  • Insulin stimulates glucose uptake in muscle and adipose cells primarily by recruiting GLUT4 from an intracellular storage pool to the plasma membrane. Dysfunction of this process known as insulin resistance causes hyperglycemia, a hallmark of diabetes and obesity. Thus the understanding of the mechanisms underlying this process at the molecular level may give an insight into the prevention and treatment of these health problems. GLUT4 in rat adipocytes, for example, constantly recycles between the cells surface and an intracellular pool by endocytosis and exocytosis, each of which is regulated by an insulin-sensitive and GLUT4-selective sorting mechanism. Our working hypothesis has been that this sorting mechanism includes a specific interaction of a cytosolic protein with the GLUT4 cytoplasmic domain. Indeed, a synthetic peptide of the C-terminal cytoplasmic domain of GLUT4 induces an insulin-like GLUT4 recruitment when introduced in rat adipocytes. Relevance of these observations to a novel euglycemic drug design is discussed.

  • This study is a basic study on the development of functional substances involved in obesity prevention, lipid metabolism, and immune regulation. Male Sprague-Dawley rats were fed a high-fat diet for 10 weeks. Allium monanthum extracts (AME) were administered orally to obesity-induced rats, and their lipid-lowering, antioxidative and various types of biological effects related to the immune system were examined. Blood free fatty acid and triglyceride concentrations decreased as the dose of AME increased. Total cholesterol and LDL cholesterol concentrations in the blood decreased as the dose of AME increased. The total cholesterol concentrations in the liver of the AME-treated groups were lower than the control group. The thiobarbituric acid reactive concentrations were lower in the plasma and liver of all AME-treated groups than the control group. Plasma AST and ALT activities did not show any significant differences among the treatment groups. IL-$1{\beta}$ and IL-6 concentrations in the liver tended to decrease as the dose of AME increased. TNF-${\alpha}$ and IL-10 concentrations did now show any significant differences compared to the control group. Lower expression levels of TNF-${\alpha}$, Apo-B and Apo-E genes were found in the AME-treated groups. Taken together, these results indicate that AME may show positive effects in lipid lowering, antioxidation and anti-inflammation.

  • While increasing the convenience of life is a high population BMI (Body Mass Index) is increasing rapidly. Accordingly, the development of the monitoring system to manage and prevent obesity is the time that is required. But most of the monitoring system, the less information it receives management and show to have only simple information calculated this was a low efficiency problem. Also Users with normal and disease Management accuracy is low. In this paper shows the user in a graph of Body Mass Index, BMR (Basal metabolic rate) divided by grade increased accuracy for users to manage their own. Also represented by recovery with exercise machines you used, select a balanced movement mechanism, expressed as a Kcal consumption. If the graph recent data show only increased the visibility. We developed an efficient web-based monitoring system for design a exercise plan.

  • The incidence of cardiovascular disease increases rapidly after 40's. The thickness of the epicardial adipose tissue was measured to analyze the risk factors affecting the thickness change. We present the cut off value for the epicardial adipose tissue thickness for high impacted variables. For the study, 547 patients underwent echocardiography, epicardial adipose tissue thickness, body mass index, abdominal subcutaneous fat thickness and diabetes mellitus were used as analytical variables. As a result, multiple regression analysis of age group showed that diabetes mellitus was highly influential in all age group, so we could predict the cut off value for the epicardial adipose tissue thickness for diabetes mellitus. As a result of ROC curve analysis, cut off value of 7.44 mm was obtained with sensitivity of 79.6% and specificity of 60.6% for diab etes variab le. Logistic regression analysis b ased on this value showed that the risk for diab etes increased 6 times with each increase in the thickness of the epicardial adipose tissue. Among the various obesity indexes, epicardial adipose tissue is used as an important index especially to prevent the risk of cardiovascular disease. Therefore, we suggest that the use of echocardiography as a routine screening method for the prevention of cardiovascular disease and metabolic syndrome, which is rapidly increasing in patients over 40's age, will enable more effective monitoring. Radiation is the emission or transmission of energy in the form of waves or particles through space or through a material medium.

