Electro-catalytic Performance of PtRu Catalysts Supported on Urea-treated MWNTs for Methanol Oxidation

  • In this work, nitrogen and oxygen functionalities was introduced to the graphite nanofibers (GNFs) and their effect on electrocatalytic performance of the GNF supports for direct methanol fuel cells (DMFCs) was invesigated. The nitrogen and oxygen groups were introduced through the urea treatments and acid treatment, respectively. And, PtRu catalysts deposited on modified GNFs were prepared by a chemical reduction method. The catalysts were characterized by means of elemental analysis, nitrogen adsorption, and X-ray photoelectron spetroscopy (XPS). The structure and morphological characteristics of the catalysts were characterized by X-ray diffraction (XRD) and transmission electron microscopy (TEM). As a result, the Pt-Ru nanoparticles were impregnated on GNFs with good formation in 3-5 nm. And, the cyclic voltammograms for methanol oxidation revealed that the methanol oxidation peak varied depending on changes of surface functional groups. It was thus considered that the PtRu deposition was related to the reduction of PtRu and surface characteristics of the carbon supports. The changes of surface functional groups were related to PtRu reduction, significantly affect the methanol oxidation activity of anode electrocatalysts in DMFCs.

고농도 질산 및 아질산성 질소 함유 폐수의 생물학적 탈질에 관한 연구 (Study for Biological Denitrification of High-Strength Nitrate and Nitrite Industrial Wastewater)

  • An economic treatment method to remove oxidized nitrogen from wastewater is biological denitrification with organic matters. Several organics can be used, however, methanol is commonly used. When methanol is provided, M:N (Methanol to Nitrogen) ratio is used to define methanol demand for denitrification. In this study, two artificial wastewaters were provided to a biological system to evaluate denitrification performance. Differences of influent total CODcr from effluent soluble CODcr were converted to methanol equivalent and oxidized nitrogen difference between influent and effluent were converted to nitrate equivalent to define M:N ratios. Modes I, II, III, I-1 and IV showed 5.1, 2.7, 3.3, 2.3 and 2.6 of M:N ratios, respectively. Since denitrifying microorganisms had to build a new metabolic system for methanol and influent organics, initial operation mode, Mode I, required more methanol and this resulted in high M:N ratios compared with later operation mode, Mode I-1. Salt in influent did not show inhibitory effects on denitrfication, although this was believed to increase effluent SS and soluble CODcr concentrations in Mode III, I-1 and IV, respectively. The concentrations of effluent soluble $COD_{Mn}$ did not changed much with influent salt.

Quantitative Analysis of Tiropramide in Human Blood by Gas Chromatography with Nitrogen-Phosphorus Detector

  • The analytical method of antispasmodic agent tiropramide {$(\pm)\alpha$-(benzoylamino)-4-[2-(diethylamino)ethoxy]-N,N-dipropylbenzenepropanamide hydrochloride} was developed by gas chromatography/nitrogen-phosphorus detector (GC/NPD) in human plasma. Two kinds of tiropramide tablets were orally administered to volunteers by Latin square crossover design, and blood was withdrawn as designed schedule. The plasma of 1 mL was loaded on Sep-pak $C_{18}$ cartridge and eluted with methanol after washing with 30% methanol. The residue dissolved in 100 $\mu$L of methanol after evaporation was analyzed by GC/NPD. Precision (CV%) of intra-day was located within 2.6% and accuracy was less than 9.7%. Inter-day precision was below 8.7% and accuracy was relatively good as less than 14%. Plasma samples obtained from human volunteers were analyzed for the determination of tiropramide concentration by using this method. The method was sensitive, rapid and suitable enough to be applied for pharmacokinetic and bioequivalence studies of tiropramide in human volunteers.

  • A gas chromatographic method was established for the simultaneous determination of urinary nicotine and cotinine. The analytes in basified urine containing a sufficient amount of Na$_2$S0$_4$were extracted into dichloromethane by vigorous shaking. Into the transferred organic phase was added a small amount of acidified methanol (0.5 N HCI in methanol), followed by concentrating the mixture to dryness using a mild stream of nitrogen gas. The concentrate was reconstituted with methanol and the final solution analyzed using the gas chromatograph equipped with the nitrogen-phosphorus detector. The reproducibility tests showed coefficients of variation less than 11% for both compounds. The percent recovery for both analytes ranged from 88 to 103%. The estimated method detection limits for nicotine and cotinine were 0.60 and 5.1 ng/mL, respectively. Extraction efficiencies for both nicotine and cotinine apparently declined without the addition of Na$_2$S0$_4$into the urine. Moreover, the absence of methanolic HCI in the extract resulted in almost complete evaporation of nicotine and partial loss of cotinine during the concentration process, indicating that the formation of nicotine-HCI and cotinine-HCI species is prerequisite to the suppression of the loss of both compounds.

Methylotrophic Yeast를 이용한 외래단백질 발현에서의 발효 변수 최적화 (The Optimization of Fermentation Parameters for Heterologous Protein Productivity Enhancement with Pichia pastoris)

  • The methylotrophic yeast, Pichia pastoris, is known to be a potential host to offer many advantages for production of recombinant proteins. Fermentation parameters were optimized to enhance the heterologous ${\beta}$-galactosidase productivity with P. pastoris. Optimum concentration of methanol, used as inducer, was observed to be 8 g/L and the extent of repression of AOX1 promoter by glycerol was lower than by glucose. The degradation of the gene product ${\beta}$-galactosidase by protease was inhibited as the pH increased from 5 to 8 and the yeast extract(1%) as nitrogen source increased expression level 4 times higher compared to yeast nitrogen base(1%) as nitrogen source increased expression level 4 times higher compared to yeast nitrogen base(1%). Induction method, in which methanol is just added to fermentation medium without centrifugation, was found to be as much effective as the one with centrifugation.

