Is catechol-o-methyltransferase gene polymorphism a risk factor in the development of premenstrual syndrome?

  • Objective: The objective of this study was to investigate whether there was a correlation between catechol-o-methyltransferase (COMT) gene polymorphism, which is believed to play a role in the etiology of psychotic disorders, and premenstrual syndrome (PMS). Methods: Fifty-three women with regular menstrual cycles, aged between 18 and 46 years and diagnosed with PMS according to the American Congress of Obstetrics and Gynecology criteria were included in this study as the study group, and 53 healthy women having no health problems were selected as the controls. Venous blood was collected from all patients included in the study and kept at $-18^{\circ}C$ prior to analysis. Results: There was no significant difference between the groups in terms of demographic features such as age, body mass index, number of pregnancies, parity, and number of children. No statistically significant difference was observed in terms of COMT gene polymorphism (p=0.61) between women in the PMS and the control groups. However, a significant difference was found between arthralgia, which is an indicator of PMS, and low-enzyme activity COMT gene (Met/Met) polymorphism (p=0.04). Conclusion: These results suggested that there was no significant relationship between PMS and COMT gene polymorphism. Since we could not find a direct correlation between the COMT gene polymorphism and PMS, further studies including alternative neurotransmitter pathways are needed to find an effective treatment for this disease.

A Challenging Study to Identify Target Proteins by a Proteomics Approach and Their Validation by Raising Polyclonal Antibody

  • This study was conducted to validate the theoretical feasibility of a technique to identify biomarkers in Korean native black pig (KNP) and a commercial Landrace breed. Using two-dimensional electrophoresis, we found six proteins (NADH dehydrogenase Fe-S protein 1, an unnamed protein product, similar to T-complex protein 1, annexin V = CaBP33 isoform, fatty acid-binding protein, and catechol O-methyltransferase), which appeared in KNP alone. We raised polyclonal antibodies (used as the primary antibody) for Western blotting to confirm the characteristics of the six KNP proteins. As a result, catechol O-methyltransferase, annexin V = CaBP33 isoform, and the unnamed protein product presented thicker bands in KNP than those in Landrace. Moreover, catechol O-methyltransferase was shown to be more feasible as a biomarker for KNP. However, cross-reactivity was observed with the polyclonal antibodies for KNP and the other three proteins (NADH dehydrogenase, a protein similar to T-complex protein 1, and fatty acid-binding protein). This study only showed limited results from a limited number of animals; however, our research suggests possibilities for future studies.

Protein Carboxylmethylation in Porcine Spleen is Mainly Mediated by Class I Protein Carboxyl O-Methyltransferase

  • The functional role of protein carboxylmethylation (PCM) has not yet been clearly elucidated in the tissue level. The biochemical feature of PCM in porcine spleen was therefore studied by investigating the methyl accepting capacity (MAC) of natural endogenous substrate proteins for protein carboxyl O-methyltransferase (PCMT) in various conditions. Strong acidic and alkaline-conditioned (at pH 11.0) analyses of the MAC indicated that approximately 65% of total protein methylation seemed to be mediated by spleen PCMT. The hydrolytic kinetics of the PCM products, such as carboxylmethylesters (CMEs), under mild alkaline conditions revealed that there may be three different kinds of CMEs [displaying half-times (T$_{1}$2/) of 1.1 min (82.7% of total CMEs), 13.9 min (4.6%), and 478.0 min (12.7%)], assuming that the majority of CME is base-labile and may be catalyzed by class I PCMT. In agreement with these results, several natural endogenous substrate proteins (14, 31 and 86 kDa) were identified strikingly by acidic-conditioned electrophoresis, and their MAC was lost upon alkaline conditions. On the other hand, other proteins (23 and 62 kDa) weakly appeared under alkaline conditions, indicating that PCM mediated by class II or III PCMT may be a minor reaction. The MAC of an isolated endogenous substrate protein (23-kDa) was also detected upon acidic-conditioned electrophoresis. Therefore, our date suggest that most spleen PCM may be catalyzed by class I PCMT, which participates in repairing aged proteins.

Molecular Cloning and Characterization of Bacillus cereus O-Methyltransferase

  • Biotransformation is a good tool to synthesize regioselective compounds. It could be performed with diverse sources of genes, and microorganisms provide a myriad of gene sources for biotransformation. We were interested in modification of flavonoids, and therefore, we cloned a putative O-methyltransferase from Bacillus cereus, BcOMT-2. It has a 668-bp open reading frame that encodes a 24.6-kDa protein. In order to investigate the modification reaction mediated by BcOMT-2, it was expressed in E. coli as a His-tag fusion protein and purified to homogeneity. Several substrates such as naringenin, luteolin, kaempferol, and quercetin were tested and reaction products were analyzed by thin layer chromatography (TLC) and high performance liquid chromatography (HPLC). BcOMT-2 could transfer a methyl group to substrates that have a 3' functional hydroxyl group, such as luteolin and quercetin. Comparison of the HPLC retention time and UV spectrum of the quercetin reaction product with corresponding authentic 3'-methylated and 4'-methylated compounds showed that the methylation position was at either the 3'-hydroxyl or 4'-hydroxyl group. Thus, BcOMT-2 transfers a methyl group either to the 3'-hydroxyl or 4'-hydroxyl group of flavonoids when both hydroxyl groups are available. Among several flavonoids that contain a 3'- and 4'-hydroxyl group, fisetin was the best substrate for the BcOMT-2.

