The objective of this study is to suggest the improvement method of the profitability in our marine fish culture business. So I investigated the actual condition for aquaculture farms in South Sea coast by the question and actual survey. 1 recognized the following facts; 1) The marine floating netcage system are a great portion in two province - Kyeongnam and Cheunnam, 2) The rockfish are reared absolutely much more than the olive flounder, 3) The polyculture by various species are more carried out than the monoculture of the olive flounder or rockfish, etc. I analyzed the profitability in olive flounder monoculture, rockfish monoculture, olive flounder and rockfish polyculture, and rockfish polyculture taking turns year by year respectively. At a result I comfirmed that the net income to net sales was the highest in the olive flounder monoculture bacouse the olive flounder,s sale price was higher than rockfish price. But internal rate of return(IRR) and net present value(NPV) were high in the olive flounder and rockfish polyculture or the rockfish taking turns polyculture. The reasons were alttributed to their high utility of a netcage and to reduction of a capital cost. So, I suggest 1) to feed the bigger product in short term, 2) to reduction of a feed and seed cost, 3) to increase a survival rate of fish and 4) to enlarge production by highly utilizing a netcage. And I recognized that a aquaculture manager should make constantly an effort to gain more profit by rearing more good products and by reducing a production cost.
Seed production of olive flounder Paralichthys olivaceus was performed in a pilot RAS. The growth of juvenile olive flounder and changes in water quality were monitored for the entire production period. The pilot RAS consisted of 8 circular culture tanks($4.0mD{\times}1.0mH$), 2 trickling biofilters($1.7mD{\times}2.0mH$), 2 protein skimmers ($0.8mD{\times}2.5mH$), and 4 sedimentation chambers($0.7mD{\times}1.5mH$). The culture surface area was about $100.5m^2$ and the actual working volume was about $106.9m^3$. As many as 300,000 fertilized olive flounder eggs were initially distributed into 2 culture tanks with the water temperature at $19.0^{\circ}C$. Live feeds such as rotifers and Artemia nauplii were fed until the 32nd day after hatching, and a commercial diet was fed from the 19th day to the end of the experiment. After 70 days, 150,256 juveniles with a body length of $65.8{\pm}3.9mm$ were produced in the RAS, with a daily growth rate for body length of 4.7%/day. At this time, the final culture density was 1,495 individuals $m^{-2}$, and 13.6 L of makeup water, 0.071 kW of electricity and 0.025 L of diesel fuel were used to produce a juvenile olive flounder. During metamorphosis of the larvae, the TAN concentration increased to 0.99 mg/L, which made the larvae sensitive to result in some mortality. However no more massive mortality occurred at the juvenile stage after metamorphosis even at a TAN concentration of 4.25 mg/L and a ${NO_2}^{-}-N$ concentration of 2.45 mg/L.
Hybrid fish were induced between olive flounder (Paralichthys olivaceus) female and spotted flounder (Verasper variegatus) male by artificial inseminations. Survival, growth and morphological changes in larval stage of hybirds were examined and compared to their parental species. Survival and growth up to 25 days after hatching of the hybrids were more similar to those of olive flounder than to those of spotted flounder (P<0.05). The ratio of yolk length to yolk width in hybrids was similar to that of female parents (P<0.05). The oil globule were resorbed within 18 days after hatching in hybrids and 14 days after hatching in olive flounder, and spotted flounder has no oil globule. These results indicated that survival, growth and morphological changes in larval stage of hybrids were similar to those of maternal olive flounder.
We determined the optimal culture conditions for obtaining the maximum number of intestinal bacteria from the olive flounder Paralichthys olivaceus, and studied bacterial diversity using both culture-dependent and culture-independent methods. Using six culture conditions, mean bacterial numbers were greater than $10^6$ per gram of gut mucus, regardless of the medium. However, the bacterial diversity, based on colony morphology, appeared much higher on Marine agar (MA) and Zobell 2216 agar than on other media. We found eight and 17 cultured bacterial phylotypes with 99% minimum similarity in gut mucus grown on MA and tryptic soy agar, respectively. Furthermore, we used genomic DNA extracted from gut mucus to generate 78 random clones, which were grouped into 25 phylotypes. Of these, six were affiliated with Firmicutes, Actinobacteria, and Verrucomicrobia, and were not found using our culture-dependent methods. Consequently, we believe that Marine agar and Zobell 2216 agar are optimal media for culturing diverse intestinal microbes; we also discovered several novel sequences not previously recognized as part of the gut microbiota of olive flounder.

