Influence of Temperature on the Photosynthetic Responses of Benthic Diatoms: Fluorescence Based Estimates (온도가 저서규조류 광합성 반응에 미치는 영향: 형광을 이용한 추정)

  • Benthic diatoms are very important primary producers in understanding estuary ecosystems and their productions are largely varied by their photo-physiological characteristics. The short-term effects of increased temperature on the photosynthetic and photo-physiological characteristics of cultured different species of benthic diatoms (Navicula sp., Nitzschia sp., Cylindrotheca closterium, and Pleurosigma elongatum) were investigated by measuring their PSII-fluorescence kinetics using a Diving-PAM. Photosynthesis versus irradiance curves were measured every two hours at six different temperatures (10, 15, 20, 25, 30, and $35^{\circ}C$) for twenty-four hour. The effective quantum yield of PSII ($\Phi_{PSII}$) for most of the species showed a decreasing trend with increased temperature. The relative maximum electron transport rate (rETRmax) was significantly increased up to the optimum temperature level and then sharply decreased. Relative to the values of other parameters, the maximum light use coefficient ($\alpha$) was not substantially changed at lower temperature levels (<$30^{\circ}C$) but significantly decreased only at higher temperatures (30 and $35^{\circ}C$). The light saturation coefficient ($E_K$) mirrored the rETRmax temperature response. In regards to the temperature acclimation abilities of the four species with time, Navicula sp. and C. closterium acclimated to short-term changes in temperature through their photo-physiological adjustments.

Development and Succession of Marine Fouling Organisms on Artificial Substrata (인조기판 위에서 해양 부착생물의 발달 및 천이)

  • Fouling communities developing in Jinhae Harbor and Masan Bay were studied by slide and panel immersion test during the period from Dec., 1982 to Nov., 1983. The total viable count of bacteria was estimated more than 1.7${\times}$ 10$\^$4/CFU/$\textrm{cm}^2$ after 15 days of immersion and 46 taxa of benthic diatoms were classified in micro-community. Progressional change of fouling communities was clearly shown and dominant diatom species are Licmophora flavellata, Navicula grevillei, and Nitzschia closterium Major macrofouling organisms are Mytilus edulis, Balanus amphitrite amphitrite, Hydroides ezoensis, and Celleporina sp. Wet weight production of macrofouling organisms exceeds 500g/100cm$\^$2/ after 5months of immersion. Regional defferences in community development are clearly shown in two study areas, and mainly due to the disparities of physicochemical stability and nutritional status of ambient water. Seasonality of larvae and the growth rate are the important factors in fouling community development. Overall process of community development is as follow : bacteria and diatoms-multicellular algae-barnacle, mussels and polychaete-sponge, anemone and ascidian.

Relative Immigration Activity of the Hibernal Diatom Community in Eutrophic Waters (저온기 부영양 수계 규조군집의 유입능)

  • To understand the relative immigration activities (RIA) of hibernal diatom community in Lake Ilgam (at the station with and without macrophyte) and River Han (at the station with different current and nutrient), a natural and artificial bed substrate was deposited from 15 to 29 January 2008. For the study period, both waters comprised huge biomass of diatom, over 75% of total abundance. Nutrients in water column, ash-free dry matter (AFDM) and chlorophyll-a (Chl-a) concentration in substrates were decreased with increasing of current in rivers, while in lakes the macrophyte largely affected the diatom community and water quality. Algal RIA in rivers was high at stations with high concentration of nutrient, AFDM and Chl-a, and in high nutrients and no macrophytes in lakes, respectively. The species showing a higher RIA was Nitzschia amphibia in rivers and Aulacoseira italica in lakes, while the dominant species were Navicula sp. in rivers and Syndera acus in lakes, suggesting no relationship between water algal biomass and its RIA. These results suggest that the RIA of stream diatom community depends exclusively on the current, while the planktonic diatom in lake was of the macrophyte.

