Production of Knockout Mice using CRISPR/Cas9 in FVB Strain

  • KO mice provide an excellent tool to determine roles of specific genes in biomedical filed. Traditionally, knockout mice were generated by homologous recombination in embryonic stem cells. Recently, engineered nucleases, such as zinc finger nuclease, transcription activator-like effector nuclease and clustered regularly interspaced short palindromic repeats (CRISPR), were used to produce knockout mice. This new technology is useful because of high efficiency and ability to generate biallelic mutation in founder mice. Until now, most of knockout mice produced using engineered nucleases were C57BL/6 strain. In the present study we used CRISPR-Cas9 system to generate knockout mice in FVB strain. We designed and synthesized single guide RNA (sgRNA) of CRISPR system for targeting gene, Abtb2. Mouse zygote were obtained from superovulated FVB female mice at 8-10 weeks of age. The sgRNA was injected into pronuclear of the mouse zygote with recombinant Cas9 protein. The microinjected zygotes were cultured for an additional day and only cleaved embryos were selected. The selected embryos were surgically transferred to oviduct of surrogate mother and offsprings were obtained. Genomic DNA were isolated from the offsprings and the target sequence was amplified using PCR. In T7E1 assay, 46.7% among the offsprings were founded as mutants. The PCR products were purified and sequences were analyzed. Most of the mutations were founded as deletion of few sequences at the target site, however, not identical among the each offspring. In conclusion, we found that CRISPR system is very efficient to generate knockout mice in FVB strain.

Effects of Di(n-butyl) Phthalate on the Developing Immune System of Fetal and Neonatal SD Rats (Di(n-butyl) Phthalate가 태자와 신생자 SD Rat의 면역계 발생에 미치는 영향)

  • Some of endocrine disruptors with sexual hormone-like effects have been increasingly reported to be immunotoxic in many species in recent several years. Phthalate esters have possible effects on the endocrine system. Prenatal exposure to di(n-butyl) phthalate (DBP) has been reported to impair the androgen-dependent development of the male reproductive tract in rat. Therefore, the immunomodulatory effect of DBP was investigated in the developing immune system of fetal and neonatal Sprague-Dawley rats. Timed-bred pregnant SD rats were given to the doses of 0, 250, 500, and 750 mg DBP/kg$\cdot$ body weight /day by gavage once a day from gestational day (GD) 5 to 18. On GD19 or GD22/postnatal day one (PD1), the dams were euthanized, and the changes in organ weights and thymus phenotypes were examined for their offsprings. At 750 mg DBP/kg$\cdot$b.w./day in maternal exposure group, GD19 fetuses showed decreases in body weight. The spleen/body weight ratios were reduced in GD 19 fetuses from the dams exposed to 500 and 750 mg DBP/kg$\cdot$b.w./day. There were no significant changes in thymus and spleen cellularities though these cellularities showed a tendency to decrease in a dose dependent way. In the DBP-exsposed GD22/PD1 offsprings, the body weights, the relative organ weights and the cellularities did not exhibit alteration. Additionally, the percentages of CD3$^{+}$(CD4$^{+}$CD8$^{+}$, CD4$^{+}$CD8$^{-}$, CD4$^{-}$CD8$^{+}$, and CD4$^{-}$CD8$^{-}$) and CD3$^{-}$(CD4$^{+}$CD8$^{+}$, CD4$^{+}$CD8$^{-}$, CD4$^{-}$CD8$^{+}$, and CD4$^{-}$CD8$^{-}$) thymocyte subsets were not changed in any DBP-treated group. The proliferative responses of splenic T cells to Con A and B cells to LPS were decreased in all DBP-exposed GD22/PD1 offsprings.

Effect of Maternal Dietary Restriction and Rehabilitation on the Muscle Protein Breakdown of Young Rats (어미 쥐의 식이제한과 식이회복이 새끼쥐의 근육단백질 분해에 미치는 영향)

  • A quantitative restriction of maternal diet was given to the Sprague Dawley rats during the lactation. The control group were fed a commercial diet ad libitum throughout the experimental period. Dietary restriction started from birth to weaning in deficient group. After weaning at 21 days, all offsprings were fed the same diet ad libitum for 4 weeks of rehabilitation. They were analyzed for body weight, muscle protein, muscle 3-methylhistidine and serum protein level of offsprings at 0, 1, 2, 3, 5, 7 weeks. Body weight of offsprings of deficient group were significantly lower than the control group, but after rehabilitation there were no significant differences between two groups. Maternal dietary restriction caused a decrease in the 3-methylhistidine, protein content in muscle and total protein level in serum, and rehabilitation after weaning induced a marked increase. These results suggest that there is a quantitative reduction in 3-methylhistidine metabolism in the undernourished rats and these reductions can be nearly recovered by 4 weeks of dietary rehabilitation after weaning.