  • Objective: This study investigated the effect of purified Carthamus tinctorius (C. tinctorius) extracted with a hot water and ethanol method on adipogenic differentiation of mouse bone marrow-derived mesenchymal stromal stem cells (mBMSCs). Methods: The C. tinctorius was extracted using hot water and ethanol. The samples were concentrated by a rotary evaporator and were then dried using a freeze-dryer. The mBMSCs were cultured and maintained in a minimum essential medium eagle alpha (${\alpha}-MEM$) supplemented with 10% FBS and 1% antibiotic antimycotic solution. To induce adipogenic differentiation, the cells were treated with Dulbecco's modified eagle's medium-low glucose (DMEM-LG) containing 1 mg/mL insulin, 1 mM dexamethasone, and 0.5 mM 3-isobutyl-1-methylxanthine. To evaluate the adipogenic differentiation ability, oil-red O staining was performed after adipogenic differentiation for 21 days. The mRNA expression and protein level of adipogenic-related genes were quantified by quantitative real-time PCR and western blotting, respectively. Results: In the results of the MTT assay, no concentrations of C. tinctorius extracts showed toxicity on mBMSCs, so we fixed the treatment concentration of the extract at 100 ng/mL. In oil-red O staining, the water-C. tinctorius extract treatment significantly decreased adipogenic differentiation compared with the control and ethanol extract groups. The water-C. tinctorius extract group in particular showed reduced mRNA and protein expression of Peroxisome proliferator-activated receptor gamma ($Ppar{\gamma}$) and CCAAT/enhancer-binding protein alpha ($C/ebp{\alpha}$), which are adipogenic-related transcription factors. Conclusion: These data suggest that extract of C. tinctorius decreased the adipogenic differentiation of mBMSCs, while only water-C. tinctorius extract had an effect on different adipogenesis in mBMSCs. The C. tinctorius will be a useful therapeutic reagent for the prevention of obesity-related diseases such as diabetes, hyperlipidemia, coronary artery disease, and osteoporosis.

  • This study investigated and compared the daily activities and energy expenditure of normally-weighted and obese elementary school children. The participants were 115 boys and 115 girls (6th grade) at ten elementary schools in Busan. Time spent on each activity was evaluated using the twenty-four hour recall method. 1) The general characteristics of the normally-weighted and obese children did not differ, although the normally-weighted children exercised and used stairs more than the obese children.2) Among their classified activities, the children spent most of their time resting, study, leisure, physiology and hygiene, commuting, and housework in that decreasing order. Normally-weighted children spent less time tying down and in conversation with family and friends than obese ones. 3) The children's average activity factors were 1.47 - 1.50. The normally-weighted children's rest energy expenditure (REE) (1391.4 kcal,1264.7 kcal) was less than that of the obese children (1711.4 kcal. 1461.0 kcal) . The normally-weighted children's daily energy expenditure was less than that of the obese, but energy expenditure per body weight was less in obese children than in the normally-weighted. In conclusion, sedentary activities and energy expenditure per body weight was less in the obese compared to the normally-weighted children. Programs to help children perceive and practice desirable physical activities should be suggested for prevention of obesity in children. (Korean J Nutrition 38(10): 847$\sim$855,2005)

  • The present study was carried out to investigate the antiobesity and hypocholesterolemic effects of Bifidobacteria animalis DY-64 (B. animalis DY-64), a lactic acid bacterium isolated from the human intestine, in rats fed a high-fat/high-cholesterol diet for 4 weeks. Forty male Sprague-Dawley rats were divided into four groups and fed either a normal (N) or high-fat/high-cholesterol (HFC) diet without or with oral administration of B. animalis DY-64 (N-BA, HFC-BA). The gain in body weight, and liver and adipose tissue weights of the HFC group were heavier than that of the HFC-BA group. Serum total cholesterol (TC), LDL-cholesterol, and leptin levels of the HFC group, which were significantly elevated compared to those of the N group, dropped by 19, 18, 21, and 13% in the HFC-BA group, respectively, whereas the serum HDL-cholesterol level markedly increased. However, serum TG, LDL-cholesterol, HDL-cholesterol, and leptin levels were not significantly different among the N groups (N, N-BA) with or without B. animalis DY-64 administration. TC and TG levels of the liver as well as the TG level of the adipose tissue were significantly reduced in the HFC-BA group. In addition, HR-LPL activity in adipose tissue was also lower in the HFC-BA group than in the HFC group. These results suggest that B. animalis DY-64 isolated from the human intestine exerts hypocholesterolemic effects by reducing serum and liver cholesterol levels and plays a role in the prevention of obesity induced by HFC diet.
