Effects of medicinal herb extract on nonspecific immune responses, hematology and disease resistance against Edwardsiella tarda in olive flounder, Paralichthys olivaceus were evaluated. Wormwood, Artemisia asiatica NAKAI and barrenwort, Epimedium koreanum NAKAI were mixed at a ratio of 7 : 3 (w/w) for 2-herbs extract and wormwood, barrenwort, Korean forsythia, Forsythia koreana NAKAI, chrysanthemum, Chrysanthemum zawadskii var. latilobum KITAMURA, peppermint, Mentha arvensis L. var, piperascens MALINV., great burnet, Snaguisorba afficinalis L., Lizard tail. Saururus chinensis BAILL., mulberry, Morus alba L., and star anise, Illicium varum HOOK, f, at the same weight for 9-herbs extract. Two-herbs of 9-herbs extract were prepared by heating after adding 10㎖ of distilled water per g of the herb mixtures. Fish (10.3$\pm$2.5g) were fed the experimental diets supplemented with the 2-herbs or 9-herbs extract at the different concentrations of 0%, 0.1%, 0.5% and 1% per kg diet for 12 weeks. Lysozyme and bactericidal activities of serum, and hematological characteristics were examined during experimental period. After feeding test period, all experimental groups were challenged with E. tarda. Lysozyme activity from the fish fed the diet supplemented with 0.1% or 0.5% of 2-herbs extract was significantly higher than the control. But there was no difference both in bactericidal activity and hematology among each group. Sixty seven % of relative percent survival values (RPS) in the group fed the diet supplemented with 0.1% of 2-herbs was higher than the other group and the control. These results suggest that supplenmentation of 0.1% of 2-herbs extract to a commercial diet may enhance disease resistance in olive flounder. Although both 0.1% and 0.5% 9-herbs extract did not improve non-specific immune reponses, they could enhance disease resistance of 53% RPS, respectively.

Subtoxic 농도의 수은 투여에 따른 Arthus 및 DTH반응의 결과는 $Arthus(T_3)$반응에 있어서 수은 투여군과 대조군 사이에 유의한 차이가 나타나지 않았던 반면, $T_{24},\;T_{48}$의 DTH반응은 수은 투여군이 대조군에 비하여 억제되었다. 수은 투여 마우스의 면양적혈구(SRBC)에 대한 항체반응을 측정한 결과 면양적혈구에 대한 총응집소가는 수은 투여 농도에 의존적으로 대조군의 값에 비해 억제되었다. 시험관 내에서 마이토젠에 대한 비장세포의 증식 반응은 수은 농도에 의존적으로 억제되었다. 수은 투여군의 IgE 생산은 농도 증가에 따라 $1.7\sim2.0$배의 수준으로 증가하였으며, $IgG_1$의 경우도 증가의 경향을 보였으나 대조군에 비해 큰 차는 아니었다. 이상의 결과를 살펴볼 때 수은 중독시 출현할 수 있는 면역반응의 변화로서, 초기에는 세포성 면역반응의 감소가 현저할 것으로 사료되나 중독이 진행됨에 따라 Ig농도의 상승을 제외한 체액성 면역반응의 전반적인 감소가 수반될 것으로 예측된다.
This study investigated the effects of dietary supplementation with hot water extracts (HE) of selfheal (Prunella vulgaris) and Lactobacillus rhamnosus culture fluid (LF) on the growth and non-specific immune responses of juvenile olive flounder. A total of 270 fish ($5.07{\pm}0.01g$, average $weight{\pm}SD$) were divided randomly into nine groups, and three groups were fed one of three isonitrogenous (51% crude protein) and isocaloric (17.6 MJ/kg) diets. The diets contained no supplement, 50 mL hot water extract, or 50 mL L. rhamnosus fluid (designated as diets CON, HE and LF, respectively). During the 8-week feeding trial, the growth, feed utilization and survival of the fish were not significantly affected by the experimental diets. There were no significant differences in the hematocrit and hemoglobin of fish fed each of the experimental diets. However, the serum alanine aminotransferase and aspartate aminotransferase activity were lower with the dietary supplements containing HE and LF. The alanine aminotransferase activity of fish fed the HE diet was significantly lower than that of fish fed the control diet. The hematocrit, hemoglobin, and nitroblue tetrazolium (NBT) activity were higher 3 hr after feeding than 24 hr afterwards. The NBT activity of fish fed the HE and LF diets were significantly higher than that of fish fed the control diet 3 hr after feeding. The findings suggest that dietary supplementation with HE and LF could enhance the nonspecific immune responses of juvenile olive flounder.
