The purpose of this study was to investigate the effects of Astragali Radix on the diet-induced hyperlipidemia in rats. Hyperlipedemia was induced in rats with high fat diet. Rats were divided into 4 groups, normal group(supplied enough water and feeds only), high fat diet administered group(supplied high fat diet for 4 weeks, Control group) and Astragali Radix administered group(supplied high fat diet and Astragali Radix lyophilization extract for 4 weeks, 178.6 mg/kg(rat) in sample A, 297.8 mg/kg(rat) in sample B). Body weight, liver weight and serum lipid levels were evaluated. The water extract of Astragali Radix decreased body weight, liver weight, total cholesterol, LDL-cholesterol, triglyceride and phospolipid in high fat diet induced hyperlipidemia in rats, and increased HDL-cholesterol.
Hypercholesterolemia is associated with many pathological states such as fat deposit in the tendon and skin, hepatomegaly, pancreatitis and heart attack. The present study was focused on the effect of Korean red ginseng water extract (RGWE) on the recovery of hepatic function that was deteriorated in high-cholesterol diet fed mice. Mice were divided into 6 groups ; normal diet group (AIN-2WK), 4 week-high-cholesterol diet group (CHOL-4WK), 2 week-high-cholesterol + 2 week-normal diet group (CHOL-2WK), 2 week-high-cholesterol + 2 week-normal diet + RGWE treated group (R100, R300, R500). RGWE treated group received normal diet and RGWE at daily doses of 100, 300, 500 mg/kg b.w. for 2 weeks after the 2 weeks feeding of high-cholesterol diet. The results are as follows : 1. The hepatic weight and total hepatic lipid weight were markedly lower in RGWE treated group than those in the CHOL-4WK or CHOL-2WK group, indicating that ginseng inhibited hepatic hypertrophy. 2. Total hepatic cholesterol and HDL-cholesterol in normal diet conversion group (CHOL-2WK) were significantly reduced compared with the high cholesterol group (CHOL-4WK). The lowering effect was significantly reinforced by RGWE administration. 3. Hepatic triglyceride level of CHOL-4WK group was markedly lower than that of the normal group (AIN-2WK). However, the level was significantly increased in CHOL-2WK and RGWE treated group. The result indicated that ginseng administration and diet conversion exert normalizing effects of hepatic function. 4. The lipid peroxide levels in RGWE treated groups were markedly lower than CHOL-2WK group. These results suggest that Korean red ginseng water extract can be useful as a dietary supplement for the treatment of obesity or hypercholesterolemia by improving lipid metabolism and hepatic function.
This study determined plasma glucose, free fatty acid(ffa), epinephrine, norepinephrine, creatine the phosphokinase(CPK), calcium (Ca), magnesium(Mg) and hydroxyproline, Ca and Mg in the heart muscle. Thirty six male Sprague Dawley rats average weighing 110g divided into two diet group : normal Mg group(0.05% Mg), Marginal Mg group(0.005% Mg). After the rats were on experimental diets for 2 weeks, rats for each group were randomly assigned to stress and no stress group and 30 minutes IMMB stress was administered to stress group every day for two weeks. The following were found : 1) Rats fed normal magnesium diet showed decreased plasma glucose and non signficant change in norepinephrine and CPK with IMMB stress, while magnesium deficient rats showed elevated plasma glucose, norepinephrine and CPK with stress. 2) Myocardial hydroxyproline and plasma epinephrine were not significantly different among groups. 3) In the rats fed adequate magnesium, stress increased plasma magnesium while rats on marginal magnesium diet did not show significant change with stress. 4) Plasma and myocardial calcium were increased in rats fed marginal Mg diet, but did not show any significant difference with stress.
Objectives : This study was performed to investigate the effects of Rhizoma Arisaematis water extract on lipid and glucose metabolism and histochemical change of obese rats. Methods and materials : 10 rats were divided into normal, control and RA (Rhizoma Arisaematis) groups. We fed the control group a high-fat diet and administered normal saline for 8 weeks. We fed the experimental group of rats a high-fat diet and administered an extract of Rhizoma Arisaematis for 8 weeks. At the end of the experiment, the rats were sacrificed to determine their chemical composition. The groups were examined for effects on blood serum lipids, blood sugar, blood insulin concentration and epididymal fat cells. Results : 1. Serum total cholesterol, triglyceride, HDL-cholesterol and glucose of the RA group decreased compared with those of the control group. These decreased rates were significant(P<0.05). 2. Serum LDL-cholesterol, total lipid, free fatty acid and the average size of epididymal fat cells of the RA group decreased compared with those of the control group. These decreased rates were significant(p<0.01). Conclusions : These results suggest that RA may be used to prevent or cure the obesity iniduced by a high-fat diet.
