To understand plant-pathogen interactions, a complete set of hot pepper genes differentially expressed against pathogen attack was isolated. As an initial step, hundreds of differentially expressed cDNAS were isolated from hot pepper leaves showing non-host resistance against bacterial plant pathogens (Xanthomonas campestris pv. glycines and Pseudomonas syringae pv. syringae) using differential display reverse transcription polymerase chain reaction (DDDRT-PCR) technique. Reverse Northern and Northern blot analyses revealed that 50% of those genes were differentially expressed in pepper loaves during non-host resistance response. Among them, independent genes without redundancy were micro-arrayed for further analysis. Random EST sequence database were also generated from various CDNA libraries including pepper tissue specific libraries and leaves showing non-host hypersensitive response against X. campestris pv. glycines. As a primary stage, thousands of cDNA clones were sequenced and EST data were analyzed. These clones are being spotted on glass slide to study the expression profiling. Results of this study may further broaden knowledge on plant-pathogen interactions.
In the speech recognition under the noisy environments, reducing the mismatch introduced between training and testing environments is an important issue. Spectral subtraction is widely used technique because of its simplicity and relatively good performance in noisy environments. In this paper, we introduce histogram method as a reliable noise estimation approach for spectral subtraction. This method has advantages over the conventional noise estimation methods in that it does not need to detect non-speech intervals and it can estimate the noise spectra even in time-varying noise environments. Even though spectral subtraction is performed using a reliable average noise spectrum by the histogram method, considerable amount of residual noise remains due to the variations of instantaneous noise spectrum about mean. To overcome this limitation, we propose a new over-estimation technique based on distribution characteristics of histogram used for noise estimation. Since the proposed technique decides the degree of over-estimation adaptively according to the measured noise distribution, it has advantages to be few the influence of the SNR variation on the noise levels. According to speaker-independent isolated word recognition experiments in car noise environment under various SNR conditions, the proposed histogram-based over-estimation technique outperforms the conventional over-estimation technique.

Background: The cause of maxillary growth restriction in patients with cleft lip and palate remains controversial. While studies have investigated the effects surgical technique and timing have on maxillary growth, few focus on patients with isolated cleft palate (ICP). The purpose of this study was to determine the impact palate repair and its associated complications may have on maxillary growth. Methods: A retrospective chart review of ICP patients who underwent palatoplasty from 1962 to 1999 at Akron Children's Hospital was performed. Patient demographics, Veau type, age at primary repair, closure technique, presence of fistula or velopharyngeal insufficiency (VPI), number of palatal operations, maxillary hypoplasia (MH) frequency, and follow-up were recorded. Exclusion criteria included patients with cleft lip, submucous cleft, or syndromes. Results: Twenty-nine non-syndromic ICP patients were identified; 62% (n = 18) had Veau type 1 and 38% (n = 11) had Veau type 2. All patients underwent 2-flap or Furlow palatoplasty with mobilization of mucoperiosteal flaps. Vomerine flaps were used in all Veau 2 cleft palate closures. Palatoplasty was performed at a mean age of 19.9 ± 8.2 months. Average follow-up was 209 ± 66.5 months. The rate of VPI was 59% (n = 17) and the rate of oronasal fistula was 14% (n = 4). Conclusions: There was a low incidence of MH despite complications after initial palate closure. Our results seem to suggest that age at palate closure, type of cleft palate, and type of surgical technique may not be associated with MH. Additionally, subsequent procedures and complications after primary palatoplasty such as VPI and palatal fistula may not restrict maxillary growth.

