The Effect of Laminaria japonica Diet on the Pharmacokinetics of Glipizide in Rats (다시마 식이가 흰쥐에서 글리피지드의 체내동태에 미치는 영향)

  • Drug interactions with food, on occasion, lead to serious nutritional and functional changes in the body as well as alternations of pharmacological effect. It, therefore, should be necessary to take drug interactions with food into consideration for effective and safe therapeutics. Diabetes mellitus is a heterogeneous group of disorders characterized by abnormal glucose homeostasis, resulting in hyperglycemia, and is associated with increased risk of micovascular, macrovascular, and neuropathic complications. However, the precise mechanism of diabetes mellitus remains unclear. Three basic objectives in the care of diabetic patients are maintaining optimal nutrition, avoiding hypo- or hyperglycemia and preventing complications. The purpose of this study was to investigate thε effect of Laminaria japonica diet on the absorption, distribution, metabolism and excretion of glipizide which are frequently used in the treatment of diabetes. Diabetic rats induced by streptozotocin were employed in this study. Blood concentrations of oral hypoglycemic agents were measured by HPLC and resultant pharmacokinetic parameters were calculated by RSTRIP. The mechanisms of drug interaction with food were evaluated on the basis of pharmacokinetic parameters such as $k_{a},\;t_{1/2},\;C_{max},\;t_{max}$ and AUC. Administration of glipizide in normal rats treated with Laminaria japonica diet showed significant increase in AUC, $k_{a},\;t_{1/2},\;t_{max}$ and decrease in $C_{max}$, compared to those without Laminaria japonica diet. This might result from adsorption of glipizide on components of Laminaria japonica, causing delayed absorption. Administration of glipizide in diabetic rats treated with Laminaria japonica diet showed significant increase in $t_{1/2}\;and\;t_{max}$, and decrease in $C_{max}$, compared to those without Laminaria japonica diet. This might also result from adsorption of glipizide on components of Laminaria japonica, causing delayed absorption and flattened blood concentration of glipizide. The oral glucose test showed that Laminaria japonica diet could lower blood glucose level probably through either inhibiting the activity of disaccharidases, intestinal digestive enzymes, or delaying the absorption of glucose. More studies should be followed to fully understand pharmacokinetic changes of glipizide caused by long-term Laminaria japonica diet.

Chronic Administration of Baicalein Decreases Depression-Like Behavior Induced by Repeated Restraint Stress in Rats

  • Baicalein (BA), a plant-derived active flavonoid present in the root of Scutellaria baicalensis, has been widely used for the treatment of stress-related neuropsychiatric disorders including depression. Previous studies have demonstrated that repeated restraint stress disrupts the activity of the hypothalamic-pituitary-adrenal (HPA) axis, resulting in depression. The behavioral and neurochemical basis of the BA effect on depression remain unclear. The present study used the forced swimming test (FST) and changes in brain neurotransmitter levels to confirm the impact of BA on repeated restraint stress-induced behavioral and neurochemical changes in rats. Male rats received 10, 20, or 40 mg/kg BA (i.p.) 30 min prior to daily exposure to repeated restraint stress (2 h/day) for 14 days. Activation of the HPA axis in response to repeated restraint stress was confirmed by measuring serum corticosterone levels and the expression of corticotrophin-releasing factor in the hypothalamus. Daily BA administration significantly decreased the duration of immobility in the FST, increased sucrose consumption, and restored the stress-related decreases in dopamine concentrations in the hippocampus to near normal levels. BA significantly inhibited the stress-induced decrease in neuronal tyrosine hydroxylase immunoreactivity in the ventral tegmental area and the expression of brain-derived neurotrophic factor (BDNF) mRNA in the hippocampus. Taken together, these findings indicate that administration of BA prior to the repeated restraint stress significantly improves helpless behaviors and depressive symptoms, possibly by preventing the decrease in dopamine and BDNF expression. Thus, BA may be a useful agent for the treatment or alleviation of the complex symptoms associated with depression.

  • Earthquakes are almost impossible to predict and take place in a short time. In addition, there is little time to take aggressive action when an earthquake occurs. Therefore, there are more casualties and property damage than with other natural disasters. Recently, earthquakes have been occurring all over the world. As the number of earthquakes increase, studies on the safety of structures are being carried out. On the other hand, there are few studies on the electric facilities, which are relatively non - structural factors. Currently, electrical equipment in Korea is often not designed for earthquake safety and is quite vulnerable to damage when an earthquake occurs. Therefore, in this study, modeling was conducted through ABAQUS similar to an actual cabinet panel and 3D dynamic nonlinear analysis was performed using a natural seismic. According to seismic zone I and normal ground rock conditions of the power transmission and transmission facility seismic design practical guide, the maximum response acceleration of the performance level was 0.157g. In this study, however, it was not safe to reach the limit state of 30% of the analytical result at 0.1g for the general cabinet panel. From the results, the seismic fragility curve was derived and analyzed. The derived seismic fragility curve is presented as a quantitative basis for determining the limit state of the cabinet panel and can be utilized as basic data in related research.

