Comparative Molecular Similarity Index Analysis on 2-(indol-5-yl)thiazolederivatives as Xanthine Oxidase(XO)inhibitors

  • Xanthine Oxidase is an enzyme, which oxidizes hypoxanthine to xanthine, and xanthine to uric acid. It is widely distributed throughout various organsincluding the liver, gut, lung, kidney, heart, brain and plasma. It is involved in gout pathogenesis. In this study, we have performed Comparative Molecular Field Analysis (CoMSIA) on a series of 2-(indol-5-yl) thiazole derivatives as xanthine oxidase (XO) inhibitors to identify the structural variations with their inhibitory activities. Ligand based CoMSIA models were generated based on atom-by-atom matching alignment. In atom-by-atom matching, the bioactive conformation of highly active molecule 11 was generated using systematic search. Compounds were aligned using the bioactive conformation and it is used for model generation. Different CoMSIA models were generated using different alignments and the best model yielded across-validated $q^2$ of 0.698 with five components and non-cross-validated correlation coefficient ($r^2$) of 0.992 with Fisher value as 236.431, and an estimated standard error of 0.068. The predictive ability of the best CoMSIA models was found to be $r{^2}_{pred}$ 0.653. The study revealed the important structural features required for the biological activity of the inhibitors and could provide useful for the designing of novel and potent drugs for the inhibition of Xanthine oxidase.

Changes in the Antioxidant Potential of Persimmon Peel Extracts Prepared by Different Extraction Methods (고종시 감껍질의 추출조건에 따른 항산화 활성)

  • Background: Astringent persimmon (Diospyros kaki Thunb. cv. Kojongsi) peels are by-products of dried persimmons. This study aimed to evaluate the antioxidant activities of Kojongsi persimmon peel (KPP) extracts prepared by 15 different extraction methods: 5 heating durations (0.5 - 2.5 h) at 3 heating temperatures (50, 70, and $90^{\circ}C$). Methods and Results: An increase in heating temperature increased the antioxidant effect of KPP extracts. Those prepared by heating at 1 h had the highest total phenol content, regardless of the heating temperature. In addition, the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity and cell-protective effects against $H_2O_2-induced$ oxidative stress were dependent on the total phenol contents of the extract. However, the KPP-induced increased in catalase expression was dependent on heating temperature and duration. Conclusions: These results suggest that extraction by heating at $90^{\circ}C$ for 1 h may enhance KPP's antioxidant effects, which mainly involve non-enzymatic antioxidant systems.

A CoMFA Study of Quinazoline-based Anticancer Agents

  • Cancer has emerged as one of the leading cause of deaths worldwide. A three-dimensional quantitative structure-activity relationship (3D-QSAR) analysis was performed on a series of quinazoline-based anticancer agents. Purpose of the study is to understand the structural basis for their inhibitory activity. Comparative molecular field analysis (CoMFA) technique was employed to develop 3D-QSAR model. Ligand-based alignment scheme was used to generate a reliable CoMFA model. The model produced statistically significant results with a cross-validated correlation coefficient ($q^2$) of 0.589 and a non-cross-validated correlation coefficient ($r^2$) of 0.928. Model was further validated by bootstrapping and progressive scrambling analysis. This study could assist in the design of novel and more potent anticancer agents.

Purification and biological activity of recombinant human bone morphogenetic protein-2 produced by E. coli expression system (E. coli 발현 시스템에 의해 생산된 recombinant human bone morphogenetic protein-2의 정제와 생물학적 활성)

  • Purpose: Bone morphogenetic protein-2(BMP-2) has been shown to possess significant osteoinducitve potential. There have been attempts to overcome a limitation of mass production, and economical efficiency of BMP. The aim of this study was to produce recombinant human BMP-2(rhBMP-2) from E. coli in a large scale and evaluate its biological activity. Materials and Methods: The E.coli strain BL21(DE3) was used as a host for rhBMP-2 production. Dimerized rhBMP-2 was purified by affinity chromatography using Heparin column. To determine the physicochemical properties of the rhBMP-2 expressed in E. coli, we examined the HPLC profile and performed Western blot analysis. The effect of the purified rhBMP-2 dimer on osteoblast differentiation was examined by alkaline phosphatase (ALP) activity and representing morphological change using C2C12 cell. Results: E. coli was genetically engineered to produce rhBMP-2 in a non-active aggregated form. We have established a method which involves refolding and purifying a folded rhBMP-2 dimer from non-active aggregates. The purified rhBMP-2 homodimer was characterized by SDS-PAGE as molecular weight of about 28kDa and eluted at 34% acetonitrile, 13.27 min(retention time) in the HPLC profile and detected at Western blot. The purified rhBMP-2 dimer stimulated ALP activity and induced the transformation from myogenic differentiation to osteogenic differentiation. Conclusion: rhBMP-2 was produced in E. coli using genetic engineering. The purified rhBMP-2 dimer stimulated ALP activity and induced the osteogenic differentiation of C2C12 cells.

Non-rigid 3D Shape Recovery from Stereo 2D Video Sequence (스테레오 2D 비디오 영상을 이용한 비정형 3D 형상 복원)

  • The natural moving objects are the most non-rigid shapes with randomly time-varying deformation, and its types also very diverse. Methods of non-rigid shape reconstruction have widely applied in field of movie or game industry in recent years. However, a realistic approach requires moving object to stick many beacon sets. To resolve this drawback, non-rigid shape reconstruction researches from input video without beacon sets are investigated in multimedia application fields. In this regard, our paper propose novel CPSRF(Chained Partial Stereo Rigid Factorization) algorithm that can reconstruct a non-rigid 3D shape. Our method is focused on the real-time reconstruction of non-rigid 3D shape and motion from stereo 2D video sequences per frame. And we do not constrain that the deformation of the time-varying non-rigid shape is limited by a Gaussian distribution. The experimental results show that the 3D reconstruction performance of the proposed CPSRF method is superior to that of the previous method which does not consider the random deformation of shape.

