화장품에 배합되는 원료 중에 피부 모공을 막아 여드름의 초기단계인 면포를 유발함으로써 염증성 여드름으로 악화시키는 사례가 보고되고 있다. 본 연구는 '여드름 피부에 사용이 적합한' 면포 비유발성 화장품의 객관적인 평가 기준을 마련하고자 하였다. 외국 임상기관에서 실시하고 있는 non-comedogenic test를 조사하고 등 상부 반복 폐쇄 첩포를 통해 면포 유발 평가방법을 확립하고자 연구를 수행하였다. 또한 안면부 사용성 시험을 동일 피험자에게 추가로 진행하였다. 등 상부 반복 폐쇄 첩포를 통해 면포를 채취하여 분석한 결과 시험시료로 사용한 보습제와 자외선 차단제가 면포를 유발하지 않음을 확인할 수 있었고, 동일한 시험제품을 안면부에 사용하여 얻은 면포 유발 결과와 Global acne grading system (GAGS)의 여드름 육안평가 결과는 상관관계가 없었다. 또한 사진 판독을 통해 분석하는 면포와 모낭의 구별을 용이하게 하기 위해 면포 채취 표본에 Oil red O staining을 실시하였다. 염색을 한 경우의 표본이 염색을 하지 않은 표본과 비교하여 결과 간 높은 일치도를 확인하였다. 본 연구에서는 Oil red O staining을 통해 객관성과 신뢰성을 증진시킨 새로운 버전의 화장품 면포 비유발성 평가법을 확립하였다.
본 연구에서는 제주도에서 자생하는 30여 종의 식물추출물에 대한 항비만 효능을 조사하였다. 항비만 효능은 in vitro oil red-O staining 방법을 이용하여 지방전구세포 3T3-L1에서 분화억제력을 측정하였다. 그 결과, 30여 종의 식물 중에서 약도라지, 호장근, 유근피 등을 포함한 6종의 식물이 지방전구세포 3T3-L1의 분화를 억제하였다. 지방전구세포의 분화를 억제하는 물질 중 유근피, 약쑥, 호장근, 후박 4종은 우수한 항산화 효능도 동시에 가지고 있음을 확인하였다. 이러한 결과를 통해, 본 실험에서 확보된 추출물이 항비만 물질로 사용될 수 있는 가능성을 확인하였다.
Background: Popeye deformity is common after rupture of the biceps muscle's long head tendon. Herein, we report on histological changes in biceps brachii muscles following tenotomy of the long head biceps tendon. Methods: Twelve Sprague-Dawley rats (12-week-old) underwent tenotomy of the long head biceps tendon in the right shoulder. At postoperative weeks 4, 7, and 10, the operative shoulders were removed by detaching the biceps brachii muscle from the glenoid scapula and humerus; the opposite shoulders were removed as controls. H&E staining was performed to elucidate histological changes in myocytes. Oil-red O staining was performed to determine fatty infiltration. Myostatin antibody immunohistochemistry staining was performed as myostatin is expressed by skeletal muscle cells during myogenesis. Results: H&E staining results revealed no changes in muscle cell nuclei. There were no adipocytes detected. Compared with that of the control biceps, the cross-sectional area of the long head biceps was significantly smaller (p=0.00). Statistical changes in the total extent of the 100 muscle cells were significant (p=0.00). Oil-red O staining revealed no fatty infiltration. Myostatin antibody immunohistochemical staining revealed no significant difference between the two sides. Conclusions: Muscular changes after tenotomy of the long head biceps included a decrease in the size of the individual muscle cells and in relative muscle mass. There were no changes observed in muscle cell nuclei and no fatty infiltration. Moreover, there were no changes detected by myostatin antibody immunohistochemistry assay.
