Effect of Heavy Metals on the Secretion of Amylase in Rat Pancreatic Fragments (중금속류가 취절편의 Amylase 분비에 미치는 영향)

  • Heavy metals which are present as trace elements in human body have been known to modify various enzymatic reaction. These metals can be essential or non-essential. Zinc, copper and calcium are essential in maintaining some biological processes, whereas non-essential metals such as cadmium, lead and mercury produce accumulatve toxic effect. Cadmium accumulated in pancreas can cause toxicity and damage of pancreatic cells, thereby influencing CHO metabolism. Lead compounds are known to produce toxic effects on the kidney, digestive system and brain fellowed by inhibition of activity of ${\rho}-aminolevulinic$ acid and biosynthesis of hemoproteins and cytochrome. Evidence has been accumulated that zinc not only acts as a cofactor in enzyme reaction but also prevents toxic effect induced by heavy metal such as copper and cadmium. To demonstrate the effect of heavy metals on pancreatic secretion, part of uncinate pancreas was taken and incubated in Krebs-Ringer bicarbonate buffer with heavy metals used. Additional treatment with CCK-OP was performed when needed. After incubation during different period of time, medium was analyzed for amylase activity using Bernfeld's method. The present study was attempted in order to elucidate the effect of several kinds of heavy metal on exocrine pancreatic secretion in vitro. The results obtained are as follows: 1) CCK-OP stimulated significantly amylase release from pancreatic fragments in vitro. 2) CCK-OP response of amylase release from pancreatic fragments was inhibited by treatmant with cadmium, especially high doses of cadmium. 3) CCK-OP response of amylase release from pancreatic fragments was inhibited when pretreated with $10^{-4}M$ copper chloride. 4) Lead chloride at the concentration of $10^{-3}M\;and\;10^{4}M$ stimulated the basal amylase release in vitro but CCK-OP response did not augment by lead chloride. 5) Zine chloride did not affect amylase release from pancreatic fragment in vitro. From the results mentioned above, it is suggested that CCK-OP response was inhibited it the amylase release from pancreatic fragments pretreated with cadmium and copper chloride.

Predictors of Relapse in Patients with Organizing Pneumonia

  • Background: Although organizing pneumonia (OP) responds well to corticosteroid therapy, relapse is common during dose reduction or follow-up. Predictors of relapse in OP patients remain to be established. The aim of the present study was to identify factors related to relapse in OP patients. Methods: This study was retrospectively performed in a tertiary referral center. Of 66 OP patients who were improved with or without treatment, 20 (30%) experienced relapse. The clinical and radiologic parameters in the relapse patient group (n=20) were compared to that in the non-relapse group (n=46). Results: Multivariate analysis demonstrated that percent predicted forced vital capacity (FVC), $PaO_2/FiO_2$, and serum protein level were significant predictors of relapse in OP patients (odds ratio [OR], 0.82; 95% confidence interval [CI], 0.70-0.97; p=0.018; OR, 1.02; 95% CI, 1.00-1.04; p=0.042; and OR, 0.06; 95% CI, 0.01-0.87; p=0.039, respectively). Conclusion: This study shows that FVC, $PaO_2/FiO_2$ and serum protein level at presentation can significantly predict relapse in OP patients.

A Study on the Mechanism of Opening-with-Pungent/Lowering-with-Bitter Method of Wenbing and the Spleen/Stomach Disease Treatment in Piweilun (온병학(溫病學) 신개고강법(辛開苦降法)과 『비위론(脾胃論)』의 비위병(脾胃病) 치료 기전에 대한 고찰)

  • Objectives : The aim of this paper is to compare the mechanisms of the OP/LB method and the SSD treatment in 『Piweilun』. Methods : Wenbing texts, articles on the OP/LB method, and the 『Piweilun』 was examined for comparison of treatment mechanisms of SSD. Results : The mechanism of the OP/LB method in treating SSD was to treat the Spleen and Stomach separately, to restore the ascending/descending pattern of qi through simultaneous use of pungent and bitter flavors which raises and lowers, respectively. Moreover, the use of medicinals with contrary properties regulate the other's biased nature, and none of the medicinals create dampness. The pungent and bitter flavors play central roles, where the pungent flavor opens and communicates and the bitter flavor clears and lowers. The treatment method of SSD in the 『Piweilun』 treat the Spleen and Stomach together, with a focus on raising and dispersing through upraising yang and reinforcing qi by means of Wind medicinals with pungent and bitter flavors added to sweet and warm medicinals. Conclusions : Owing to the expansion of the OP/LB method concept by modern-contemporary scholars, the potential for treating SSD as can be seen in the 『Piweilun』 with the OP/LB method has emerged. The similarity between the OP/LB method and treatment of SSD in the 『Piweilun』 is that the objective of the formulas is to lower fire heat and communicate qi to help qi movement. This common objective allows for treatment of SSD within Wenbing and those in 『Piweilun』 with the said formulas through appropriate modification fit for each situation.