Seasonal Variation of Phytoplankton and Environmental Factors in Kyungpo Lake (경포호의 식물플랑크톤과 환경요인의 계절적 변동)

  • In order to elucidate chatracteristics of water quality and seasonal dynamics of phytoplankton community in Kyungpo lake, the weekly variation of algal population and 11 environmental factors were investigated from March 1998 to February 1999. A total 121 species of phytoplankton belonged to 57 genera, 4 genera 7 species of the Euglenophyceae, 1 genera 2 species of the Chrysophyceae, 2 genera 3 species of the Cryptophyceae, 1 genera 1 species of the Dinophyceae, 6 genera 9 species of the Cyanophyceae, 28 genera 78 species of the Bacillario-phyceae and 15 genera 21 species of the Chlorophyceae, was identified. Cryptomonas sp., Chamydomonas sp., Nitzschia frustulum, Oscillatoria sp., Lyngbya sp, and Dictyosphaerium pulchellum occurred successively as dominant phytoplankton species from March 1998 to February 1999. Density of phytoplankton cell number showed the peak in early April when Chlamydomoas sp. was dominated. The diversity index of phytoplankton community showed low values in the dominating period of Cryptomonas sp. and Chlamydomonas sp. The concentration of chlorophyll a fluctuated between 1.09 and 107.7$\mu\textrm{g}$ㆍ1$^{-1}$. Water temperature, salinity, DO, conduc tivity and SS were ranged 0.5-27.8$^{\circ}C$, 2.8-25.2 ppt, 4.62-19.21 mgO$_2$ㆍ1$^{-1}$, 5.33-34.76 mS/cm and 0.017-0.184 mgㆍ1$^{-1}$, respectively. Annual means of Secchi disc transparency was 36cm and thai of pH value was 8.5. The concentrations of PO$_4$-P was ranged 0.0012-0.077 mgㆍ1$^{-1}$ , and showed high value from August to October. The concentrations of NO$_2$-N, NO$_3$-N and NH$_4$-N was ranged 0.0011-0.1 mgㆍ1$^{-1}$, 0.074-3.33mgㆍ1$^{-1}$ and 0.0024-0.259 mgㆍ1$^{-1}$, respectively. Among environmental factors, water temperature was negatively correlated with DO, salinity and conductivity, DO was positively correlated with salinity and conductivity. Chlorophyll a, one of the indicies of eutropication in lake, was negatively correlated with NH$_4$-N. The cell density of Cryptomonas sp., which was the most dominant species in this lake, was negatively correlated with transparency. The Characteristics on the Spatial and Temporal Distribution of Phytoplankton in the Western Jinhae Bay, Korea (진해만 서부해역에서 식물플랑크톤의 시.공간적 분포특성)

  • We studied spatial and temporal distributions of the phytoplankton and their relationships to physico-chemical environmental factors in the western Jinhae Bay, Korea from November 2003 to August 2004. In most cases, physico-chemical environmental factors showed homogeneous distribution. The phytoplankton communities were composed of mainly diatoms and dinoflagellates, and their standing crops ranged from $16{\times}10^3\;cells\;l^{-1}\;to\;5,845{\times}10^3\;cells\;l^{-1}$ (with a mean value of $555{\times}10^3\;cells\;l^{-1}$). The bloom of phytoplankton was observed in Gohyun Port in the summer. Seasonal variation of phytoplankton standing crops was higher in winter and summer than in spring and autumn. The dominant species were Skeletonema costatum, Akashiwo sanguinea, Pseudo-nitzschia pungens, Dactyliosolen sp., Leptocylindrus danicus, cryptomonads and etc. Especially, S. costatum was predominant in the summer and A. sanguinea (spring and autumn), Pseudo-nitzschia sp. (summer), Guinardia striata (spring), unidentified flagellates (summer) and cryptomonads (spring) appeared to be an opportunistic species. Concentrations of Chl a ranged from $0.6{\mu}g{\cdot}l^{-1}\;to\;16.7{\mu}g{\cdot}l^{-1}$ (with a mean value of $3.4{\mu}g{\cdot}l^{-1}$). The results of the canonical correspondence analysis implies the study area was grouped into the 2 water masses (inner and outer waters of Gohyun Port) and inner waters had higher abundance and Chl a concentration than outer waters. Also, phytoplankton sanding crops were related with temperature, DO and nutrients ($SiO^2$, TN, TP and etc.) in inner waters. Inner water-mass of Gohyun Port expanded between Gacho Is. and Chilchon Is. during the winter.

