Establishment of HPLC-UV Analysis Method Validation for Simultaneous Analysis of Standard Compounds of Oplopanax elatus Nakai Stem (HPLC-UV를 이용한 땃두릅나무 줄기의 지표 성분 동시 분석법 확립)

  • In our previous study, we found uracil, adenosine, protocatechuic acid, syringin (eleutheroside B) and scoparone (6, 7-dimethoxycoumarin) in the Oplopanax elatus Nakai Stem. High-performance liquid chromatography (HPLC) -UV was used to quality and quantify the internal marker compounds in the O. elatus extract after validation of method with linearity, limit of detection (LOD), limit of quantitation (LOQ), accuracy and precision. The specificity assessment visually confirmed that the substance was detected without the introduction of other substances. The established method showed high linearity of the calibration curve and coefficient of correlation ($R^2$) of over the 0.999. HPLC was reported as five standard compounds equivalent using the following linear equation based on the calibration curve. The accuracy of measurement was 84.34 ~ 119.74% and the relative standard deviation (RSD) value was 0.28 ~ 1.60%. In addition, our established method showed high repeatability. The RSD value was 1.10 ~ 6.81%. So, we found the amount of the internal marker compounds in the O. elatus extract. These results demonstrated that can be used to quality evaluation of the O. elatus.

  • This study aimed to evaluate the safety of Oplopanax elatus (Nakai) Nakai hydrothermal extract powder. It was conducted using male and female Sprague-Dawley (SD) rats. The test group was established with dose of 500 (low-dosage group), 1,000 (medium- dosage group), and 2,000 (high- dosage group) mg/day. These are investigated that number of dead animals, general symptoms, weight changes, food consumption, ophthalmological examination, urinalysis, urine volume, hematological values, plasma coagulation time values, serum biochemical values, absolute organ weight, relative organ weight and histopathological finding during the experiment. As a result of the above, toxicological changes were not observed. Therefore, the non-toxic content of Oplopanax elatus (Nakai) Nakai hydrothermal extract powder is determined to be 2,000 mg/kg/day, and target organ was not observed.

  • This study was conducted to investigate the toxicity symptoms and approximate lethal dose (ALD) of Oplopanax elatus (Nakai) Nakai hydrothermal extract powder by single oral dose toxicity and 4 weeks of repeated oral dose determination. The Sprague-Dawley (SD) male and female rats were treated with 1,250 (low- dosage group), 2,500 (medium- dosage group) and 5,000 (high- dosage group) mg/kg. In the single oral dose toxicity test, no dead animals and toxic symptoms were observed during the experiment. And there were no related with anomalies in normal weight changes and autopsy results. In the four-week repeated oral dose determination test, no death animals and toxicity symptoms were observed during the experiment, and there were no abnormal results in weight changes, feed and negative intake measurements. Results of eye examination, urinalysis, hematological values and serum biochemical values, gross findings and absolute organ were not of singularity. These result demonstrated that no toxic symptoms were observed by the test substance Oplopanax elatus (Nakai) Nakai hydrothermal extract powder under this test condition, and the non-toxic content is determined to be 5,000 mg/kg/day.

Establishment of Suspension Culture System to Induce Somatic Embryo in Oplopanax elatus Nakai (현탁배양을 통한 땃두릅나무의 체세포배 유도)

  • This study was carried out to select the appropriate medium (especially, carbon and nitrogen source, potassium phosphate and pH) for somatic embryogenesis in order to develop the rapid mass production system in suspension culture of Oplopanax elatus Nakai. Direct somatic embryos were obtained from root explants in the hormone free suspension culture (MS). Combination of $NH_4NO_3$ and $KNO_3$ at the ratio of 1650 (mg/${\ell}$) : 1900 (mg/${\ell}$) obtained the better result to produce somatic embryo in suspension culture. MS medium supplemented with $170mg/{\ell}$ $KH_2PO_4$. The addition of 1 and 3% sucrose was effective for formation of embryogenic callus. Therefore, this report will be helped to improve the establishment for suspension culture in Oplopanax elatus Nakai.

