I-SSR 표지자에 의한 눈측백나무 남한 잔존집단의 유전변이와 구조 (Genetic Variation and Structure of the Relict Populations of Korean Arborvitae (Thuja koraiensis Nakai) in South Korea, Employing I-SSR Markers)

  • 본 연구에서는 눈측백나무(Thuja koraiensis Nakai) 4개 천연집단을 대상으로 84개체를 선발한 뒤 다형성을 보인 29개의 I-SSR amplicons를 이용하여 유전변이를 조사하였다. 6개의 I-SSR primer로 유전다양성을 추정한 결과 평균 유효대립유전자의 수($A_e$)는 1.44개, 이형접합도의 기대치($H_e$)는 0.258, Shannon의 다양성 지수(S.I.)는 0.385로 크게 높지는 않은 것으로 추정되었다. 이를 다양한 표지자를 이용하여 측백나무과(Cupressaceae)에 속하는 지금까지 연구된 수종들과 비교하여보면 국내의 눈측백나무는 유사하거나 다소 높은 유전변이량을 보유하고 있는 것으로 나타났다. 조사된 4개의 집단을 대상으로 AMOVA를 수행한 결과 전체 유전변이 가운데 13%가 집단간 차이로부터 기인하는 것으로 나타났고, 나머지 87%는 집단내 개체간 차이로부터 기인한 것으로 나타났다. 유전적 거리에 의한 UPGMA 유집분석을 실시한 결과 지리적인 경향은 나타나지 않았다. 유전변이량이 가장 높은 장산집단과 유전적 조성에서 가장 이질적인 방태산집단이 보전 가치가 큰 것으로 판단된다.

Polymorphism, Expression of Natural Resistance-associated Macrophage Protein 1 Encoding Gene (NRAMP1) and Its Association with Immune Traits in Pigs

  • Natural resistance-associated macrophage protein 1 encoding gene (NRAMP1) plays an important role in immune response against intracellular pathogens. To evaluate the effects of NRAMP1 gene on immune capacity in pigs, tissue expression of NRAMP1 mRNA was observed by real time quantitative polymerase chain reaction (PCR), and the results revealed NRAMP1 expressed widely in nine tissues. One single nucleotide polymorphism (SNP) (ENSSSCG00000025058: g.130 C>T) in exon1 and one SNP (ENSSSCG00000025058: g.657 A>G) in intron1 region of porcine NRAMP1 gene were demonstrated by DNA sequencing and PCR-RFLP analysis. A further analysis of SNP genotypes associated with immune traits including contain of white blood cell (WBC), granulocyte, lymphocyte, monocyte (MO), rate of cytotoxin in monocyte (MC) and $CD4^-CD8^+$ T lymphocyte subpopulations in blood was carried out in four pig populations including Large White and three Chinese indigenous breeds (Wannan Black, Huai pig and Wei pig). The results showed that the SNP (ENSSSCG00000025058: g.130 C>T) was significantly associated with level of WBC % (p = 0.031), MO% (p = 0.024), MC% (p = 0.013) and $CD4^-CD8^+$ T lymphocyte (p = 0.023). The other SNP (ENSSSCG00000025058: g.657 A>G) was significantly associated with the level of MO% (p = 0.012), MC% (p = 0.019) and $CD4^-CD8^+$ T lymphocyte (p = 0.037). These results indicate that the NRAMP1 gene can be regarded as a molecular marker for genetic selection of disease susceptibility in pig breeding.

PKC inhibitors RO 31-8220 and Gö 6983 enhance epinephrine-induced platelet aggregation in catecholamine hypo-responsive platelets by enhancing Akt phosphorylation

  • Impaired responsiveness of platelets to epinephrine (epi) and other catecholamines (CA) has been reported in approximately 20% of the healthy Korean and Japanese populations. In the present study, platelet aggregation induced by epi was potentiated by RO 31-8220 (RO) or G$\ddot{o}$ 6983 (G$\ddot{o}$). Phosphorylated Akt (p-Akt) was very low in epi-stimulated PRP from CA-hypo- responders (CA-HY), whereas it was detected in those from CA-good responders (CA-GR). RO and G$\ddot{o}$ increased p-Akt, one of the major downstream effectors of phosphoinositol-3 kinase (PI3K), in epi-stimulated PRP from both groups. Wortmannin, a PI3K inhibitor, attenuated the RO or G$\ddot{o}$-induced potentiation of p-Akt in epi-stimulated PRP, suggesting positive effects for RO and G$\ddot{o}$ on PI3K. $TXA_2$ formation was increased by the addition of either RO or G$\ddot{o}$ in epi-stimulated platelets. The present data also suggest that impaired Akt phosphorylation may be responsible for epinephrine hypo-responsiveness of platelets.