In this study, we monitored the status of marine environment of olive flounder, Paralichythys olivaceus, culture ground in Jeju-do, Republic of Korea. It reported the consumption of raw olive flounder meat containing of Kudoa, Kudoa septempunctata, could induce vomitting and diarrhea in Japan. The Kudoa is a new mycosporean species, researchers found Kudoa from the muscles of olive flounder cultured at western Japan and imported from Jeju-do. We choose two Kudoa positive farms and two negative farms in the mid of concern about the relation between Kudoa and habitat. We found two marine invasive species at the water-pumping seabed at one of the Kudoa positive farm. The concentration of pH and DO at on-growing flounder farms showed the decreasing pattern along side the raceway (influent sea waters > on growing sea waters > outfluent sea waters). The TN and TP values increased gradually following to the raceway (influent sea waters < on growing sea waters < outfluent sea waters). The concentration of COD and SS were in the range of $0.100-2.581mg{\cdot}L^{-1}$, $1.00-12.70mg{\cdot}L^{-1}$, respectively. The calculated residence time was 4hr 32minutes at F1, 11hr 21minutes at F2, and 9hr 50minutes at F3, respectively. It was calculated same distance of 4 km away from effluent pipes. Although direct relation between Kudoa and marine environment could not define well based on this study result, the more studies on marine environmental stressors for olive flounder are required to conduct as a reliable method including socio-economic group and environmental group.
A nationwide survey was conducted to study the rate of Kudoa septempunctata parasitization in cultured olive flounder, Paralichthys olivaceus, in Korea in 2013. Of 1107 olive flounder sampled randomly from 89 culture farms in five different regions in Korea, K. septempunctata was detected only in Jeju, where 10 of 318 fish (3.14%) were PCR positive, with genomic concentrations of $4.67{\times}10^5$ to $1.48{\times}10^{11}$ rDNA copies/g by real-time PCR. Of the ten Kudoapositive fish, K. septempunctata spores with 5-7 polar capsules were detected only in four fish. No samples of K. septempunctata were detected in olive flounder from the other regions surveyed. Furthermore, K. septempunctata was not detected in 326 samples of olive flounder seeds sampled from 39 hatcheries in seven different regions in Korea. Therefore, the parasite infection is restricted to Jeju and K. septempunctata infection is not spread from hatcheries.

To successful molecular breeding, identification and functional characterization of breeding related genes and development of molecular breeding techniques using DNA markers are essential. Although the development of a useful marker is difficult in the aspect of time, cost and effort, many markers are being developed to be used in molecular breeding and developed markers have been used in many fields. Single nucleotide polymorphisms (SNPs) markers were widely used for genomic research and breeding, but has hardly been validated for screening functional genes in olive flounder. We identified single nucleotide polymorphisms (SNPs) from expressed sequence tag (EST) database in olive flounder; out of a total 4,327 ESTs, 693 contigs and 514 SNPs were detected in total EST, and these substitutions include 297 transitions and 217 transversions. As a result, 144 SNP markers were developed on the basis of 514 SNP to selection of useful gene region, and then applied to each of eight wild and culture olive flounder (total 16 samples). In our experimental result, only 32 markers had detected polymorphism in sample, also identified 21 transitions and 11 transversions, whereas indel was not detected in polymorphic SNPs. Heterozygosity of wild and cultured olive flounder using the 32 SNP markers is 0.34 and 0.29, respectively. In conclusion, we identified SNP and polymorphism in olive flounder using newly designed marker, it supports that developed markers are suitable for SNP detection and diversity analysis in olive flounder. The outcome of this study can be basic data for researches for immunity gene and characteristic with SNP.

Olive flounder and Black rockfish comprises around 90% of the total fish culture in Korea. Most of the olive flounder is consumed as raw fish meat “saengseonhoe”, as like a sashimi in Japan. Also, texture is particularly a important factor which determine the quality of raw fish meat in Korea. Viral hemorrhagic septicemia (VHS) is one of the most serious viral diseases affecting farmed olive flounder (Paralichthys olivaceus) in Asian countries. VHS, caused by viral hemorrhagic septicemia virus (VHSV), occurs in over 80 different cultured and wild fish species worldwide. Our previous study demonstrated that VHSV infection can be restricted by adjusting the water temperature to over 17℃ from the host optima. We confirmed that the effective VHSV immersion vaccine treatment was a tissue culture infection dose (TCID) of 105.5 TCID50/mL at 17℃. However, the safety of live VHSV immersion vaccines remains unclear. The objectives of this study were to 1) demonstrate the safety of the live VHSV immersion vaccine under co-habitant conditions and 2) estimate the pathogenicity of VHSV in live VHSV-vaccinated flounder at 10℃. No mortality was observed in olive flounder treated with the live VHSV immersion vaccine, and the vaccinated flounder challenged with VHSV did not transfer VHSV to naïve fish at 10℃ through cohabitation. VHSV titration was below the detection limit (< 1.3 log TCID50/mL) in live VHSV immersion vaccine-treated flounder challenged with VHSV at 10℃. This study demonstrated that flounder treated with the live VHSV immersion vaccine were resistant to VHSV infection, and the live vaccine was also safe for naïve fish even at a water temperature known to be VHS infectious.
This study was conducted to evaluate the dietary supplementation of citrus byproduct (CBP) on the growth performance, feed utilization, and innate immune responses of juvenile olive flounder Paralichthys olivaceus under low water temperatures (11-15℃). Dietary L-ascorbyl-2-polyphosphate was replaced with graded CBP levels at 0 (Con), 25 (CBP25), 50 (CBP50), 75 (CBP75), and 100% (CBP100). Triplicate groups of juvenile olive flounder were handfed with one of the diets twice a day for 42 days. The growth performance and feed utilization of fish fed with diet containing levels of CBP75 or CBP100 increased significantly compared to those of fish with fed Con. Dietary CBP supplementation increased the protein efficiency ratio in fish. There was no significant differences in innate immune responses between groups, even though CBP supplementation tended to increase. These findings indicate that CBP could be used as a vitamin C source and improve the growth performance of juvenile olive flounder under low water temperatures.
