate of rose was 100%, 100%, 92%, and 92%, respectively. Two different drainpipe height (0, 3 cm) and 7 media composition (pure coir and pelite, and mixed with two media 3 : 1, 2 : 1, 1 : 1, 1 : 2, 1 : 3 v/v) was treated to determine of media composition related to drainpipe height. The supply of nutrient solution was controlled by the signal of water potential at -5 kPa using frequency domain reflectometry (FDR) sensor in mixed coir with pelite 3 : 1, 1 : 1, 1 : 3 (v/v), respectively. Irrigation frequency reduced in high ratio of coir media and 3 cm height of drainpipe. Quality of cut rose except for flower weight and yield until 2nd harvest was not significantly affected by drainpipe height, but yield after 3rd was higher at 3 cm than 0 cm height of darinpipe. In the media composition, yield and qulity of cut rose was increased at high ratio of coir media.

The effects of $CO_2$ concentration, nutrient levels, and irradiance on the growth of germlings and juveniles, and on the photosynthesis of adults were examined in a green tide alga, Ulva australis. We used a factorial experimental design with two $CO_2$ concentrations (380 and 750 ppm), two nutrient levels (control and PES medium), and two irradiance levels (50 and $100{\mu}mol$ photons $m^{-2}s^{-1}$). Germlings grew best ($664.15{\pm}61.45{\mu}m$ in length) under conditions of 750 ppm, PES, and $100{\mu}mol$ photons $m^{-2}s^{-1}$ after 10 days in culture. Relative growth rates (RGR) of the juveniles were greatest (4.41% $day^{-1}$) under conditions of 750 ppm, PES, and $50{\mu}mol$ photons $m^{-2}s^{-1}$ after 5 days in culture. Photosynthetic efficiency ($F_v/F_m$) of the adult discs was $0.73{\pm}0.05$ before the experiment and reached a maximum ($0.83{\pm}0.01$) under conditions of 750 ppm, control, and $50{\mu}mol$ photons $m^{-2}s^{-1}$ after 5 days in culture. Growth (germlings and juveniles) and photosynthesis (adult discs) of Ulva australis increased when $CO_2$ levels were 750 ppm. Additionally, the optimal irradiance for growth and photosynthesis differed among stages, wherein germlings grew best at $100{\mu}mol$ photons $m^{-2}s^{-1}$, juveniles grew best at $50{\mu}mol$ photons $m^{-2}s^{-1}$, and adults photosynthesized most at $50{\mu}mol$ photons $m^{-2}s^{-1}$. The performance of Ulva australis at all examined life stages was enhanced under the PES nutrient treatment. In conclusion, the physiological responses of U. australis to varying $CO_2$, nutrient, and irradiance levels differed slightly among life stages. However, growth and photosynthesis always increased with elevated $CO_2$ and nutrient concentrations. These results indicate that U. australis green tide blooms might occur more frequently in coastal areas if $CO_2$ and nutrient concentrations increase.
This experiment was conducted to introduce PAT (phosphinothricin acetyltransferase, non-selective herbicide bialaphos resistant gene) gene into potato (Solanum tuberosum. cv. Desiree). Optimal shoot regeneration from leaf discs and stem segments was obtained in MS medium supplemented with 0.1 mg/L IBA and 0.5 mg/L BA, and the frequency of shoot regeneration was 54% in left discs and 46% in stem segments. In this condition, leaf discs and stem segments of potato were co-cultivated with A. tumefaciens MP90 which contained binary vector with GUS: :NPTII gene and PAT gene. Transgenic shoots were regenerated from leaf and stem-derived calli on selection medium with 100mg/L kanamycin. The 100${\mu}M$ acetosyringone treatment during the co-cultivation highly enhanced(4 times than the control) the shoot regeneration on selection medium. When the putative transgenic plants were transferred to medium with 10mg/L basta, all of them were survived. After PCR. GUS test, and Southern blot analysis of the survived plant, we confirmed that the gene was stably integrated into the potato genome and expressed. After the transgenic plants were transplanted in soil, and the transgenic plants were sprayed with the herbicide basta (300ml/10a), the transgenic plants remained green but control plants were died.

This experiment was conducted to determine optimal hot-air drying time for producing antioxidative calendula tea. The edible calendula was dried in hot air at $60^{\circ}C$ for 5, 10, 15 hours and Hunter's color value was measured and extracted with 95% ethanol as a solvent. The contents of total polyphenol, total flavonoid, total carotenoid and DPPH (2,2 Dipheny-1-picrylhydrazyl) radical scavenging activity were investigated. The correlation between the amount of antioxidant and antioxidant activity was analyzed. As the treatment time increased, the Hunter values of L, a, and b tended to decrease, and the L value and b value decreased significantly at 15 hours. The color change was occurred in all treatments compared with the control, and the total color difference (${\Delta}E$) was the largest at 15 hours treatment. The contents of total flavonoids, total carotenoids and DPPH scavenging activity increased in the 5 hours, but decreased in the 15 hours compared to the 10 hours. The content of total polyphenol was not changed with different treatment time. A positive correlation ($p{\leq}0.01$, r = 0.610) between carotenoids and DPPH radical scavenging activity was demonstrated. The DPPH radical scavenging activity was increased at 5 and 10 hours compared to the control and decreased at 15 hours. Therefore, 5 hours or 10 hours of treatment is appropriate and further studies are needed to determine the specific treatment time.