  • Decarburized zone of metal tongue which is used in seat belt for automobiles was carbon-restoration quenched and tempered using nitrogen-methanol gaseous atmosphere. The effects of microstructure and mechanical properties of metal tongue on the effectiveness of carbon-restoration during tempering was studied. Metal tongue showed $20{\sim}30{\mu}m$ decarburized zone. However, after carbon-restoration, it has uniform microstructure and thus hardness without decarburized zone. Carbon-restoration quenching and tempering process resulted in better wear and corrosion resistances than quenching and tempering process.

  • As host for the production of eucaryotic heterologous proteins, methylotrophic yeast Pichia pastoris and Hansenula polymorpha are the most highly developed of a small group of alternative yeast species chosen for their perceived advantages. This paper describes the method to enhance the recombinant protein productivity with P. pastoris and H. Plymorpha. In these experiments, the effects of methanol induction timing, induction method, pH, culture temperature and kinds of nitrogen sources on foreign protein production were tested with P. pastoris and compared with H. polymorpha.. In addition, optimum methanol concentration as inducer and the effects of carbon sources on AOX1 or MOX promoter repression and secretion efficiency were also studied in both cases.

Assay of Midazolam in Human Plasma by Gas-Liquid Chromatography with Nitrogen-Phosphorus Detection

  • A sensitive and specific method is described for the determination of midazolam in human plasma. The drug was extracted from 1 mL of carbonate buffered plasma (pH 9.6) with 8 mL of diethyl ether. Famprofazone was used as internal standard. The organic phase was evaporated to dryness. The residue was dissolved in methanol for the direct analysis by gas chromatograph-nitrogen phosphorus detector system. In the concentration range of 1-5000 ng/mL, the calibration curve was linear. The coefficients of variation from the precision test were <6% at the range of the concentration of 0.10-2.00 μg/mL and the detection limit for midazolam in 1 mL of plasma was o.5 ng. This assay is more sensitive, selective, simple and rapid than earlier methods. Plasma midazolam concentrations were determined by this method after administration of midazolam.

  • To prevent surface and underground water pollution, wastewater treatment is essential. Four bench-scale activated sludge units (10 L operational volumes) were operated at 5, 10 and $20^{\circ}C$ for evaluation of treatment efficiencies with typical wastewater from swine housing. The units were set for a 24-hour cycle. As compared to the conventional process, high removal efficiencies for organic substances, nitrogen and phosphorus in swine wastewater were obtained simultaneously with an intermittent aeration process (lAP). The NOx-N produced during an aeration period was immediately reduced to nitrogen gas (e.g. $N_2$ or $N_2O$) in the subsequent non-aeration periods, and nitrification in aeration periods occurred smoothly. Under these conditions, phosphorus removal occurred with the release of phosphorus during the non-aeration periods followed by the excess uptake of phosphorus in the activated sludge during aeration periods. It was confirmed that the lAP had a better ability to remove pollutants under both low temperatures and high nitrogen loading conditions than the ordinary method did. In addition to that, the total emission of $N_2O$ from lAP was reduced to approximately 1/50 of the conventional process for the same loading. By adopting an adequate aeration programme for individual swine wastewater treatment, this system will provide a promising means for nitrogen and phosphorus control without pH control or addition of methanol.

보관 방법에 따른 Rotifer Brachionus plicatilis 내구란의 부화 (The Hatching Rate of Resting Eggs of the Rotifer Brachionus plicatilis according to Preservation Method)

  • The rotifer Brachionus plicatilis is one of the most important food organisms in aquaculture. The resting eggs produced by mictic female rotifers are easily stored and hatched, making them useful as the starter for the mass culture of rotifers in marine larval culture. This study examined the optimum preservation method for resting eggs to ensure a high hatching rate. To produce resting eggs, the marine rotifer B. plicatilis was cultured with Nannochloris oculata (KMMCC 16). The resting eggs were harvested and cryopreserved using 5% and 10% methanol (MeOH), dimethylsulfoxide (DMSO), and glycerol as cryoprotectant agents (CPAs). The cryopreservation comprised slow or rapid freezing and the resting eggs were stored for one month in liquid nitrogen ($-196^{\circ}C$). The resting eggs were also dried at different temperatures (30, 40, and $50^{\circ}C$) and for different times (1, 2, and 3 h). In general, the hatching rates of the resting eggs preserved with CPA were higher than those without CPA and the slow freezing method was better than the rapid freezing method. However, the optimum CPA concentration for the hatching rate of the resting eggs varied with the freezing method and kind of CPA, and the CPA also affected the viability of the resting eggs. Dried resting eggs had a high, rapid hatching rate over 80%. The moisture content of the resting eggs cryopreserved in liquid nitrogen affected the hatching rate. Drying at $30^{\circ}C$ for 1 hour resulted in a high hatching rate of the resting eggs. In conclusion, drying at $30^{\circ}C$ for 1 hour and preservation in liquid nitrogen with the slow freezing method, without CPA, is recommended for a high hatching rate (ca. 95%) of rotifer resting eggs.
