The results of our observation in May 2000 indicated that the SeaWiFS algorithm (O'Reilley et al., 1998), which was adopted for OSMI data processing, overestimated the actual chlorophyll values. This was rather unexpected in that there were good reasons to expect that the bio-optical properties of East/Japan Sea belonged to Case 1 water and in such case, the OC2 algorithm would give unbiased estimates of actual chlorophyll a values. In November 2000, a cruise conducted bio-optical surveys in the same area. This time we added HPLC (High Performance Liquid Chromatography) method for measuring chlorophyll a concentration to the standard fluorometric method, which we hale been using during the past Fluorometric method with acidification is known to result in under/overestimation of chlorophyll values in many parts of the world oceans, while it is easier and cheaper than HPLC method. To our surprise, the comparison of HPLC chlorophyll and fluorometric chlorophyll values show that fluorometric values gave an underestimation up to 50%. This error was due to the presence of accessory pigments such as chlorophyll b. Considering this error, our precious result of May 2000(Yoo et al., 2000) might have to be reinterpreted. Calculation of reflectance at 490 and 555nm, however, indicated that this is not still enough to explain the discrepancies.

Carbon dioxide capture and storage (CCS) technology is recognizing one of method responding the climate change with reduction of carbon dioxide in atmosphere. In Korea, due to its geological characteristics, sub-seabed geological $CO_2$ storage is regarded as more practical approach than on-land storage under the goal of its deployment. However, concerns on potential $CO_2$ leakage and relevant acidification issue in the marine environment can be an important subject in recently increasing sub-seabed geological $CO_2$ storage sites. In the present study effect data from literatures were collected in order to conduct an effect assessment of elevated $CO_2$ levels in marine environments using a species sensitivity distribution (SSD) various marine organisms such as microbe, crustacean, echinoderm, mollusc and fish. Results from literatures using domestic species were compared to those from foreign literatures to evaluate the reliability of the effect levels of each biological group and end-point. Ecological effect guidelines through estimating level of pH variation (${\delta}pH$) to adversely affect 5 and 50% of tested organisms, HC5 and HC50, were determined using SSD of marine organisms exposed to the $CO_2$-induced acidification. Estimated HC5 as ${\delta}pH$ of 0.137 can be used as only interim quality guideline possibly with adequate assessment factor. In the future, the current interim guideline as HC5 of ${\delta}pH$ in this study will look forward to compensate with supplement of ecotoxicological data reflecting various trophic levels and indigenous species.
The coral symbiotic algae zooxanthellae is often expelled from the host as the host coral is under physiological stress, causing the coral to turn completely white. Such coral bleaching events are occurring more frequently with the increase in the global warming, ocean acidification and increased level of anthropogenic impacts such as eutrophication. In the present study, we investigated the effects of inorganic nutrients including ammonium, nitrate, phosphate and elevated water temperature on the symbiotic zooxanthellae density in the fragment of branching coral Acropora nobilis. Zooxanthellae density in the host coral decreased 8 hrs after the experiment at a given elevated water temperature ($32^{\circ}C$, p < 0.05). In contrast, no clear coral bleaching or decrease in the symbiotic algae density was observed from the branching coral exposed to a normal water temperature of $30^{\circ}C$ and high levels of nutrients such as 20 ${\mu}M$ of $NH_4Cl$, 20 ${\mu}M$ of $NaNO_3$ and, 10 ${\mu}M$$KH_2PO_4$. Accordingly, the data indicated high water temperature is one of the stressful factors to cause bleaching in A. nobilis, whereas the high levels of nutrients is not a factor. It is believed that the results obtained in the present study are useful as baseline information in the management of the coral reefs.
