New record of an alien plant, Verbena bracteata (Verbenaceae) in Korea

  • Verbena bracteata Cav. ex Lag. & Rodr. (Verbenaceae), native to North America and an alien plant in China and Japan, was newly found in central and southern areas of Korea. This species can be distinguished from other Verbena taxa in Korea by having larger floral bracts and stems that are prostrate or procumbent to decumbent-ascending. Verbena bracteata grows in dry and sunny grasslands or along roadsides. A precise description, illustration, photographs, and a key to related taxa are provided.

Effect of Cold Stratification and Gibberellin Treatment on Androsace septentrionalis L. Seed Germination

  • Androsace septentrionalis is a grass species restricted to North Korea; however, it is at the brink of extinction due to habitat loss and environmental changes caused by natural disasters and anthropogenic activities. This study was conducted to characterize the dormancy conditions of A. septentrionalis in an effort to conserve this North Korean plant resource. For this purpose, the morphological characteristics and vigor of A. septentrionalis seeds were examined, and its germination characteristics under different temperature conditions (15/6℃ and 25/15℃ ), low-temperature stratification, and gibberellin (GA3) treatment were determined. The results revealed that A. septentrionalis exhibits non-deep type morpho-physiological dormancy, and low-temperature stratification treatment was not effective in breaking the dormancy of A. septentrionalis seeds. Meanwhile, GA3 treatment significantly increased the mean germination time, rate, and speed of the seeds. The optimal conditions for the germination of A. septentrionalis seeds were 25/15℃ fluctuating temperature and 500 mg·L1 GA3 treatment. The results of this study are useful for the mass propagation of A. septentrionalis.

울릉도 자생 큰연령초의 분포와 자생지의 생태적 특성 (Ecological Characteristics and Distribution of Native Trillium tschonoskii in Ulleung Island)

  • Trillium tschonoskii Maxim. is a kind of 64 endangered plant species designated by the Ministry of Environment in Korea. It is very a rare native plant throughout the country. Therefore, this study was conducted to investigate the native T. tschonoskii in Ulleung Island for their distribution and ecological characteristics. They were mostly distributed valley, mid-slope of the mountain from 186 to 458 m altitude. They had a little low light conditions as they faced west, north west, south west, south-south west, north-north east and east-east north. They were provided with slightly humid condition. Aerial humidities of these quadrates were variable as 21.5-71%. They were located on the variable slopes of 5 to 43 degrees. Soil humidity was slightly dry. Native sites were semi-shaded because tree layer such as Tilia insularis, Zelkova serrata, okamotoanum etc.and sub-tall tree layer such as Ligustrum foliosum, Comellia japonica, Tilia insularis, Callicarpa japonica and so on were grown near by. Woody plants such as A. Okamotoanum, Sorbus commixta, Fagus japonica var. multinervis etc. were frequently observed. These environmental conditions were suitable that T. tschonoskii grows up naturally because of high Percentage as 52.78% of blooming individuals in this study. These native sites were classified with three communities, T. tschonoskii-Majanthemum dilatatum community, T. tschonoskii-Anthriscus sylvestris community and T. tschonoskii-Hepatica maxima community in accordance with altitude and aerial humidity. The best way ffr the preservation of T. tschonoskii habitats and maintainance of its recent population is maintained in recent environmental condition and ecological condition without any management by human-being.

감자 바이러스 Y 저항성 형질전환 감자 개발 (Development of Potato Virus Y-Resistant Transgenic Potato)

  • ND860-2, Norchip, Russet Norkotah, Goldrush 및 Norgueen Russet 등 5개 품종의 감자 잎 조직을 감자 바이러스 Y(PVY)의 외피단백질 (CP) 유전자를 운반하는 Agrobacterium tumefaciens을 이용하여 형질전환시켰다. ND860-2와 Norchip품종과 같은 흰색 감자에서 재분화율과 형질전환율이 높게 나타났다. 형질전환체를 선발한 후 CP의 생성을 western blot으로 확인한 결과 30 kD의 밴드가 나타났다. 온실에서 재배된 형질전환체는 한 개체를 제외하고는 표현형에 있어서는 기존의 품종과 다르지 않았다. 인위적으로 감자 바이러스 Y를 감염시킨 15일 후의 검사결과 control구는 80%의 감염율을 보인 반면 형질전환체들은 38-58%의 감염율을 보였다. 30일 후의 검사 결과는 모든 5품종의 control구에서 100%의 감염을 보였으나 형질전환체는 7개체가 저항성을 보였다. 마지막 85일 후의 검사 결과 4개의 저항개체를 발견하였으며 이 개체들은 현재 계속 연구가 진행 중에 있다.

