Study of Anti-inflammatory Effect of Ohmae-hwan Water Extract (오매환(烏梅丸) 물추출물의 항염증 활성에 관한 연구)

  • Ohmae-hwan has been used to treat the symptoms of loose stools mixed with blood accompanied by abdominal pain due to heat dysentery in Korean medicine. However, its mechanism of action on inflammatory disease using macrophages was not known. The author was to find out about the influence of Ohmae-hwan water extract on expressions of NO, cytokine IL-6 and TNF-α in LPS-stimulated RAW 264.7 cell, on expressions of mRNA of iNOS, IL-6 and COX-2 and on suppression of expressions of protein of iNOS and COX-2, and was to find out about the mechanism. As a result of the study, it was found that Ohmae-hwan suppressed generation of NO activated by LPS depending on its concentrations, suppressed expressions of cytokine IL-6, and TNF-α and suppressed expressions of mRNA of iNOS, IL-6, and COX-2 and suppressed expressions of protein of iNOS, COX-2. Considering the above results, it can be known that Ohmae-hwan water extract has anti-inflammatory effect against inflammatory response mediated by macrophages by suppressing generation of NO, TNF-α, and IL-6 and expressions of iNOS, COX-2 as it acts on macrophages.

Anti-inflammatory and Anti-oxidant Studies of Osung-tang Extracts in LPS-Induced RAW 264.7 Cells (LPS로 유도된 RAW 264.7 세포에서 오성탕(五聖湯) 추출물의 항염증 및 항산화 연구)

  • Objectives : The purpose of this study was to investigate the anti-inflammatory and anti-oxidative effects of Osung-tang(OST) extract. Methods : MTT assay was performed to confirm the survival rate of RAW 264.7 cells treated with OST extract(50-400㎍/㎖) and the production of NO from LPS-induced RAW 264.7 cells was confirmed. The mRNA expression of iNOS, COX-2, and NF-κB were measured by real-time PCR. The protein expression of iNOS, COX-2 and p-IκB were measured by western blot and the anti-oxidant activity of OST extract(50-400㎍/㎖) was investigated by measuring DPPH scavenging activity. Results : OST extract showed a cell survival rate of 90% or more at 50-400㎍/㎖. The NO production was inhibited in a dose-dependent manner in RAW 264.7 cells treated with LPS on OST treated group. mRNA expression levels of iNOS, COX-2 and NF-κB decreased in a concentration-dependent manner after treatment with OST(50-400㎍/㎖). Protein expression levels of iNOS, COX-2 and p-IκB decreased in a concentration-dependent manner after treatment with OST(50-400㎍/㎖). It was found that OST has a high free-radical scavenging ability. Conclusions : These results suggest that OST extract can be used as a treatment for various skin diseases by demonstrating its anti-inflammatory and anti-oxidant effects.

Effects of Butanol extract from Rhois Vernicifluae Cortex (RVC) in lipopolysaccharides-induced macrophage RAW 264.7 cells (칠피(漆皮) 부탄올 분획물이 LPS로 유도된 RAW 264.7 대식세포에 미치는 영향)

  • Objectives : RVC has long been used for a useful natural agent ameliorating inflammation related symptoms in the folk medicine recipe. This study was performed to investigate effects of RVC on the inflammation and oxidation in RAW 264.7 cells. Methods : The RVC was extracted with 80% ethanol and sequentially partitioned with solvents in order to increase polarity. With the various fractions, we determined the activities on the inflammation and oxidation in RAW 264.7 cells. Results : 1. Among the various solvent extracts of RVC, the butanol fraction showed the most powerful inhibitory ability against nitric oxide (NO) production in lipopolysaccharide (LPS)-induced RAW 264.7 cells without affecting cell viability. 2. Butanol fraction showed a oxidation inhibition effect by decreasing the DPPH and OH radicals. 3. Butanol fraction exhibited the inhibitory avilities against iNOS and COX-2. 4. Reverse transcriptase polymerase chain reaction (RT-PCR) and Westem blotting analysis revealed that the BuOH fraction provided a primary inhibitor of the iNOS protein and mRNA expression in LPS-induced RAW 264.7 cells. Among the up-regulater molecules of iNOS and COX-2, the BuOh fraction of RVC was shown the inhibitory activity of phoshporylation of c-Jun N-terminal kinase (JNK) 1/2 and threonine protein kinase (AKT), the one of the MAPKs pathway. Conclusion : Thus, the present study suggests that the response of a component of the BuOH fraction to NO generation via iNOS expression provide a important clue to elucidate anti-inflammatory and anti-oxidation mechanism of RVC.

