The purpose of this study was to investigate the gene profiles that were up- or down-regulated in the livers of high-fat diet-induced obese mice and $db_-/db_-$ mice with deficient leptin receptor. C57/BL6 normal mice and $db_-/db_-$ mice, respectively, were divided into two groups and fed a standard or high-fat diet for four weeks. Liver weight was unchanged in the normal mice but the high-fat diet led to a 10% weight increase in the $db_-/db_-$mice. Adipose tissue mass increased by about 88% in the normal mice that were fed a high-fat diet and by about 17% in the $db_-/db_-$mice on the high-fat diet. In terms of serum lipids, total cholesterol significantly increased in mice on the high-fat diet. Microarray analysis was carried out using total RNA isolated from the livers of standard or high-fat diet-fed mice of the normal and $db_-/db_-$ strains. The change of gene expression was confirmed by RT-PCR. About 1.6% and 6.8% of total genes, respectively, showed different expression patterns in the normal mice fed the high-fat diet and $db_-/db_-$ mice. As a result of microarray, many genes involved in metabolism and signal pathways were shown to have different expression patterns. Expression of Mgst3 gene increased in the livers of normal and $db_-/db_-$ mice that were fed a high-fat diet. Wnt7b and Ptk9l were down-regulated in the livers of the normal mice and $db_-/db_-$ mice that were fed a high-fat diet. In conclusion, a high-fat diet induced obesity and affected gene expression involved in metabolism and signal pathway.
120 mice of both sexes weighing from 18 to 22 g. were used. These mice were divided nito 12 groups : control (non-restraint and restraint) , acclinmation (non-restraint and restraint , acclimation for 5 and 10 days) , adrenalectomized (non-restraint and restraint0 and acclimation-adrenalectomized (non-restraint and restraint, acclimation for 5 and 10 days) groups. The rectal temperature of each group were measured at 10, 30 and 50 min under environment of $0^{\circ}C$.Measurements were done with a microphrometer of B.T.-32 thermocorpules. The results obtained may be summarized as follows : 1) The rectal temperature in normal mice without restraint was higher than that of normal mice with restraint under environment of $0^{\circ}C$. 2) The rectal temperature in normal mice was higher than that of adrenalectomized mice at 20$^{\circ}$ C. But the rectal temperature in adrenalectomized mice without restraint was higher than that of adrenalectomized mice with restraint under cold-stress. This difference was in the order of 1.65$^{\circ}$ under environment of 20$^{\circ}$ and 10 $^{\circ}$ at 50 min under environment of 0$^{\circ}$. 3) The rectal temperature of normal mice with restraint was less than that of adrenalectomized mice (non-restraint) under cold-stress. 4) The rectal temperature of normal and adrenalectomized mice that acclimated under cold stress ($0^{\circ}C$) for 5 and 10 days was usually higher than that of normal and adrenalectomized mice of non-acclimation under environment of $0^{\circ}C$. 5) The tolerance in mice under cold-stress was increased on the acclimtion of cold.
Eosinohil and IgE responses of interleukin IL(-5 transgenic and normal C3H/HeN mice were studied after experimental infection with Nippostrongylus brasiliensis 9Nb). Intestinal worms were recovered at day 5 post-infection (PI), and numbers of total white blood cells (WBC) and eosinophils, and total serum IgE and anti-hapten (dinitrophenyl)(DNP) specific IgE titers, were measured at days 0,14 and PI. IL-5 mice appeared resistant to Nb infection showing a significantly ower worm recovery rate than normal mice (P<0.05). Total WBC and eosinophil counts (/mm3) were significantly increased in Nb infected normal mice (p<0.05), but unchanged (total WBC) or decreased (eosinophils) in IL-5 mice at day 21 PI. The total serum IgE level remarkably increased in normal mice, but only a little in IL-5 mice at days 14 and 21 PI. Priming with DNP brought about more remarkable increases of the total and anti-DNP specific IgE in normal mice than in IL-5 mice. The results show that IL-5 mice are resistant to Nb infection, and that eosinophil and IgE responses in these mice are not augmented by N infection.
Effect of the butanol fraction of Astragali Radix (BFAR) on the humoral immune response were investigated in ICR mice. Mice were divided into 4 groups and BFAR at doses of 5,25 and 125 mg/kg were administered orally to mice daily for 3 weeks, and the normal animals were given vehicle. The results of this study are summarized as follows; the relative weight of spleen was markedly increased by BFAR treatment, compared with that in normal mice. However, the body weight gain and the relative weight of liver were not affected. Splenic plaque forming cells and hemagglutination titers to sheep red blood cells, and the secondary IgG antibody response to bovine serum albumin were also dose-dependently enhanced by BFAR treatment. In these mice, BFAR did not increase serum alanine aminotransferase total protein, sect albumin and albumin/globulin ratio when compared with those in normal mice. Thus, these findings indicate that BFAR significantly enhances humoral immune response to antigen in concentrations that do not affected liver function.