본 연구에서는 다양한 면역 유도제를 주사하여 갯지렁이의 면역을 유도시킨 후, 갯지렁이의 항균 펩타이드를 가장 많이 발현시킬 수 있는 최적의 면역 유도제를 선정하였으며, 선정된 면역 유도제로 면역을 유도한 갯지렁이를 분말화 하여 사료에 첨가하였을 때 양식 어류의 면역 활성을 증가시킬 수 있는지에 대한 여부를 조사하였다. 넙치와 조피볼락은 면역활성을 위한 갯지렁이 분말의 사료 첨가율이 서로 달랐으며, 넙치의 경우 0.1%의 사료 첨가율이 넙치의 라이소자임 활성, 혈청 내 코르티졸 농도뿐만 아니라 E. tarda에 대한 항병성에 가장 높은 효과를 보였다. 조피볼락은 0.5%의 사료 첨가율이 조피볼락의 라이소자임 활성을 증가시키고 혈청 내 코르티졸 농도를 감소시켰으며 S. iniae에 대한 항병성 또한 증가시켰다. 따라서 넙치와 조피볼락은 각각 면역 유도된 갯지렁이 분말이 0.1%와 0.5% 첨가된 사료를 급이하였을 때 면역 활성을 가장 많이 유도 할 수 있을 것으로 생각되지만, 반면 그 이상의 농도로 면역 유도된 갯지렁이 분말을 사료에 첨가하여 급이할 시 오히려 면역 활성에 부정적인 영향을 미칠 수 있을 것으로 생각된다. 이번 연구에서는 주간 8 갯지렁이 분말 첨가 사료를 급이한 것에 대한 양식 어류의 면역 활성을 검증 하였으며, 추후 연구에서는 갯지렁이 첨가사료의 급이 기간을 고려한 연구가 진행되어야 할 것으로 생각된다.
본 연구는 양식 넙치 Paralichthys olivaceus 사료 내 생균제 첨가가 넙치의 성장, 면역반응 및 병저항성에 미치는 영향을 평가하였다. 실험사료는 넙치용 배합사료(조단백질 52%, 조지방 11%, 조섬유 3%, 조회분 14%, 인 2.7%, 칼슘 1.5%, Suhyup Co., Korea)에 Bacillus sp. IS-2를 첨가하여 $10^3$, $10^5$, $10^7$ CFU/kg의 실험사료를 제작하였다. 2주간의 예비사육 후, 평균무게 $210{\pm}13g$인 실험어를 1,000 L 원형수조에 실험구 당 70마리씩 무작위로 배치하여 실험사료를 1일 2회 어체 중의 2%씩 12주 동안 공급하였다. 성장도 조사 결과 모든 실험구에서는 일반사료를 투여한 대조구에 비해 높은 성장률을 나타내었으며 $10^5$ CFU/kg 실험구에서는 대조구에 비해 약 13% 정도 높은 성장률을 나타내었다. 혈액분석 결과에는 glucose를 제외한 GOT, GPT, 단백질, 총콜레스테롤 등에서 실험구간 유의적인 차이를 나타내지 않아 생균제로 인한 간독성이나 어체 내 문제가 발생하지 않은 것으로 사료된다. Glucose인 경우 실험 종료 시점에서 증가되는 변화를 확인 할 수 있었는데 이는 수온, 수질에 의한 일시적인 현상이라 사료된다. Respiratory burst activity (NBT assay)에 있어서는 실험사료를 공급한 실험구가 대조구에 비해 높은 값을 확인할 수 있었으며 특히 $10^5$ CFU/kg 실험구와 $10^7$ CFU/kg 실험구에서 유의적으로 높은 값을 확인하였다. 혈청의 lysozyme 및 백혈구 활성에 있어서도 실험사료를 투여한 실험구에서 대조구에 비해 높은 활성을 확인 하였다. 공격실험 결과, Streptococcus inae를 접종한지 5일째부터 폐사가 시작되어 7일째 일반사료를 투여한 대조구에서는 100% 폐사율을 보인 반면, $10^3$ CFU/kg 실험구에서는 73%의 폐사율을 $10^5$ CFU/kg 실험구에서는 53%의 폐사율을 나타냈으며, $10^7$ CFU/kg 실험구에서는 45%의 폐사율을 보여 대조구에 비해 많게는 55% 이상의 높은 생존율을 나타내었다. 사료 내 첨가한 Bacillus sp. IS-2의 장내 생존 확인을 위해 실험이 종료 된 후 모든 실험구와 대조구 실험어의 장을 분리하여 배양 된 균체를 DNA를 분리 한 후에 제작 된 ditection primer를 이용한 PCR 결과 일반사료를 투여한 대조구를 제외한 모든 실험구에서 1,465 bp의 PCR product를 확인 할 수 있었다. 상기 결과를 토대로 양식넙치 사료 내 Bacillus sp. IS-2의 첨가는 양식넙치의 성장 및 면역증강, S. iniae에 대한 병정항성에 좋은 효과를 나타내어 사료첨가제로써의 이용 가능성이 클 것이라 사료된다.
