To investigate the effect of hyperoxia on EKG findings and to evaluate the applicability of EKG as noninvasive monitoring index of oxygen toxicity, 38 rabbits were continuously exposed to 6 different conditions-3 hyperbaric oxygenations (HBO-2.5, 3.5 and 5ATA, 100% $O_2$), normobaric oxygenation (NBO,100% $O_2$), hyperbaric aeration (HBA-5ATA, 21% $O_2$) and normobaric aeration (NBA, 21% $O_2$)-for 120 minutes and their EKG and time to dyspnea and convulsion were recorded. Dyspnea and death were observed in exposure conditions of HBO-3.5 and HBO-5 (Positive rate of dyspnea 10%, 100%, death : 10%, 25%, respectively) only, and convulsion in 4 oxygenation groups (NBO;20%, HBO-2.5;20%, HBO-3.5;20%, HBO-5;88%). Abnormal EKG findings included arrhythmia and ST-T changes and the incidences was increasing with doses(partial pressure of oxygen). In addition to EKG change, findings observed during exposure were dyspnea and convulsion in the order of appearance and when non specific ST-T change was accepted as positive(abnormal) finding, the frequency of abnormal EKG was statistically significant(p<0.01), but when it was excluded from positive results, the frequency of EKG change was not significant(p>0.05). These results suggest that the effect of hyperoxia on heart is myocardial ischemia and arrhythmia, that oxygenation more than 3.5ATA causes myocardial damage in 120 minutes exposure, and that EKG is valuable as monitoring index of oxygen toxicity.
Zinc oxide nanoparticles (ZnONPs) are widely used in a variety of products and cosmetic products including paper, paints, plastics and sunscreen. However, information on the safety of ZnONPs are not enough and many publications suggest possible toxic effects on environmental and human health. Furthermore, physico-chemical characteristics of nanoparticles makes it hard to test toxicity using the test guidelines of chemicals adopted by regulatory bodies. In this study, stability of ZnONPs was investigated using different types of isotonic solution, which is important in the toxicity study of intravenous route. Precipitation, aggregation, size, zeta potential and morphology of ZnONPs were evaluated with different times and concentrations. Precipitation of ZnONPs were observed in ionic isotonic solution including phosphate-buffered saline, Kreb's-Ringer solution, physiological salt solution and cell culture media of DMEM (Dulbecco's Modified Eagle's Medium) with 10% fetal bovine serum. On the other hand, they were stable without precipitation in non-ionic isotonic solution such as 5% glucose and 2% glycerol, respectively, which are biocompatible for intravenous injection. The average size of ZnONPs in 5% glucose and 2% glycerol was stably maintained, which is less than 30 nm and very similar as that in water dispersion of ZnONPs, provided by the manufacturer. The stability was maintained during the experimental period of 5 days and diluted state up to 15,000 ppm. These data suggest that 5% glucose and 2% glycerol solution can be used for the vehicles of ZnONPs in the toxicity study of intravenous injection route.
Protein and peptide candidates are screened to apply therapeutic application as a drug. Ensuring that these candidates are delivered and maximized effectiveness is still challenging and a variety of studies are ongoing. As drug delivery system vehicles, cell-penetrating peptide (CPP) can deliver various kinds of cargo into the cell cytosol. In a previous study, we developed Ara27 CPP, which are a zinc knuckle family protein of Arabidopsis, and confirmed internalization in human dermal fibroblasts and human dental pulp stem cells at low concentration with short time treatment condition without any toxicity. Ara27, an amphipathic CPP, could be modified and utilized in the biomedical field excluding the risk of toxicity. Therefore, we would like to confirm the non-toxic induced penetrating ability of Ara27 in various cell lines. The purpose of this study was to screen the cell internalization ability of Ara27 in various cell lines and to confirm Ara27 as a promising core CPP structure. First, Ara27 was screened to confirm non-toxicity concentration. Then, fluorescence-labeled Ara27 was treated on human normal cell lines, cancer cell lines and animal cell lines to identify the cellular internalization of Ara27. Ara27 was well intracellular localized in all cell lines and the intensity of fluorescence was remarkably increased in time pass manner. These results indicate that Ara27 has the potential as a core structure for applications in various drug delivery systems.