This study investigated the effect of the addition of pomegranate concentrate to yogurt on the growth of pathogenic and lactic acid bacteria. The concentration of the MRS broth was adjusted to one-half and used for an experiment. Pomegranate concentrate was added at concentrations of 4%, 2%, 1%, and 0.5%, which significantly promoted the growth of Lacto-coccus cremoris, Weissella cibaria, Weissella paramesenteroides, Lactobacillus plantarum, Lactobacillus acidophilus, Streptococcus thermophillus, Lactobacillus bulgaricus, and Lactobacillus lactis. The growth of lactic acid bacteria increased with higher concentrations of pomegranate. However, the addition of pomegranate concentrate inhibited the growth of Escherichia coli KCCM11587, E. coli KCCM11591, E. coli KCCM11596, and E. coliKCCM11600. Yogurt with added pomegranate concentrate demonstrated optimal conditions compared to that of the control without the addition. Particularly, the viable cell count of lactic acid bacteria was significantly higher in the yogurt with pomegranate concentrate. Furthermore, the viability of the lactic acid bacteria in the yogurt with pomegranate concentrate was higher than that of the control without the addition of concentrate during storage.
King oyster mushrooms are one of the major fresh agricultural products which their exports are increasing every year in Korea. Lycoriella ingenua, is notorious insect pest in agriculture, especially in mushroom production. Larvae of L. ingenua cause mainly direct crop damage and adults are vectors of several dangerous fungal pathogens. In this study, the effects of electron beam, X-ray, and gamma-ray irradiation on the development and reproduction of L. ingenua were evaluated. In addition, to find the optimal dose to control L. ingenua in a box filled with king oyster mushrooms, an empirical experiment was conducted for each radiation. As a result, the development and reproduction of L. ingenua were inhibited at 50 Gy for all electron beam, X-ray, and gamma-ray irradiation. Additionally, at the top, middle, and bottom of the export box filled with king oyster mushrooms, the development and reproduction of L. ingenua were inhibited by electron beam with 150 Gy, X-ray with 100 Gy, and gamma-ray with 50 Gy. These results can be provided as basic data for establishing an integrated quarantine management system when exporting mushrooms. It will also contribute to the safety of agricultural products and the strengthening of export competitiveness.
Pleurotus eryngii is one of the most promising mushrooms produced on the domestic farms. The quality as well as quantity of Eryngii is sensitively affected by micro climate factors such as temperature, relative humidity, $CO_2$ concentration, and light intensity. To safely produce high-quality Eryngii all the yew round, it is required that the environmental factors be carefully controlled by well designed structures equipped with various facilities and control systems. At the commercial mushroom cultivation houses of permanent frame type (A, B), this study was carried out to find out reasonable range of each environmental factor and yield together with economic and safe structures influencing on the optimal productivity of Eryngii. This experiment was conducted for about two-year ken Nov. 2003 to Dec. 2005 in cultivation house. Ambient temperature during the experiment period was not predominantly different from that of a normal year. The capacity of the hot water boiler and the piping systems were not enough. Because the capacity of electric heater and air circulation were not enough, air temperatures in cultivation house before improvement of system were maintained somewhat lower than setting temperature, and maximum air temperature difference between the upper and lower growth stage during a heating time period was about 5.1. But the air temperatures after system improvement were maintained within the limits range of setting temperature without happening stagnant of air. Air temperature distribution was generally distributed uniform. Relative humidity in cultivation house before , improvement was widely ranged about $44{\sim}100%$. But as the relative humidity after improvement was ranged approximately $80{\sim}100%$, it was maintained within the range of relative humidity recommended. And $CO_2$ concentration was maintained about $400{\sim}3,300mg{\cdot}L^{-1}$ range. The illuminance in cultivation house was widely distributed in accordance with position, and it was maintained lower than the recommended illuminance range $100{\sim}200lx$. The acidity of midium was some lower range than the recommend acidity range of pH $5.5{\sim}6.5$. The yield was relatively ununiform. In case of bottle capacity of 1,300cc, the mushroom of the lowest grade was less than 3%. The consumption electric energy was quite different according to the cultivation season. The electric energy consumed during heating season was much more than that of cooling season.

