순환식 상추 양액재배시 양액재활용 기술 (Reusing Techniques of Nutrient Solution for Recycling Hydroponic Culture of Lettuce)

  • 수경재배에서 양액의 재활용이 상추의 생육이나 수량에 어떠한 영향을 미치는지 알아보고자 Control, 양액보충, EC조절, 양액검정보정, 양액검정보정+humus첨가 등의 5처리를 두어 실험을 수행한 결과는 다음과 같다. 양액의 pH에서 양액검정보정구와 양액 검정보정.+Humus참가구가 4차재배까지 7.0이하로 안정된 경향을 나타내었고, EC는 양액보충구가 재배횟수가 늘어날수록 현저히 떨어졌고 다른 처리는 대조구와 유사한 경향이었다. 생육 및 수량은 양액검정보정 처리가 양액내 무기이온들의 균형으로 대조구와 유사한 경향을 나타내었으며 양액보충구 및 EC조절구는 재배횟수가 늘어날수록 떨어지는 경향이었다. 양액의 무기성분중 N $O_3$-N와 N $H_4$-N는 재배후 처리에 관계없이 농도가 크게 떨어졌고 특히 Ca과 Mg은 모든 처리구에서 재배횟수에 관계없이 재배전에 비해 재배 후에 양액내에 축적되는 경향을 보였다. 상추엽중 무기성분함량은 T-N와 P$_2$ $O_{5}$ 의 경우 1차재배후 양액검정보정+Humus 첨가구에서 다른 처리에 비해 함량이 적었고 재배횟수가 늘어날수록 처리간에 차이가 없었다. K과 Mg은 재배횟수나 처리간에 차이가 없었고, Ca의 경우 양액검 정보정+Humus 첨가구에서 1차와 2차재배 후 다소 많았으나, 재배횟수가 늘어날수록 처리간에 차이가 없었다.

순환식 수경재배시 무기이온 조절이 Single-Node Cutting 장미의 양분흡수, 생육 및 품질에 미치는 영향 (Effect of Mineral Nutrient Control on Nutrient Uptake, Growth and Yield of Single-Node Cutting Rose Grown in a Closed Hydroponic System)

  • 본 실험은 장미 식물공장에서 single-node cutting 'Versillia'의 양분흡수 특성을 구명하고 순환식 수경재배 시스템에 적합한 배양액 내 무기이온 조절방식을 구명하고자 수행하였다. 실험 기간동안 각 처리별 배양액 내 무기이온함량 변화를 살펴본 결과 EC 제어구의 경우 $NO_3$-N은 생육 후반이 되면 그 함량이 적정 범위 이상으로 증가 하였고 P와 Mg은 감소하였으며 배양액 첨가구는 전체적인 배양액 내 무기이온 함량이 증가하였다. 이에 비해 무기이온 제어구는 생육 기간동안 근권 내 적정 범위를 유지하였다. 광합성 효율을 나타내는 지표 중 하나인 Fv/Fm는 무기이온제어구와 배양액 첨가구에서 높았고 절화장, 생체중 등은 무기이온 제어구에서 높았다. 그러나 뿌리의 활성 정도를 측정해본 결과 무기이온 제어구에서 높았으나 배양액 첨 가구에서는 생육 초기에 배양액 내 pH 변화폭이 커서 가장 낮게 나타났다. 위의 결과를 종합하였을 때, 무기이온 조절(다량 미량원소 제어구와 다량원소 제어구)을 통해 배양액을 관리하는 것이 생육 전 기간 동안 근권 환경을 적절히 유지해 줄 수 있었고 이에 따라 절화 품질도 향상되므로 장미(single-node cutting) 순환식 수경재배에 적합한 근권 환경 제어방식이라고 할 수 있다.