Bujeonghangamtang(扶正抗癌湯) has been used for cure of tumor as a traditional medicine without any experimental evidence to support the rational basis for its clinical use. This study was carried out to evaluate the possible therapeutic or antitumoral effects of Bujeonghangamtang extract against tumor, and to carry out some mechanisms responsible for its effect. Some kinds of tumor were induced by the typical application of 3-methylcholanthrene. (MCA) or by the implantation(s.c) of malignant tumor cells such as leukemia cells(3LL cells) or sarcoma cells(Sl80 cells). Treatment of the Bujeonghangamtang water-extract (dailly 1mg/mouse, i. p.) was continued for 7 days prior to tumor induction and after that the treatment was lasted for 20 days. Against squamous cell carcinoma induced by MCA, Bujeonghangamtang decreased not only the frequency of tumor production but also the number and the weight of tumors per tumor bearing mice (TBM). Bujeonghangamtang also significantly suppressed the development of 3LL cell and S180 cell-implanted tumors in occurrence-frequency and their size, and some developed tumors were regressed by the continuous treatment of Bujeonghangamtang extract into TBM. In vitro, treatment of Bujeonghangamtang extract had no effect on the growth of some kinds of cell line such as FsaII, A431 strain but significantly inhibited the proliferation of 3LL, S180 cells and augmented the DNA synthesis of mitogen-activated lymphocytes. Bujeonghangamtang also stimulated the migrative ability of leukocyte, the MIF and IL-2 production of T lymphocytes, but not IL 6 production of B cells. Bujeonghangamtang-administration to mice enhanced NK cells activities. These results demonstrated that Bujeonghangamtang extract exhibited a significant prophylactic benefits against tumors and its antitumor activity was manifested depending on the type of tumor cells. And these results also suggested that effect of Bujeonghangamtang might be chiefly due to nonspecific enhancement of NK cell activities and cell-mediated immune responses.
This study was conducted to replace fish meal (FM) with three plant proteins (soybean meal, soy protein concentrate, and wheat gluten) in diets for growing olive flounder Paralichthys olivaceus. The control diet was formulated to contain 65% sardine FM and four other replacement diets were formulated to replace FM with the plant proteins by 25, 30, 35 and 40% (designated FM25, FM30, FM35 and FM40, respectively). The replacement diets were added with three essential amino acids (lysine, methionine and threonine) to meet their requirements for the fish. Olive flounder (initial average weight, 96.8±0.2 g) were randomly distributed into 20 tanks (425 L each) at a density of 25 fish per tank. Four replicate groups of fish were fed one of the diets two times daily for 15 weeks. At the end of the feeding trial, no significant differences were found among all the fish groups in growth performance, feed utilization, nonspecific immune responses and hematological health parameters. Thus, this result indicates that the plant proteins with the three limiting amino acids could replace FM up to 40% in diets for growing olive flounder.
Background: In the thymus, developing thymocytes continually interact with thymic epithelial cell components. Self MHC restriction of mature T cells are imposed in the thymus through interaction of immature double positive thymocytes and thymic cortical epithelial cells. The site of negative selection, however, is a matter of debate. Through systemic injection of anti-TCR antibody or antigenic peptides, investigators suggested that most of the negative selection occurs in the thymic cortex. But the requirements for negative selection, i.e cellular counterparts and costimulatory molecules are more available in the medulla or cortico-medullary junction rather than in the thymic cortex. Methods: The direct and indirect pathways of thymocyte death after systemic anti-TCR antibody injection were separated through several experimental systems. B6 mice were either adrenalectomized or sham-adrenalectomized to evaluate the role of endogenous glucocorticoids from adrenal gland. Role of TNF were evaluated through using TNF receptor double knockout mice. Results: We found that without indirectly acting mediators such as $TNF-\alpha$ or corticosteroid, double positive thymocyte death were minimal by systemic injection of anti-TCR antibody in TNF receptor double knockout neonatal mice. Also by analyzing neonatal wild-type mice with adoptively transferred mature T cells, only peripheral activation of mature T cells could induce extensive double positive thymocyte death. Conclusion: Thus, systemically injected anti-TCR antibody mediated thymocyte death are mostly induced through indirect pathway.
Objective: Currently available therapies for human malignant gliomas have limited efficacy. Toxoplasma gondii, an obligate intracellular protozoan parasite, and Quil-A are nonspecific, potent immune stimulants. T. gondii is shown to have antitumor activity in some types of cancers. Therefore, this study is undertaken to evaluate the antitumor effect of Toxoplasma lysate antigen (TLA), alone or in combination with Quil-A, on human glioma U373MG and U87MG cells. Methods: The in vitro effects of TLA alone or in combination with Quil-A on the proliferation, invasion, and apoptosis of glioma cells were tested using MTT, Matrigel invasion, and DNA fragmentation assays, and the in vivo effects on the growth of gliomas were evaluated in athymic nude mice transplanted with glioma cells. Results: Treatment with TLA resulted in the suppressed proliferation and invasion of both U373MG and U87MG cells, in a dose-dependent manner. In addition, at high concentration, TLA induced glioma cell apoptosis. When TLA was administered in the mouse glioma model, malignant glioma growth was decreased. The combined treatment of TLA with Quil-A significantly inhibited the proliferation and invasion of cultured cells as well as tumor mass of implanted mice. Conclusion: TLA inhibits the proliferation and invasion of glioma cells in vitro and in vivo, and these antitumor effects of TLA are significantly enhanced by the addition of Quil-A.
