DNA Sequence analysis and rfbM gene amplification using PCR for detect salmonella C1 serogroup (살모넬라 C1 serogroup 특이 rfbM 유전자 증폭과 염기서열 분석)

  • The Salmonella rfb gene encoding for the biosynthesis of the oligosaccharide-repeating units of the O-antigenic determinants was cloned and sequenced. A set of nucleotide primers(a forward and reverse) was selected to target a defined region of the guanosine diphospho-mannose(GDP-Man) pyrophosphorylase synthase gene : rfbM of Salmonella C serogroup. The primer set was used to develop a PCR-based rapid and specific detection system for Salmonella C1 serogroup. Amplification bands of predicted size(1,422bp) were generated from 11 different Salmonella C1 isolates. The bands were verified to be specific for the C1 serogroup by Southern blot analysis using reference homologous DNA specificity was further confirmed by the lack of reactivity with heterologous DNA derived from non-salmonella members of the family enterobacteriaeceae. A specificity of 100% was deduced along with a very high sensitivity shown by a detection limit of 1fg of a purified DNA template. The isolated DNA sequence was found to be 99.8% homologous to S montevideo but the related primers amplified with the predicted band sizes with all the Salmonella C1 serogroups tested. It is concluded that the PCR protocol based on the rfbM gene from S cholerasuis is optimal fast and specific for the detection of Salmonella C1 serogroup and also the corresponding probe is suitable for rapid detection of all Salmonella C1 serogroup DNA tested. This technology should facilitate the identification of contaminated pig products and for any other products contaminated with the Salmonalla C1 serogroup. The immediate impact of this developed method will be in the area of food safety of pig products with the potential prospect for adaptation to other food inspection technologies.

Outcomes and Patency of Complex Configurations of Composite Grafts Using Bilateral Internal Thoracic Arteries

  • Background: It is generally agreed that using a bilateral internal thoracic artery (BITA) composite graft improves long-term survival after coronary artery bypass grafting (CABG). Although the left internal thoracic artery (LITA)-based Y-composite graft is widely adopted, technical or anatomical difficulties necessitate complex configurations. We aimed to investigate whether BITA configuration impacts survival or patency in patients undergoing coronary revascularization. Methods: Between January 2006 and June 2017, 1,161 patients underwent CABG at Seoul National University Bundang Hospital, where the standard technique is a LITA-based Y-composite graft with the right internal thoracic artery (RITA) sequentially anastomosed to non-left anterior descending (LAD) targets. Total of 160 patients underwent CABG using BITA with modifications. Their medical records and imaging data were reviewed retrospectively to investigate technical details, clinical outcomes, and graft patency. Results: Modifications of the typical Y-graft (group 1, n=90), LITA-based I-graft (group 2, n=39), and RITA-based composite graft (group 3, n=31) were used due to insufficient RITA length (47%), problems using LITA (28%), and target vessel anatomy (25%). The overall 30-day mortality rate was 1.9%. Among 116 patients who underwent computed tomography or conventional angiography at a mean interval of 29.9±33.1 months postoperatively, the graft patency rates were 98.7%, 95.3%, and 83.6% for the LAD, left circumflex artery, and right coronary artery territories, respectively. Patency rates for the inflow, secondary, and tertiary grafts were 98.2%, 90.5%, and 80.4%, respectively. The RITA-based graft (group 3) had the lowest patency rate of the various configurations (p<0.011). Conclusion: LITA-based Y composite graft, showed satisfactory clinical outcomes and patency whereas modifications of RITA- based composite graft had the lowest patency and 5-year survival rates. Therefore, when using RITA-based composite graft, other options should be considered before proceeding atypical configurations.

  • In recent years, there is growing concern about the potential use of biological agents in war or acts of terrorism accompanied an increased realization that rapid preparedness and response are needed to prevent or treat the human damage that can be caused by these agents. The threat is indeed serious, and the potential for devastating numbers of casualties is high. The use of agents as weapons, even on a small scale, has the potential for huge social and economic disruption and massive diversion of regional and national resources to combat the threat, to treat primary disease, and to clean up environmental contamination. Biological weapons are one of weapons of mass destruction (or mass casualty weapons, to be precise. since they do not damage non-living entities) that are based on bacteria, viruses, rickettsia, fungi or toxins produced by these organisms. Biological weapons are known to be easy and cheap to produce and can be used to selectively target humans, animals, or plants. Theses agents can cause large numbers of casualties with minimal logistical requirements (in wide area). The spread of disease cannot be controlled until there is awareness of the signs of infection followed by identification of agents; and if the organism is easily spread from person to person, as in the case of smallpox, the number of casualties could run into the tens of thousands. Biological weapons could be used covertly, there can be a lot of different deployment scenarios. A lot of different agents could be used in biological weapons. And, there are a lot of different techniques to manufacture biological weapons. Terrorist acts that make use of Biological Agents differ in a number of ways from those involving chemicals. The distinction between terrorist and military use of Biological Weapon is increasingly problematic. The stealthy qualities of biological weapons further complicate the distinction between terrorism and war. In reality, all biological attacks are likely to require an integrated response involving both military and civilian communities. The basic considerations when public health agencies establish national defence plan against bioterrorism must be 1) arraying various laws and regulations to meet the realistic needs, 2)education for public health personnels and support of concerned academic society, 3)information collection and cooperative project with other countries, 4)Detection and surveillance(Early detection is essential for ensuring a prompt response to biological or chemical attack, including the provision of prophylactic medicines, chemical antidotes, or vaccines) and 5) Response(A comprehensive public health response to a biological or chemical terrorist event involves epidemiologic investigation, medical treatment and prophylaxis for affacted persons, and the initiation of disease prevention or environmental decontamination measures). The purpose of this paper is providing basic material of preparedness and response for biological terrorism in modern society.

