The protection of dry matter and nitrogen from rumen degradation of dhaincha seed (Sesbania aculeate) meal was studied using nylon bag technique. The dhaincha seed meal was subjected to various heat treatments that included oven drying, autoclaving and boiling. Similar experiment was conducted with fish meal as reference for comparison. The oven-dried dhaincha meal was found to retain more dry matter and nitrogen than was found boiled or autoclaved meal. While autoclaving appeared to improve nitrogen and dry matter retention to some extent, boiling seemed to cause more loss of dry matter from nylon bag. Heat treatment caused high retention of nitrogen by fish meal. The calculated effective protein degradation was 80.4% and 83.2% for the oven dried fish meal and dhaincha seed meal whereas same values were 74.2% and 86.7% for autoclaved fish and dhaincha seed meal respectively at the outflow rate of 4.4% per hour. The in vitro study revealed higher digestibility for heat treated samples by pepsin. The dry matter, nitrogen and ash content of dhaincha seed meal were 85.93%, 5.93% and 7.31% respectively.
Ruminal degradation characteristics and intestinal availability of crude protein (CP) in four animal-origin feeds (fish meal, meat meal, viscera meal, feather meal) were estimated by mobile nylon bag technique. Three ruminally and duodenally cannulated Holstein dairy cows (average body wt. 550kg) fed a diet containing 40% concentrate and 60% orchard grass hay on a dry matter (DM) basis. Assuming that the outflow rate of diet in rumen is 5% per hour (k =0.05), contents of quickly degradable CP (QDP), slowly degradable CP (SDP), and undegradable CP (UDP) in the rumen were 27.6%, 9.4%, 63.0% for fish meal, 34.3% 28.1%, 37,6% for meat meal, 43.9%, 12.5%, 43.6% for viscera meal, and 14.4%, 15.8%, 69.8% for feather meal, respectively. Intestinal CP degradability was 51.0% for fish meal, 27.2% for meat meal, 37.9% for viscera meal and 56.2% for feather meal. Available UDP in the intestinal tract was contained 288 g, 217 g, 246 g and 423 g per kilogram DM of diet in fish meal, meat meal, viscera meal and feather meal, respectively.
A series of experiments was conducted to determine the influence of various pepsin-HCL pretreatment factor, hereby the factors of duration of washing for the retrieved bags, inherent to the mobile nylon bag technique (MNBT), on apparent ileal digestibility of crude protein (AIDCP) and apparent ileal digestibility of dry matter (AIDDM). At last, the AIDCP and apparent ileal digestibility of amino acids (AIDAA) in maize, barley, wheat, rapeseed meal, cottonseed meal and three mixed diets were determined with the MNBT and ileo-rectal anastomis pigs (IRAT). For the MNBT techniques, bag measuring $25{\times}40$ MM and containing 0.75 g feedstuff samples, after pre-digestion in vitro, were introduced into the ileo-rectal anastomis pigs (IRAT) gastrointestinal tract through a duodenal cannula and recovered in the ileal digesta between 6 and 12 h. later. 1. The apparent ileal digestibility of dry matter (AIDDM) and crude protein (AIDCP) of the tested samples, with the exception of fish meal, determined by MNBT were not affected by the different pepsin-HCL pretreatment times in vitro between 2.5 h. and 4 h. 2. There was no significant (p > 0.05) difference of the AIDCP and AIDDM of maize determined by the MNBT among different pepsin concentration (0.03%, 0.07% and 0.1 %) treatment in vitro. 3. The AIDCP determined with the MNBT was affected by the washed and unwashed recovered bags from the ileal digesta. 4. The AIDCP and AID amino acids (AIDAA) of maize, barley, wheat, rapeseed meal, soya-bean meal, cottonseed meal and three mixed diets from the MNBT, with a solution of 0.01N HCL (PH 2) and 0.1% of pepsin concentration, a pepsin-HCL pretreatment time in vitro or 4h. and a washing time of the recovered bag from the ileal digesta compared well with those from the IRAT. The linear regression analysis showed a significant correlation (p < 0.01) of AIDCP and AIDDA between the IRAT and MNBT.