Background : Obesity, the syndrome caused by a high fat diet, is a disease. At the same time, obesity causes diabetes mellitus, hyperlipidemia, and cardiovascular disease. Recently, its prevalence rate is increasing. Yagwan-cheunghyeoltang (YCT) used in this experiment is the prescription of Yagwanmoon added to Cheunghyeol-tang which is reported to be very effective in weight loss controlling and serum cholesterol. It is also reported that Yagwanmoon has significant antioxidant effects and YCT has a significant effect on blood glucose control. Objectives : This study was conducted to experimentally evaluate the effects of YCT on obesity in rats induced by high fat diet. Methods : The experiment was conducted with 4-week-old male rat s divided into 5 groups. They were a normal diet group, a high fat diet group, a positive drug control group, a 1% YCT group, and a YCT 3% group, and were tested for eight weeks. After four weeks of inducing obesity by a high fat diet, rats were allowed to lose weight by following the normal diet group, approximately 30% compared with 10 rats in each group were determined as still obese. Changes in body weight and organ weight and serum cholesterol, triglyceride, glucose-density, low density lipoprotein cholesterol, antioxidant activity were checked. Results : In the experimental groups, we observed weight loss and visceral fat reduction, improvement of liver function, reduction of serum glucose, activation of HMG-CoA reductase inhibitor, reduction of concentrations of leptin and it showed a significant effect on antioxidants and lipid peroxidation. Conclusions : YCT has significant effects on the regulation of hyperlipidemia and lipid peroxidation associated with obesity and has significant effects on, antioxidants and lipid peroxidation, too. Additional clinical studies are needed.
The effects of the garlic preparation on the response of normal and cancer-stricken mice were studied. Food consumption of skin cancer-stricken group decrease more rapidly than that of normal group as cancer of skin develops, but here was no observable change in garlic groups. The rate of weight of increase was the highest in skin cancer-stricken group whereas that of garlic groups was reduced. The feed efficiency ratio was found to be very high in the normal group, whereas that in other goups after inculation of cancer cell was lower. The total protein content of serum was the highest in skin cancer-stricken group was observed to have more conspicuous tumor than other groups, the simultaneous administration of experimental diet and but inoculation of cancer cell produced more severe tumours. It appears that the garlic may exhibit the tendency to reduce the propagation of skin cancer of mice.

The present study was designed to elucidate the effects of the Co-60 γ irradiation and/or calcium-deficient diet on the periodontal tissue formation in rat pups. The pregnant three-week old Sprague-Dawley rats were used for the study. The experimental group was divided into two groups, irradiation/normal diet group (Group 2) and irradiation/calcium-deficient diet group (Group 3). The control group was non-irradiation/normal diet group (Group 1). The abdomen of the rats at the 19th day of pregnancy were irradiated with single absorbed dose of 350 cGy. The rat pups were sacrificed on the 14th day after delivery, and the maxillae including molar tooth germ were taken. The specimens including the 1st molar tooth germ were prepared to make tissue sections for light and transmission electron microscopy. Some of tissue sections for light microscopy were stained immunohistochemically with anti-fibronectin and anti-osteonectin antibodies. The results were as follows; 1. In the periodontal ligament forming area, the fibroblasts of Group Z showed irregular arrangement and low activity. The immunoreactivity between the fibroblasts and collagen fibers was decreased, compared with Group 1. The fibroblasts of Group 3 showed atrophic change and clumped nucleus. The collagen fibers showed cystic change and low immunoreactivity to the fibronectin. 2. In the cementum forming area, the cementoblasts of Group 2 showed decrease of number and atrophic change. The cementoblasts of Group 3 showed edematous change, atrophy of cytoplasm, and clumping of nucleus. 3. In the alveolar bone forming area, the bone of Group 2 was thin and various degree of immunoreactivity to the osteonectin. Group 3 showed edematous osteoblasts, fibrous degeneration of bone marrow, and weak immunoreactivity to the osteonectin.