  • Multiple species of muridae and arvicolidae rodents serve as the natural reserviors of hantaviruses. Hantaviruses are distributed in rodent populations world-widely even in geographical areas where hemorrhagic fever with renal syndrome (HFRS) has not been reported. Serologic diagnosis of infection, using hantaviral antigen, indicates that hantaviruses are widey distributed in wild rodents. This study was designed to intended the hantavirus infection among wild rodents captured in Kyebang mountain, Kangwon-do in Korea. A total of 216 wild rodents in 3 species were trapped in July and September in 1995. Serological evidence for hantaviruses infection were tested against five hantavirus antigens by indirect immunofluorescent antibody technique (IFA). Among 100 Eothenomys regulus, 78 Apodemus peninsulae and 38 Apodemus agrarius; 12 C. regulus, 15 A. peninsulae and 6 A. agrarius were IF antibody positive against hantaviruses. This data suggest that Eothenomys regulus and Apodemus peninsulae would be a natural reservoir of hantaviruses.

Melanocyte-stimulating Hormone Receptor (MC1R) Genotype and Its Effects on Coat Color in Korean Jindo Dogs

  • The Jindo dog is a Korean natural monument and is recognized by the Fédération Cynologique Internationale. A prominent feature is the diverse coat color within the breed. To analyze the genetic basis of variation in the Jindo coat color, we sequenced the protein-coding regions of the melanocortin 1 receptor gene (MC1R). The MC1R coding sequence was determined from 154 dogs in five breeds (Jindo, Labrador Retriever, English Springer Spaniel, Belgian Malinois, and German Shepherd). To confirm the genetic structure of sampled populations, we tested for Hardy-Weinberg equilibrium (HWE) and computed $F_{st}$ The sample populations did not significantly deviate from HWE. $F_{st}$ was 0.02 between white and fawn Jindo dogs; this was lower than $F_{st}$ between breeds. Six single nucleotide polymorphisms (SNPs) were detected in the MC1R coding region. Among the six SNPs, five were non-synonymous (S90G, T105A, Q159P, M264V, and R306ter) and one was synonymous SNP (Y298Y). From the SNPs, we predicted four haplotypes (H1, H2, H3, and H4) for Jindo MC1R. Jindo dogs had different haplotypes corresponding to different coat colors. H1 was frequently observed in white Jindo dogs with an odds ratio of 5.03 (95% CI: 2.27-11.18, p<0.0001), whereas H2 and H4 were observed only in fawn Jindo dogs. Our findings indicate that SNP haplotype can influence coat color. Knowledge of MC1R haplotypes can help discriminate white and fawn coats in Jindo dogs. We hope this report will trigger more research into the genetics of this traditional Korean dog and will be a reference for dogs of Asian origin. Also, our results will provide a useful genetic marker for Jindo dog breeders who have selected for specific colors.

Stainless steel에 접종된 식중독 미생물에 대한 천연항균제 및 유기산 분무 살균효과 (Efficacy of Aerosolized Natural Antimicrobial and Organic Acids as a Sanitizer against Foodborne Pathogens on Stainless Steel)

  • 본 연구는 Escherichia coli O157:H7, Salmonella Typhimurium 그리고 Listeria monocytogenes에 대하여 천연항균제와 유기산을 이용한 분무 살균제 (aerosol sanitizer)의 살균효과를 검토하였다. 실제 가공공장의 주요 기계나 기구로 사용하는 stainless steel을 coupon으로 이용하여 인위적으로 접종한 식중독 미생물을 model cabinet에서 5분 동안 자몽종자추출물(grapefruit seed extract), acetic acid, citric acid 그리고 lactic acid로 처리 하였다. 3가지 식중독 미생물의 수는 GEF와 유기산 단독 처리시 0.34-3.77log의 균 감소를 나타내었고, 병행 처리시 1.72-3.89 log의 균 감소를 나타내었고 GEF, 유기산 그리고 알코올을 병행 처리시 1.46-5.05 log의 균 감소효과를 나타냈다. 세 종류의 유기산 중 항균력이 가장 높은 것으로 측정된 lactic acid를 이용하여 scale-up 모델에서 10분 동안 처리 한 결과는 E. coli O157:H7, S. Typhimurium 그리고 L. monocytogenes는 각각 3.42, 2.72, 2.30log의 균 감소를 나타내었다. 따라서 천연항균제와 유기산을 이용한 분무 살균제는 안전한 식품에 대한 소비자의 수요를 충족시키고, 환경 위생 방법으로 사용할 수 있다고 생각되었다.