암면배지내 인공 zeolite 형 각종 이온교환제 처리가 온실멜론의 이온교환능에 미치는 영향 (Effects of Artificial Zeolite and Various ion Exchangers Supplemented in Rockwool on the Ion Exchangability of Muskmelon)

  • 본 실험은 인공 zeolite 및 각종 이온교환제를 암면슬라브내에 첨가하므로서 완형능력을 부여하여 온실멜론의 암면재배에 대한 이온 교환능 및 흡착능의 작용기작을 비교검사하고자 수행하였다. 1. 이온교환제의 처리에 따른 멜론의 생육특성중 초장은 대조구인 원시 1/2단위에서 124cm인데 대해 Ca형 인공zeolite에서는 131,2cm로 켰으며 경 및 엽의 생체중에서도 같은 경향이 보였다. 2. 과실중은 인공 zeolite를 첨가한 경우에 무거워지는 경향을 나타냈으며, 당도 및 외관은 처리에 따른 차이가 거의 보이지 않았다. 3. Zeolite의 첨가에 의해 배양액의 pH나 이온교환능을 안정시키는 것이 가능하고, 양분흡수를 순조롭게 하며, 양분흡수의 증가를 촉진하고 생육촉진도 기대할 수 있었다. 이상의 결과는 인공 zeolite를 암면배지내에 첨가하므로서 이온 보유능, 흡착능을 갖게하여 배지내의 생육환경이 안정되고 암면배지는 인공 zeolite 첨가로 배지의 CEC를 대폭적으로 개선시키는 효과가 컸다.

국내 시판 이유식류의 영양성분규격 설정 방안 (Study on the Establishment of Nutrient Requirements for Commercial Supplementary Foods for Infants and Young Children)

  • This study was conducted to evaluated the nutrition quality of the commercial supplementary foods for infants and young children and to seek a solution to the establishment of standards of nutrient requirements for supplementary foods in Korea. Information on food ingredients, nutrient contents, claims about usefulness of food components and instructions for feeding preparation were obtained from the labels of 33 commercial supplementary foods manufactured by 4 different domestic companies. According to the standard of supplementary foods for infants and young children described in the Korean Food Code, the commercial supplementary foods were categorized into two different types, weaning food and baby food. All the commercial weaning foods were in powder form and mainly composed of cereals, whereas all the baby foods were mainly composed of fruits in the form of canned juice. The weaning foods contained more nutrients than the baby foods did, and the nutrient levels of the weaning foods expressed as nutrient density on energy basis were higher than the RDA for infants aged 5 to 11 months, suggesting that the commercial weaning foods provide adequate amounts of nutrients. If one followed the instructions for feeding preparation appearing on the label, however, recommended amounts of intake of the weaning foods would provide too much energy as well as nutrients. There were many differences in nutrient standards of weaning foods between the Korean Food Code and Codex international food standard. In conclusion, the establishment of standards for nutrient requirements for the supplementary foods requires significant scientific studies on what nutrients are the most inadequate in Korean infants and young children feeds and what levels of nutrients should be added to the foods in order to supplement their nutrition. In addition, it is very important to have a strong scientific basis to support our standard when discrepancies exist between our standard and the international standard. 순환식시스템을 이용한 토마토 양액재배에서 배액의 재순환 방법 (Supplement Method of Drained Solution in Tomato Cultivation Using Recycling Systems)

  • 본 실험은 토마토의 순환식 시스템 양액재배에서 배액의 재순환 시 안정된 pH와 균형적인 양분 공급 방법을 구명하고자 수행되었다. 배액의 pH나 무기성분을 교정하지 않는 구(대조구), EC측정 및 양분 분석 교정구를 처리내용으로 두었다. 초기생육인 초장은 처리간에 차이가 없었으나 생체중 및 건물중은 양분분석구가 대조구에 비해 무거웠다. 후기생육에서는 양분분석구가 대조구에 비해 과중이 무겁고, 착과수가 많아 수량이 증가하였다 배액 EC는 대조구에 비해 EC측정구와 양분분석구가 전반적으로 낮았다. 배액의 pH는 전 생육기간에서 양분 분석구 및 EC측정구는 6.2∼6.5를 유지하였으나 대조구는 지속적으로 높아져 후기에는 7.2정도 되었다. 배액의 무기성분에서 N, P 및 K의 함량은 모든 처리구에서 셍육이 경과함에 따라 적어졌으며, 대조구에 비해 양분분석구가 적었다 Ca 및 Mg의 함량은 모든 처리구에서 생육초기에는 급격히 감소하였으며 생육후기에는 앙분 분석구와 EC측정구는 적은 상태로 유지되었으나 대조구는 급격히 증가하는 경향이었다. 이상의 결과에서 순환식 시스템을 이용한 토마토의 양액재배시 배액의 양분을 분석한 후 교정하는 것이 생육에 효과적이었다.

