Lactic acid bacteria (LAB), belonging to the Firmicutes phylum, lack heme biosynthesis and, thus, are characterized as fermentative and catalase-negative organisms. To verify the hypothesis that heme-rich-nutrients might compensate the heme-biosynthesis incapability of non-respiratory LAB in animal gut, a heme-rich-nutrient was fed to a dog and its fecal microbiome was analyzed. Firmicutes abundance in the feces from the heme-rich-nutrient-fed dog was 99%, compared to 92% in the control dog. To clarify the reason of increased Firmicutes abundance in the feces from the heme-rich-nutrient-fed dog, Lacobacillus gasseri were used as model anerobic LAB to study a purified heme (hemin). The anaerobic growth of L. gasseri in the medium with 25 µM hemin supplementation was faster than that in the medium without hemin, while the growth in the 50 µM hemin-supplemented medium did not vary. Cellular activities of the cytochrome bd complex were 1.55 ± 0.19, 2.11 ± 0.14, and 2.20 ± 0.08 U/gcell in the cells from 0, 25, and 50 µM hemin-supplemented medium, while intracellular ATP concentrations were 7.90 ± 1.12, 11.95 ± 0.68, and 12.56 ± 0.58 µmolATP/gcell, respectively. The ROS-scavenging activities of the L. gasseri cytosol from 25 µM and 50 µM hemin-supplemented medium were 68% and 82% greater than those of the cytosol from no hemin supplemented-medium, respectively. These findings indicate that external hemin could compensate the heme-biosynthesis incapability of L. gasseri by increasing the cytosolic ROS-scavenging and extra ATP generation, possibly through increasing the electron transfer. Increase in the number of anaerobic bacteria in heme-rich-nutrient-fed animal gut is discussed based on the results.
The in vitro culture of rice panicles is a culturing technique only panicle without other organs in culture solution containing organic substance, so that would be useful to study how assimilate supply affects grain development and maturation. To find the optimum stage for in vitro culture, rice panicles grown in greenhouse were sampled periodically after anthesis and cultured in nutrient medium. The panicles older than 1 weeks after anthesis had produced normal grains. Grain-filling was apparently dependent upon sucrose concentration (8-12 %) in medium, but not affected by nitrogen concentration supplied with glutamine. As far as rice panicle was supplied with sucrose and N in nutrient medium, grains continued accumulation of dry matter and maturation regardless to light condition. Considerably, grain-filling was improved when panicles were positioned horizontally inside flask, so that each grain was partially submerged to nutrient medium.

Food waste causes various economic losses and environmental pollution problems such as soil pollution and groundwater pollution. Food waste has been used as a resource in various forms and has been used mostly for feed and composting. This study compared microalgal nutrient medium (BG-11) with food waste compost to determine the possibility of using it as a culture medium. Scenedesmus quadricauda was isolated and cultured in an eutrophic reservoir and incubated for 3 days in distilled water before laboratory use. Food waste compost was produced in two food waste processing facilities, and hot water was extracted in the laboratory to be used for microalgae cultivation. The growth curve of the microalgae was analyzed based on the Chl-a concentration measured during the experiment, and the growth rate of the microalgae grown in the food waste compost was compared with the growth rate of those grown in the nutrient medium. Food waste compost showed a similar growth rate to that of the nutrient medium, and there was a difference depending on the manufacturing facility. The growth of microalgae in such food waste was further amplified when trace elements were added and showed better growth than that of the nutrient media. Particularly, when trace elements were added, the growth rate increased, and the growth period was further extended. Therefore, food waste compost can be sufficiently utilized as a microalgal culture medium, and if trace elements are added, it is considered that microalgae can be more effectively cultured compared to the existing nutrient medium.
Most commonly occurring predacious fungus Arthrobotrys oligospora showed great variation in size and shape of conidia on some media. The formation of larger conidia was recorded on beef extract and nutrient agar media. The length of conidia in Richard's YPSS, Sabouraud's, PDA and com meal agar media was of medium size while smaller conidia were produced on Czapek's, Jensen's, Martin's medium. Maximum width of conidia was recorded on YPSS medium followed by Sabouraud's medium. The average size of spores on nematode infested com meal agar medium was slightly increased than those on com meal agar medium.