Screening of cryoprotectants (CPAs) for cryopreservation in the Nitzschia sp. of marine microalgae (해양 규조류 Nitzschia sp.의 초저온동결보존을 위한 보존제의 영향 분석)

  • Biodiversity has continued to degrade in the $21^{st}$ century due to global warming occasioned by destruction of the environment around the world.. The Nagoya protocol places Korea in a unique position to effectively develop and protect its domestic genetic resources. Microalgae under study in this research contains large amount of antioxidant substances such as beta carotene and astaxanthin, that can be used as biological resource owing to the large amounts of biomass that can be secured through photosynthesis. However, it is difficult to preserve it since cryopreservation method used for long-term preservation is yet to be developed. A basic study for long term cryopreservation was carried out on Nizschia frustulum and Nitzschia amabilis which belong to marine diatoms. As cryoprotectants (CPAs), glycerol, DMSO, and methanol which penetrate into cells were prepared at 5%, 10%, and 15% concentrations each, in case of methanol, it was tested at concentrations of 5%, 10% and 12% by its nature. Two kinds of microalgae, N. frustulum and N. amabilis, were diluted with $10^2$, $10^3$ and $10^4cells\;ml^{-1}$, respectively. The highest survival rate was shown at12% concentration of methanol, and the figures were $6.94{\pm}0.31%$ in N. frustulum and $8.85{\pm}0.16%$ in N. amabilis. As a result of 3 weeks cultivation of thawed microalgae after freezing, the result is shows that N. frustulum increased about 10 times faster and N. amabilis increased about 12 times the original concentration.

A study on water pollution of the physio-chemical conditions and phytoplankton of the Gumho River. (금호강의 이화학적 조건과 식물성 Plankton에 따른 수질오염에 관한 연구)

  • A study on the water pollution of Gumho river by the relationship between physio-chemical conditions and water quality level by phytoplankton was examined at 7 sampling positions during the period from Aug. 1 to Nov. 30, 1981. Examination of physio-chemicat water analysis such as temperature, pH, DO, BOD, and biological analysis are as follows: 1. pH was in the range of 6.6-7.3. 2. At all positions DO was0.5-11.9 mg/l. But at Shinchun bridge and Gangchang was 0.5-3.9 mg/l. 3. BOD was in the range of 3.4-29.2 mg/l. Banyawol, Dongchon and Gumdan was shown good condition. But at Shinchun bridge was 21.1-29.2 mg/l. 4. The plankton identification in this study period showed, Cyanophyceae is 7 genera 13 species, Bacillariophyceae is 11 genera 32 species, and Chlorophceae is 17 genera 27 species: total 35 genera 72 species. 5. In the point of phytoplankton classification, upper stream of Banyawol, Dongchon and Gumdan which BOD was 3.4-8.7 mg/l, dominant phytoplanktons were Synedra ulna, Ulothrix sp., Oscillatoria sp. and Frusturia rhomboides. At Shinchun bridge which BOD was 21.1-29.2 mg/l, Microcystis aeruginosa, Closterium acerosum and Oscillatoria sp were found a small. At 3rd gongdan which BOD was 9.2-12.5 mg/l, dominant species were Synedra ulna, Hormidium sp and Actinastrum hantzschii. At Paldal which BOD was 7.8-9.2 mg/l, dominant species were Nitzschia palea, Synedra ulna and Scenedesmus bijuga. At Gangchang of down stream which BOD was 6.9-9.2 mg/l, dominant phytoplanktons were Closterium acerosum, Microcystis aeruginosa and Actnastrum hantzschii. 6. The results of biological water analysis by saprobic system were as follows: Banyawol was from oligosaprobic to $\beta$-mesosaprobic, Dongchon and Gumdan was from $\beta$-mesosaprobic to $\beta$-mesosaprobic, Shinchun bridge was polysaprobic, 3rd gongdan was from $\alpha$-mesosaprobic to $\beta$-polysaprobic, Paldal was $\beta$-polysaprobic and Gangchang was $\alpha$-polysaprobic.

Seasonal Variations of Microphytobenthos and Growth of Ruditapes philippinarum at Jeongsanpo and Hwangdo Tidal flat, Taean, Korea (한국 태안 정산포와 황도갯벌에서 저서미세조류의 계절적 변동과 바지락의 생장)