The present study was conducted to find out effect of PFOS on survival, activity, growth and organ structure of the melania snail, Semisulcospira gottschei. Experimental groups were composed of one control condition and four PFOS exposure condition (0.1, 0.5, 1.0 and 5.0 mg/L). After 16 week exposure, survival rate and activity were not significantly influenced at the two lower exposure groups, 0.1 and 0.5 mg/L, but they were significantly reduced in 1.0 and 5.0 mg/L groups. Total weight and meat weight rate (MWR) were reduced in PFOS exposure group in comparison to control group. Also, histological degenerations such as acidification of mucous, necrosis and split of muscular fiber bundle, atrophy of anterior pedal gland were recognized in the foot. Hepatopancreas showed the atrophy and degeneration of the digestive cell, vacuolation of digestive gland and closure of lumen in digestive gland.
We tested the combined effects of increased partial pressure of $CO_2$ ($pCO_2$) and temperature on the reproduction and survival of the copepod Calanus sinicus from Asan Bay, the Yellow Sea under laboratory conditions to understand the impact of acidification on copepods. Egg production rate, survival rate, and fecal pellet production of C. sinicus were not affected by 1305 ppm $pCO_2$ or with combined treatments of temperature and $pCO_2$, including $8^{\circ}C$ and 289 ppm $pCO_2$ (ambient), $8^{\circ}C$ and 753 ppm $pCO_2$ (high $pCO_2$), $12^{\circ}C$ and 289 ppm $pCO_2$ (high temperature), and $12^{\circ}C$ and 753 ppm $pCO_2$ (greenhouse), for 5 or 10 d of exposure. However, egg hatching success of C. sinicus decreased significantly in the greenhouse treatment compared with the ambient or the high $pCO_2$ treatments. These results suggest that a combined treatment ($pCO_2$ and temperature) affected egg viability more than a single treatment($pCO_2$).
DNA damage such as genotoxicity was identified with comet assay, which blood cell of a marine parrot fish (Oplegnathus fasciatus) was exposed to an acidified seawater, lowered pH gradient making of $CO_2$ gas. The gradient of pH were 8.22, 8.03, 7.81, 7.55 with control as HBSS solution with pH 7.4. DNA tail moment of fish blood cell was $0.548{\pm}0.071$ exposed seawater of pH 8.22 condition, on the other hand, DNA tail moment $1.601{\pm}0.197$ exposed acidified seawater of pH 7.55 lowest condition. The approximate difference with level of DNA damage was 2.9 times between highest and lowest of pH. DNA damage with decreasing pH was significantly increased with DNA tail moment on blood cell of marine fish (ANOVA, p < 0.001). Ocean acidification, especially inducing the leakage of sequestered $CO_2$ in geological structure is a consequence from the burning of fossil fuels, and long term effects on marine habitats and organisms are not fully investigated. The physiological effects on adult fish species are even less known. This result shown that the potential of dissolved $CO_2$ in seawater was revealed to induce the toxic effect on genotoxicity such as DNA breakage.

The pH of water is one of the main environmental factors exerting selective pressure on marine and freshwater organisms. Here, we focus on the influence of pH on an organism's ability to maintain homeostasis and investigate the effects of acidification on immunity-related genes and osmotic pressure during early development of the olive flounder, Paralichthys olivaceus. The aim of our study was to determine the influence of various pH levels on the fertilized eggs and larvae of P. olivaceus. Gametes of P. olivaceus were artificially introduced and the resulting fertilized eggs were incubated at pH 4.0 (low), 6.0, and 8.0 (equivalent to natural sea water; control). We found that all eggs sank from the water column at pH 4.0. After 38 h, these eggs showed slow development. Hatching occurred more slowly at pH 4.0 and 6.0 and did not occur at all at pH 4.0. Result of gene expression, caspase and galectin-1 were expressed from the blastula to pre-hatch stages, with the exception of the two-cell stage. HSP 70 was also steadily expressed at all pH levels over the five days. The osmolality of fertilized eggs differed marginally at each stage and across pH levels. So, this results demonstrates that low pH level is detrimental to P. olivaceus fertilized eggs.