  • Different Different from a single attack, in DDoS Attacks, the botnets that are distributed on network initiate attacks against the target server simultaneously. In such cases, it is difficult to take an action while denying the access of packets that are regarded as DDoS since normal user's convenience should also be considered at the target server. Taking these considerations into account, the DDoS botnet detection system that can reduce the strain on the target server by detecting DDoS attacks on each user network basis, and then lets the network administrator to take actions that reduce overall scale of botnets, has been implemented in this study. The DDoS botnet detection system proposed by this study implemented the program which detects attacks based on the database composed of faults and abnormalities collected through analyzation of hourly attack traffics. The presence of attack was then determined using the threshold of current traffic calculated with the standard deviation and the mean number of packets. By converting botnet-based detection method centering around the servers that become the targets of attacks to the network based detection, it was possible to contemplate aggressive defense concept against DDoS attacks. With such measure, the network administrator can cut large scale traffics of which could be referred as the differences between DDoS and DoS attacks, in advance mitigating the scale of botnets. Furthermore, we expect to have an effect that can considerably reduce the strain imposed on the target servers and the network loads of routers in WAN communications if the traffic attacks can be blocked beforehand in the network communications under the router equipment level.

Effect of Birth Weight and Litter Size on Productive Performance of Rabbits

  • This study chiefly aims to ascertain the effect of birth weight and litter size on productive performance in rabbits. After delivery kits were removed from their mothers for individual weighing and randomly assigned to one of the following three experimental treatments: Group (KT 6) with six tiny kits (about 45 g) per litter (21 litters); Group (KT 8) with eight tiny kits (about 45 g) per litter (17 litters); Group (KN8) with eight normal kits (about 70 g) per litter (20 litters). The litters, separated from their mothers by putting them in a closed cage, suckled one time a day for 15 minutes. Daily milk production was measured by weighing the does immediately before and after suckling; body weight of kits was measured on a weekly basis. At weaning 60 rabbits per group were raised for six weeks to ascertain post-weaning performance and health. Compared with the two other groups, the kits of group KT8 ingested smaller quantities of milk both during the first 3 weeks (497.7 vs. 520.0 vs. 544.3 g, respectively for groups KT8, KN8, KT6) and the following period (261.3 vs. 285.0 vs. 302.7 g, respectively for KT8, KN8, KT6). The differences between groups KT8 and KT6 were statistically significant (p<0.01) in the first period while in the second period also KN8 group showed milk intake different in respect of KT8 (p<0.01) and KT6 (p<0.05) groups. Solid feed intake was significantly (p<0.05) lower in group KT6 (23.1 vs. 26.2 vs. 26.8 g/d, respectively for groups KT6, KN8, KT8). At weaning the kits of group KN8 always weighed more (966 vs. 894 vs. 888 g; p<0.01, respectively for groups KN8, KT6, KT8). After weaning, no parameter considered (daily weight gain, feed intake, feed conversion ratio, mortality and risk sanitary index) was affected by birth weight and litter size.

Effect of Dietary Grape Pomace on Lipid Oxidation and Related Enzyme Activities in Rats Fed High Fat Diet (포도박이 고지방식이를 섭취한 흰쥐의 지질 산화와 항산화 효소 활성에 미치는 영향)

  • The present study was conducted to investigate the effect of dietary supplementation of grape pomace on lipid peroxidation and related enzyme activities of rats fed high fat diet. Male Sprague-Dawley rats weighing about 90 g were assigned to 4 experimental groups of 8 rats on the basis of their body weight. The high fat diet contained additional 15% lard to AIN 93-based diet. Rats were fed experimental diets containing 5% grape pomace for 4 weeks. Dietary supplementation of grape pomace reduced serum concentration of lipid peroxide in rats fed high fat diet. Hepatic concentration of lipid peroxide tended to be lower by feeding grape pomace. Hepatic total glutathione content and GSH/GSSG ratio were increased by grape pomace feeding in normal or high fat diet groups. Hepatic superoxide dismutase activity of grape pomace group with high fat diet was induced significantly compared with high fat diet group without grape pomace. Hepatic catalase activity of high fat fed rats was induced by feeding grape pomace. Grape pomace diet increased glutathione-S-transferase and glutathione peroxidase activities in rat liver fed high fat. Hepatic glucose-6-phosphatase activity was not affected by dietary supplementation of grape pomace in rats fed high fat. These results suggest that dietary supplementation of grape pomace may alleviate lipid peroxidation through antioxidant effect in rats fed high fat.

