Induction of Lysozyme Gene Expression During Involution of Mouse Mammary Gland

  • To understand molecular mechanisms of mouse mammary gland involution, clones were isolated by differential screening of a cDNA library. Partial sequences of a clone showed 100% identity to cDNA sequences of mouse lysozyme P gene. Northern analysis was performed to examine expression levels of lysozyme mRNA in mammary gland at several physiological states. Expression of lysozyme gene was induced at involution day 5 compared with lactating stage. High levels of lysozyme mRNA were also detected at virgin tissues. Two types of separate genes, P and M lysozyme, have been known in mouse, and we found that both lysozyme P and M genes were expressed in mammary tissues by reverse transcriptase-polymerase chain reaction. The lysozyme enzyme activity determined by lysoplate assay was also higher in involuted mammary tissues compared with lactating tissues, showing a similar trend to its mRNA levels. Lysozyme is an antimicrobial protein and involved in host defense mechanism. The increase in lysozyme gene expression may help to prevent microbial infection during mammary gland involution at which stage the residual milk in the mammary gland provides good nutritional sources for microbial growth.

Nutritional Value of a Heterotrichous Ciliate, Fabrea salina with Emphasis on Its Fatty Acid Profile

  • Fabrea salina is a hypersaline ciliate having importance as a live food source for juvenile stages of aquatic animals including smaller invertebrates. The analysis of this ciliate for proximate and biochemical composition was carried out. The moisture, protein, fat, carbohydrate and ash content of F. salina from natural sources were 86.66$\pm$0.380, 56.66$\pm$0.494%, 36.66$\pm$0.614%, 1$\pm$0.073% and 4$\pm$0.182%, respectively. Gas chromatographic analysis (percent area below the curve) revealed that the presence of oleic acid was higher over other fatty acids in both natural and cultured F. salina. The absolute content of oleic acid was higher in natural (18.91% area) than in the cultured (10.74% area) F. salina. Linoleic and linolenic acids were also among major fatty acids with the percentage area of 16.29 and 14.58, respectively. The number of fatty acids in cultured Fabrea was less as compared to the natural ones and the oleic acid was followed by palmitic acid, palmitoleic acid, linoleic acid and stearic acid.

한국인의 식이섬유 섭취 실태 (Dietary Fiber Intake of Korean)

  • According to the data published in 1990s on the intake of dietary fiber(DF) of Korean, DF intake of Korean has been gradually decreasing in the last two decades. Mean daily intake of DF in 1990s is estimated to be in the rage of 17-20g per capita and over seventy five percent of the subjects examined appeared to consume DF less than 20g per day. The major food sources of Df in Korean diet include cereals, vegetables, fruits, seaweeds and seasonings. The present level of DF intake of Korean is lower than the tentatively recommended minimum intake of DF. It is predicted that, if the present trend of change on food consumption pattern is maintained, the gradual increases in the incidence of chronic degenerative diseases will be continuing. Therefore, the beneficial effects of DF on health care and disease control should be emphasized through the nutritional education and high level consumption of DF needs to be strongly recommended in the dietary guideline. In order to raise the daily consumption of DF, more whole grain cereals need to be used as a part of staple and more frequent intake of legumes and seaweeds is required as well as the development of high-fiber diet recipes and natural high-fiber products.

Estimated dietary isoflavone intake among Korean adults

  • This study estimated the isoflavone intake level in Koreans using Food Frequency Questionnaire and analyzed related variables. The results showed that the average daily intake of isoflavone in adults was shown as 23.1 mg. The isoflavone intake level at 50 percentile was 16.9 mg ($0{\sim}90\;mg$), and 10% of adults took almost 50 mg of isoflavone a day and 10% took about 5 mg a day. The major food sources for isoflavone in Koreans were in the order of soybean, soybean paste, soy milk, soybean curd (tofu), and bean sprouts; the intake was different depending on age, educational background, occupation, economic standard, and family type. The result showed higher isoflavone intake levels in the group over 30 years old and the highest isoflavone intake in subjects working in farming/fishery, followed by housemakers. According to the differences by families the families with elderly members showed 50% higher isoflavone intake than young families with friends or siblings. Depending on related ecological variables, therefore, various nutrition education programs should be developed for a variety of intakes of soybean foods, along with easy and simple cooking methods as parts of continuous research.

도시가계조사 자료를 이용한 소득계층별 식생활 패턴분석 (Analysis of Food Consumption Patterns by Income Levels Using Annual Report on the Family Income and Expenditure Survey)

  • Korea has not tried any food consumption survey so far except the national nutrition survey, which does not show food consumption patterns of different income stratas. The results of the family income and expenditure survey(FIES) by the national statistical office can be precious sources which show household food consumption patterns due to large, random. Samples, year-round survey period and socioeconomic background data. This study analyzed the FIES data to find out food consumption patterns including nutrient intakes and frequently consumed foods by households among different monthly income levels. Big difference was found in food consumption patterns among the quartile-income groups especially the amount of consumed foods, food expenditure, and nutrient intakes. For every food item, the higher the monthly invomr, yhr motr og goof yhry vondumrf. The monthly food expenditure of higher higher income strata was composed with higher percentage of relatively expensive foods compared to other stratas. Nutrient intake levels of lower income strata were 50-60% of the RDA, which showed the necessicity of food assistance programs for those high risk groups to complement the nutritional difficiency. Isolation, Identification and Optimal Culture Conditions of Streptomyces albidoflavus C247 Producing Antifungal Agents against Rhizoctonia solani AG2-2

