Effects of nitrogen and organic carbon sources on growth and lipid production of Chlorella sp. KR-1 in flask cultures (플라스크 배양에서 Chlorella sp. KR-1의 균체 성장 및 지질 생산에 대한 질소원 및 유기탄소원의 영향)

  • Recently microalgae have been proposed as a promising biodiesel feedstock, owing to their higher lipid productivity and non-arable land based cultivation system. Biomass and lipid productivities of microalgae are largely affected by various environmental and nutritional factors. In this study, the effects of nitrogen (nitrate and ammonium) and organic carbon (glucose and glycerol) sources on the cell growth and lipid production of Chlorella sp. KR-1 were examined in flask cultures. Under autotrophic culture conditions for 15 days, overall cell growth and lipid (fatty acid methyl ester, FAME) production with nitrate were better than those of ammonium, resulting in 1.06 g cell/L and 333 mg FAME/L, respectively. Maximal intracellular lipid contents (348 - 352 mg FAME/g cell) were observed at low concentrations of 1 mM for both nitrate and ammonium. In the supply of light, addition of glucose in the range of 1 - 20 g/L showed higher cell densities than the autotrophic cell growth condition. Higher lipid accumulation of 375 mg FAME/g cell could achieved at 5 g glucose/L albeit of relatively short incubation of 7 days. With glycerol, intracellular lipid contents were ~1.9 times lower than glucose cases although similar cell growths were observed for both carbon sources.

  • Pressurization system in a liquid-propellant launcher supplies the controlled gas into the ullage volume of propellant tanks to feed propellants to combustion chamber by pressurizing propellants stored in propellant tanks. The ullage part of propellant tank should be constantly pressurized to supply the propellants stored in propellant tanks to turbo-pump or combustion chamber by pressurant pressurization system. Pressurant used to pressurize propellants is generally stored in a series of tanks at cryogenic temperature and high preassure inside an oxidizer tank. The reason is to store the quantity of pressurant as much as possible and to make pressurant tanks as small as (i.e. as light as) possible. However for test convenience pressurant tank is located at STP (standard temperature and pressure) environment in this study. Orifices are widely adapted to several pressurization systems in liquid rocket propulsion systems. Discharge coefficients of orifices are essentially needed for the optimized design of pressurization system in liquid rocket propulsion system. For this study gaseous nitrogen was served as pressurant and rounded entry orifices were employed. The forty-two (42) rounded entry orifices (the radii of curvatures are 0.5 and 1.0) have been tested experimentally in the supersonic flow region. The discharge coefficients of rounded entry orifices with inside diameters ranging from about 1.4 to 5.0mm was measured with 0.95 ${\sim}$ 0.99.

  • The process of separating oxygen and nitrogen from the air is mainly performed by electric liquefaction, which consumes a lot of electricity, resulting in higher operating costs. On the other hand, when used for cold energy of LNG, electric power can be reduced compared to the electric Linde cycle. Currently, LNG cold energy is used in the cold refrigeration warehouse, separation of air-liquefaction, and LNG cold energy generation in Japan. In this study, the system using LNG cold energy and the Linde cycle process system were simulated by PRO/II simulators, respectively, to cool the elevated air temperature from the compressor to about $-183^{\circ}C$ in the air liquefaction separation process. The required amount of electricity was compared with the latent heat utilization fraction of LNG, the LNG supply pressure, and the LNG cold energy usage. At the air flow rate of $17,600m^3/h$, the power source unit of the Linde cycle system was $0.77kWh/m^3$, compared with $0.3kWh/m^3$.

Effects of Green Manure Crops, Hairy vetch and Rye, on N Supply, Redpepper Growth and Yields (질소공급, 고추의 생육 및 수량에 대한 녹비작물 환원 효과)

  • Winter annual green manure crops may be an effective tool for environmental-friendly agriculture system. The effect of legume (hairy vetch), non-legume (rye) and N fertilization ($190kg\;N\;ha^{-1}$) was examined and compared on red-pepper yield, nitrogen uptake, carbohydrate composition, and soil N and C contents. We monitored soil N and C for 120 days after incorporation (DAI) of green manures or mineral fertilizer. The mineralization of nitrogen reached the maximum around 30 DAI. The amount of inorganic nitrogen supplied by mineralization of hairy vetch residue was greater with than chemical N or rye. Photosynthetic rate was similar by 70 DAT in all treatments however, it in rye-incorporated red-pepper presented a sharp decline at later growth period. Leaf total nitrogen was greater with hairy vetch and chemical N than rye throughout the experiment. The soluble sugar increased steadily in all treatments from 40 to 110 days after transplanting (DAT) whereas starch showed a tendency of great decrease. Hairy vetch greatly promoted red-pepper growth by the later period however, chemical N showed the highest fruit yields.