  • To examine the relationship between microphytobenthos biomass and the condition index of Ruditapes philippinarum (manila clam), field observations were conducted seasonally at the tidal flats of Jeongsanpo and Hwangdo, Taean, Korea from February to November, 2012. A total of 122 species of microphytobenthos were identified over the study period with 85 species (30-45 species in season) at Jeongsanpo and 92 species (32-57 species) at Hwangdo. Chlorophyll a concentrations and cell number of microphytobenthos were $79.75mg/m^2$ and $3,255cells/cm^2$ at Jeongsanpo, and $151.50mg/m^2$ and $15,943cells/cm^2$ at Hwangdo, respectively. The dominant species were slightly different: Cylindrotheca closterium, Fallacia forcipata, Fogedia sp., Gyrosigma sp., and Navicula sp. at Jeongsanpo and C. closterium, Detonula pumila, Diploneis sp., Navicula sp. and Merismopedia sp. at Hwangdo tidal flat. Paralia sulcata was the representative species based on cell number at the two study sites. The number of microphytobenthos identified from the digestive organs of manila clams seasonally varied from 18 to 31 species at Jeongsanpo and dominant genus were Amphora, Navicula, Nitzschia and Paralia sulcata. At Hwangdo, the species number of microphytobenthos found in the digestive organs of manila clams were in the range of between 19 and 25 species in season and the dominant genus were Actinocyclus, Amphora, Coscinodiscus, Diploneis, Gyrosigma, Navicula, and Diploneis. The condition index of manila clams were greater at Hwangdo (0.57) than at Jeongsanpo (0.42). Present results could support that the condition index of manila clams is positively correlated with the species richness and chlorophyll a contents of microphytobenthos.

The Distribution and Standing Crop of Phytoplankton of Lagoons in the East Coast of Korea (동해안 석호의 식물플랑크톤 분포와 현존량)

  • The distribution and standing crop of phytoplankton were investigated at 12 stations of Songjiho, Ssangho, Maeho and Hyangho as of four lagoons in the east coast from May to November, 2001. It turned out to be total of 164 taxa, and classified as four phylums, four classes, 14 orders, 20 families, 59 genera, 139 species, 22 varieties, two forms and 1 unidentified species by Engler's classification system. Among 104 taxa, 16 taxa including Oscillatoria chlorina were identified as indicators of water pollution and only Cocconeis placentula was the indicator of the clean water. Standing crops of all stations investigated appeared to be relatively high values. Based on the present study upon the distribution and standing crop of phytoplankton, it is regarded as the state of the eutrophication in Songjiho, Ssangho, Maeho and Hyangho as of four lagoons in the east coast.

Seasonal distribution and primary production of microphytobenthos on an intertidal mud flat of the Janghwa in Ganghwa Island, Korea (강화도 장화리 갯벌에서 저서미세조류의 계절적 분포 및 일차 생산력)

  • We studied seasonal distribution of the microphytobenthos and their primary production with $C^{14}$ method and carried out pigment analysis with HPLC in an estuarine mudflat of the Ganghwa Island, Korea from May 2002 to April 2004. The abundances of microphytobenthos were higher at the middle than upper part and lower part of intertidal flat. Abundances of microphytobenthos ranged from $2.3{\times}10^5\;cells\;cm^{-2}$ to $140.9{\times}10^5\;cells cm^{-2}$. The bloom of microphytobenthos was observed in the early spring and then it decreased from spring to summer and autumn. The pennate diatom was a predominated group among the microphytobenthos in this area. The dominant species were Paralia sulcata, Cylindrotheca closterium and Nitzschia sp.. Nitzschia sp. and Cylindrotheca closterium were predominant in February. The results of pigment analysis suggest the presence of diatoms, euglenophytes, chlorophytes, cyanobacteria, cryptophytes, chrysophytes, prymnesiophytes, dinoflagellates and prasinophytes. The biomass of microphytobenthos ranged from 1.18 to 34.25 mg chl-a $m^{-2}$, with a mean of 7.60 mg chl-a $m^{-2}$. The mean ratio of Fuco/Chl a was 0.7 which indicates that most of biomasses of microphytobenthos were due to diatoms. The ratios of Chl b/Chl a ranged from 0 to 0.82(with a mean of 0.17), implying that euglenophytes and chlorophytes lived together in special period seasonally. Temporal variation of primary production ranged from 4.2 to 113.0 $mgC{\cdot}m^{-2}{\cdot}hr^{-1}$(mean value was 33.9 $mgC{\cdot}m^{-2}{\cdot}hr^{-1}$ and initial slope$({\alpha})$ was measured from 0.002-0.005$(mgC\;mgchl-a^{-1}\;hr^{-1}){\cdot}({\mu}E\;m^{-2}\;s^{-1})^{-1}$. Assimilation number$(P_m)$ was in the range of 0.50-1.32 $mgC{\cdot}mgChl-a{\cdot}hr^{-1}$ and daily primary production ranged from 20.9 to 678.1 $mgC{\cdot}m^{-2}{\cdot}d^{-1}$(mean value was 206.72 $mgC{\cdot}m^{-2}{\cdot}^{-1}$).