The synovial tissues are a valuable MSCs source for cartilage tissue engineering because these cells are easily obtainable by the intra-articular biopsy during diagnosis. In this study, we isolated and characterized the canine MSCs derived from synovial fluid of female and male donors. Synovial fluid was flushed with saline solution from pre and post-puberty male (cM1-sMSC and cM2-sMSC) and female (cF1-sMSC and cF2-sMSC) dogs, and cells were isolated and cultured in advanced-DMEM (A-DMEM) supplemented with 10% FBS in a humidified 5% $CO_2$ atmosphere at $38.5^{\circ}C$. The cells were evaluated for the expression of the early transcriptional factors, such as Oct3/4, Nanog and Sox2 by RT-PCR. The cells were induced under conditions conductive for adipogenic, osteogenic, and chondrogenic lineages, then evaluated by specific staining (Oil red O, von Kossa, and Alcian Blue staining, respectively) and analyzed for lineage specific markers by RT-PCR. All cell types were positive for alkaline phosphatase (AP) activity and early transcriptional factors (Oct3/4 and Sox2) were also positively detected. However, Nanog were not positively detected in all cells. Further, these MSCs were observed to differentiate into mesenchymal lineages, such as adipocytes (Oil red O staining), osteocytes (von Kossa staining), and chondrocytes (Alcian Blue staining) by cell specific staining. Lineage-specific genes (osteocyte; osteonectin and Runx2, adipocytes; PRAR-${\gamma}2$, FABP and LEP, and chondrocytes; collagen type-2 and Sox9) were also detected in all cells. In this study, we successfully established synovial fluid derived mesenchymal stem cells from female and male dogs, and determined their basic biological properties and differentiation ability. These results suggested that synovial fluid is a valuable stem cell source for cartilage regeneration therapy, and it is easily accessible from osteoarthritic knee.
본 연구에서는 3T3-L1 전지방세포를 이용하여 발효 삼정환의 지방 분화 억제 효과를 확인하고자 하였다. 삼정환을 5가지 주요 유산균으로 각각 발효시킨 후, Oil Red O 염색법으로 지방세포 분화 억제 여부를 알아보고, 이의 기전을 알기 위해서 지방생성에 핵심적인 역할을 하는 transcription factor 및 지질 조절 효소의 유전자 발현량을 비교하여 보았다. Oil Red O 염색 분석에서는 Lactobacillus plantarum, Enterococcus faecium으로 발효한 삼정환에서 지방구 수의 감소를 확인할 수 있었다. 또한 transcription factor로 $C/EPB{\alpha}$, $PPAR{\gamma}$, UCP-2 및 콜레스테롤 조절과 관련된 효소인 HMG-CoA reductase의 4가지 유전자 발현 정도를 분석하였는데, 발효시키지 않은 삼정환의 경우 농도가 $100{\mu}L/mL$ 이상일 때 네 가지 항목에서 억제 효과를 나타냈으며, 발효시켰을 때는 유산균의 종류에 따라 효과 억제효과가 있었는데, Lactobacillus plantarum으로 발효한 삼정환이 효과가 가장 좋은 것으로 나타났다. Lactobacillus plantarum으로 발효한 삼정환은 동일 농도의 발효하지 않은 삼정환과 비교했을 때, Oil Red O 실험에서 염색된 지방구 수가 더욱 감소하였으며, $PPAR{\gamma}$, UCP-2, HMG-CoA reductase의 경우 농도가 $200{\mu}L/mL$일 때 각각 35%, 57%, 54%로 5가지 발효 삼정환 중 가장 높은 억제율을 나타내었다. 이에 지방분화 억제 효과를 가진 삼정환을 발효시키면 그 효과가 더욱 증가하는 것으로 나타났는데, 특히 Lactobacillus plantarum으로 발효한 삼정환은 $PPAR{\gamma}$, UCP-2, HMG-CoA reductase의 유전자 발현조절을 통해 지방 분화를 억제시키는 것으로 밝혀져 발효 삼정환은 항비만 혹은 항고지혈증 약물로 개발 가능성이 있는 것으로 사료된다.