  • I. Subject of the Study: Studies on the economical nutrition supplement of cereal foods for the improvement in our notional eating habits. II. Purpose and Importance of the Study: 1. Our nation is confronted with the situation that the rice, a principal food, short of some essential amino acids, lysine and threonine, leads to imbalanced meals insufficient in the nutrient of protein, to bring many difficulties in the elevation of our national physique. 2. The shortage of even the rice imperfect in the nutrient of protein makes the import of lots of foreign rice inevitable. It is considered that the protein supplement and decrease in the consumption amount, of rice, is a serious key to the solution of our food difficulty, and then a way of the proetin supplement of rice through the addition of essential amino acid is to be rarely applied in the view of the our present finance and situation. 3. In the present experiment, therefore, it aims to the suggestion of an aspect of the improvement in our national eating habits guiding in the nutrition elevation which our nation can afford economically through the development of first, a way of the protein supplement by the mixture of cereals producted plentifully in our country, and second, a way of the decrease in the consumption amount and the improvement in the nutrition of rice through the substitution of the other cereals for rice. III. Contents of Scope of the Study: 1. Objects of the study: Objects of the study are the following three items; a) The nutrition supplement of rice through the mixture of cereals. Our nation makes mainly rice as a principal food, but practically many kinds of cereal are produced in our country. They contain different levels and qualities of each nutrient and they are different from one another in the kinds of essential amino acid consisting protein. For that reason, the mutual complement efficeincy of insufficient nutrients is observed through the mixture of cereals. b) The nutrition supplement of rice through the addition of superior protein sources to rice, a principal food. The development of rice as superior foods in the sense of nutrition is conducted through the risement in protein quality by the addition of protein sources in good quality, for example, fish flour (anchovy flour), egg powder, milk powder, and so on, and through the supplement of vitamins and minerals. c) The decrease in the consumption amount of rice through the substitution of the other cereals for rice, as a principal food. The compensation for the short amount of rice is made by the reduction in the consumption of rice through the discovery of a way of substitution of the other cereals for rice, as a principal food and of the settlement of problems in nutrition and finance subsequent to this. 2. Contents of the study: a) An ideal mixture-ratio of cereals is established for rats by feeding mixed foods(rice-barely or rice-wheat) containing 5%, 15%, 25%, 35% and 45% level of either barely or wheat. b) The nutritive value is determined in the whole subsititution of other foods for rice, and then, a way of the complement of over and under nutrients is devised. c) The ideal combination is investigated for rats through feeding mixed foods of main food, rice and supplement foods of protein sources, soy bean, fish flour, egg powder and milk Powder. d) According to results from the above three experiments, the concise functional test for men and the examination of economical property are made. 3. Scope of the study: a) The observation of the effect of each diet on the growth rate for rats. The growth rate of rats was observed for 15 groups of mixed foods of a main food, rice, and wheat flour, barley powder or soy bean powder, respectively, and 12 groups of wheat flour diets supplemented with $1{\sim}3%$ milk powder, and rice or wheat flour diets supplemented with 5% of milk powder, egg powder, fish flour or soy bean powder, respectively. b) The determination of food consumption. The food consumption was determined at weekly intervals for 27 kinds of diet described in a) item. c) The determination of food efficiency rate. The food efficiency rate for each diet was determined by calculation from the gained body weight and the food consumption amount at the same intervals described in b) item. d) The determination of protein efficiency rate. The protein efficiency rate for each diet was determined by calculation form gained body weight and the protein amount of the food consumption amount at the same intervals described c) item. e) The determination of the body component. The hematocrite and hemoglobin levels in the blood, total nitrogen in the serum, blood sugar, and lipids and glycogen in the liver were determined. f) The observation of nitrogen balance. As a means of the observation of nitrogen balance, the total nitrogen in the urine was determined. g) The analysis of economical property. The economical property was analyzed as the gained body weight to the amount equivalent to one won through the conversion of the food consumption amount into money. h) The functional test for men. The concise functional test for men was made in order to establish if the best diet for experimental animals can be applied to men. IV. Results of the Study: The national food product plan, nationwide nutritive enlightment and the improvement activities in our country eating habits, especially, mixed and powder food problems are to be significantly referred, and the following results must be applied. a) In the mixed foods of cereals, the mixed food of the rice-barley containing $5{\sim}15%$ level of barley is best in terms of nutrition. b) The addition of superior protein sources, egg, Bilk, soy bean, or fish, respectively to either rice or wheat flour makes a great risement in the nutritive value. c) The animal protein is more effective in the elevation of nutritive value of cereals. d) Rice takes the most nutritive operation and has the highest preference, among rice, wheat flour and barley. e) Wheat flour is more economical than rice in evaluation of the gained body weight to the regular money, and the addition of fish or soy bean is more economical than that of any other supplement food. But the above results are true of the range of nutrition and economical property. f) The study on the nutrition composition and barley will lead to the improvement in our national eating habits as mixed food of the rice-barley containing $5{\sim}15%$ level of barley is more nutritive. g) This study on the nutrition only for the growing animal can not be considered as a perfect and entire evaluation. Consequently, the perfect data for our national nutrition can be obtained from the experiment similar to this for the much longer period examining, in details, the growth rate, change of physical strength, mental and bodily change, average life span, and resistance ability to infectious diseases.