Hybrid MBE Growth of Crack-Free GaN Layers on Si (110) Substrates

  • Two main MBE growth techniques have been used: plasma-assisted MBE (PA-MBE), which utilizes a rf plasma to supply active nitrogen, and ammonia MBE, in which nitrogen is supplied by pyrolysis of NH3 on the sample surface during growth. PA-MBE is typically performed under metal-rich growth conditions, which results in the formation of gallium droplets on the sample surface and a narrow range of conditions for optimal growth. In contrast, high-quality GaN films can be grown by ammonia MBE under an excess nitrogen flux, which in principle should result in improved device uniformity due to the elimination of droplets and wider range of stable growth conditions. A drawback of ammonia MBE, on the other hand, is a serious memory effect of NH3 condensed on the cryo-panels and the vicinity of heaters, which ruins the control of critical growth stages, i.e. the native oxide desorption and the surface reconstruction, and the accurate control of V/III ratio, especially in the initial stage of seed layer growth. In this paper, we demonstrate that the reliable and reproducible growth of GaN on Si (110) substrates is successfully achieved by combining two MBE growth technologies using rf plasma and ammonia and setting a proper growth protocol. Samples were grown in a MBE system equipped with both a nitrogen rf plasma source (SVT) and an ammonia source. The ammonia gas purity was >99.9999% and further purified by using a getter filter. The custom-made injector designed to focus the ammonia flux onto the substrate was used for the gas delivery, while aluminum and gallium were provided via conventional effusion cells. The growth sequence to minimize the residual ammonia and subsequent memory effects is the following: (1) Native oxides are desorbed at $750^{\circ}C$ (Fig. (a) for [$1^-10$] and [001] azimuth) (2) 40 nm thick AlN is first grown using nitrogen rf plasma source at $900^{\circ}C$ nder the optimized condition to maintain the layer by layer growth of AlN buffer layer and slightly Al-rich condition. (Fig. (b)) (3) After switching to ammonia source, GaN growth is initiated with different V/III ratio and temperature conditions. A streaky RHEED pattern with an appearance of a weak ($2{\times}2$) reconstruction characteristic of Ga-polarity is observed all along the growth of subsequent GaN layer under optimized conditions. (Fig. (c)) The structural properties as well as dislocation densities as a function of growth conditions have been investigated using symmetrical and asymmetrical x-ray rocking curves. The electrical characteristics as a function of buffer and GaN layer growth conditions as well as the growth sequence will be also discussed. Hydrogeochemistry and Statistical Analysis of Water Quality for Small Potable Water Supply System in Nonsan Area (논산지역 마을상수도 수질의 수리지화학 및 통계 분석)

  • This study was carried out to provide proper management plans for small portable water supply system in the Nonsan area through water quality monitoring, hydrogeochemical investigation and multivariate statistical analyses. Nonsan area is a typical rural area heavily depending on small water supply system for portable usage. Geology of the area is composed of granite dominantly along with metasedimentary rocks, gneiss and volcanic rocks. The monitoring results of small portable water supply system showed that 13-21% of groundwaters have exceeded the groundwater standard for drinking water, which is 5 to 8 times higher than the results from the whole country survey (2.5% in average). The major components exceeding the standard limits are nitrate-nitrogen, turbidity, total coliform, bacteria, fluoride and arsenic. High nitrate contamination observed at southern and northern parts of the study area seems to be caused by cultivation practices such as greenhouses. Although Ca and $HCO_3$ are dominant species in groundwater, concentrations of Na, Cl and $NO_3$ have increased at the granitic area indicating anthropogenic contamination. The groundwaters are divided into 2 groups, granite and metasedimentary rock/gneiss areas, with the second principal component presenting anthropogenic pollution by cultivation and residence from the principal components analysis. The discriminant analysis, with an error of 5.56% between initial classification and prediction on geology, can explain more clearly the geochemical characteristics of groundwaters by geology than the principal components analysis. Based on the obtained results, it is considered that the multivariate statistical analysis can be used as an effective method to analyze the integrated hydrogeochemical characteristics and to clearly discriminate variations of the groundwater quality. The research results of small potable water supply system in the study area showed that the groundwater chemistry is determined by the mixed influence of land use, soil properties, and topography which are controlled by geology. To properly control and manage small water supply systems for central and local governments, it is recommended to construct a total database system for groundwater environment including geology, land use, and topography.