BACKGROUND: For the food safety of cultivated mushroom, information on the safety of agricultural by-products imported as medium substrates for mushroom cultivation is urgently needed. Therefore, this study was performed to detect the presence of heavy metals, residual pesticides, and nutrient component in the imported medium substrates. METHODS AND RESULTS: Six kinds of medium substrates imported from nine countries from 2015 to 2017 were investigated. A mercury analyzer MA-2000 and an inductively coupled plasma spectrometer OPTIMA 7000DV were used to analyze mercury, lead, arsenic, copper, nickel and cadmium. All of these heavy metals were detected at lower level than heavy metal tolerance standard level of by-product fertilizer in Korea. When 246 kinds of residual pesticides were examined by GC and HPLC, imidacloprid, thiamethoxam and carbendazim were detected from Egyptian beet pulp, Indian cottonseed meal and cottonseed hull, respectively. The content of nutrient components (water, crude ash, crude fat, crude protein and crude fiber) varied among imported countries and the medium substrates. CONCLUSION:The presence of heavy metals and residual pesticides in imported medium substrates for mushroom cultivation was confirmed. For the safe production of mushroom, this study shows that imported medium materials for mushroom cultivation need to be managed through continuous monitoring.
A factorial experiment was conducted to assess nutrient utilization by growing lambs maintained on three levels each of digestible energy (high: HE, medium: ME, low: LE) and protein (high: HP, medium: MP, low: LP) in nine combinations (HEHP, HEMP, HELP, MEHP, MEMP, MELP, LEHP, LEMP, LELP). The experiment was conducted during the hot season in a semiarid location. Daily dry matter intake (DMI) was similar in all the groups in terms of unit body weight or metabolic body size. Digestibility of DM and nitrogen free extract increased (p<0.01) from low to medium and high energy regimen while the CF digestibility followed a reverse trend. The digestibility of crude protein (CP) decreased from high to medium and low protein regimens while it was similar in terms of energy variation. Nitrogen intake was higher in high followed by medium and low protein regime while fecal and urinary nitrogen loss were similar in all the treatment groups. Lambs in all the three levels of protein were in positive N balance and percent N retention was higher (p<0.01) in high followed by medium and low protein levels whereas it was similar in terms of energy variation. Initial body weight was similar in all the groups while final weight, total gain in the experiment and average daily gain (ADG) were higher in high than medium and low energy regimens. It is concluded that crossbred lambs required 75.1 g DM, 9.6 g CP, 6.3 g DCP and 711 KJ DE/kg W $^{0.75}$or 11.0 g CP/MJ DE or 7.2 g DCP/MJ DE for 93 g average daily gain in a hot semiarid environment.
Ebb and flow 시스템을 사용한 분화 칼랑코에 재배에서 광도와 양액 농도가 생육, 양분 흡수율 및 배지 내 무기 이온의 집적에 미치는 영향을 구명하기 위하여 본 연구를 수행하였다. 연구 목적을 달성하기 위하여 광도 3수준(PPF 4.26, 5.51, $9.75mol{\cdot}m^{-2}{\cdot}d^{-1}$)과 양액 농도 3수준(EC 1.2, 1.8, $2.4dS{\cdot}m^{-1}$)을 조합하여 요인실험으로 수행하였다. 분화 재배한 칼랑코에의 생육은 광도와 양액 농도에 뚜렷하게 영향을 받았으며, 광도가 높아질수록 생육이 양호하였다. 각 광도 내에서는 양액의 EC를 $1.8\sim2.4dS{\cdot}m^{-1}$의 범위로 조절할 경우 칼랑코에 지상부 생육이 비교적 양호하였지만, EC를 $2.4dS{\cdot}m^{-1}$로 조절한 처리의 생체중, 건물중 및 엽면적이 가장 크게 나타났다. 광도와 양액 농도는 ebb and flow 시스템의 양액 탱크로부터 양분 및 수분 흡수량과 배지 내 무기 이온의 집적에 유의하게 영향을 미쳤지만, 식물체의 무기물 함량에는 뚜렷한 영향을 미치지 않았다. 광도는 양액의 EC에 비해 이온의 흡수율에 미치는 영향이 상대적으로 적었으며, 양액 EC가 높을 수록 $NO_3^-$, P, K, Mg 이온의 흡수율은 높았다. 이상의 연구결과를 고려할 때 ebb and flow 저면 관수 시스템에서 배지 내 무기 이온 집적에 영향을 주는 주 요인은 양액 농도임을 확인하였으며, 광도와 양액의 EC를 이용하여 배지 내 무기 이온의 집적량을 예측하기 위한 회귀식을 제시하였다.
