5톤/일 규모 SRF 전용 연소보일러에서의 질소산화물의 생성과 저감에 대한 연구 (A Study on the Formation and Reduction of NOx in 5TPD SRF Boiler)

  • The emission of nitrogen oxides has a great environmental impact. It leads to Los Angeles type smog, and it recently has attracted attention as a source of ultrafine dust. The main sources of nitrogen oxides are internal combustion engines and industrial boilers. These emission sources are processes that are essential for human industrial activities, so the regulation of original use is impossible. Therefore, special control methods should be applied to reduce NOx emissions into the atmosphere. In this study, we investigated how the supply of ER and urea influences the removal of nitrogen oxides from SRF combustion boilers. Experimental results show that the removal efficiency of nitrogen oxides can be up to 80% under the conditions of ER 2.0 and a urea feed of 0.5 LPM.

충주댐 유역의 오염원에 따른 오염부하량 발생 특성 (Characteristics of Pollutant Loads according to Types of Sources for the Chungju Dam Watershed)

  • Soil and Water Assessment Tool (SWAT) model was selected as a tool for assessing the effect of pollutant sources on the total loads from the Chungju Dam upstream watershed. The model was constructed through calibration of parameters related to nitrogen (N) and phosphorus (P), which was based on the runoff and sediment modeling performed in the previous research. Using this, the spatial and temporal pollutant loadings by source type were investigated. Results of this study indicated that in most forested upstream sub-watersheds, pollutant loadings from point sources were very low, and total loadings by point and non-point sources were also insignificant. On the other hand, in #14 sub-watershed including Jecheon city, the loadings by point source were relatively considerable. For the whole watershed, non-point sources accounted for 99% of sediment, 97% of N, and 93% of P loads. And monthly non-point source loadings were concentrated on rainy summer season, while point source loadings of N and P kept nearly constant throughout the year and were high on dry winter season relative to non-point source.

Enhanced Biodegradation of 2,4,6-Trinitrotoluene (TNT) with Various Supplemental Energy Sources

  • The biodegradation of 2,4,6-trinitrotoluene (TNT) was performed on a laboratory scale using P. putida originally isolated from explosive-contaminated soil. One hundred mg/1 of TNT was completely degraded within 20 h under optimum conditions. Various supplemental energy sources (carbon sources, nitrogen sources, and surfactant) were tested, with the main objective of identifying an inexpensive source and enhancing the degradation rate for large-scale biodegradation. Based on the degradation rate, molasses was selected as a possible supplemental carbon source, along with NH$_4$Cl and Tween 80 as a nitrogen source and surfactant, respectively. The degradation rate increased about 3.3 fo1d when supplemental energy sources were added and the degradation rate constant increased from 0.068 h$\^$-1/ to 0.224 h$\^$-1/. These results appear to be promising in application of the process to TNT-contaminated soil applications.

Effect of C/N Ratio on the Production of Poly(3-hydroxyalkanoates) by the Methylotroph Paracoccus denitrificans

  • Two series of carbon sources, linear primary $C_1$~$C_9$ alcohols and linear $C_2$~$C_{10}$ monocarboxylic acids were tested for PHA synthesis in Paracoccus denitrificans. The results showed that the growth-associated synthesis of PHA could be referred only to the carbon sources with odd number of carbon except methanol. For all carbon sources with even number of carbon, nitrogen limitation was required to induce PHA synthesis in P. denitrificans. Poly(3-hydroxyvalerate)[P(3HV)] homopolymer was synthesized from $C_5$, $C_7$, and $C_9$ while growing in the presence of nitrogen, but the nitrogen depletion in the later growth period incorporated 3-hydroxybutyrate(3HB) unit into the polymer chain. The optimum C/N ratio for P(3HV) homopolymer production was found to be 10 when the strain was grown on 10 ml/l of valeric acid for 96 h. P. denitrificans synthesized P(3HB-co-3HV) copolymers from n-hexanoic and n-octanoic acid. The microstructural characterics of the P(3HB-co-3HV) copolymer from n-propanol was investigated using $^13C$-nuclear magnetic resonance spectroscopy, showing a structural heterogeneity.

Sucrose를 탄소원으로 재조합 대장균에 의한 Poly(3-Hydroxybutyric Acid) 합성시 복합질소원이 미치는 영향 (Effect of Complex Nitrogen Sources on Poly(3-Hydroxybutyric Acid) Synthesis by Recombinant Escherichia coli Using Sucrose as a Carbon Source)

  • Sucrose를 탄소원으로 이용할 수 있는 대장균 W 를 A. eutrophus PHA 합성 유전자가 cloning된 높 은 복제수의 plasmid pSYL105로 형질전환하여 단 순배지에서 배양시 $0.21g/\ell$ 의 낮은 PHB 농도를 얻었다. 이에 107}지의 복합질소원을 소량 첨가하여 PHB 합성에 미치는 영향을 살펴 보았다. Tryptone, casamino acids, casein hydrolysate, 혹은 soy bean hydrolysate의 첨가시 PHB 합성 이 10배 이 상 증대되었다. 또한 이때 sucrose로부터의 PHB 수율도 10배 이상 증가하여 이 네 가지 복합질소원 을 척정 농도로 첨가함으로써 sucrose로부터 효과적인 PHB 생산이 가능함을 알았다.