Docosahexaenoic acid reduces adenosine triphosphate-induced calcium influx via inhibition of store-operated calcium channels and enhances baseline endothelial nitric oxide synthase phosphorylation in human endothelial cells

  • Docosahexaenoic acid (DHA), an omega-3-fatty acid, modulates multiple cellular functions. In this study, we addressed the effects of DHA on human umbilical vein endothelial cell calcium transient and endothelial nitric oxide synthase (eNOS) phosphorylation under control and adenosine triphosphate (ATP, $100{\mu}M$) stimulated conditions. Cells were treated for 48 h with DHA concentrations from 3 to $50{\mu}M$. Calcium transient was measured using the fluorescent dye Fura-2-AM and eNOS phosphorylation was addressed by western blot. DHA dose-dependently reduced the ATP stimulated $Ca^{2+}$-transient. This effect was preserved in the presence of BAPTA (10 and $20{\mu}M$) which chelated the intracellular calcium, but eliminated after withdrawal of extracellular calcium, application of 2-aminoethoxy-diphenylborane ($75{\mu}M$) to inhibit store-operated calcium channel or thapsigargin ($2{\mu}M$) to delete calcium store. In addition, DHA ($12{\mu}M$) increased ser1177/thr495 phosphorylation of eNOS under baseline conditions but had no significant effect on this ratio under conditions of ATP stimulation. In conclusion, DHA dose-dependently inhibited the ATP-induced calcium transient, probably via store-operated calcium channels. Furthermore, DHA changed eNOS phosphorylation suggesting activation of the enzyme. Hence, DHA may shift the regulation of eNOS away from a $Ca^{2+}$ activated mode to a preferentially controlled phosphorylation mode.

Studies on the NO Production and Expression Induction Effect of NOS Gene by Salviae Radix (단삼에 의한 NO 생성 및 NOS 유전자의 발현 유도 효과에 관한 연구)

  • Objective : This experiment was performed in order to study the effect of an aqueous extract of Salviae radix root(SRRAE) on NO production and NOS gene induction from macrophages Methods : To investigate dose-dependent effects of SRRAE for NO release on the $rIFN-{\gamma}-treated$ macrophages, the cells were incubated for 6 hrs in a medium containing $rIFN-{\gamma}$ (5 U/ml), stimulated with SRRAE and incubated in a CO2 incubator. The cells were treated with 5 U/ml $rIFN-{\gamma}$ plus 100 g/ml of SRRAE, Then, the cells were incubated with various concentrations of NGMMA at $37^{\circ}C$ for 48 hrs, Results : SRRAE had no effect on NO production by itself, whereas recombinant $interferon-{\gamma}(rIFN-{\gamma})$ alone showed modest activity, When SRRAE was used in combination with $rIFN-{\gamma}$, there was a marked cooperative induction of NO production in a dose-dependent manner. The optimal effect of SRRAE on NO production was shown at 6hrs after treatment with $rIFN-{\gamma}$. The SRRAE-induced production of NO was inhibited by NG-monomethyl- L-arginine(NGMMA) and arginase. $rIFN-{\gamma}$ in combination with SRRAE showed a marked increase of the expression of the inducible NOS(iNOS) gene. In addition, the effect of SRRAE was mainly dependent on the SRRAE-induced tumor necrosis $factor-{\alpha}(TNF-{\alpha})$ secretion. Conclusions : SRRAE induces NO production from macrophages as a result of SRRAE-induced $TNF-{\alpha}$ secretion. SRRAE may provide a second signal for synergistic induction of NO production in macrophages already induced to express iNOS gene by $rIFN-{\gamma}$.