The frequency of chromosomal abnormalities and the prenatal cytogenetic analyses for couples with recurrent abortions

  • Between 1988-1998, cytogenetic analyses were performed for 1,476 couples and 162 women with recurrent abortions. We applied GTG-banding, high resolution-banding and FISH (fluorescent in situ hybridization) techniques in this study. The frequency of balanced translocations was 3.6% (112/3114). Of them, 74 cases (2.38%) were reciprocal translocations and 38 (1.22%) were robertsonian translocations. Chromosome aberrations were more frequent in women (80 cases) than in men (32 cases). No phenotypical abnormalities were found in all carriers who had experienced recurrent spontaneous abortions or experienced giving birth to malformed offsprings. Prenatal cytogenetic analyses were carried out on 40 subsequent pregnancies for carrier couples with balanced translocation. The fetal karyotypes showed that 13 cases (32.5%) were normal, 25 (62.5%) were balanced translocations, and two (6%) were unbalanced translocations. It is believed that the frequency of chromosomal abnormalities in patients with recurrent spontaneous abortion is higher than that of the normal population. Most of the fetal samples showed normal karyotypes or balanced translocations matching that of one of their parents. Although the incidence of chromosomal imbalance in the fetuses was relatively low in prenatal cytogenetic analysis, individuals with balanced translocations are predisposed to giving birth to malformed offsprings with partial trisomy or monosomy. Therefore, we recommend the cytogenetic and the prenatal cytogenetic analysis for those who experiences recurrent abortion as well as in case they become pregnant, to prevent the birth of offsprings with chromosomal abnormalities.

The Study of Intergenerational Support Exchange between Older parents and Adult Children (노년기 부모-성인자녀간 지원유형에 관한 연구)

  • The authors aimed to identify the patterns of mutual support between aged parents and their adult offsprings and to analyze the demographic and social characteristics of each pattern and the quality of living for the aged parents. The data of the study were drawn from the 2008 Survey of the Living Conditions and Welfare for the Elderly published by the Ministry of the Health and Welfare. The sample of our analysis was 14,843 aged persons with their adult offsprings. A cluster analysis produced four types of mutual support patterns: High mutual supporters, parents as beneficiaries, parents as benefactors, ans low mutual supporters, with parents as benefactors as the most frequent pattern of mutual support. The study showed that the group of high mutual supporters was highest in the quality of life while the group of parents as beneficiaries lowest. This findings imply that the support relationship between old people and their adult offsprings may undergo transformation towards a two-way relation of mutual support.

Studies on the Reproductive Toxicant and Blood Metabolite in Pups Born After Bisphenol A Administration in Pregnant Mice (임신중인 생쥐에 Bisphenol A 투여 후 태어난 차산자의 생식독성과 혈액성분에 관한 연구)

  • Bisphenol A (BPA) is a monomer used in the manufacture of a multitude of chemical products, including epoxy resins and polycarbonate. The objective of this study was to evaluate the effects of BPA administration on reproductive characteristics and blood hematological and chemical values in offspring of pregnant dams treated with BPA. BPA was administrated to pregnant mice by intraperitoneally injection with 0, 0.05, 0.5 and 5.0 mg/kg B.W. for 5 times at 3 days interval on gestation days 1-16. There were no treatment-related effects of BPA on reproductive organ weight in male offsprings at 45 days-of-age, but body weight was the lowest in 5.0mg BPA group when compared to other groups (P<0.05). No differences in semen characteristics (sperm concentration, viability, motility and abnormality) were observed between the control and BPA treatment groups. The WBC, HB, HT, MCV, MCH, MCHC, albumin, BUN and total protein of blood hematological and chemical values in male offsprings were not difference for any treatment groups, but RBC value in BPA groups was significantly increased comparing to the control group (P<0.05). The PLT value was slightly higher in 5.0mg BPA groups than in any other group, but not significantly difference among the experimental groups. In female offsprings, the effects of BPA didn't affect to the body and ovary weight, but the uterus weight in 5.0mg BPA group was slightly heavier than that of control group (P>0.05). No statically significant difference in blood hematological values in female offsprings were observed between the control group and BPA groups, but the concentration of albumin and BUN were significantly higher in 0.5mg BPA group when compared to control and other BPA treatment groups (P<0.05). The histological evaluation of testis and ovary in growing offspring at 45 days-of-age was not difference between the control group and BPA groups, but endometriosis of the uterus in female offspring was dramatically increased in 0.5 and 5.0mg BPA groups. These founding suggest that low concentration of BPA might not have a important role on reproductive ability or blood metabolite in offspring of pregnant dams treated with BPA.