Objective: This study aimed to identify genes regulated by cyclin dependent kinases 5 (CDK5) that participate in hair pigmentation in mice. Methods: The mRNA expression profiles of skin samples from CDK5-knockdown mice were constructed using high-throughput RNA sequencing and compared with those of wild-type mice. Results: In total, 8,002 known genes were differentially expressed between CDK5-knockdown and wild-type mice. Of these, 3,658 were upregulated and 4,344 were downregulated in the skin of CDK5-knockdown mice. An additional 318 previously unknown genes were also differentially expressed, with 171 downregulated and 147 upregulated genes in the skin of CDK5-knockdown mice. Of the known genes expressed in mouse skin, 80 were associated with hair color, with 61 showing lower expression and 19 exhibiting higher expression in skin of CDK5-knockdown mice. Importantly, the expression of the tyrosinase-related protein 1 (TYRP1) and the calcium signaling pathway were also found to be regulated by CDK5, suggesting that pigmentation is regulated by CDK5 via the calcium signaling pathway and TYRP1. Conclusion: The transcriptome profiles obtained from the skin of CDK5-knockdown mice compared to wild-type mice provide a valuable resource to help understand the mechanism by which CDK5 regulates melanogenesis in mice and other animals.
It has well studied that immune cells are strongly related to tumor progression and tumor suppression. To identify the difference of immune cell between tumor bearing mice and normal mice, we examined systemically the immune cell of CT26 tumor bearing mice on 21 days after tumor cell administration. As previously reported, CD4+ and CD8+ T cells population of tumor bearing mice significantly decreased 38% and 30% on day 21 compared to that of normal mice, respectively. All subpopulation of CD4 and CD8+ T cell significantly decreased, except CD49b+ T cell subpopulation. But, myeloid cell population ($CD11b^{high}$ and all Gr-1+ subpopulation) of tumor bearing mice significantly increased on day 21. Especially, all subpopulation of CD11b+Gr-1+ cell of tumor bearing mice significantly increased on day 21. Also, Foxp3+$CD25^{high}$ CD4 T cell (regulatory T cells) population significantly increased on day 21. These results suggest that tumor can induce the decline of T lymphocyte and the expansion of myeloid cells and regulatory T cells, and provide the basic information for the study of tumor immunology.
Mouse models of chronic toxoplasmosis and atopic dermatitis (AD) were combined to clarify the effect of opportunistic Toxoplasma gondii infection on the development of AD. AD was induced as a chronic contact hypersensitivity (CHS) with repeated challenge of 2,4,6-trinitro-1-chlorobenzene (TNCB) on the dorsal skin of mice. TNCB induced skin thickness increases in both normal and toxoplasmic mice. The changing patterns were different from the sigmoidal which saturated at 20 days in normal mice to the convex saturated at 12 days in toxoplasmic mice with the crossing at 18 days. Compared to normal mice, toxoplasmic mice presented CHS more severely in earlier times and then moderately in later times. These data suggest that host immune modification by T. gondii infection enhances CHS in early times of atopic stimulation but soothes the reaction of CHS in later times in mouse model.

Effects of endosulfan (EN), an insecticide, and bleomycin (BL), an antibiotic, on the body weight in the normal mice, and the in vivo cell growth, tumor weight, and hematological parameters of the Ehrlich ascites carcinoma (EAC) cell-bearing Swiss albino mice Mus musculus were evaluated. EN and BL were respectively administered orally and intraperitoneally to the experimental mice; the control group consisted of EAC cell-bearing untreated mice only. EN reduced the body weight in normal mice, whereas BL resulted in a steady body weight compared to the control. EN increased the EAC cell count significantly by reducing the growth of normal viable cells. In contrast, BL reduced the cell count by increasing the proportion of viable cells in the body. The tumor weights induced by EN were significantly higher than those of the EAC control and the BL-treated animals. In comparisons with the control and the BL mice, hematological parameters such as hemoglobin (%) and the number of RBC and lymphocytes were lowered, while counts of WBC, neutrophils, and monocytes were elevated after EN treatments. These results show that BL is capable of reducing the EN-induced neoplastic and haematological alterations in the mice under laboratory conditions.
CJ-50001 is a recombinant granulocyte-colony stimulating factor (rG-CSF) developed by Cheil Jedang R&D Center. The effects of CJ-50001 on the increase of peripheral neutrophil count following intravenous and subcutaneous single administration at a dose of 20$\mu$g/kg in normal ICR mice and SD rats, respectively, were compared with those of Grasin, a control drug. Both CJ-50001 and Grasin significantly increased the peripheral neutrophil number in four treatment groups and the maximum number of neutrophil was achieved at 12 to 18 h in rats and mice, respectively. The dose dependency test was studied for CJ-50001 only in normal mice by intravenous or subcutaneous administration. When administered i.v or s.c at the various doses in normal mice, CJ-50001 significantly increased the neutrophil number over the dose of 160 ng/kg, compared with the vehicle control group. From these results, it was concluded that CJ-50001 showed efficacy similar to Grasin in the peripheral neutrophil count increase.
Calcitonin gene-related peptide (CGRP) expressing neurons are distributed widely throughout the central and peripheral nervous systems. Due to its distribution and pharmacological studies, CGRP has been implicated to be involved in anxiety, fear and depression. In this study, ${\alpha}CGRP-knockout$ mice were used to assess the consequences of removing this neuropeptide to the mice behaviors. ${\alpha}CGRP-knockout$ mice performed equally as well as wild type mice in the light-dark transition test and in the elevated plus maze test of anxiety. ${\alpha}CGRP-null$ mice behaved similarly as wild-type mice in the Porsolt swim test of depression. They also exhibited normal learning and memory in the fear conditioning tasks. It is concluded that ${\alpha}CGRP$ is not essential for mice to be able to perform these tests, despite the presence of ${\alpha}CGRP$ in the relevant regions of the brain.