Inhibitory Effect of Salvia officinalis on the Inflammatory Cytokines and Inducible Nitric Oxide Synthasis in Murine Macrophage RAW264.7 (RAW 264.7 Cell에서 세이지에 의한 염증성 Cytokine 및 iNOS억제 효과)

  • Primary pro-inflammatory cytokines are a trio: tumor necrosis- $\alpha$ (TNF-$\alpha$), interleukine-$\beta$ (IL-$\beta$), and interleukine-6 (IL-6). These cytokines initiate and regulate the acute-phase inflammatory response during infection, trauma, or stress and appear to play an important role in the immune process. Nitric oxide (NO) is a multi-functional mediator, which plays an important role in regulating various biological functions in vivo. NO production by inducible nitric oxide synthase (iNOS) in macrophages is essential for the defense mechanisms against microorganisms and tumor cells. However, over-expression of iNOS by various stimuli, resulting in over-production of NO, contributes to the pathogenesis of septic shock and some inflammatory and auto-immune disease. Solvent fractions of sage ( Salvia officinalis L.), which is cultivated in Jeju-Do, was assayed for their effects on TNF-$\alpha$ and IL-6 production in LPS-stimulated RAW 264.7 macrophages. Hexane and ethylacetate (EtOAc) fraction of sage inhibited the protein and mRNA expression of TNF-$\alpha$ and IL-6 in LPS stimulated RAW 264.7 cells at the concentration of 100 $\mu\textrm{g}$/$m\ell$. Also, incubation of RAW 264.7 cells with the fraction of hexane or EtOAc (50 $\mu\textrm{g}$/$m\ell$) inhibited the LPS induced nitrite accumulation and the LPS/IFN-${\gamma}$ induced iNOS protein. And this inhibition of iNOS protein is concordant with the inhibition of iNOS mRNA expression. Above results suggest that extract of sage may have anti-inflammatory activity through the inhibition of pro-inflammatory cytokines (TNF-$\alpha$, IL-1$\beta$, IL-6), iNOS and NO.

Selective iNOS Inhibition Attenuates Skeletal Muscle Reperfusion Injury (선택적 iNOS 억제에 의한 골격근 재관류 손상의 감소)

  • The purpose of this study is to determine the effects of selective inducible nitric oxide synthase (iNOS) inhibitor N-[3-aminomethyl]benzyl]acetamidine (l400W) on the reperfused cremaster muscle. The extracellular superoxide dismutase knockout ($EC-SOD^{-/-}$) mice was used to make the experimental window for ischemia-reperfusion injury. The muscle was exposed to 4.5 h of ischemia followed by 90 min of reperfusion and the mice received either 3 mg/kg of 1400W or the same amount of phosphate buffered saline (PBS) subcutaneously at 10 min before the start of reperfusion. The results showed that 1400W treatment markedly improved the recovery of the vessel diameter and blood flow in the reperfused cremaster muscle compared to that of PBS group. Histological examination showed reduced edema in the interstitium and muscle fiber, and reduced nitrotyrosine formation (a marker of total peroxinitrite ($ONOO^-$) in 1400W-treated muscle compared to PBS. Our results suggest that iNOS and $ONOO^-$ products are involved in skeletal muscle I/R injury. Reduced I/R injury by using selective inhibition of iNOS is perhaps via limiting cytotoxic $ONOO^-$ generation, a reaction product of nitric oxide (NO) and superoxide anion ($O_2^-$). Thus, inhibition of iNOS appears to be a good treatment strategy in reducing clinical I/R injury.

Immunomodulatory Effects of β-sitosterol and Daucosterol Isolated from Dioscorea batatas on LPS-stimulated RAW 264.7 and TK-1 Cells (산약에서 분리한 β-sitosterol과 daucosterol의 RAW 264.7 세포와 TK-1 세포에서의 면역 활성 조절 효능)