The nutrient recovery in phosphate crystallization process was investigated by using laboratory scale uptlow reactors, adopting sequencing batch type configuration. The industrial waste lime was used as potential cation source with magnesium salt($MgCl_2$) as control. The research was focused on its successful application in a novel integrated sludge treatment process, which is comprised of a high performance fermenter followed by a crystallization reactor. In the struvite precipitation test using synthetic wastewater first, which has the similar characteristics with the real fermentation effluent, the considerable nutrient removal(about 60%) in both ammonia and phosphate was observed within $0.5{\sim}1$ hr of retention time. The results also revealed that a minor amount(<5%) of ammonia stripping naturally occurred due to the alkaline(pH 9) characteristic in feed substrate. Stripping of $CO_2$ by air did not increase the struvite precipitation rate but it led to increased ammonia removal. In the second experiment using the fermentation effluent, the optimal dosage of magnesium salt for struvite precipitation was 0.86 g Mg $g^{-1}$ P, similar to the mass ratio of the struvite. The optimal dosage of waste lime was 0.3 g $L^{-1}$, resulting in 80% of $NH_4-N$ and 41% of $PO_4-P$ removal, at about 3 hrs of retention time. In the microscopic analysis, amorphous crystals were mainly observed in the settled solids with waste lime but prism-like crystals were observed with magnesium salt. Based on mass balance analysis for an integrated sludge treatment process(fermenter followed by crystallization reactor) for full-scale application(treatment capacity Q=158,880 $m^3\;d^{-1}$), nutrient recycle loading from the crystallization reactor effluent to the main liquid stream would be significantly reduced(0.13 g N and 0.19 g P per $m^3$ of wastewater, respectively). The results of the experiment reveal therefore that the reuse of waste lime, already an industrial waste, in a nutrient recovery system has various advantages such as higher economical benefits and sustainable treatment of the industrial waste.

The midwater pair trawl which is being used at present in Korea have several problems. Firstly, it is difficult to control the net height on high towing speed. Secondly, net breaking often occurs owing to floats and thirdly, the volume of net on the net drum is so large. This study is aiming for examining the possibility of application for the Korean midwater pair trawl through the model experiment of non-float midwater pair trawl. The model of non-float midwater pair trawl was manufactured as 1/100 of the full scale net which is being used in bottom pair trawl for 850ps class considering the Tauti's Similarity law. The model experiment was carried out to analyze the opening efficiency according to the variation of lower warp length and the opening efficiency was investigated between th proto type and non-float type. The results obtained can be summarized as follows ; 1. The hydrodynamic resistance of non-float type was about 10~20% smaller than that of the proto type and it increased about 1ton according to the increase of dL at the condition of the same flow speed. The resistance acting on the lower warp decreased about 5% but that of the upper warp increased according to the increase of lower warp length (dL) at the condition of the same flow speed. 2. The net height of the non-float type decreased almost linearly according as the increased of flow speed and it increased in a logarithmic functional form with the increase of the lower warp length at the condition of the same flow speed. On the decreasing rate of the net height, the non-float type was lower than the proto type and the difference of the decreasing rate was about 12% at 3.0 knot, 25% at 4.0 knot, 25% at 4.0 knot respectively when dL was 30m. 3. The net width of non-float type was not varied so much as only 2m range and was larger than that of proto type. 4. The mouth area of non-float type decreased in a exponential functional form. On the decreasing rate of the mouth area, the non-float type was lower than the proto type. The filtering volume increased in a logarithmic functional form with increasing flow speed and the filtering volume of proto type decreased steeply over 3.0knot, but that of non-float type increased until 4.0knot. 5. The optimal length of lower warp was when the value of dL was about 30m and the optimal position of front weight was at the connection point of four net pendants.
The experiment was conducted to investigate the nutrition absorption pattern in the growth stages and develope the optimal nutrient solution hydroponically grown the tomato in closed substrate culture system with the nutrient solution of National Horticultural Research Station in Japan into 1/2S, 1 S, and 2S. When plant was grown in 1/2 S, the growth and yield were high and the pH and EC in the rooting zone were stable. Suitable composition of nutrient solution for tomato was $NO_3-N$ 7.1, $PO_{4}$-P 2.1, K 4.0, Ca 3.1, Mg 1.2, and $SO_{4}-S\;1.2\;me{\cdot}L^{-1}$ in the early growth stage and $NO_3-N$ 6.5, $PO_4-P$ 2.3, K 3.4, Ca 3.1, Mg 1.1, and $SO_4-S\;1.1\;me{\cdot}L^{-1}$ in the late growth stage by calculating a rate of nutrient and water uptake. To estimate the suitability for the nutrient solution of tomato in a development of optimum nutrient solution of tomato developed by Wonkwang university in korea (WU), plant was grown in perlite substrate supplied with different solution and strengths(S) by research station for greenhouse vegetable and floricultuin in the Netherlands (Proefstation voor tuinbouw onder glas te Naaldwijk; PTG) of 1/2 S, 1 S and 2 S, respectively, The growth was good at the PTG and WU of 2 S in early growth stage, and at the WU 2S in late growth stage. The highest yield of tomato obtained in the WU of 2 S, although blossom-end rot was appeared in all treatments. pH and EC in root zone of WU of 2 S were stable during the early and late growth stage. Therefore when plant was grown in WU of 2 S, N and P content in the nutrient solution need to low, according N and P content of their leaves were high in WU of 2 S.