Analysis of Sorption and Desorption Behaviors of Radionuclides (Cobalt and Strontium) in Natural Soil (자연 토양에서의 방사성 핵종(Co, Sr)의 흡/탈착 거동 특성 평가)

  • This study was conducted to investigate sorption and desorption behaviors of radionuclides (Cobalt and Strontium) in natural soil. Sorption kinetics and isotherms were analyzed to predict sorption behaviors of radionuclides in natural soil and the experimental data were fitted to several sorption models. Desorption experiments were also performed with or without CMCD at constant pH and ion strength conditions. The results showed that $Sr^{2+}$ was more strongly sorbed than $Co^{2+}$ in natural soil. Both $Co^{2+}$ and $Sr^{2+}$ followed a pseudo-second order kinetics and Sips model. The desorption-resistance of $Co^{2+}$ and $Sr^{2+}$ was estimated using a natural surfactant Carboxymethyl-${\beta}$-cyclodextrin(CMCD) or non-desorbing fraction. Desorption of radionuclides was partially irreversible and $Sr^{2+}$ was more resistant than $Co^{2+}$ Addition of CMCD facilitated desorption of $Co^{2+}$ and $Sr^{2+}$ from soil.

Effects of Protease-resistant Antimicrobial Substances Produced by Lactic Acid Bacteria on Rumen Methanogenesis

  • Effects of protease-resistant antimicrobial substances (PRA) produced by Lactobacillus plantarum and Leuconostoc citreum on rumen methanogenesis were examined using the in vitro continuous methane quantification system. Four different strains of lactic acid bacteria, i) Lactococcus lactis ATCC19435 (Control, non-antibacterial substances), ii) Lactococcus lactis NCIMB702054 (Nisin-Z), iii) Lactobacillus plantarum TUA1490L (PRA-1), and iv) Leuconostoc citreum JCM9698 (PRA-2) were individually cultured in GYEKP medium. An 80 ml aliquot of each supernatant was inoculated into phosphate-buffered rumen fluid. PRA-1 remarkably decreased cumulative methane production, though propionate, butyrate and ammonia N decreased. For PRA-2, there were no effects on $CH_4$ and $CO_2$ production and fermentation characteristics in mixed rumen cultures. The results suggested that PRA-1 reduced the number of methanogens or inhibited utilization of hydrogen in rumen fermentation.

3D QSAR Studies on Cinnamaldehyde Analogues as Farnesyl Protein Transferase Inhibitors

  • Three-dimensional quantitative structure-activity relationship (3D-QSAR) studies on 59 cinnamaldehyde analogues as Farnesyl Protein Transferase (FPTase) inhibitors were investigated using comparative molecular field analysis (CoMFA) with the PLS region-focusing method. Forty-nine training set inhibitors were used for CoMFA with two different grid spacings, $2{\AA}\;and\;1{\AA}$ Ten compounds, which were not used in model generation, were used to validate the CoMFA models. After the PLS analysis, the best predictive CoMFA model showed that the cross-validated value $(r^2_{cv})$ and the non-cross validated conventional value$(r^2_{ncv})$ are 0.557 and 0.950, respectively. From the CoMFA contour maps, the steric and electrostatic properties of cinnamaldehyde analogues can be identified and verified.

Study of High Temperature Corrosion Behavior of Fe-Cr Steel in Sewage Sludge-(SO2-O2-H2O-bal. CO2) mixed Gas Environment (하수슬러지-(SO2-O2-H2O-bal. CO2) 혼합 가스 분위기에서 Fe-Cr 강의 고온부식거동 연구)

  • Two Fe-Cr steels of T22 steel and STS430 steel were corroded at 650 and 750℃ for 100hr in sewage sludge-(0.3% SO2-6% O2-10% H2O-balance CO2) mixed gas environment. T22 steel corroded faster than STS430, indicating that the Cr content significantly influence the corrosion rates. T22 formed thick and non-protective Fe2O3 as the major oxide and Fe3O4 as the minor one. With an increase in corrosion temperature, their corrosion rates increased, being accompanied with formation of pores and cracks in the thickened oxide scales that were non-adherent. STS430 steel formed Fe2O3, Fe3O4 as the outer scale and (Fe, Cr)-O as the inner layer by which its corrosion rate is greatly reduced. Both the T22 and STS430 steel samples formed multi-layered scales by outward diffusion of Fe ions and inward diffusion of oxygen and sulfur ions at high-temperature more than 650℃.

Study on Regrouping of Gray Gases in spectral WSGGM for Arbitrary Mixtures of CO2 and H2O Gases (이산화탄소-수증기 혼합가스에 대한 파장별 회색가스가중합법에서 회색가스재조합에 대한 연구)

  • The WSGG-based narrow band model was employed to solve the radiative transfer equations along isothermal and non-isothermal paths through $CO_2-H_2O-N_2$ gas mixtures at 1 atm. When the WSGGM is applied for arbitrary gas mixtures by considering the multiplication property of transmissivity in overlapping bands, the number of gray gases is significantly increased. To reduce the computation time, three different regrouping methods for the gray gases are tested in obtaining the mean absorption coefficient for each gray gas group. Among them, the regrouping method by minimizing the regrouping error shows the best results. For the isothermal media, 10 gray gases show fairly good agreement with the results by statistical narrow band(SNB) model which are regarded as reference solutions. For non-isothermal media, 20 gray gases show good agreement with reference solutions.
