Bacillus licheniformis was grown in minimal nutrient medium containing 1% (w/v) of distillers dried grain with soluble (DDGS), palm kernel meal (PKM), wheat bran (WB) or copra meal (CM), and the enzyme activity of endoglucanase, ${\beta}$-glucosidase, xylanase and reducing sugars was measured to investigate a possibility of using cost-effective agricultural residues in producing cellulolytic and hemicellulolytic enzymes. The CM gave the highest endoglucanase activity of 0.68 units/mL among added substrates at 48 h. CM yielded the highest titres of 0.58 units/ml of ${\beta}$-glucosidase, compared to 0.33, 0.23, and 0.16 units/mL by PKM, WB, and DDGS, respectively, at 72 h. Xylanase production was the highest (0.34 units/mL) when CM was added. The supernatant from fermentation of CM had the highest reducing sugars than other additional substrates at all intervals (0.10, 0.12, 0.10, and 0.11 mg/mL respectively). It is concluded that Bacillus licheniformis is capable of producing multiple cellulo- and hemicellololytic enzymes for bioethanol production using cost-effective agricultural residues, especially CM, as a sole nutrient source.

To establish an efficient acclimation system for regenerated plantlets of soybean, we used various media with hydroponic nutrient solutions before regenerants were transplanted into soil. The hydroponic nutrient solution was essential for the survival of the plantlets. The vermiculite with nutrient solution at pH 5.5 was found to be the best medium with 97-100% survival rate and better growth of regenerants plantlets. Regeneraed grew best in the following order of solutions: Yoshida solution, modified Yoshida solution, SoyI, Soy II, and MS medium. However, Soy I solution (EC 2.9 mS/cm), developed by the Honam Agricultural Research Institute proved to be the most effective for acclimation in terms of the time required for vigorous growth and economical use of chemicals.
Microalgae is considered as one of environmentally sustainable and potential feedstocks to produce biodiesels. However, recent studies on life cycle assessments (LCA) of microalgal buidiesels have shown that energy requirement is not small to produce biodiesel from microalgae, especially during cultivation stage. The costs for carbon sources, nutrients like nitrogen or phosphorous, and water for cultivation can contribute up to 80% of the total medium costs. In the present article, recent trends on the utilization of several promising nutrient sources such as municipal wastewaters, organic fertilizers, combustion exhaust emissions and organic solid wastes were reviewed, and the potential strategies to be used as substitutes of artificial culture media, especially for the biodiesel production, were discussed.
Microbial oxidation of D-sorbitol to L-sorbose by Acetobacter suboxydans is of commercial importance since it is the only biochemical process in vitamin C synthesis. The main bottleneck in the batch oxidation of sorbitol to sorbose is that the process is severely inhibited by sorbitol. Suitable fed-batch fermentation designs can eliminate the inherent substrate inhibition and improve sorbose productivity. Fed-batch sorbose fermentations were conducted by using two nutrient feeding strategies. For fed-batch fermentation with pulse feeding, highly concentrated sorbitor (600g/L) along with other nutrients were fed intermittently in four pulses of 0.5 liter in response to the increased DO signal. The fed-batch fermentation was over in 24h with a sorbose productivity of 13.40g/L/h and a final sorbose concentration of 320.48g/L. On the other hand, in fed-batch fermentation with multiple feeds, two pulse feeds of 0.5 liter nutrient medium containing 600g/L sorbitol was followed by the addition of 1.5 liter nutrient medium containing 600g/L sorbitol at a constant feed rate of 0.36L/h till the full working capacity of the reactor. The fermentation was completed in 24h with an enhanced sorbose productivity of 15.09g/L/h and a sorbose concentration of 332.60g/L. The sorbose concentration and productivity obtained by multiple feeding of nutrients was found to be higher than that obtained by pulse feeding and was therefore a better strategy for fed-batch sorbose fermentation.
Sporulation of Streptomyces griseus NRRL B-2682 occurs at 26~28 hours during incubation while shaking at 3$0^{\circ}C$ in a defined medium. The time of sporulation is the same when the levels of each nutrient is increased ten times. The levels of the carbon, nitrogen and phosphate source are at a high level when sporulation begins. Sporulation of S. griseus B-2682 is clearly not caused by nutrient deprivation. It appears that a clock mechanism is involved instead. Once spores are germinated, the time of sporulation is programmed. Sporulation of S. griseus is repressed by high levels of casein hydrolysate. A study of the effect of individual amino acids revealed that L-valise when added to the normal growth medium causes an inhibition or repression of sporulation without affecting growth.