This study was conducted to compare the fluoride concentrations in urine of preschool children aged 3${\sim}$6 years between residing in community water fluoridation area(Kwangju City) and non-fluoridation area(Sungnam City). The acid-diffusible fluoride in the urine and drinking water was isolated by the acid-diffusion technique and measured with a fluoride electrode. The mean daily fluoride excretion to urine of children residing in Kwangju and Sungnam were $1.27{\pm}0.75mgF^-$/g creatinine and $0.87{\pm}47 mgF^-$/g creatinine, respectively. It is concluded from this investigation that the $F^-$concentration in urine sample of kindergarten and drinking water of children living in Kwanju(fluoridated areas) were significantly higher than that of children living in Sungnam(non-fluoridated areas).
This study aimed to determine the fluoride intakes in 120 preschool children aged 3 to 6 residing in Jumunjin (community water fluoridation area) and Gangneung (non-fluoridation area). The parents were asked to collect 24-hour urine samples and to duplicate the samples of all the diets that their children ingested in the day of urine collection. The acid-diffusible fluoride in the food and non-carbonate beverages were isolated by the acid-diffusion technique and then measured with a fluoride electrode. The fluoride in carbonate beverages, drinking waters and urine samples were measured directly with a fluoride electrode. The geometric mean (geometric standard deviation) of daily fluoride intakes from all kinds of diet was 5.99 (2.27) $\mu$g/kg/day in the children in Gangneung and that of the children in Jumunjin was 18.36 (2.69). The amount of fluoride intake by food and drinking water in fluoridation area were significantly larger than that in non-fluoridation area but the statistical difference of fluoride intake by beverages between two areas was not observed. The GMs (GSDs) of daily fluoride excretion by urine of children in non-fluoridation area and in fluoridation area were 8.39 (1.73) and 18.62 (1.77) $\mu$g/kg/day, respectively. The correlation between fluoride intake from diet excluding beverage and urinary excretion was statistically significant. It is concluded that the amount of fluoride intake of children living in fluoridation area did not exceed the upper intake level to avoid the risk of dental fluorosis (2.2 mg/day in 4- to 8-year-olds) and urinary excretion of fluoride was good indicator of fluoride intake from diets.
In this paper, a new high step up DC/DC converter with a modified super-lift technique is presented. The coupled inductor technique is combined with the super-lift technique to provide a tenfold or more voltage gain with a proper duty cycle and a low turn ratio. Due to a high conversion ratio, the voltage stress on the semiconductor devices is reduced. As a result, low voltage ultra-fast recovery diodes and low on resistance MOSFET can be used, which improves the reverse recovery problems and conduction losses. This converter employs a passive clamp circuit to recycle the energy stored in the leakage inductance. The proposed convertor features a high conversion ratio with a low turn ratio, low voltage stress, low reverse recovery losses, omission of the inrush currents of the switch capacitor loops, high efficiency, small volume and reduced cost. This converter is suitable for renewable energy applications. The operational principle and a steady-state analysis of the proposed converter are presented in details. A 200W, 30V input, 380V output laboratory prototype circuit is implemented to confirm the theoretical analysis.

A method to quantitatively analyze the defects formed by the hydrogen evolution during electroplating was suggested based on the theoretical approach of the small angle neutron scattering technique. In case of trivalent chrome layers, an isolated defect size due to the hydrogen evolution was about 40 nm. Direct and pulse plating conditions gave the average defect size of about 4.9 and $4.5{\mu}m$ with rod or calabash shape, respectively. Current density change of the pulse plating from $1.5A/dm^2$ to $2.0A/dm^2$ enlarged the average defect size from 3.3 to $7.8{\mu}m$. The defect morphology like rod or calabash was originated by inter-connecting the isolated defects. Small angle neutron scattering was useful to quantitatively evaluate defect morphology of the deposit.
A family of non-isolated DC-DC three-port converters (TPCs) that allows for a more flexible power flow among a renewable energy source, an energy storage device and a current-reversible DC bus is introduced. Most of the reported non-isolated topologies in this area consider only a power consuming load. However, for applications such as hybrid-electric vehicle braking systems and DC microgrids, the load power generating capability should also be considered. The proposed three-port family consists of one unidirectional port and two bi-directional ports. Hence, they are well-suited for photovoltaic (PV)-battery-DC bus systems from the power flow viewpoint. Three-port converters are derived by combining different commonly known power converters in an integrated manner while considering the voltage polarity, voltage levels among the ports and the overall voltage conversion ratio. The derived converter topologies are able to allow for seven different modes of operation among the sources and load. A three-port converter which integrates a boost converter with a buck converter is used as a design example. Extensions of these topologies by combining the soft-switching technique with the proposed design example are also presented. Experiment results are given to verify the proposed three-port converter family and its analysis.
In vertical seismic isolation (VSI), a building is partitioned intentionally by vertical layers into two dynamically different substructures for seismic response reduction. Initially, a 1-story frame was partitioned into two substructures, interconnected by viscous and visco-elastic links, and seismic responses of the original and the vertically isolated structures (VIS) were obtained, considering a large number of stiffness and mass ratios of substructures with respect to the original structure. Color contour graphs were defined for presentation and investigation of large amounts of output results. Dynamic characteristics of the isolated structures were studied by considering the non-classical damping of the system, and then the effects of viscous and visco-elastic link parameters on the modal damping ratios were discussed. On this basis, three states of mass isolation, interactional state, and control mass were differentiated. Response history analyses were performed by Runge-Kutta numerical method. In these analyses, interaction of isolation ratios and link parameters, on response control of VIS was studied and the appropriate ranges for link parameters as well as the optimal ranges for isolation ratios were suggested. Results show that by using the VSI technique, seismic response reduction up to 50% in flexible substructure and even more in stiff substructure is achievable.
Two kinds of mutants were isolated to clone a cluster of genes essential for zooglan biosynthesis. Zoogloea ramigera 115 strains produce capsular polysaccharide. To achieve conjugation in strain 115 and to facilitate recovery of product, a capsule non-forming strain was isolated via successive centrifugation and screening. The other kind of mutants devoid of or producing altered exopolysaccharides were obtained using classical transposon(Tn5) technique and screened for altered colony morphology and celluflour binding properties. Complementation of these mutants was achieved with Z. ramigera 115 slime gene library constructed in a broad host range cosmid vector and helper plasmid by triparental conjugation.