Background: Platinum-based regimens are effective treatments for advanced non-small cell lung cancer (NSCLC), but the five-year survival rate is still less than 20%. One possible factor appears to be resistance involving polymorphisms in the CTR1 gene which plays an importance role in accumulation of platinum in the cytoplasm. Purpose: To establish both prevalence of CTR1 polymorphism and its impact on treatment related toxicity in Thai advanced NSCLC patients. Materials and Methods: Thirty-two advanced NSCLC participants received carboplatin and gemcitabine during January to June 2016 at King Chulalongkorn Memorial Hospital (KCMH) were recruited for analysis of the CTR1 rs12686377 genotype. These participants were planning to be treated with platinum-based chemotherapy for at least two cycles. Results: Allele frequency of CTR1 polymorphism $G{\rightarrow}T$ was found to be 25%. The results showed that genetic polymorphism at CTR1 rs12686377 was associated with emesis side effects (P = 0.020) and neuropathic symptoms (P = 0.010). In addition, hematologic side effects in terms of anemia also tended to be related to this polymorphism. Conclusions: This is the first study suggesting that polymorphism at CTR1 rs12686377 may be associated with toxicity from platinum-based regimens. Therefore, it could be a factor to aid in treatment decision-making.

Stink bug, Canthecona furcellatta (Hemiptera: Pentatomidae), is an important predator of silkworm larvae. Nymphs and adult attack the early stage silkworm larvae and causes about 10-15 per cent loss to silk industry. Synthetic organic pesticides has tremendous impact on minimizing the pest population but repeated and frequent use has created problems of residual toxicity, development of resistance to insecticides, pest resurgence and out break, phyto-toxicity and hazards to non target species and beneficial organism. Silkworms are very sensitive to pesticides; therefore, attempt has made to control the bug population through introduction of its native natural enemies in the silkworm-rearing field. Biological control has tremendous scope in sericulture because it is eco-friendly in nature and non-harmful farmers. Native natural enemies have been screened. Psix striaticeps, Trissolcus spp. and Telenomus spp. have been recorded as the most potential parasitoid against pentatomid bug. Life cycle, sex ratio and other various attributes of the par-asitoids have been recorded. The parasitization potential of the parasitoid is very high and they have the ability to discriminate between parasitized and unpar-asitized host. Mass propopagation technique under laboratory condition has been standaydized.
Two dogs were presented with melena, vomiting and depression after accidental swallowing of candy form of Strepsils (flurbiprofen), which is one of non-steroidal anti-inflammatory drugs used in human medicine for controlling a sore throat. These dogs had common signs of anemia induced by gastrointestinal ulceration and hemorrhage with azotemia and leukocytosis. The dogs were treated with blood transfusion, fluid therapy, proton-pump inhibitor, antiemetics, mucus protectant and antibiotic. Although most of clinical signs of two dogs were resolved, azotemic problem with evidence of renal injury have remained.
This study was undertaken to evaluate the antioxidant potential of A. lanata on oxalate mediated free radical toxicity in ethylene glycol induced calcium oxalate urolithic rats. Calcium oxalate (CaOX) stone was induced by 0.75% ethylene glycol in drinking water for 28 days. From $29^{th}$ day onwards, the CaOX urolithic rats were treated with A. lanata aqueous suspension (2,000 mg/kg body weight/dose/day) orally for another 28 days. At the end of experimental periods the animals were sacrificed, samples were collected and analyzed the lipid peroxidation product, protein oxidation product, enzymatic and non-enzymatic antioxidants in normal and experimental groups. Lipid peroxidation and protein oxidation products were significantly elevated while enzymatic and non-enzymatic antioxidant levels were significantly decreased in ethylene glycol induced CaOX urolithic rats when compared with control rats. The above alterations were reverted to near control in rats treated with aqueous suspension of A. lanata. This study suggests that A. lanata could prevent the free radical formation from calcium oxalate urolithiasis in rats and protecting the renal cells from oxidative injury.
Objective : To observe the efficacy and safety of CheungYeolYiSeup-tang and Hwangbaek external dressings on dampness-heat pattern atopic dermatitis(AD) in a non-comparative study. Methods : 10 patients with AD were included for 4 weeks of treatment. Efficacy and safety assessment included the scoring atopic dermatitis index(SCORAD), typical signs and symptoms of AD, results of some laboratory tests related to toxicity, and the incidence of adverse events. Results : Improvements in efficacy parameters were observed and produced no significant changes in laboratory tests related to toxicity in these patients. Their SCORAD results significantly decreased after 4weeks(P value<.01, according to the Wilcoxon sum of ranks test). Similarly, significant reductions from baseline in subjective pruritus scores and (P value<.05 by the Wilcoxon sum of ranks test) and the mean average of individual signs and symptoms of AD were reported after 4 weeks(P value<.05, P<0.01 by the Wilcoxon sum of ranks test). There were no significant changes in eosinophil, neutrophil, lymphocyte, immunoglobulin E and ESR in blood serum by paired ttest. Conclusion :CheungYeolYiSeup-tang administration and Hwangbaek external dressings are an effective and safe treatment for the management of dampness-heat pattern atopic dermatitis.
