황기의 혈압강하 작용에 관한 연구 (A Study on the Hypotensive Action of Astragali Radix Water Extract in the Rabbit)

  • Effects of Astragali Radix Water Extract (ARWE) on the blood pressure were investigated in the whole rabbit and the spinalized rabbit. ARWE, when administered into the ear-vein or lateral ventricle, produced a fall in the blood pressure in the whole rabbit, but intravenous ARWE in the spinalized rabbit did not elicite the hypotensive action. Pretreatments with chlorisondamine, guanethidine, phentolamine and cyproheptadine in the whole rabbit weakened the depressor action of ARWE. The hypotensive action of the whole rabbit to ARWE was not influenced by the pretreatment of the animals with diphennylhydramine, propranolol, and vagotomization, whereas inhibited by atropine. ARWE did not affect the pressor response by angiotensin. However, it enhanced the hypertensive action by norepineprine and reduced the elevation in the blood pressure by carotid occlusion in the whole rabbit. These experimental observations suggest that ARWE may cause the depressor response via mechanisms of the central sympathetic blocking action, cholinergic action by peripheral origin and serotonin-like action.

  • The aim of this study was to implement the system that could evaluate the function of urinary tract with noninvasive and comfort methods. There for in this study hardware device and computer software have been developed that enables the analysis of the urine flow rate and vesical pressure signal measured during voiding. These signals were recorded simultaneously and transmitted to the PC. For the measurement system evaluation, the model system for the lower urinary system of men was designed. From the evaluation of the model system, vesical pressure was correlated with the occlusion degree. In a pilot study on 5 male subjects, means of standard deviation was 1.06, average error rate was 2.09 and coefficient variation was 2.09.

상지(桑枝)가 흰쥐의 혈압 및 Streptozotocin유발 당뇨에 미치는 영향 (The Study on Anti-hypertensive and Anti-diabetic Effect of Mori Ramulus)

  • Objectives : This study was performed to prove the anti-hypertensive and anti-diabetic effect of Mori Ramulus(MR). Methods : The examination was done on the femoral artery occlusion of the rat, and check the blood pressure. In addition, we observed therapueutic effects on diabetes rats induced by streptozotocin. Results : MR MeOH Ex, showed few effect on descending blood pressure in SD rat model. MR CHC13 layer (MRC) showed significant effect on descending blood pressure. MR was effective on the streptozotocin-induced diabetes SD rats. Especially MRC reduced the amount of serum glucose, BUN, triglyceride and insulin level. Conclusions : MRC could induce descending blood pressure in the rat model. Also, it is expected to be effective in diabetes treatment.

백부자(白附子)가 혈압(血壓), 호흡(呼吸) 및 시상하부(視床下部)의 전기자극(電氣刺戟)에 의(依)한 심맥관계반응(心脈管系反應)에 미치는 영향(影響) (Effects of Ethanol Extracts of Aconite Root on Respiration, Arterial Blood Pressure and Pressor Responses Elicited by Electrical Stimulation of the Hypothalamus in Cats)

  • The effects of ethanol extacts of aconite root (Aconitum koreanum) on mean arterial pressure, heart rate, pulse pressure, and respiration were investigated and also studied the effect on electrical activation of the hypothalamus in cats. From the present experiment the following results were obtained. 1) On administering 5 mg or 10 mg aconite extracts per kg of body weight, the mean arterial blood pressure declined markedly possibly as the result of negative inotropic and chronotropic effects of aconite. 2) From the enhanced pressor responses to intravenously injected epinephrine, the existance of vasodilatory effect of the aconite was suggested. 3) After administration of aconite extract, no significant differences were observed in the presser responses to carotid occlusion and to electrical stimulation of the hypothalamus. It is, therefore, concluded that the aconite extract exerts no significant effect on the excitability of hypothalamus as well as medullary cardiovascular center of cats. 4) After administration of $5{\sim}10$ mg/kg aconite extracts, respiratory rate was increased while depth of respiration decreased. On the otherhand, respiratory rate was markedly decreased by injection of 20 mg/kg aconite into animal.