  • In a Broadcast Encryption System, a sender sends an encrypted message to a large set of receivers at once over an insecure channel and it enables only users in a target set to decrypt the message with their private keys. In 2005, Boneh et al. proposed a fully collusion-resistant public key broadcast encryption in which the ciphertext and the privatekey sizes are constant. In general, pairing-based broadcast encryption system is efficient in bandwidth and storing aspects than non-pairing based broadcast encryption system, however, it requires many computational costs that resource-constrained devices is not suit to be applied. In this paper, we propose a Broadcast Encryption scheme(called BEWD) that user can decrypt a ciphertext more efficiently. The scheme is based on Boneh et al.scheme. More precisely, it reduces receiver's computational costs by delegating pairing computation to a proxy server which computation is required to receiver in Boneh et al.scheme. Furthermore, the scheme enables a user to check if the proxy server compute correctly. We show that our scheme is secure against selective IND-RCCA adversaries under l-BDHE assumption.

A construction method for IP-based Fixed and Personalized A/V Mosaic EPG service (IP 기반 고정형 및 맞춤형 동영상 모자익 EPG 서비스 구축방법)

  • As accelerates the technical evolution of high-speed network and progresses the digitalization of broadcasting network, TV channel service through satellite/cable/terrestrial networks becomes more stable and mature. However, TV channel service using IP network such as IPTV is recently emerging. Especially, when it comes to current mosaic EPG(Electronic Program Guide) as a channel guide, the implementation of EPG via IP network is under developing. Furthermore, the personal target mosaic EPG is not provided at all in the IPTV. This paper proposes a construction method of mosaic system which can support fixed and personalized mosaic EPG using IP network for viewers. The fixed mosaic EPG is made several steps as follows ; First, H/E generates several mosaic A/V streams. Then, which are transmitted to the STB in terms of multicasting via IP network. Finally, mosaic EPG is displayed on TV through STB. In addition, this paper describes a construction model of the personalized A/V mosaic EPG that represents each person's favorite channels according to their tastes and interests. As for the contributions. The TV channel guide using IP network enable viewer to select channel more easily with practical adaptation of multi-channel expansibility and sufficient usability. In addition, through personal mosaic EPG, a number of viewers can compose their own mosaic EPG and enjoy a variety of channel easily in accordance with their preferences. Finally, the personal mosaic EPG can prohibit non-adult users from connecting adult-only contents more efficiently.

  • The purpose of this study was to evaluate the effects of self-efficacy that impacts on personal goal setting.. In addition, in the process of going to achieve the goal, this study was to evaluate the role of self-regulation resources. Specifically, this paper is divided into two groups of self-regulation resources, to analyze the relationship between self-efficacy and goal-setting theory. To achieve this goal, by manipulating the self-adjusting resources to target college students divided into two groups, this measured the performance of the self-efficacy and goal setting. In the test, the a mediated effect of goal setting between self-efficacy and performance was meaningful, and evidence of enhancing the self-regulatory resource by self-efficacy was found. As a result, this paper provides guideline that manages the performance by using the self-efficacy and self-regulatory resource.

Biotope Type Classification based on the Vegetation Community in Built-up Area (시가화지역 식물군집 특성에 기초한 비오톱 유형분류)

  • This study aims to classify the biotope types based on the vegetation community in built-up areas by different land use and to map the plant communities. By classifying biotopes according to a taxonomic system, the characteristics of a biological community can be well-represented. The biotope classification indexes for the target area include human behavioral factors such as land use intensity, land-use patterns and land-cover types. The type classification was divided into four hierarchic ranks starting with Biotope Class, next by Biotope Group and Biotope Type and lastly by Biotope Sub-Type. The Biotope Class was first divided into two areas: the areas improved by humans and the areas unimproved by humans. The improved areas were again divided into permeable and non-permeable regions on the Biotope Group level. In the Biotope Type level, permeable paving areas were divided into areas with wide gap pavers and those with narrow gap pavers. The differential species of each biotope type are Lindera glauca, Conyza canadensis, Mazus pumilus, Vicia tetrasperma, Crepidiastrum sonchifolium, Zoysis japonica, Potentilla supina and Festuca arundinacea. The results of this study suggest that the biotope classification methodology, using a subjective phytosociological approach, is a useful and valuable tool and the results also suggest the possibility of applying more objective and scientific methods in mapping and classifying various environments.