The objectives of this study were to determine an optimal cultivation time for populations of yeast and lactic acid bacteria (LAB) co-cultured in fermented milk and effects of soybean meal fermented milk (SBMFM) supplementation on rumen degradability in beef cattle using nylon bag technique. The study on an optimal cultivation time for yeast and LAB growth in fermented milk was determined at 0, 4, 8, 24, 48, 72, and 96 h post-cultivation. After fermenting for 4 days, an optimal cultivation time of yeast and LAB in fermented milk was selected and used for making the SBMFM product to study nylon bag technique. Two ruminal fistulated beef cattle ($410{\pm}10kg$) were used to study on the effect of SBMFM supplementation (0%, 3%, and 5% of total concentrate substrate) on rumen degradability using in situ method at incubation times of 0, 2, 4, 6, 12, 24, 48, and 72 h according to a Completely randomized design. The results revealed that the highest yeast and LAB population culture in fermented milk was found at 72 h-post cultivation. From in situ study, the soluble fractions at time zero (a), potential degradability (a+b) and effective degradability of dry matter (EDDM) linearly (p<0.01) increased with the increasing supplemental levels and the highest was in the 5% SBMFM supplemented group. However, there was no effect of SBMFM supplement on insoluble degradability fractions (b) and rate of degradation (c). In conclusion, the optimal fermented time for fermented milk with yeast and LAB was at 72 h-post cultivation and supplementation of SBMFM at 5% of total concentrate substrate could improve rumen degradability of beef cattle. However, further research on effect of SBMFM on rumen ecology and production performance in meat and milk should be conducted using in vivo both digestion and feeding trials.
Previous studies conducted with swine have reported that the mobile nylon bag technique (MNBT) does not always accurately predict in vivo nutrient digestibilities. Therefore, in this study, the MNBT was modified so that nutrient digestibilities would more closely resemble those from conventional (Con) digestibility studies obtained using the indicator method. A total of 19 feeds were tested including five cereal grains, five legumes, three high protein sources and six mixed diets. The principle changes to the MNBT included the use of a fecal collection harness which minimized the number of bags lost. In addition, previous protocols involved pooling of bags within pig while in the present experiment all bags were analyzed separately to increase the precision of the test. Finally, chemical analyses were done using the entire nylon bag plus residue rather than opening.the bags and scraping out the contents. With the exception of the barley sample (p=0.01), dry matter digestibility (DMD) coefficients obtained with the MNBT were not significantly different from those obtained with the indicator method. The linear regression equation relating the MNBT to the indicator method was Con DMD=-O.77+1.02 MNBT DMD ($r^2=0.93$: p<0.0001). There was no significant (p>0.05) difference in gross energy digestibility (GED) coefficients determined using the MNBT or the indicator method for any of the 19 feeds. The regression line equation relating the MNBT to the indicator method was Con GED=-5.68+1.06 MNBT GED ($r^2=0.94$: p<0.0001). The MNBT was less effective in predicting in vivo crude protein digestibility (CPD) than it was in predicting dry matter and energy digestibility. Differences greater than five percentage units were observed for two of the legumes, Kabuli chickpeas (p=0.02) and the extruded pea-canola seed mixture (p=0.01) as well as for three of the mixed diets including the unheated hulled barley-based diet (p=0.01), the unheated hulless-barley based diet (p=0.08) and the barley-soybean meal based diet (p=0.008). The regression equation relating the MNBT to the indicator method was Con CPD=5.75 + 0.90 MNBT CPO ($r^2=0.76$; p<0.0001). This study indicates that the modified MNBT can be used for the rapid determination of dry matter and energy digestibility in a wide variety of ingredients. For the measurement of crude protein digestibility, the technique produces results similar to conventional digestibility studies for cereal grains and high protein feeds but tends to overestimate protein digestibility for legumes and mixed diets.