  • Although many studies on immune modulatory materials have used RAW 264.7 cells, few have used T cell-derived TK-1 cell lines. Moreover, although some studies have investigated the efficacy of plant-derived β-sitosterol, few have examined the immunomodulatory activity of its analogue, daucosterol. In this study, β-sitosterol and daucosterol were isolated from D. batatas and identified by nuclear magnetic resonance spectroscopy. To evaluate the immune-enhancing or inhibitory effects of the isolated phytosterols, the expression levels of the inflammatory response genes COX-2, TNF-α, IL-6, and iNOS were analyzed by RT-PCR. The relative expression levels of TNF-α and iNOS in RAW 264.7 cells were increased more than threefold with β-sitosterol treatment comparing to those of untreated control. In the case of TK-1 cells, the expression level of TNF-α was decreased and the expression level of iNOS was increased in a β-sitosterol concentration-dependent manner. The expression levels of COX-2, TNF-α, and IL-6 increased by approximately 0.7-1.2 times in RAW 264.7 cells treated with daucosterol compared to those of untreated control, but iNOS expression decreased by 0.8-0.18 times. In the case of daucosterol-treated TK-1 cells, the expression levels of TNF-α, IL-6, and iNOS were markedly reduced from those of TK-1 cells treated only with lipopolysaccaride. As a conclusion, β-sitosterol treatment increased TNF-α and iNOS expression levels in RAW 264.7 cells, thus exerting an immune- boosting effect. However, in TK-1 cells, iNOS expression increased while TNF-α expression decreased, indicating an immunosuppressive activity of β-sitosterol. Daucosterol appears to exert an immunosuppressive effect in both macrophages and T cell lines by inhibiting iNOS expression in RAW 264.7 cells and greatly inhibiting the expression of TNF-α, IL-6, and iNOS in TK-1 cells.

Changes in Cytochrome c Oxidase and NO in Rat Lung Mitochondria Following Iron Overload

  • In this study, the effects of iron on cytochrome c oxidase (CcO) in rat lung mitochondria were examined. Similar to liver mitochondria, iron accumulated considerably in lung mitochondria (more than 2-fold). Likewise, the reactive oxygen species and nitric oxide (NO) content of mitochondria were increased by more than 50% and 100%, respectively. NO might be produced by nitric oxide synthase (NOS), eNOS and iNOS type, with particular contribution by NOS in mitochondria. The respiratory control ratio of iron overloaded lung mitochondria dropped to nearly 50% due to increased state 4. Likewise, cytochrome c oxidase activity was lowered significantly to approximately 50% due to excess iron. Real-time PCR revealed that the expression of isoforms 1 and 2 of subunit IV of CeO was enhanced greatly under excess iron conditions. Taken together, these results show that oxidative phosphorylation within lung mitochondria may be influenced by iron overload through changes in cytochrome c oxidase and NO.

Development and Application of NOS Education Program according to Analyzing the Characteristics of Nature of Science(NOS) in Exhibits of Science Museum (국내 과학관 전시물에 반영된 과학의 본성(NOS) 특징 분석에 따른 프로그램 개발 및 이의 적용)

  • The purpose of this study is to identify the status of the science museum reflected in the exhibition, and develop and apply Nature of Science(NOS) education programs based on the science museum exhibition. The analyse tool was developed to understand the NOS in the science museum. The researchers recognized the nature of science reflected in four exhibition halls in Korea. Based on the analysis, we developed the NOS education program. NOS education programs were developed and applied to supplement the NOS that appears to be limited to scientific exhibitions based on the prior analysis of science exhibition. The results of the study were as follows. First, we analyzed the nature of science reflected in the exhibition of two main science museums, and it was mostly implicit and most were to understand the relationship among STS(Science-Technology-Society). And also we analyzed the NOS reflected in the exhibition of two national history museum, and it was also mostly implicit and most were about the way of how to find out the knowledge, inference. Second, in order to supplement the NOS of the science museum, we developed the NOS education program based on the exhibits. After applying it to the science museum, we conducted a qualitative study. As a result, there was a positive change only in the aspects of NOS (science is tentative, science is from creativity and imaginative, science is the produce of social and culture, science is from the scientific method) that reflected explicitly. The conclusions derived from this study are as follows : For the cultivation of science in the scientific museum, various factors are needed depending on the theme of the science museum. Also, it is helpful to actively implement the NOS educational programs that utilize the exhibit. Therefore, the exhibit planners' and developers' competencies are critical to develop explicit NOS education programs in its expertise.