Effects of 2-deoxy-D-glucose and quercetin on the expression of osteonectin and osteopontin during the differentiation of irradiated MC3T3-El osteoblastic cells

  • Purpose : To characterize the effects of 2-deoxy-D-glucose (2-DG) and quercetin (QCT) on gene expression of osteonectin (ON) and osteopontin (OP) in irradiated MC3T3-El cells. Materials and Methods : When MC3T3-El osteoblastic cells had reached 70-80% confluence, cultures were transferred to a differentiating medium supplemented with 5 mM 2-DG or 10 ${\mu}M$ QCT and then irradiated with 2, 4, 6, and 8 Gy. At various times after irradiation, the cells were analyzed for the expression of bone mineralization genes such as ON and OP. Results : The mRNA expression of both ON and OP was increased according to the culture time in the differentiation medium, and the increase of the genes peaked at 14 days after the differentiation induction. In the case of OP, the increase of mRNA expression was maintained to 28 days after the differentiation, while the mRNA level of ON was reduced to the basal level at the same time. Irradiation adding 2-DG showed a significant peak value in the expression pattern of ON at 4 Gy 7 days after irradiation. Irradiation adding QCT increased the mRNA expression of ON and OP in a dose-dependant manner, but irradiation adding 2-DG did not show any differences between the control and experiments 14 days after irradiation. Irradiation adding QCT increased significantly the expression patterns of ON 21 days after irradiation. Conclusion : The results showed that QCT acted as a radiosensitizer in the gene expression of ON and OP during differentiation of the late stage of irradiated MC3T3-E1 osteoblastic cells in vitro. A Study on the PP/PS Blends with Nylon 6 Reactive Compatibilizers (Nylon6계 반응 상용화제에 의한 PP/PS 블렌드에 관한 연구)

  • RPP(maleic-anhydride grafted PP)and OPS(oxazoline grafted PS) do not react to each other, and thus show immiscibility. In this study, Nylons was added to RPP/OPS blend systems, as a reactive compatibilizer for enhancing the miscibility of the blends. When Nylon6 was added to the blends of RPP and OPS, RPP/Nylon6/OPS was produced. The effects of the molar ratio of Nylon6 on the RPP-Nylon6-OPS reaction were studied. Torque test and FT-IR analysis have been carried out to investigate the reaction of RPP/Nylon6/OPS system. The reaction torgue ratio and reaction efficiency show the maximum values at 1 : 0.66 : 1 and 1 : 1 : 1 (in moles) for RPP/Nylon6/OPS. In the RPP/Nylon6/OPS blends, their mechanical properties were changed with the molar ratio of Nylon6 and showed the highest value at molar ratio of 1.5. Physical properties and compatibility of RPP/Nylon6/OPS were compared with those of PP/Nylon6/OPS. Consequently, RPP/Nylon6/OPS plays a proper role as a reactive compatibilizer to the PP/PS blend system.

Induction of In Vitro Vitellogenin Synthesis by Bisphenol, Nonylphenol and Octylphenol in Chinese Minnow(Phoxinus oxycephalus) Hepatocytes

  • Bisphenol A (BPA), nonylphenol (NP), and 4-tert-octylphenol (OP) are known endocrine disrupting chemicals (EDCs) with estrogenic activity in fish. This study compared the effects of BPA, NP and OP on in vitro vitellogenin (VTG) synthesis in primary cultures of hepatocytes of the Chinese minnow Phoxinus oxycephalus. The VTG secreted into the culture medium was measured using enzyme-linked immunosorbent assay (ELISA), which we developed in this study using an antibody prepared from homogenates of Chinese minnow egg. VTG synthesis was induced by estradiol-17$\beta$ ($E_2$) and phenols (BPA, NP and OP) treatment. $E_2$ at concentrations of 10$^{-6}$ M or higher increased VTG levels significantly (P < 0.05). Exposure to 10^5\;M\;BPA\;or\;10^-4$M NP and OPinduced in vitro VTG synthesis (P < 0.01). However, $10^-3$ M BPA, NP or OP did not induce VTG synthesis. These results suggest that SPA has the highest estrogenic potential in Chinese minnow hepatocytes. Tamoxifen, an anti-estrogen, drastically blocked the production of VTG by phenols (BPA, NP and OP) suggesting that phenols (BPA, NP and OP) may act via binding to estrogen receptor (ER) in Chinese minnow hepatocytes.