We examined changes in the physiological responses of gray mullet Mugil cephalus exposed to acidic seawater (pH 6.0, 6.5, 7.0) and normal seawater (pH 8.0, control) for 15 days. As pH decreased, survival rate and body weight also decreased. Levels of aminotransferase, total protein and triglycerides also differed significantly with changes in pH, presumably due to stress caused by exposure to acidic water. The level of osmotic pressure was significantly higher in the pH 6.0 group than in other groups. Superoxide dismutase was significantly higher in the pH 6.5 and 7.0 groups than in the pH 8.0 group, and glutathione level was lowest in the pH 6.0 group. We conclude that decreasing the pH level of seawater induces a stress response in fish, damaging their ability to control their hematological and osmotic pressure. Antioxidant enzymes are generally sensitive to osmotic stress; in this study, antioxidant activity significantly changed with pH level. These results indicate that physiological stress induced by exposure to acidification reduces survival rates and inhibits growth in M. cephalus.
We examined the effects of ocean acidification (OA) and eutrophication on the physiology of a red alga, Gracilariopsis chorda, using specimens collected at Wando Island, Korea, in July of 2015. The samples were transported to a laboratory and placed on growth media for treatments involving low or high levels of ammonium ($4{\mu}M$ or $60{\mu}M\;NH_4{^+}$) and low or high pH(7.5 or 8.2). The control treatment used filtered seawater (pH 8.2 and $4{\mu}M\;NH_4{^+}$). All experiments were conducted at $20^{\circ}C$ and under a lighting intensity of $80{\mu}mol\;photons\;m^{-2}\;s^{-1}$, with or without an injection of $CO_2$ (pH 7.5). In addition, we calculated rates of respiration under darkness, at a pH of 7.5 and $60{\mu}M\;NH_4{^+}$. Fluctuations in pH as well as the evolution of photosynthetic oxygen and $NH_4{^+}$ uptake rates were monitored for 6 h. The greatest increase in pH levels, from 7.50 to 8.65, occurred in response to $60{\mu}M\;NH_4{^+}$, whereas the largest decrease, from 7.50 to 7.42, was associated with elevated respiration rates. At a pH of 7.5, rates of oxygen evolution were higher (236% saturation) for samples treated with $60{\mu}M\;NH_4{^+}$ than for the control (121% saturation). Ammonium uptake was highest at pH 7.5 and $60{\mu}M\;NH_4{^+}$, with a rate of $0.526{\pm}0.002{\mu}mol\;g^{-1}\;FW\;h^{-1}$, followed in order by the treatments of $pH\;8.2/60{\mu}\;NH_4{^+}$, $pH\;7.5/4{\mu}M\;NH_4{^+}$, and the control ($pH\;8.2/4{\mu}M\;NH_4{^+}$). We speculated that the rates of photosynthesis and $NH_4{^+}$ uptake could be enhanced at a higher ammonium concentration and lower pH because $CO_2$ concentrations were increased through greater photosynthetic activity. Therefore, these findings suggest that the physiology of G. chorda populations can be improved by the interaction of optimized $CO_2$ concentrations and an adequate supply of essential nutrients such as ammonium.
Carbonic anhydrase (CA) is a key controller of the carbon concentrating mechanism (CCM), and is known to be affected by ambient pH and CO2 compositions. Herein, we characterized three novel CAs genes (PmCA1, 2, and 3) from the marine dinoflagellate Prorocentrum minimum, and evaluated the relative expressions of the PmCAs and photosynthetic genes PmatpB and PmrbcL under different pH conditions. Each PmCA was predicted to have amino acid residues constituting the zinc binding site. With signal peptide, PmCA1 and PmCA2 were predicted to be intracellular CAs located in the cytoplasm and chloroplast membrane, respectively. On the other hand, PmCA3 was predicted to be extracellular CA located in the plasma membrane. Also, PmCA1 was classified into the beta family, and PmCA2 and PmCA3 were classified into the alpha family via phylogenic analysis. The photosynthesis efficiency of P. minimum was similar at pH 7 to 9, and decreased significantly at pH 6 and pH 10. Overall, relative gene expression levels of the three PmCAs decreased at low pH, and increased as pH increased. Photosynthesis related genes, PmatpB and PmrbcL, showed similar expression patterns to those of PmCAs. These results suggest that changes in seawater pH may affect photosynthesis and CO2 metabolism in marine dinoflagellates.