This research was conducted to determine the plant growth and nutrient contents of potted chrysanthemum 'Lima Honey' as influenced by application rate of fused superphosphate (FSSP) in three root media, peatmoss+vermiculite (1:1, v/v; PV), peatmoss+composted rice hall (1:1, v/v; PR), and peatmoss+composted pine bark (1:1, v/v; PB). All root media contained polyacrylic acid sodium salt at a rate of $4.5g L^{-1}$. The treatment of $1.4g L^{-1}$ in PV and those of $0.7g L^{-1}$ in PR and PB had the greatest fresh and dry weights in each root medium at both 43 and 80 days after transplanting. Elevated application rates of FSPP increased tissue contents of N, P, and K at both 43 and 80 days after transplanting in PV medium. However, the differences in tissue contents of N, P and K in PR medium were less significant among treatments of FSPP. The pre-planting FSPP also less affected the tissue contents of nutrients at 80 days after transplanting as compared to those at 43 days after transplanting. Elevated application rates of FSPP in PV medium increased EC and the concentrations of $NO_3,\;P_2O_5$, K, Ca, and Mg in soil solution of root media at 43 days after transplanting. The EC in PV medium at 80 days after transplanting was higher than that at 43 days after transplanting. The EC in all root media at 80 days after transplanting was not different among treatments of FSPP.
Herbs and plants widely used for the ecological restoration were selected for germination rate analysis under treatment of microorganisms to determine ideal treatment conditions and medium for enhanced germination rate. Albizzia julibrissin, when submerged in a nutrient medium or distilled water, presented a decrease in germination period rather than increase in germination rate. When treated with microorganism culture solution (JM-2) for 24 hours, 90% germination was achieved in two days, which is sufficient evidence to conclude that such treatment accelerates the germination of Albizzia julibrissin. Germination period decreased for Lespedeza cyrtobotrya samples submerged in microorganism solution for 15 and 48 hours, however, increases in germination rates were not observed. Sample treated in the solution for 24 hours had increased germination rate and enhanced germination period. Microorganism solution treatment had a negative effect on germination for Lespedeza cuneata, unlike Lespedeza cyrtobotrya and Albizzia julibrissin. Microorganism treated seeds of Lepsedeza cuneata had a lower germination rate than that of the control with no treatment. However, submerging treatments in a nutrient medium or distilled water for 24 to 48 hours were proven effective with higher germination rates than control sample with no treatment. It is fundamental to control individual condition of every seedling. Automatic individual control is used by data control and analysis at on-line. As a result the best condition system was build without all waste. This system uses one of new technology irrigation system. This irrigation system supply accurate quantity of nutrient solution in the shortest time. The system named the upward injection irrigation system. First of all it is necessary to be considered whether the soil is proper or improper for upward injection irrigation system. It is important that root absorb nutrient solution as fast as possible. The ability of spreading, storing water, contamination of environment and cost were considered when choose the medium. The soil of organic culture is developed recently. The soil consists of paper pulp and vermiculite. The new soil is more suitable than ordinary medium for growing plant because this medium is made of paper pulp. The ability of store and spread of water is it's feature. We can make paper tray of this paper pulp's raw material. It is possible that pulp tray replaced plastic tray. The original plug tray of growing seedling system can make which consist of pulp medium and pulp tray. In this study, it was examined whether the plug seedling of paper pulp medium grow with upward injection irrigation system in this seedling plant system. At the same time, examine ability of store and spread of water and how to grow plant on the paper pulp medium.