Crystallization and Preliminary X-Ray Diffraction Analysis of BcOMT2 from Bacillus cereus: A Family of O-Methyltransferase

  • O-Methyltransferases (OMTs), one of the ubiquitous enzymes in plants, bacteria, and humans, catalyze a methyl-transfer reaction using S-adenosylmethionine and a wide range of phenolics as a methyl donor and acceptor, respectively. Substrates for most bacterial OMTs have largely remained elusive, but recent investigation using BcOMT2, an OMT from Bacillus cereus, suggested that ortho-dihydroxyflavonoids could serve as substrates. To elucidate the functional and structural features of BcOMT2, we expressed, and purified BcOMT2, and crystallized an apoenzyme and its ternary complex in the presence of a flavonoid and S-adenosylhomocysteine. Each crystal diffracted to $1.8{\AA}$ with its space group of C2 and $P2_{1}2_{1}2_{1}$, respectively. Structural analysis of apo-BcOMT2 and its ternary complex will provide the structural basis of methyl transfer onto (iso)flavonoids in a regiospecific manner.

Anatomical, Chemical, and Topochemical Characteristics of Transgemic Poplar Down-regulated with O-methyltransferase

  • The present work was undertaken to investigate the anatomical and chemical characteristics of transgenic poplar down-regulated with antisense OMT gene. Also the distribution of lignin in transgenic poplar trees was investigated at cellular level. No visible abnormal phenotype was observed in the fibers and vessel elements of transgenic poplar. Any marked differences in the staining intensities of Wiesner and Mäule color reaction were not identified in the transgenic poplar. TEM micrographs did not show any staining intensities in the cell walls stained with KMnO4. Interestingly, the UV spectroscopy of semi-thin sections exhibited a distinct decrease of lignin absorption at 280 nm in the vessel walls, indicating transgenic poplar wood with lower amount of guaiacyl lignin in vessel elements. Chemical composition of antisense OMT poplar was almost identical to that of wild-type poplar. Klason lignin content of transgenic poplar did not show any significant difference from that of the controls. The solid state NMR spectra revealed the transgenic poplar with only slightly more syringyl lignin than the control. The present work showed that antisense OMT gene constructed in the poplar was not enough to reduce the overall content of Klason lignin, and suggested that the expression of transformation was confined to vessel walls.

In silico discovery and evaluation of phytochemicals binding mechanism against human catechol-O-methyltransferase as a putative bioenhancer of L-DOPA therapy in Parkinson disease

  • Levodopa (L-DOPA) therapy is normally practised to treat motor pattern associated with Parkinson disease (PD). Additionally, several inhibitory drugs such as Entacapone and Opicapone are also cosupplemented to protect peripheral inactivation of exogenous L-DOPA (~80%) that occurs due to metabolic activity of the enzyme catechol-O-methyltransferase (COMT). Although, both Entacapone and Opicapone have U.S. Food and Drug Administration approval but regular use of these drugs is associated with high risk of side effects. Thus, authors have focused on in silico discovery of phytochemicals and evaluation of their effectiveness against human soluble COMT using virtual screening, molecular docking, drug-like property prediction, generation of pharmacophoric property, and molecular dynamics simulation. Overall, study proposed, nine phytochemicals (withaphysalin D, withaphysalin N, withaferin A, withacnistin, withaphysalin C, withaphysalin O, withanolide B, withasomnine, and withaphysalin F) of plant Withania somnifera have strong binding efficiency against human COMT in comparison to both of the drugs i.e., Opicapone and Entacapone, thus may be used as putative bioenhancer in L-DOPA therapy. The present study needs further experimental validation to be used as an adjuvant in PD treatment.

연부조직육종 환자에서 $O^6$-MGMT 와 촉진자 과메틸화의 예후적 중요성 (Prognostic Significance of $O^6$-MGMT and Promotor Hypermethylation in Patients with Soft Tissue Sarcomas)

  • 서론: $O^6$-methylguanine-DNA methyltransferase (MGMT)는 DNA 염기 손상에 의해 형성된 $O^6$-methylguanine에서 알킬기를 제거하여 DNA 염기 손상을 복구하는 역할을 한다. MGMT의 후생유전적 불활성화가 인체 종양에서 보고되고 있으며, 항암 치료에 대한 저항성에 영향을 주는 요소로 알려져 있다. 본 연구에서는 연부조직육종에서 이러한 MGMT 불활성화가 미치는 영향에 대해 살펴보고자 하였다. 재료 및 방법: 총 62예의 연부조직육종조직에서 메틸화 특이 중합효소연쇄반응을 이용하여 MGMT 유전자의 촉진자 부위 메틸화 정도를 알아보고, 면역조직화학염색을 통하여 MGMT 단백 발현의 소실 양상을 살펴보았다. 결과: MGMT 단백 발현 소실은 진행성 병기(p=0.000), 조직학적 고등급(p=0.005), 종양의 재발 또는 전이(p=0.011), 그리고, 낮은 생존률(p=0.017)과 통계학적 유의성을 보였다. MGMT 유전자 촉진자 부위 메틸화는 조직학적 고등급, 종양의 재발 또는 전이, 그리고, 낮은 생존률과 관련성을 보였다. 또한, MGMT 단백 발현의 소실은 MGMT 유전자 촉진자 부위 과메틸화와 높은 상관성을 나타내었다(p=0.000). 결론: 본 연구는 MGMT 단백 발현의 소실과 MGMT 유전자 촉진자 부위 과메틸화가 연부조직 육종에서 흔히 발생하며 종양의 공격적인 양상 및 나쁜 예후와 관련성이 있다는 것을 보여준다. 또한 MGMT 단백 발현의 소실은 대개 MGMT 유전자 촉진자 부위 과메틸화로 인해 발생한다는 사실을 보여주고 있다.