This study was conducted to examine of rice straw after chemical treatments and ensiling on its feeding value, in situ studies using a rumen fistulated Korean cow and nylon bag technique. NaOH treatment greatly improved the degradation the Dry matter and Neutral detergent fiber degradation in the rumen but the intake was not affected. Ammonia treatment did not improve the degradation rate of rice straw in the rumen, but remarkably increased the rice straw digestibility and intake by sheep. Making silage of rice straw did not affect its rumen degradation rate, but the digestibility and its take by sheep were greatly improved especially when a little molasses together with Lactobacillus were supplemented. Degradation rate of rice straw in the rumen measured by nylon bag technique was influenced by various treatments but did not appear to coincide with digestibility by sheep. This would be due to the fact that feed intake affect digestibility as well as the degradation in rumen. Therefore, it can be said that making silage with some molasses and Lactobacillus is one of the easest way of using rice straw for animal feed. (Key words : NaOH, Digestibility, Silage, Molasses )
A study was conducted to estimate nutritive value of forage sources used in deer diets. Bags containing feedstuffs were incubated four times for periods up to 72 hours in two chemostats filled with ruminal fluid from deer or cattle. Parameters estimated were water-soluble and $65{\mu}m$ filterable plus insoluble digestible fractions(No. = 4) and extent of disappearance(No. = 8) of feed neutral detergent fiber(NDF). Among tested feeds, the ranking of values of these parameters were soybean hulls > alfalfa pellets > corn cobs or rice straw > cottonseed hulls or rice hulls > sawdust, indicating that soybean hulls and alfalfa pellets were more fermentable than other agricultural residues. It is possible to utilize variability among tested feeds of fraction, disappearance rate and (or) extent of DM and NDF when these feeds are used as roughage sources in deer diets.
The colonization pattern and extent of decay produced in paddy stubble by soil inhabiting mycoflora were done by using nylon net bag technique. Among the three methods used for isolation of fungi, dilution plate technique recorded the highest number of fungi followed by damp chamber and direct observation method. Nutrient availability and climatic conditions (temperature, humidity and rainfall) influenced the occurrence and colonization pattern of fungi. Maximum fungal population was recorded in October ($48.99{\times}10^4/g$ dry litter) and minimum in May ($11.41{\times}10^4/g$ dry litter). Distribution of Deuteromycetous fungi was more in comparison to Zygomycetes, oomycetes and ascomycetes. In the early stage of decomposition Mucor racemosus, Rhizopus nigricans, Chaetomium globosum and Gliocladium species were found primarly whereas at later stages of decomposition preponderance of Aspergillus candidus, Torula graminis, Cladosporiun cladosporioides and Aspergillus luchuensis was recorded.
An experiment was conducted with three ruminally and intestinally cannulated non-lactating cows of Finnish Ayrshire breed, to assess the ruminal degradation characteristics of oat (Avena sativa), rye (Secale cereale) and rice (Oryza sativa) straw by the nylon bag technique, and the subsequent post-ruminal degradation of their rumen-undegraded residues by using the mobile bag technique, respectively. The straw samples were untreated or treated with aqueous $NH_3$ or with urea solution in cold or hot water. The untreated straw samples were milled or chopped, and the treated straw samples were chopped. The constant values a, b, and c were computed according to the exponential equation, where a = intercept of degradation curve at time 0, b = potentially degradable material, c = rate of degradation of band (a+b) = maximum potential degradability (asymptote). It was found that nitrogen contents of chemically treated straw were markedly increased by both $NH_3$ and urea treatments. Milling the samples attributed to a remarkable loss at 0 h incubation time as compared to chopping of the respective samples. However, chemical treatment markedly improved the b value and the subsequent (a+b) values for dry matter, organic matter, neutral-detergent fiber, and acid-detergent fiber of the samples. Furthermore, temperature of the water used in the urea solutions was considered essential, since urea in hot water rather than in cold water seemed to enhance the overall degradability. The disappearance of rumen-incubated straw residues from the mobile bags ranged from 4.5 to 9.6% for the parameters measured. On average, the OM disappearance from bags was clearly higher for the residues of urea treated straw compared to those of ammonia treated straw, but the disappearance of NDF tended, however, to be higher on the ammonia treatment.