  • Streptomyces albidoflavus C247 was isolated from the soil of the Gyeongsan golf course in Korea. Physiological, biochemical and 16S rDNA gene sequence analysis strongly suggested that the isolate belonged to Streptomyces albidoflavus. Preliminary screening revealed that the isolate was active against fungi and bacteria. Self-directing optimization was employed to determine the best combination of parameters such as carbon and nitrogen source, pH and temperature. Nutritional and culture conditions for the production of antibiotics by this organism under shake-flask conditions were also optimized. Maltose (5%) and soytone (5%) were found to be the best carbon and nitrogen sources for the production of antibiotics by S. albidoflavus C247. Additionally, 62.89% mycelial growth inhibition was achieved when the organism was cultured at $30^{\circ}C$ and pH 6.5. Ethyl acetate (EtOAc) was the best extraction solvent for the isolation of the antibiotics, and 100 ${mu}$/ml of EtOAc extract was found to inhibit 60.27% of the mycelial growth of Rhizoctonia solani AG2-2(IV) when the poison plate diffusion method was conducted.

Evaluation of Vitamin $B_{6}$ Status and Korean RDA in Korean College Students Following a Uncontrolled Diet

  • The vitamin $B_{6}$ status of 49 healthy college student (women, aged 20-26 y) was estimated for evaluation of vitamin $B_{6}$ status and the Korean Recommended Dietary Allowance (RDA) for vitamin $B_{6}$. The average daily vitamin $B_{6}$ intake of the subjects was 0.86 $\pm$ 0.289 mg/d or 61.43 $\pm$ 24.10% of Korean RDA. The average ratio of vitamin $B_{6}$ intake to daily protein intake was 0.014 $\pm$ 0.003 mg/g protein. Foods from animal and plaint sources provided 34.25 $\pm$ 18.62% and 65.78 $\pm$ 18.72%, respectively, of total vitamin $B_{6}$. Plasma pyridoxal 5'-phosphate (PLP) concentration was significantly (p<.01 - p<.001) positively correlated to intakes of all other nutrients except vitamin C. However, no significant correlation was found between plasma PLP and nutrient intake. Vitamin $B_{6}$ intake only tended to have a positive correlation with plasma PLP concentration. Plasma total cholesterol was correlated to plasma PLP concentration (p<.05). Plasma PLP had no correlation with levels of glucose, triglyceride, and albumin. These results confirm that the present Korea RDA for vitamin $B_{6}$ of 1.4mg/d based on 0.02 mg/g protein is adequate.

  • Aspergillus fumigatus IFO 5840을 공시균주로 하여 cytosine deaminase의 생성 최적조건을 밝히기 위하여 각종 탄소원, 질소원과 같은 영양소 및 배양 온도, 초기 pH, 배양시간, 통기성의 영향을 조사하였다. 본 공시균의 cytosine deaminase의 생성을 위한 최적 배지의 조성은 2% 포도당, 1% 효모추출물로, 배지의 초기 pH는 8.5이었다. 배양 최적 조건은 $30^{\circ}C$에서 28시간 진탕 배양이 양호했다. 이러한 최적조건에서 500ml 용량의 진탕 flask에 배지 100ml를 넣었을때 효소생성이 최고에 달하였다. 공시균주의 세포내 효소의 생성이 무기질소원에 의해 억제 되었으나 유기질소원인 yeast extract에 의해서는 촉진되었다.

Increased Amino Acid Absorption Mediated by Lacticaseibacillus rhamnosus IDCC 3201 in High-Protein Diet-Fed Mice

  • The use of dietary protein products has increased with interests in health promotion, and demand for sports supplements. Among various protein sources, milk protein is one of the most widely employed, given its economic and nutritional advantages. However, recent studies have revealed that milk protein undergoes fecal excretion without complete hydrolysis in the intestines. To increase protein digestibility, heating and drying were implemented; however, these methods reduce protein quality by causing denaturation, aggregation, and chemical modification of amino acids. In the present study, we observed that Lacticaseibacillus rhamnosus IDCC 3201 actively secretes proteases that hydrolyze milk proteins. Furthermore, we showed that co-administration of milk proteins and L. rhamnosus IDCC 3201 increased the digestibility and plasma concentrations of amino acids in a high-protein diet mouse model. Thus, food supplementation of L. rhamnosus IDCC 3201 can be an alternative strategy to increase the digestibility of proteins.

Enzymatic preparation and antioxidant activities of protein hydrolysates from hemp (Cannabis sativa L.) seeds

  • Hemp (Cannabis sativa L.) seeds have recently been attracting attention as a new high-value-added food material owing to their excellent nutritional properties, and research on the development of functional food materials using hemp seeds is actively progressing. This study aimed to evaluate the antioxidant properties of hemp seed protein hydrolysates. Protein hydrolysates were prepared from defatted hemp seed powder (HS) by enzymatic hydrolysis using five different proteases (alcalase, bromelain, flavourzyme, neutrase, and papain). 2,4,6-trinitrobenzene sulfonic acid (TNBS) assay and SDS-PAGE analysis revealed that HS showed a high degree of hydrolysis after treatment with each enzyme except papain. The total polyphenol content of the protein hydrolysates (<3 kDa) and the RC50 values obtained from two different antioxidant tests showed that alcalase hydrolysate (HSA) had a relatively high level of antioxidant capacity. In addition, treatment with HSA (25-100 ㎍/mL) significantly inhibited linoleic acid peroxidation. These results suggest that hemp seed protein hydrolysates are potential sources of natural antioxidants. Future studies will focus on the identification of active peptides from HSA.