양액재배 시 selenium의 첨가가 basil의 저장에 미치는 영향 (Effect of Selenium on Storability of Basil (Ocimum basilicum) in Hydroponics)

  • 본 연구는 수경재배 바실의 MA 저장시 적정 저장 온도를 선정하고 기능성 배가를 위해 양액 내 첨가한 Se이 바실의 저장성에 미치는 영향을 조사하고자 수행하였다. 바실은 European Vegetable R & D Center에서 개발한 허브 배양액 1배액을 사용하여 담액 순환식으로 재배하였으며, Se을 sodium selenate($Na_2SeO_4$)의 형태로 수확 3주 전 2, 4, 6, $8mg{\cdot}L^{-1}$의 농도로 양액 내 첨가하였다. 바실의 MA 저장 시 $5^{\circ}C$에서는 저온장해가 발생하여 전해질 용출량이 크게 증가하였다. 그러나 Se 처리에 의해 저장기간을 무처리구에 비해 3일간 연장시킬 수 있었다. $10^{\circ}C$ 저장 시 $5^{\circ}C$보다 생체중 감소율이 적었으며, 비타민 C 함량도 더 높았다. Se 처리는 작물 내 비타민 C 함량을 증가시키고 전해질 용출량은 감소시켰다. 작물 내 Se 함량은 Se 처리 농도에 따라 증가하였으며 저장 전 $2mg{\cdot}L^{-1}$ 처리구에서는 $67{\mu}g{\cdot}g^{-1}DM$, $8mg{\cdot}L^{-1}$ 처리구에서는 $423{\mu}g{\cdot}g^{-1}DM$을 함유하였다. 이상에서 바실의 MA 저장 시 저온장해를 줄이고 여러 가지 내적 품질을 유지하기 위해서는 $10^{\circ}C$ 저장 온도가 적합하다. 또 보다 안전하게 Se 함유 바실을 섭취하기 위한 적정 Se 처리농도는 $Na_2SeO_4\;2mg{\cdot}L^{-1}$ 이하가 효과적이라고 생각된다.