  • In this paper, we introduce modulo ( 2$^n$-1) parallel-processing residue multipliers, using Op Amp circuits, and their technological application to designing binary multipliers. The limit of multiplying speed in computational processing is a serious harrier in the advances of VLSI technology. To solve this problem, we implement a class of modulo ( 2$^n$-1) parallel multipliers having superior time complexity to O( log$_2$( log$_2$( log$_2$$^n$))) by applying Op Amp circuits, while investigating their technological application to binary multipliers. Since they have excellent time & area complexity compared with previous parallel multipliers, and are applicable to designing binary multipliers of the same efficiency, such parallel multipliers possess high academic value.   • Objectives : The cortex and root of Acanthopanax sessiliflorus (AR), a herbal medicine, have been used for several diseases including cancer in Oriental countries. Recently, we reported that AR has an immune-potentiating action. Oga-Power(OP) was made using extract from AR. For these reasons, we hypothesized that OP can potentiate the immune system in terms of accelerating proliferation rates of immune cells such as thymocytes and splenocytes. Methods : In this experiment, proliferation rates of thymocytes and splenocytes were measured using modified 3-[4,5-dimethy -lthiazol-2-yl]-2,5-diphenyltetrazolium bromide (MTT). No production levels in macrophages isolated from normal mice were measured using Griess method. Results : In our results, treatment with OP accelerated proliferation rates of splenocytes, but did not affect those of thymocytes in vitro. On the other hand, proliferation rates of thymocytes was elevated in vivo. In addition, level of NO production from macrophage separated from abdominal cavity of normal mice was elevated by treatment with OP. Conclusions : In conclusion, OP has immune-potentiating action, by acceleration of splenocyte proliferation and elevation of NO production level from macrophages.

Effects of Safflower Seed on New Bone formation (홍화씨가 신생골 형성에 미치는 영향)

  • Korean safflower seed has been known to have healing effects on both bone fracture and osteoporosis. On the base of such a notice, this experiment was carried out to explore the effects of safflower seed on bone formation and bone repair. The toxicity test and the effect of Korean safflower seed were evaluated with 60 rats, 3-month old. Forty Sprague-Dawley rats composed of 20 male and 20 female were underwent unilateral tibial defect and then fastened with unilateral fixators. The operated rats were divided into two groups depending on the composition of diet, such as positive control group fed normal diet(C-OP group) and safflower seed group fed 30% of safflower seed diet and 70% of normal diet(S-OP group). Another 20 rats without operation were maintained, each 10 rats were fed either normal diet or 30% of safflower seed diet and 70% of normal diet, and observed the toxicity of safflower seed by measuring weight and urine parameters. Postoperative radiography were taken once in 2 weeks to evaluate callus formation for operated groups and blood collection via heart puncture were carried out once in 3 weeks for 3 groups. The concentration of Ca and Pi in serum were measured using both auto Kit and $^{31}$ P Nuclear Magnetic Resonance(NMR). At present study, no toxic effect was observed from both weight increment and urine index after feeding the safflower seed diet. The comparison of the radiography between C-OP and S-OP group were showed that the safflower seed diet appeared to stimulate the formation of callus in the rat. The ratio of Ca/P in serum was low in S-OP group compared to C-OP group with the auto Kit, but there were no significant differences between two groups (p < 0.05). In addition, the variations of Pi values in NMR examination were also confirmed based on the result of auto Kit. In conclusion, this study implied that safflower seed might influence to bone formation and shorten the periods of remedy by stimulating the calcification of bone

Model test on slope deformation and failure caused by transition from open-pit to underground mining

  • Open-pit (OP) and underground (UG) mining are usually used to exploit shallow and deep ore deposits, respectively. When mine deposit starts from shallow subsurface and extends to a great depth, sequential use of OP and UG mining is an efficient and economical way to maintain mining productivity. However, a transition from OP to UG mining could induce significant rock movements that cause the slope instability of the open pit. Based on Yanqianshan Iron Mine, which was in the transition from OP to UG mining, a large-scale two-dimensional (2D) model test was built according to the similar theory. Thereafter, the UG mining was carried out to mimic the process of transition from OP to UG mining to disclose the triggered rock movement as well as to assess the associated slope instability. By jointly using three-dimensional (3D) laser scanning, distributed fiber optics, and digital photogrammetry measurement, the deformations, movements and strains of the rock slope during mining were monitored. The obtained data showed that the transition from OP to UG mining led to significant slope movements and deformations that can trigger catastrophic slope failure. The progressive movement of the slope could be divided into three stages: onset of micro-fracture, propagation of tensile cracks, and the overturning and/or sliding of slopes. The failure mode depended on the orientation of structural joints of the rock mass as well as the formation of tension cracks. This study also proved that these non-contact monitoring technologies were valid methods to acquire the interior strain and external deformation with high precision.