For investigating the effect of amount of applied fertilizer with pig and chicken manures in the field, the number of soil bacteria were counted on the full strength conventional nutrient broth (NB) medium and its 100-fold dilution (DNB) medium. From the control soil samples without organic amendments, the number of bacteria on DNB medium was 5 to 10 times higher than that on NB medium. However, population density on NB medium was higher than on DNB medium from the treated soil with 60 and 120 Mg/㏊ for 3 years. Most of isolates from DNB plates did not grow on the NB plates. There were only grown on the 100-fold dilution medium, so it was called as DNB organisms. The DNB organisms were occupied as dominant group over 60% fo isolates in control soil. However, their occupation rates were rapidly decreased in the treatment soil with pig and chicken manures above 60 and 120 Mg per ha. These DNB organisms (oligotrophs) were significantly low population desities in the treatment soil with organic materials.
This study was conducted to identify the growth characteristics of melon cultivated with mixture medium of scoria and perlite in capillary system. The growth and fruit characteristics of all cultivars cultivated under the nutrient solution capillary supplying system were comprehensively favorable. Fresh fruit weight of the three cultivars was similar to weight criterion (1.8 - 2.0 kg/fruit) of melon. In melon of non-net type ‘Aris’ there were no differences in fruit characteristics between nutrient solution supply methods, but total soluble solid (TSS) was higher in drip and capillary systems showing $15.6^{\circ}$Brix and 15.5$^{\circ}$Brix, respectively, than that of mist spray system showing $14.4^{\circ}$Brix. In melon of net type, ‘Earl's elite’, drip and capillary systems showed better fruit characteristics and higher values of TSS compared to mist spray system. The change in pH in the medium before and after melon cropping was not significant between systems, but the pH of upper layer within medium in all system was higher than that of lower layer. The mineral content within lower layer showed higher value than that of the upper layer in drip system, but reversed in capillary system.
This Research was the standardization of nutrient sources by reviewing the possibility of cultivation and analysis of imported nutrient sources used in Pleurotus ostreatus culture. As a first result, the concentration of heavy metals was not detected or trace, and the total nitrogen content of cotton seed pomace was 6.22% in Australia, which was higher than that of Chinese or USA. As a second result of physical properties of nutrient sources, the water holding capacity of beet pulp from Egypt was higher with increasing pressure. The volume water content of US was highest at 400hpa and 500hpa in case of cottonseed meal. Third, the content of total nitrogen was the highest at pH 5.51 and the total nitrogen content was the highest at 2.45, and the C/N ratio was the highest at 28.18. Last result shows that first germination day, yield and biological effciency of all treatments were not significant differnet except T2. These results shows that australian cootton was inappropriate to Pleurotus ostreatus culture. Nevertheless, the safety and standardization of the Pleurotus ostreatus culture medium should be assured by reviewing the media low material importing season, mushroom yield and nutrient sources.
S. coelicolor A3(2) cells were treated with various redox-cycling agents on nutrient agar plates and examined for their effect on the growth and differentiation. When treated with plumbagin, severe effect on cell viability was observed at concentrations above 250 $\mu$M. However, the surviving colonies differentiated normally. When treated with 100 $\mu$M paraquat, growth rate was decreased and morphological differentiation was inhibited, while the survival rate was maintained at about 100% even at 5 mM paraquat. Menadione or lawsone did not cause any visible changes at concentrations up to 1 mM. The effect of paraquat was also observed when it was added to nutrient agar plate before spore inoculation. Three paraquat-resistant mutants were isolated with a frequency of 3 $\times$10${-5}$. Their mutation sites were determined by genetic crossings. All three mutations were mapped to a single locus near arg4 at about 1 o'clock on the genetic map of S. coelicolor A3(2).
A model for the role of serum was proposed to develop a low serum medium for the large scale cu1ture of mammalian cell. The strategy of medium development adopted in this study facilitated the understanding of the role being carried out by the serum in the culture of hybridoma KAl12 cell line. In this model, the serum components were divided into two main groups : the first group encompasses the nutrient factors that determine the maximum cell density and the second group includes the growth factors that regulate the cell growth rate, The model prediction was compared with the experimental results. The model enabled us to find out several useful aspects of medium composition for cell growth. 1) One particular component in the basal medium became limiting factor when serum concentration level was more than 7%. 2) The growth regulating factors and nutrient factors limited the cell growth at 3% and 5% serum concentration levels respectively.