  • The purposes of this research were to understand the realities of health management and to analyze the subjective, functional, psychological, and social health effects of the application of an exercise and education program to manage health on an elderly population residing in an agricultural community. The specific purposes of this research were: 1. to determine the subjective, functional, psychological health conditions, and performance level of instrumental activities of daily living of both the experimental and control groups before introducing a health management program, and then 2. to compare these with both the experimental and control groups' subjective, functional, psychological health condition and performance level of instrumental activities of daily living after applying the health management program. The health management program was introduced to the experimental group (50 people) but not to the control group (50 people). The health management program was composed of a health exercise program and a health education program. A comparative study of subjective health conditions was done to examine the effect of the health management program and the characteristics of health related variables before and after the program application.   • Objective : In the present study, we examined whether Canavalia gladiata D.C. (CG) and Arctium lappa L., Redix (AL) mixture (CGAL), their components, lupeol and chicoric acid, regulate immune system and suppress the tumor in vitro and in vivo. Methods : LPS-induced reactive oxygen species (ROS) and nitric oxide (NO) were measured after treatment with CG extract (CGE), CGAL, lupeol, chicoric acid and lupeol and chicoric acid mixture (lupeol+CA) in Raw264.7 cell. To determine the effect of CGE on immune responses, immune cell population and IgG production were assessed in mice. To investigate the effect of CGAL and their component on anti-tumor activity, tumor volume and weight were measured, cell cycles and immune cell population were analyzed in MC38 injected tumor bearing mice. Also, NK cell activity was determined in splenocyte isolated from tumor bearing mice. Results : CGE, CGAL, lupeol, chicoric acid and lupeol+CA decreased the LPS-induced ROS and NO production without cell toxicity in RAW264.7 cells. CGE increased the immune cell populations of $CD4^+T$, $CD8^+T$ and macrophages in various immune organ of mice. In tumor bearing mice, CGAL, lupeol, chicoric acid and lupeol+CA suppressed tumor volume and weight. In cell cycle analysis, they decreased the percentages of S phase. In addition, CGAL, lupeol, chicoric acid and lupeol+CA immune cell populations of $CD4^+T$, $CD8^+Tcell$, NK cell and macrophage in tumor as well as NK cell activity. Conclusion : CGAL and its compounds may enhance immune responses and suppress tumor growth, and may be capable of developing health functional foods.

Polyphenol-rich Sargassum horneri alleviates atopic dermatitis-like skin lesions in NC/Nga mice by suppressing Th2-mediated cytokine IL-13

  • Atopic dermatitis (AD) is one of major skin inflammatory diseases characterized by excessive Th2-mediated immune responses. Recent evidence provides that interlukin-13 (IL-13) plays the role of a key Th2 cytokine that drives the inflammation underlining AD. Due to adverse effects of commercially available synthetic drugs, the need for treatments based on natural products is gaining much attention. Sargassum horneri is an edible brown algae known for beneficial bioactivities including anti-inflammation. We investigated if polyphenol-rich S. horneri extracts (SHE) could suppress AD-like skin lesions in NC/Nga mice and if that involved inhibition of the infiltration of Th2-mediated cytokine IL-13. We observed markedly increased infiltration of IL-13 positive cells in AD-like skin lesions of mice but SHE treatments decreased it. Also, the dermal expression of IL-13 was sufficient to cause inflammatory responses in mice skin resembling human AD. SHE suppressed the dermal infiltration of inflammatory cells where IL-13 plays a crucial role in skin tissues and in the recruitment of inflammatory cells. Furthermore, it was confirmed that SHE reduced T cell, dendritic cell, and macrophage populations in spleen. Moreover, SHE decreased the collagen deposition in skin and ear dermis resulting in reduced fibrosis that occurs in AD due to excessive collagen. Taken together, our results reveal that SHE suppressed the infiltration of inflammatory cells into skin dermis by decreasing the infiltration of IL-13 positive cells. Therefore, SHE could be taken as a useful therapeutic agent to alleviate AD.

세뿔투구꽃의 자생지 식생과 환경특성 (Environment and Vegetation Characteristics of Aconitum austrokoreense Koidz. habitats)

  • 본 연구는 희귀 및 특산식물과 멸종위기야생식물 II급으로 지정되어 있는 세뿔투구꽃의 자생지 환경을 조사하여 보전 및 복원 시 기초자료를 제공하고자 한다. 조사결과 세뿔투구꽃의 자생지는 해발고도 260-728m범위와 경사 $4-39^{\circ}$의 계곡부에 주로 생육하는 것으로 조사되었다. 식생 및 관속식물 조사결과 7개 지역의 20개 방형구내에서 조사된 관속식물은 총 147분류군이 출현하였으며, 각 조사구내 초본층 피도와 빈도를 기초로 한 세뿔투구꽃의 중요치를 산출한 결과 평균 24.3%로 나타났다. 종다양도는 1.23로 산출되었으며, 우점도와 균등도는 각각 0.22와 0.77로 확인되었다. 토양분석 결과 토성은 미사질양토(4site)와 사질양토(2site), 양토(1site) 순으로 나타났으며, 유기물함량은 7.83%, 전질소함량은 0.35%, pH는 5.69으로 측정되었다. 환경요인과 개체군특성에 기초한 상관분석에서는 세뿔투구꽃의 자생지에서 엽록소와 경사도간의 강한 부의상관이 인정되었고, 개화율과 토양습도는 부의상관이 인정되었다. 자생지의 안정적인 유지를 위해서는 서식지 보전이 중요하며 서식지 주변에 보호구역을 설정이 필요하다고 판단된다.