  • 희첨(Siegesbeckiae Herba)은 진득찰(Siegesbeckiae pubescens Makino)의 잎을 말하며 한방에서 중풍, 전신마비, 반신불수, 부종등에 사용되어 왔다. 이에 관한 연구로는 한등이 diterpene계의 불합물을 분리하였고 이 diterpene계중의 한 성분이 실험적 신성 고혈압을 유기한 백서에 항고혈압작용이 있음을 보고한바 있으나 이의 고혈압 활료제로의 실용성 여부에 관하여서는 확인된 바가 없어 고혈압 치료제 개발의 일환으로서 화학적 성분 연구에 우선하여 약리학적으로 그 본태를 파악하고저 수성 및 알콜성 엑기스를 축출하여 가토에 대하여 이 실험을 시행하였다.

  • Transcranial Doppler(TCD) is an important diagnostic tool for evaluating the patients with stroke. It has some advantages and unique role when compared with other neuroimaging modalities. Recent development of transcranial color-coded Doppler(TCD) improves the limitation and pitfalls of TCD. The current indications of TCD are as follows: 1. Screening and evaluation of the intracranial major vessels 2. early detection and follow-up of vasospasm due to SAH 3. emboli detection (high-imtensity transient signals, HITs) 4. dignosis and follow-up of subclavian steal 5. evaluation of intracranial collaterals when the extracranial ICA has severe stenosis or occlusion 6. evaluation of cerebral perfusion pressure (intracranial pressure) 7. evaluation of arteriovenous malformation 8. diagnosis and follow-up of arterial dissenction 9. diagnosis and follow-up of venous sinus thrombosis (experimental).

자음건비탕가지각ㆍ천마가 뇌세포 및 뇌혈류역학 변동에 미치는 영향 (Effects of Jaeumgenby-tang adding Aurantii FructusㆍGastrodae Rhizoma on the Brain Cell and Changes of Cerebral Hemodynamics)

  • Jaeumgenby-tang(JGT) have been used in oriental medicine for many centuries as a therapeutic agent of vertigo caused by deficiency of qi(氣) and blood(血). Effect of Aurantii Fructus(AF) take off the phlegm by promoting the circulation of qi, Gastrodae Rhizoma(GR) has effects treating for headache, vertigo by calming the liver and suppressing hyperactivity of the liver-yang(陽). And, I designed to investigate whether injection of JGT adding AFㆍGR extract(JGTAG) affects cytotoxicity in vitro, cerebral hemodynamics [regional cerebral blood flow(rCBF), pial arterial diameter(PAD), mean arterial blood pressure(MABP)] in normal and cerebral ischemia rats by MCA occlusion method. The changes of rCBF and MABP were determinated by laser-doppler flowmetry(LDF), and the change of PAD was determinated by video microscope and width analyzer. The results were as follows in normal rats; JGTAG was not cytotoxicity in brain cells. And JGTAG was significantly increased rCBF, PAD and MABP. This results suggest that JGTAG increased significantly rCBF by dilating PAD. And the results were as follows in cerebral ischemic rats; The changes of rCBF and PAD were increased stably by treatment with JGTAG(10mg/kg, i.v.) during the period of cerebral reperfusion, and pretreatment with propranolol and indomethacin were increased JGT AG induced increase of rCBF and PAD during the period of cerebral reperfusion. We suggest that JGTAG has an anti-ischemic effect through the improvement of cerebral hemodynamics.

Protective Effect of Defibrotide on Splanchnic Injury following Ischemia and Reperfusion in Rats