Assessment on Antioxidant Properties of Oplopanax elatus Nakai in vitro

  • Effects of Oplopanax elatus have known to various pharmaceutical therapies. However, chemical prosperities in the plant are rarely investigated. In order to detect biological activity, we evaluated the antioxidant activity of five fractions from methanolic extracts in each part of O. elatus. Also, contents of polyphenols and polysaccharides were measured. Five fractions were of sub-fractions using n-hexane, ethyl-acetate, n-butanol, and water from methanolic extracts. Ethylacetate and n-butanol fractions from stem and root exhibited strong antioxidant activity and high total phenolics content. On the HPLC analysis, ten free phenolics, including p-hydroxybenzoic acid, chlorogenic acid, caffeic acid, syringic acid, p-cou maric acid, ferulic acid, naringin, hesperidin, quercetin and trans-cinnamic acid, were identified from the fractions and were shown to different quantitative proportions. Furthermore, ethylacetate and n-butanol fraction had the highest amount of chlorogenic acids, one of the cinnamic acid derivatives possessing pharmacological properties. These results indicated that the fractions of O. elatus, as well as methanolic extracts, could be used as natural antioxidative ingredients.

The Enhanced Effect of Oplopanax elatus Nakai on the Immune System and Antitumor Activity (땃두릅(Oplopanax elatus Nakai) 추출물의 면역자극 활성 및 항암 증진 효과)

  • The present study is designed to explore an anti-tumor activity on crude extracts of Oplopanax elatus. Water extractions of Oplopanax elatus were performed at $100^{\circ}C$(OeE-100). OeE-100 doses up to $62.5{\mu}g/m{\ell}$ had no cytotoxicity on the tumor cell lines in vitro. In experimental lung metastasis of colon26-M3.1 carcinoma or B16-BL6 melanoma, the prophylactic intravenous ($4{\sim}100{\mu}g/mouse$) or oral (2 mg/mouse) administration of OeE-100 significantly inhibited tumor metastasis as compared with tumor controls. Peritoneal macrophages stimulated with OeE-100 produced various cytokines such as TNF-${\alpha}$, IL-6 and IL-12. In an analysis of NK-cell activities, i.v. administration of OeE-100 ($10{\sim}100{\mu}g/mouse$) significantly augmented the cytotoxicity to YAC-1 tumor cells. Vaccination of mice with boiling-treated tumor cells (BT-vaccine) in combination with OeE-100 ($100{\mu}g/mouse$) showed higher inhibitions in tumor metastasis when compared with the mice of BT-vaccine treatment. In addition, the splenocytes from OeE-100 admixed BT-vaccine immunized mice secreted a higher concentration of Th1 type cytokine such as IFN-${\gamma}$. These results suggested that the OeE-100 stimulated immune system and was a good candidate adjuvant of anti-tumor immune responses.

Establishment of Acclimatization System and Growth Characteristics for Regenerated Plants of Oplopanax elatus Nakai (땃두릅나무 재분화 유식물체의 순화 체계 및 생육 특성)

  • Background: Oplopanax elatus is widely distributed at high altitudes (about 1,100 m) in China, Russia and Korea. It is hard to propagate, breed, and difficult to grow. Hence, it has been designated as a rare and endangered medicinal plant. A study was conducted to establish a system for large scale seedling production of Oplopanax elatus in vitro and to find the ideal environment for its seedling growth. Methods and Results: In this study, the explants produced under in vitro conditions during our previous study were grouped into three categories (under 10 mm, 10 mm - 30 mm and above 30 mm) based on plant height and were transferred to the growth-chamber and greenhouse for two weeks in each setting for acclimatization. The plantlet category of above 30 mm showed good performance, and was further evaluated under three acclimatization methods as follows: three different growth media (commercial soil, commercial soil + perlite, commercial soil + sand), four shading levels (0%, 50%, 70%, 90%) and four altitude levels (157 m, 218 m, 601 m, 870 m) in Gangwon province of South Korea. As results, O. elatus seedlings showed better growth characteristics at 870 m of altitude, 70% shading level and in the commercial soil compared to other treatments. Conclusions: The regenerated seedlings of Oplopanax elatus obtained through plant tissue culture would be advantageous for use in large scale seedling production systems paired with a good acclimation method. For obtaining optimal results, it is recommended that seedling be acclimatized in a high altitude environment.