Layer 2 Routing with Multi-Spanning Tree Per a Node (노드 당 다중 스패닝 트리를 이용한 2계층 라우팅)

  • Carrier Ethernet backbone network integrates distributed layer-2 based metro networks. In this networks, Multiple Spanning Tree Protocol (MSTP) has been uscd as a main routing protocol that allows multiple spanning trees in a network. A better routing protocol called IEEE802.1aq - Shortest Path Bridging (SPB) is recently proposed, that generates the shortest spanning tree per a destination node. As SPB provides a routing path per a destination node, there is no way to adapt network traffic at normal condition. If we are free from the principle of "a spanning tree per a destination node", we can achieve adaptive routing. Based on this philosophy, we propose a new spanning tree based protocol - Edge Node Divided Spanning Tree (ENDIST). ENDIST divides an edge node into sub-nodes as many as connecting links from the node and each sub-node generates a single shortest path tree based on SPB. Depending on network or nodal status, ENDIST chooses a better routing path by flow-basis. This added traffic engineering ability contributes to enhanced throughput and reduced delay in backbone networks. The simulation informs us that ENDIST's throughput under heavy load performs about 3.4-5.8 and 1.5-2.0 times compared with STP's and SPB's one respectively. Also, we verified that ENDIST's throughput corresponds to the theoretical upper bound at half of cases we investigated. This means that the proposed ENDIST is a dramatically enhanced and the close-to-perfect spanning tree based routing schemes.

A Study of Real Ship Experiments to Estimate the Heeling Angle of Passenger Type Ship when Turning (여객선형의 선회 중 횡경사 추정에 관한 실선 실험 연구)

  • Passenger ships and training ships have a common feature in that they serve many passengers. Thus, safe navigation is very important. During normal sailing, a ship may turn using various types of steering, including maneuvers to avoid collisions with dangerous target. When a ship turns, a heeling angle occurs. If trouble arises during sailing, a dangerous heeling angle may result or a capsizing accident. In this study, the heeling angle during turning was measured through experimentation with two training ships similar to passenger ships. These findings were compared with theoretical formulas for heeling angle when turning. We confirmed that the limit of the maximum heeling angle estimation using heeling angle formula when turning presented in IMO stability criteria. In addition, it was confirmed that the maximum estimated heeling angle can be reached by applying the result calculated in the theoretical formula 1.4 times when turning right and 1.1 times when turning left to reflect sailing speed when of rudder hard over. It is expected that this study will provide basis data for establishing safe operation standards for the prevention of dangerous heeling angles when turning.

Effects of Differential Distribution of Microvessel Density, Possibly Regulated by miR-374a, on Breast Cancer Prognosis

  • Background: The discovery that microRNAs (miRNAs) regulate proliferation, invasion and metastasis provides a principal molecular basis of tumor heterogeneity. Microvessel distribution is an important characteristic of solid tumors, with significant hypoxia occurring in the center of tumors with low blood flow. The distribution of miR-374a in breast tumors was examined as a factor likely to be important in breast cancer progression. Methods: Breast tissue samples from 40 patients with breast cancer were classified into two groups: a highly invasive and metastatic group (HIMG) and a low-invasive and metastatic Group (LIMG). Samples were collected from the center and edge of each tumor. In each group, six specimens were examined by microRNA array, and the remaining 14 specimens were used for real-time RT-qPCR, Western blot and immunohistochemical analyses. Correlation analysis was performed for the miRNAs and target proteins. Follow-up was carried out during 28 months to 68 months after surgery, and survival data were analyzed. Results: In the LIMG, the relative content of miR-374a was lower in the center of the tumor than at its edge; in the HIMG, it was lower at the edge of the tumor, and miR-374a levels were lower in breast cancer tissues than in normal tissues. There was no difference between VEGF-A and VCAM-1 mRNA levels at the edge and center of the tumor; however, we observed a significant difference between VEGF-A and VCAM-1 protein expression levels in these two regions. There was a negative correlation between miR-374a and target protein levels. The microvessel density (MVD) was lower in the center of the tumor than at its edge in HIMG, but the LIMG vessels were uniformly distributed. There was a significant positive correlation between MVD and the number of lymph node metastases (Pearson correlation, r=0.912, P<0.01). The median follow-up time was 48.5 months. LIMG had higher rate of disease-free survival (100%, P=0.013) and longer median survival time (66 months) than HIMG, which had a lower rate of 75% and shorter median survival time (54 months). Conclusions: Our data demonstrated miR-374a to be differentially distributed in breast cancer; VEGF-A and VCAM-1 mRNA had coincident distribution, and the distribution of teh respective proteins was uneven and opposite to that for the miR-374a. These data might explain the differences in the distribution of MVD in breast cancer and variation in breast cancer prognosis.