Source-Sink Relations in North American Ginseng Seedlings as Influenced by Leaflet Removal

  • Seedlings of North American ginseng (Panax quinquefolius L.) were grown to full canopy establishment and then leaflet or leaf removal at different times applied to determine the effects on plant growth and performance. Leaf removal at 47, 57, 69 and 78 days after seeding resulted in 82.1, 59.8, 41.3 and 29.8% reduction, respectively, in root dry matter (economic yield) ; this indicates that leaf removal during the early root growth period causes greatest reduction in root yield. Removal of 1, 2, and 3 leaflets at 42, 52, 62 and 70 days from seeding reduced root weight at harvest (80 days from seeding) linearly, particularly at earlier removal dates. The perennating bud formed on all roots and was not influenced by treatment. This would suggest that if leaf loss occurs after canopy establishment the plant will re-grow the next year after the obligatory dormancy period.

A LysM Domain-Containing Protein LtLysM1 Is Important for Vegetative Growth and Pathogenesis in Woody Plant Pathogen Lasiodiplodia theobromae

  • Lysin motif (LysM) proteins are reported to be necessary for the virulence and immune response suppression in many herbaceous plant pathogens, while far less is documented in woody plant pathogens. In this study, we preliminarily characterized the molecular function of a LysM protein LtLysM1 in woody plant pathogen Lasiodiplodia theobromae. Transcriptional profiles revealed that LtLysM1 is highly expressed at infectious stages, especially at 36 and 48 hours post inoculation. Amino acid sequence analyses revealed that LtLysM1 was a putative glycoprotein with 10 predicted N-glycosylation sites and one LysM domain. Pathogenicity tests showed that overexpressed transformants of LtLysM1 displayed increased virulence on grapevine shoots in comparison with that of wild type CSS-01s, and RNAi transformants of LtLysM1 exhibited significantly decreased lesion length when compared with that of wild type CSS-01s. Moreover, LtLysM1 was confirmed to be a secreted protein by a yeast signal peptide trap assay. Transient expression in Nicotiana benthamiana together with protein immunoblotting confirmed that LtLysM1 was an N-glycosylated protein. In contrast to previously reported LysM protein Slp1 and OsCEBiP, LtLysM1 molecule did not interact with itself based on yeast two hybrid and co-immunoprecipitation assays. These results indicate that LtLysM1 is a secreted protein and functions as a critical virulence factor during the disease symptom development in woody plants.

Overexpression of human erythropoietin in tobacco does not affect plant fertility or morphology

  • Human erythropoietin (EPO) is a leading product in the biopharmaceutical market, but functional EPO has only been produced in mammalian cells, which limits its application and drives up the production costs. Using plants to produce human proteins may be an alternative way to reduce the cost. However, a recent report demonstrated that overexpression of the human EPO gene (EPO) in tobacco or Arabidopsis rendered males sterile and retarded vegetative growth, which raises concern whether EPO might interfere with hormone levels in transgenic plants. In the present study, we demonstrated that overexpressing EPO with additional 5'-His tag and 3' ER-retention peptides in tobacco did not cause any developmental defect compared to GUS plants. With our method, all 20 transgenic plants grew on selective medium and, further confirmed by PCR, were fertile. Most of them grew similarly compared to GUS plants. Only one transgenic plant (EPO2) was shorter in plant height but had twice the life span compared to other transgenic plants. When 11 randomly selected EPO plants, along with the abnormal plant EPO2, were subjected to RT-PCR analysis, all of them had detectable EPO transcripts. However, their protein levels varied considerably; seven of them had detectable EPO proteins analyzed by western blot. Our results indicate that overexpressing human EPO protein in plants does not have detrimental effects on growth and development. Our transformation systems allow us to further explore the possibility of glycoengineering tobacco plants for producing functional EPO and its derivatives.

ISSR marker-assisted selection of male and female plants in a promising dioecious crop: jojoba (Simmondsia chinensis)

  • Simmondsia chinensis (Link) Schneider, a multipurpose and monogeneric dioecious shrub from arid zones, has emerged as a cash crop all over the globe. Its seed propagation poses severe problems due to its male-biased population: the male:female ratio is 5:1. Investigations have been carried out to generate a sex-specific Inter-simple sequence repeat (ISSR) marker for the early detection of male and female plants. Of the 42 primers analysed with a bulk sample of pooled male DNA and a bulk sample of pooled female DNA, only one primer, UBC-807, produced a unique ~1,200 base-pair fragment in the male DNA. To validate this observation, this primer was re-tested with individual male and female samples from eight cultivars. A similar unique ~1,200 bp fragment was present in the male individuals of all eight cultivars and completely absent in the female individuals tested. This is the first report of the use of ISSR markers to ascertain sex in physiologically mature S. chinensis plants.