  • A splanchic artery occlusion for 90 min followed by reperfusion of the mesenteric circulation resulted in a severe form of circulatory shock, characterized by endothelial dysfunction, severe hypotension, marked intestinal tissue injury, and a high mortality rate. The effect of defibrotide, a complex of single-stranded polydeoxyribonucleotides having antithrombotic effect, was investigated in a model of splanchnic artery occlusion (SAO) shock in urethane anesthetized rats. Occlusion of the superior mesenteric artery for 90 min produced a severe shock state, resulting in a fatal outcome within 120 min of reperfusion in many rats. Defibrotide (10 mg/kg body weight) 10 min prior to reperfusion significantly improved mean arterial blood pressure in comparison to vehicle treated rats (p<0.05). Defibrotide treatment also significantly attenuated in the increase of plasma amino nitrogen concentration, intestinal myeloperoxidase activity, intestinal lipid peroxidation, infiltration of neutrophils in intestine and thrombin induced adherence of neutrophils to superior mesentric artery segments. Superoxide anion and hydrogen peroxide production in $1{\mu}M$ formylmethionylleucylphenylalanine (fMLP)-activated PMNs was inhibited by defibrotide in a dose-dependent fashion. Defibrotide effectively scavenged hydrogen peroxide, but not hydroxyl radical. Treatment of SAO rats with defibrotide inhibited tumor necrosis factor-${\alpha}$, and interleukin-1${\beta}$ productions in blood in comparison with untreated rats. These results suggest that defibrotide partly provides beneficial effects by preserving endothelial function, attenuating neutrophil accumulation, and antioxidant in the ischemic reperfused splanchnic circulation

허혈성 뇌졸중에서의 항혈전 치료 (Antithrombotic Therapy for Ischemic Stroke)

  • Ischemic stroke is among the principal causes of death and disability in the elderly. Although control of blood pressure, decreased cigarette smoking, and modified dietary habits are among important reasons for stroke decline, the use of antithrombotic therapy, rigorously prescribed. Several antiplatelet agents are approved to reduce the risk of recurrent stroke. Aspirin is the best-studied and most widely used antiplatelet agent for stroke prevention; it provides approximately 15% to 25% relatively risk reduction for secondary prevention of stroke or the major vascular death. Combining 2 antiplatelet agents with different mechanism of action was demonstrated to provide a substantial increase in efficacy in several studies. Anticoagulation should be considered first with potential cardiac sources of embolism. Heparin reduces development of erythrocyte-fibrin thrombi that form in regions of vascular stasis especially within the heart, in severely stenosed arteries sometimes engrafted on white thrombi, in acute arterial occlusion. Heparin should not be indiscriminately given to all acute brain ischemia patients, but may contribute to treatment of large artery occlusion and severe stenosis, cardiogenic embolism with a high acute recurrence risk, and dural sinus and cerebral venous thromobosis.

실혈 후 및 혈압상승 후의 소화기 조직 혈액량 및 산소 섭취량 -제 1 편 정맥혈압과 소화기 조직 혈액량- (Gastrointestinal Tissue Blood Volume Affected by Venous Pressure Change)

  • Changes in gastrointestinal tissue blood volume induced by variations of venous pressure between 6 and 40 mmHg were studied in 32 rabbits. Venous pressure lowering was produced by withdrawal of appropriate volume of blood and venous pressure elevation was obtained by partial occlusion of intra-thoracic vena cava inferior. Estimation of regional tissue blood volume was performed by means of regional distribution of injected $Cr^{51}-labeled$ red blood cells. The following results were obtained. 1. At the normal control venous pressure value of 18 mmHg, spleen showed the highest value of tissue blood volume expressed on weight basis, namely, $111{\mu}l/gm$, Liver tissue blood volume was $95\;{\mu}l/gm$, small intestine 24 and stomach $21\;{\mu}l/gm$, respectively. 2. Linear relationships were observed between venous pressure change and gastrointestinal tissue blood volume. The coefficients of correlation were: in spleen r=0.723; in liver r=0.791; in stomach r=0.704, respectively. In small intestine the relationship was less clear and r=0.358. Tissue blood volume of extrabdominal tissue, such as M. gastrocnemius was not influenced by venous pressure change. 3. The highest change in tissue blood volume expressed on weight basis was observed in spleen. The liver tissue showed the next highest change. Change in total tissue blood volume, however, was greatest in liver and next greatest in small intestine. This was interpreted by the fact that total weight of these two organs was much greater than that of spleen. 4. The mechanism that the change in tissue blood volume lies in the venous system which has a great compliance was discussed.