Pyridoxine Deficiency on Neurotransmitters in the Developing Rat Brain - Catecholamine Metabolism- (Pyridoxine결핍이 뇌의 신경전달물질에 미치는 영향 - Catecholamine 대사 -)

  • Pregnant rats were fed a pyridoxine deficient diet during the gestation and lactation. DEF I group received the deficient diet from delivery ; DEF II group, from the 15 th day of gestation. Body and brain weights, brain protein, DNA, RNA, plasma GOT and GPT, and catecholamines were measured. Effect of MAO inhibiting drug, pargyline, was determined. Brain protein, DNA, and RNA of offsprings of deficient groups were significantly lower than the control group, but RNA/ DNA, brain weight/DNA, and protein/DNA show that cell number were more affected than cell size by the pyridoxine deficiency during the 3rd week of gestation and lactation. Plasma GOT activities were more significantly different than plasma GPT between the control and deficient group. Brain norepinephrine of offsprings of deficient group were significantly lower than the control, but brain dopamine content was not significantly different from the control. At 2nd and 3rd week, norepinephrine was significantly depressed in deficient groups. Pargyline treatment affected a 1.2 fold increase in catecholamines in 3hr while the control had a 1.5 fold increase. Thus norepinephrine and dopamine synthesis was depressed in the deficient groups. Dopaminergic neurons may be less dependent on pyridoxine level than neurons from norepinephrine. Pyridoxine deficiency in maternal diet is not so critical to brain catecholamines of offspring except to the neonatal rats.

Jindo dogs-Korean native dogs : A historical review (진도견(珍島犬)- 우리나라의 고유견(固有犬))

  • The origin of Jindo dogs has been controversial. It has been informed by words of mouth that they are the offsprings of either the digs of Sung dynasty (960~1279 A.D.) in China or the Mongolian dogs. Some breeds of dogs in Japan now have almost same appearance as Jindo dogs. The authors reviewed the history of Japan and found that Korean hunting dogs were sent to Japan in early 5th century, which was before Sung dynasty of China. Dr. Jae-Keun Ryu of National Institute of Environmental Research has done some work on serums of dogs of both Korea and Japan; the results of his serological and molecular biological studies indicated that Korean dogs were sent to Japan 1,300~2,500 years ago. Jindo dogs appear to be the descendants of the dogs of the Stone Age. They have adapted themselves to the unfavorable environment of Jindo island so that they can maintain pure line-age and wild nature until today. It was concluded that they are Korean native dogs, not the offsprings of the dogs of Sung dynasty or the Mongolian dogs.

A Teratogenicity Study on Original Woo-Whang-Chung-Sim-Won in Rats (원방우황청심원의 랫드 최기형성에 관한 연구)

  • A teratogenic study on Original Woo-Whang-Chung-Sim-Won was carried out in SpragueDawley rats. Original Woo-Whang-Chung-Sim-Won suspended in distilled water was administered to pregnant dams by oral gavage during organogenesis period (from 7th to 17th day of gestation) at daily doses of 1/9, 1/3 and I pill/kg. About two-thirds of dams were sacrificed at 20th day of gestation to scrutinize the pregnant performances and fetal development, and the remaining dams were allowed to deliver. The growth, reflex, behaviour and reproductive function of F1 offsprings were examined. There was no treatment-related difference in body weight, food consumption and necropy findings of dams. No gross, skeletal and visceral abnormalities was observed in F1 fetuses from dams treated with Original Woo-Whang-Chung-Sim-Won. F1 offsprings did not show any treatment-related difference in growth, reflex, behaviour and reproductive peuformance. At caesarean section of F1 dams, no growth retardation and gross abnormality was observed in F2 fetuses. In conclusion, Original Woo-Whang-Chung-Sim-Won did not show any potential teratogenic activity in rats.

  • The purpose of this thesis is to study an effect for the relation between internet addiction of adolescent and family & school background in information society, and for this study I did a factor analysis. This study surveyed with questionnaire, and analyzed 666 valid samples through SPSS program. As a result of factor analysis, in family background, ‘educational method of parents is a democratic’, ‘concerning of parents and offsprings’ and ‘intimacy of parents and of offsprings’ have main factors of family background for internet addiction of adolescent. In school background, ‘intimacy of teachers and students’, ‘satisfaction of school's educational system’, and ‘relationship among the friends in school’have main factors of school background of internet addiction of adolescent.