Use of Hydrogen Peroxide with Ozone to Simultaneously Reduce MIB and Quench Ozone Residual in Existing Water Treatment Plants Sourcing Water from the Han River (한강을 원수로 하는 오존/과산화수소 고도정수처리공정에서의 MIB제거 및 잔류오존 농도에 관한 연구)

  • This paper provides a detailed account of pilot testing conducted at South Lake Tahoe (California), the Ddukdo (Seoul) water treatment plant (WTP) and the Bokjung (Seongnam) WTP between February, 2010, and February, 2012. The objectives were first, to characterize the reactions of ozone with hydrogen peroxide (Peroxone) for Han River water following sand filtration, second to determine empirical ozone and hydrogen peroxide doses to remove a taste-and-odor surrogate 2-methylisoborneol (MIB) using an advanced oxidation process (AOP) configuration and third, to determine the optimum dosing configuration to reduce residual ozone to a safe level at the exit of the process. The testing was performed in a real-time plant environment at both low- and high seasonal water temperatures. Experimental results including ozone decomposition rates were dependent on temperature and pH, consistent with data reported by other researchers. MIB in post-sand-filtration water was spiked to 40-50 ng/L, and in all cases, it was reduced to below the specified target level (7 ng/liter) and typically non-detect (ND). It was demonstrated that Peroxone could achieve both MIB removal and low effluent ozone residual at ozone+hydrogen peroxide doses less than those for ozone alone. An empirical predictive model, suitable for use by design engineers and operating personnel and for incorporation in plant control systems was developed. Due to a significant reduction in the ozone reaction/decomposition at low winter temperatures, results demonstrate the hydrogen peroxide can be "pre-conditioned" in order to increase initial reaction rates and achieve lower ozone residuals. Results also indicate the method, location and composition of hydrogen peroxide injection is critical to successful implementation of Peroxone without using excessive chemicals or degrading performance.

Influences on Health Behaviors Execution and Self Rated Health as Socioeconomic Class by the Age Bracket (연령층별 사회경제적 계층에 따른 건강행위 실천과 주관적 건강수준에 미치는 영향)

  • The purpose of present study was to observe the path and influencing effects between socioeconomic class (SEC), health practices and self-rated health(SRH) by the age bracket. The subjects were 4,987 adults over 25 years old who participated in the 2008 Korean National Examination Health and Nutrition Survey and could be classified into SEC in terms of the three characteristics: education, income and occupation. Path analysis was conducted with the effects of health behaviors execution on the differences in SRH, and the complex samples analysis executed by chi-square test, t-test, ANOVA. As the result, lower SRH level paralleled with the lower SEC, and more health behaviors had differed by SEC in the younger and middle aged group. The lower SEC, the lower SRH: non-smoking and weight control for younger women and exercise for aged men had indirect effects as parameters. In conclusion, when planning a health promotion program, to select the correct target populations with consideration of the age bracket, gender and SEC and to establish tailored contents fit for each of the population would be important.

Aging Deterioration for Electric Power Transmission Tower on Offshore Through Periodic Inspections (해상송전철탑 구조물의 주기점검을 통한 경년열화 변화특성)

  • In electric power transmission tower structures on offshore, implementation of life management using the event data of regular safety inspections for structural and material damages is strongly recommended. In this study, six tower structures in Sihwa Lake around Yeoungheung island were target bodies for the safety inspections. safety inspections for deterioration about each of six towers were performed about three items for steel member, five items for concrete foundation, and four items for steel-pipe pile in seawater and seawater itself. Safety inspections for steel members included the visual observations of surface appearances, the measurements of member thicknesses, and the checks of painting states. Also safety inspections for concrete foundations comprised the estimation of crack features, the evaluation of non-destructive compression strengths, and the measurements of neutralization depths and chlorides contents. For steel-pipe piles in seawater the inspections comprised the surveys of corrosion states in accordance with potential levels tests and anode tests, the analyses of photos taken on surfaces of the piles as well as the evaluation of seawater quality. A set of deterioration inspections was performed at the same positions around october of each year for three consecutive years. As a result in this study, Newly developed deterioration indexes have been applied profitably to maintain structural safety for electric power transmission towers by utilizing these event data systematically.