Water-soluble fraction (WSF) from edible mushroom hinmogi (Tremella fuciformis) were obtained by water extraction, and polysaccharides in the WSF were separated by ethanol precipitation. The inhibitory effects of the polysaccharides on 3T3-L1 adipocyte differentiation were evaluated by the reduction of peroxisome proliferators-activated receptor ${\gamma}$ ($PPAR{\gamma}$) translation, triglyceride accumulation, Oil Red-O staining, and expression levels of $PPAR{\gamma}$, CCAAT/enhancer binding protein a (C/$EBP{\alpha}$), and leptin. The $PPAR{\gamma}$ translation in 3T3-L1 cells was inhibited by the treatment with polysaccharide precipitated by 80% ethanol (P80) which showed highest inhibitory activity among polysaccharides tested. In addition, treatment of P80 to 3T3-L1 cells significantly inhibited the triglyceride accumulation, Oil Red-O staining, and mRNA expression of $PPAR{\gamma}$, C/$EBP{\alpha}$, and leptin in a dose-dependent manner. Based upon these results, P80 from edible mushroom hinmogi shows the inhibitory activity on the differentiation of 3T3-L1 adipocytes. Therefore, it might be employed as a potential anti-obesity material.
The pathological features of a mass in the back skin region of an 8-year-old castrated male dog are described herein. The cut section of the tumor was white to tan with a soft multilobulated mass containing hemorrhagic and necrotic foci and a mucinous-like composition. Microscopically, the tumor was composed of a mixture of lipocytes, lipoblasts, spindle cells and stellate cells and had a myxoid background. Oil red O staining revealed that the cytoplasm of neoplastic cells contained large numbers of lipid droplets. Immunohistochemically, tumor cells were positive for vimentin and S-100 protein. The skin mass was diagnosed as myxoid liposarcoma.
BACKGROUND/OBJECTIVES: Psidium guajava L. (guava) leaves have been shown to exhibit hypoglycemic and antidiabetic effects in rodents. This study investigated the effects of guava leaf extract on adipogenesis, glucose uptake, and lipolysis of adipocytes to examine whether the antidiabetic properties are mediated through direct effects on adipocytes. MATERIALS/METHODS: 3T3-L1 cells were treated with 25, 50, 100 ㎍/mL of methanol extract from guava leaf extract (GLE) or 0.1% dimethyl sulfoxide as a control. Lipid accumulation was evaluated with Oil Red O Staining and AdipoRed assay. Immunoblotting was performed to measure the expression of adipogenic transcription factors, fatty acid synthase (FAS), and AMP-activated protein kinase (AMPK). Glucose uptake under basal or insulin-stimulated condition was measured using a glucose analog 2-[N-(7-nitrobenz-2-oxa-1,3-diazol-4-yl) amino]-2-deoxy-D-glucose. Lipolysis from fully differentiated adipocytes was measured by free fatty acids release into the culture medium in the presence or absence of epinephrine. RESULTS: Oil Red O staining and AdipoRed assay have shown that GLE treatment reduced lipid accumulation during adipocyte differentiation. Mitotic clonal expansion, an early essential event for adipocyte differentiation, was inhibited by GLE treatment. GLE inhibited the expression of transcription factors involved in adipocyte differentiation, such as peroxisome proliferator-activated receptor 𝛄 (PPAR𝛄), CCAAT/enhancer-binding protein α (C/EBPα), and sterol regulatory element-binding protein-1c (SREBP-1c). FAS expression was also decreased while the phosphorylation of AMPK was increased by GLE treatment. In addition, GLE increased insulin-induced glucose uptake into adipocytes. In lipid-filled mature adipocytes, GLE enhanced epinephrine-induced lipolysis but reduced basal lipolysis dose-dependently. CONCLUSIONS: The results show that GLE inhibits adipogenesis and improves adipocyte function by reducing basal lipolysis and increasing insulin-stimulated glucose uptake in adipocytes, which can be partly associated with antidiabetic effects of guava leaves.
An adult female ringed seal died suddenly and was subsequently examined for diagnostic purposes. The animal's lungs demonstrated mild non-collapsibility and multifocal white to yellow patches. Histopathological examination revealed multifocal pulmonary histiocytosis. Alveoli were filled with numerous foamy macrophages cytoplasm and scattered multinucleated giant cells containing cholesterol clefts. The foamy cytoplasm of the macrophages stained with oil red O stain. Further, lipid droplets within the cytoplasm were detected by electron microscopy. To the author's knowledge, this is the first case report describing the histochemical staining and electron microscopic findings associated with endogenous lipid pneumonia in ringed seal.