Interrelation between N and S Nutrition on Accumulation of Storage Protein in Soybean Seed

  • Soybean is an important crop because its seed has very high protein relative to others. The quality of soy protein is limited by the concentration of the sulfur-containing amino acids in the amino acid profile. Among the supply of various forms of 0.4mM sulfur as S nutrition during seed fill. only 0.4mM L-methionine can inhibit ${\beta}$-subunit synthesis completely and produce the highest glycinin-containing seeds. Compared to 0.4mM sulfate control, seeds supplied by 0.4mM L-methionine have lower ${\alpha}$-, no ${\beta}$-subunit, and highly increased glycinin without altering total protein concentration. Supply of 0.2mM cystine (0.4mM S) did not affect the accumulative pattern of seed storage protein (SSP) subunits. In the supply of L-methionine, 0.2mM treatment showed higher glycinin in seeds but 0.05mM resulted in lower glycinin than tile sulfate control. The relative abundance of ${\alpha}^`$-subunit was not altered by any N or S nutrition. Under 5mM nitrogen, protein concentration was increased about 3-5% by substituting ammonia for nitrate during seed fill independent of nutrition. The increase resulted in the only increase of 7S protein, mainly ${\beta}$-subunit. Our data suggest that the regulatory system of SSP genes responds to the balance between N and S assimilates supplied from mother plant. and controls the di fferential synthesis of their subunits for the maximum protein accumulation in developing soybean seed.

Changes of Soil Nitrogen Supply and Production of Upland Forage Crops by Cattle Manure during Conversion from Paddy to Upland Condition in Paddy Field (논의 밭전환 연차간 우분시용에 의한 질소공급 및 밭사료 작물의 생산력 변화)

  • The effect of cattle manure with the rates of 2 and 4 ton $l0a^{-1}$ for winter rye and summer corn cultivation, respectively, on the dry matter (DM) yield and nitrogen (N) uptake were investigated during successive three­year conversion period from paddy to upland condition in paddy field. The changes in soil properties and soil N sup­plying capacity during repetitive manure application were a1so examined. Growth and DM yield of upland forage crops, especially. winter rye were hindered highly by poor soil condition in the first year after conversion from paddy to upland condition, so apparent recovery of cattle manure N by crops was very low in the first conversion year. But, DM yield and N uptake of upland forage crops were increased linearly by accumulative input of cattle manure along with mineral N enrichment in soil, which also increased apparent recovery of cattle manure-No It seemed that those increases were mainly due to the improvement of soil properties such as soil mineral N, soil organic matter (soil carbon), potentially mineralizable N and bulk density by accumulative input of cattle manure rather than the increase of soil N supply according to accumulative conversion period from paddy to upland condition. It was derived that conversion period from paddy to upland condition over 2 years is needed to obtain proper DM yield in paddy field and accumulative inputs of cattle manure during the conversion period is more influential to the continuous increment of DM yield and N uptake of upland crop as well as of potential N supplying capacity of soil.

Squeeze Film Dampers for High Temperature Superconducting Radial Magnetic Bearings

  • Squeeze film dampers (SFDs) are designed and analyzed for radial superconducting bearings. The designed SFDs are mounted on the superconductors submerged in liquid nitrogen such that the dampers should supply additional damping to the relatively underdamped superconducting bearing support . Basic theory of SFD assembled with superconductors is introduced. Rotordynamic simulations are provided to support the feasibility of the superconducting bearings mounted on SFDs for a horizontal flywheel energy storage system.