Effect and Nutrient Content of Fermented Aloe Saponaria as Pigs Feed Additive Food

  • Aloe gel layer is well known as raw materials of medicines and cosmetics due to their antioxidant and anti-inflammatory properties. In aloe gel extracting process, the outer part of the leaf was removed. It contains high quality of fiber and many nutrients. However, this part is thrown away and generally used as fertilizer. The purpose of this research was to examine the important nutrient of Aloe saponaria. Moreover, the feasibility of using aloe as a dietary supplement by feeding fermentation treatment of aloe was investigated. To do this, the aloe leaf was divided into several parts including leaf skin, bottom of the leaf, tip of the leaf, middle of the leaf, and leaf flesh. Then the saponin content were analyzed from each part. The extraction method was used to clarify the saponin content. The aloe then fermented to improve it benefit. The fermented Aloe then given as dietary food to group of pig. Finally, the appropriate feed level was determined and the pork meat quality was analyzed. The extraction of saponin shows that the highest concentration of saponin located on the skin of the leaf. The feeding experiment shows that there is no significant difference in pig growth without aloe dietary food and groups with aloe as dietary food. It was conclude that fermented aloe can replace the pigs normal feeder as an alternative feeding solution.

고추재배를 위한 시설하우스 폐양액의 재활용 (Recycling of Hydroponic Waste Solution for Red Pepper (Capsicum annum L.) Growth)

  • 본 연구에서는 시설하우스 폐양액이 고추의 생육과 양분 흡수에 미치는 영향을 평가하고 폐양액 관주시 부가적으로 얻을 수 있는 관개 효과를 산출함으로써 폐양액의 토양 시비에 따른 농업적 가치를 평가하였다. 시설하우스 폐양액의 토양 관주시 Ca, Mg K 등 염류의 증가로 인하여 pH와 EC는 상승하였고, 총질소는 암모늄태 질소와 질산태 질소의 증가와 함께 처리 후 100 mg $kg^{-1}$ 이상 증가하였다. 고추 생육은 화학비료 70% 및 폐양액 30% 혼합처리구와 화학비료 50% 및 폐양액 50% 혼합처리구에서 양호하였으며 양분 흡수도 동일한 경향을 나타내었다. 고추 수확량은 화학비료 70% 및 폐양액 30% 혼합처리구에서 가장 높은 것으로 조사되었다. 1일 발생되는 폐양액(288 L 10 $a^{-1}\;day^{-1}$)을 토양에 시용하여 증발산되는 수분을 보충할 경우 최고 $409.86m^2$의 면적을 관개할 수 있는 것으로 산출되었다. 이상의 결과를 통해 폐양액의 토양처리는 화학비료 대체 효과와 폐양액의 정화 효과를 동시에 얻을 수 있으며 적정량 시용시 주변 환경에 미치는 영향을 최소화하면서 지속적인 처리가 가능할 것으로 판단되었다.

Expression and Purification of Biologically Active Human Bone Morphogenetic Protein-4 in Recombinant Chinese Hamster Ovary Cells

  • Bone morphogenetic protein-4 (BMP-4) is considered to have therapeutic potential for various diseases, including cancers; however, the high expression of biologically active recombinant human BMP-4 (rhBMP-4) needed for its manufacture for therapeutic purposes has yet to be established. In the current study, we established a recombinant Chinese hamster ovary (rCHO) cell line overexpressing rhBMP-4 as well as a production process using 7.5-l bioreactor (5 L working volume). The expression of the mature rhBMP-4 was significantly enhanced by recombinant furin expression. The combination of a chemically defined medium and a nutrient supplement solution for high expression of rhBMP-4 was selected and used for bioreactor cultures. The 11-day fed-batch cultures of the established rhBMP-4-expressing rCHO cells in the 7.5-L bioreactor produced approximately 32 mg/l of rhBMP-4. The mature rhBMP-4 was purified to homogeneity from the culture supernatant using a two-step chromatographic procedure, resulting in a recovery rate of approximately 55% and a protein purity greater than 95%. The N-terminal amino acid sequences and N-linked glycosylation of the purified rhBMP-4 were confirmed by N-terminal sequencing and de-N-glycosylation analysis, respectively. The mature purified rhBMP-4 has been proved to be functionally active, with an effective dose concentration of $EC_{50}$ of 2.93 ng/ml.