This experimental study was designed to investigate the effects of Yukgunja-tang(YGJT) on the change of weight and serum total cholesterol, HDL-cholesterol, LDL-cholesterol, triglyceride, free fatty acid, total lipid, phospholipid level in obese mice induced by high fat diet. I fed normal group fed normal diet and administered DDW 0.2㎖ during 7 weeks, control group fed high fat diet and administered DDW 0.2㎖ during 7 weeks, sample A group fed high diet and administered YGJT 300mg/kg 0.2㎖ during 7 weeks, sample B group fed high diet and administered YGJT 500mg/kg 0.2㎖ during 7 weeks. The results were as follows ; 1. Sample A and Sample B were significantly decreased body weight(4weeks) and serum free fatty acid level in comparison with control group. 2. Sample A was significantly decreased body weight(7weeks), serum total cholesterol level and serum total lipid level, but significantly increased serum HDL-cholesterol level in comparison with control group. 3. Sample A was decreased serum LDL-cholesterol level, serum triglyceride level and serum phospholipid level in comparison with control group. 4. Sample B was increased serum HDL-cholesterol level in comparison with control group. 5. Sample B was decreased body weight(7weeks), serum total cholesterol level, serum LDL-cholesterol level, serum triglyceride level, serum total lipid level and serum phospholipid level in comparison with control group. According to above results, I suggest YGJT is able to be used for the herbal medication of obesity.
This study was performed to investigate antioxidative and anti-aging action of extracts from Sancho (Zanthoxylum schinifolium) leaves. Two extracts were obtained by 80% methanol extraction followed by subsequent fractionation with methylene chloride (MC) and n-butanol (B) and fed at one or three levels to rats on normal level (5%) of fat (control) and high fat(20%) in diets. Male Sprague-Dawley rats weighing about 100 g were divided into ten groups such as control diet group(C), control diet+0.50%B group (CB), control diet+0.50%MC group (CMC), high-fat diet group (HF), high-fat diet+0.25%B group (HBL), high-fat diet+0.50%B group (HBM), high-fat diet+0.75%B group (HBH), high-fat diet+0.25%MC group (HMCL), high-fat diet+0.50%MC group (HMCM) and high-fat diet+ 0.75%MC group (HMCH) and fed each diet for four weeks. The effects of the extracts on antioxidant enzyme activities and indices of lipid peroxidation and aging were seen only in high fat diet groups. Hepatic superoxide dismutase and aryleaterase activities were not changed by Sancho extracts. But glutathione peroxidase, catalase and paraxonase activities were significantly restored by both MC and B at the level of 0.75% lipid peroxide which was increased by high fat diet was significantly reduced by B and MC at the level of 0.25% and over. Lipofuscin fluorescence and cabonyl value were increased by high fat diet were reduced by B and MC at the level of 0.5% and over. It is concluded that the Sancho extracts can be utilized as functional ingredients of health foods for reducing oxidative stress.

Increased oxidative stress in obese diabetes may have causal effects on diabetic complications, including dyslipidemia. Lipopolysccharides (LPS) along with an atherogenic diet have been found to increase oxidative stress and insulin resistance. Cranberry has been recognized as having beneficial effects on diseases related to oxidative stress. Therefore, we employed obese diabetic animals treated with an atherogenic diet and LPS, with the aim of examining the effects of cranberry powder (CP) on diabetic related metabolic conditions, including lipid profiles, serum insulin and glucose, and biomarkers of oxidative stress. Forty C57BL/KsJ-db/db mice were divided into the following five groups: normal diet + saline, atherogenic diet + saline, atherogenic diet + LPS, atherogenic diet + 5% CP + LPS, and atherogenic diet + 10% CP + LPS. Consumption of an atherogenic diet resulted in elevation of serum total cholesterol and atherogenic index (AI) and reduction of high density lipoprotein (HDL)-cholesterol. However, with 10% CP, the increase in mean HDL-cholesterol level was close to that of the group with a normal diet, whereas AI was maintained at a higher level than that of the group with a normal diet. LPS induced elevated serum insulin level was lowered by greater than 60% with CP (P < 0.05), and mean serum glucose level was reduced by approximately 19% with 5% CP (P > 0.05). Mean activity of liver cytosolic glutathione peroxidase was significantly increased by LPS injection, however it was reduced back to the value without LPS when the diet was fortified with 10% CP (P < 0.05). In groups with CP, a reduction in mean levels of serum protein carbonyl tended to occur in a dose dependent manner. Particularly with 10% CP, a reduction of approximately 89% was observed (P > 0.05). Overall results suggest that fortification of the atherogenic diet with CP may have potential health benefits for obese diabetes with high oxidative stress, by modulation of physical conditions, including some biomarkers of oxidative stress.