  • Among many microalgae cultivation types, heterotrophic culture with low cost carbon sources and energy saving culture method is crucial. A result of estimating the effects of pH on wastewater treatment using heterotrophic growing microalgae Chlorella sorokiniana shows that there was no difference in microalgae growth amount and nitrogen, phosphorus removal rate by wide range of pH(5 ~ 9). From pH 5 to 9, total nitrogen, phosphorous and glucose removal rates were 10.5 mg-N/L/d, 2 mg-P/L/d, 800 ~ 1000 mg/L respectively. This study reveals that C. sorokiniana cannot metabolite glycerol heterotrophically, however, glucose and acetate were proper carbon sources for growth and T-N, T-P and TOC removal. This research highlights the potential of heterotrophic microalgal growth with wastewater treatment plant with wide range of pH and carbon sources.

Growth and Cultural Characteristics of Ophiocordyceps longissima Collected in Korea

  • We investigated the effect of nutritional and environmental factors on Ophiocordyceps longissima mycelial growth. The longest colony diameter was observed on Schizophyllum (mushroom) genetics complete medium plus yeast extract, Schizophyllum (mushroom) genetics minimal medium, and Sabouraud dextrose agar (SDA); however, malt-extract yeast-extract agar, SDA plus yeast extract, yeast-extract malt-extract peptone dextrose agar, SDA, oatmeal agar, and potato dextrose agar showed higher mycelia density. A temperature of $25^{\circ}C$ was optimum and 7.0 was the optimum pH for mycelial growth. Colony diameter was similar under light and dark conditions. Maltose and yeast extract showed the highest mycelial growth among carbon and nitrogen sources respectively. The effect of mineral salts was less obvious; however, $K_3PO_4$ showed slightly better growth than that of the other mineral salts tested. Among all nutrition sources tested, complex organic nitrogen sources such as yeast extract, peptone, and tryptone were best for mycelial growth of O. longissima. Ophiocordyceps longissima composite medium, formulated by adding maltose (2% w/v), yeast extract (1% w/v), and $K_3PO_4$ (0.05% w/v) resulted in slightly longer colony diameter. In vitro mycelial O. longissima growth was sustainable and the production of fruiting bodies could be used for commercial purposes in the future.

Rhizopus의 아밀라제에 관한 연구 3 (Studies on the Amylase of Rhizopus(III))

  • In order to clarify the best cultural conditions of Rhizopus niveus the effects of aeration, pH and various nutrients, such as different carbon and nitrogen sources, vitamins, and growth substances, on the mycelial growth were studied through liquid culture, and amylase activities of the fungus at different cultural periods were measured. Soluble starch, xylose and galactose are excellent sources of carbon for growth of the fungus. Sorbose and lactose are utilized slightly for growth. peptone, ammonium sulfate and alanine are excellent nitrogen sources for growth, tyrptophane nad potassium nitrate are utilized slightly for growth and sodium nitrite is not utilized. Thiamine nad gibberellin are excellent growth substances for the fungal growth, and biotin, nicotinamide and indole acetic acid (IAA) are also effective. Rhizopus niveus grows better at rotatory culture than at stationary culture and earlier growth of the fungus increases remarkably at rotatory culture. Optimum pH than at pH3. Growth increases linerly with an increase of soluble starch content up to 100g per liter medium, but 5 grams of ammonium sulfate per liter is the optimum nitrogen concentration for growth, if Pfeffer's medium is employed. Amylase activities of Rhizopus at different cultural periods showed that the maximum amylase production takes place after the cell population has reached its peak in the culture. Dextrinogenic amylase production has reached maximum at stationary phase, and maximum saccharogenic maylase production takes place in the pahse of negative gorwth acceleration.

공단지역과 시골지역 주택 실내, 실외 및 개인의 이산화질소 노출평가 (Indoor, Outdoor, and Personal Exposure to Nitrogen Dioxide Comparing Industrial Complex Area with Country Area)

  • Indoor air quality can be affected by indoor sources, ventilation, decay, and outdoor levels. Various indoor and out-door combustion sources produce nitrogen dioxide ($NO_2$), which is a by-product of high temperature fossil fuel combustion. Especially, the presence of gas ranges and smoking have been identified as major factors contributing to indoor $NO_2$ exposures. In this study, we compared an industrial complex area with a country area by assessing the personal exposure to $NO_2$with measurements of indoor and outdoor $NO_2$ levels in residences and by house characteristics and questionnaire. Personal exposure concentrations were significantly correlated with indoor $NO_2$ concentrations of residences in both the industrial complex area and the country area with correlation coefficients of 0.561 and 0.664, respectively, compared to outdoors. Multiple regression analysis, indicated that indoor $NO_2$ levels in residences were only affected by outdoor levels (p = 0.000) in spite of higher indoor sources such as smoking. Therefore, it is suggested that outdoor air quality as well as indoor air quality should be considered in the reduction of the personal exposure to air pollutants.

Nitrogen Control in Corynebacterium glutamicum: Proteins, Mechanisms, Signals

  • In order to utilize different nitrogen sources and to survive in a situation of nitrogen limitation, microorganisms have developed sophisticated mechanisms to adapt their metabolism to a changing nitrogen supply. In this communication, the recent knowledge of nitrogen regulation in the amino acid producer Corynebacterium glutamicum is summarized. The core adaptations of C. glutamicum to nitrogen limitation on the level of transcription are controlled by the global regulator AmtR. Further components of the signal pathway are GlnK, a $P_{II}-type$ signal transduction protein, and GlnD. Mechanisms involved in nitrogen control in C. glutamicum regulating gene expression and protein activity are repression of transcription, protein-complex formation, protein modification by adenylylation, change of intracellular localization, and proteolysis.
