Physiological Activities of Ethanol Extracts from Different Parts of Allium hookeri (삼채부위별 에탄올 추출물의 생리활성탐색)

  • Biological compounds such as crude saponin and total polyphenol were determined in Allium hookeri roots and leaves. In addition, activities of DPPH radical scavenging activity, hydroxyl radical scavenging activity, ${\alpha}$-amylase inhibition activity and nitrite scavenging were detected in concentrates of 70% ethanol extracts of Allium hookeri roots and leaves. The crude saponin content of the roots and leaves were 4.28 mg/g, and 4.17 mg/g, respectively. Total polyphenol and total flavonoid content was higher in leaves than roots. Furthermore the DPPH radical scavenging activity and hydroxyl radical scavenging activities of leaves was higher than roots. For ${\alpha}$-glucosidase inhibitory activity, 70% ethanol extracts of Allium hookeri roots and leaves showed 60% activity inhibition at a 10 mg/mL concentration. Nitrite scavenging abilities under acidic conditions were most effective for Allium hookeri leaves extracts. These results suggested that Allium hookeri leaves might be used as the components of health functional food.

Functional Characteristics from the Barley Leaves and its antioxidant mixture - Study on the Nitrite Scavenging Effect - (맥엽 및 항산화 조성물의 생리 기능적 특성에 관한 연구 - 아질산염 소거능을 중심으로 -)

  • This study was conducted to estimate the biofunctional characteristics, namely nitrite scavenging effect of barley leaves (BL) and their mixture (AM) with other antioxidants. Aqueous or methanol extracts were obtained from BL and AM. Aqueous and methanol extracts from barley leaves had relatively higher nitrite scavenging effects and its activities and contents increased in a concentration-dependent manner. The activities and contents of methanol extracts obtained from BL and AM were higher than those of aqueous extracts. Especially, AM containing BL and other antioxidant mixture had the highest activities and contents increased in a concentration-dependent manner. BL or AM were added to macrophage cells of RAW 264.7. Survival rates of the cells treated with BL and AM were measured to be different. $IC_{50}$ value decreased in a concentration-dependent manner by the addition of aqueous or methanol extracts from BL and AM. Especially, methanol extracts of AM had the highest nitrite scavenging effects. Thus, BL and AM may have protective effect against carcinogen and immune ability against reactive molecules through NO (nitric oxide) signal pathway. From the above results, barley leaves appear to contain natural anticancer and immune-related agents and may have potentiality to be used as functional food ingredients.

Nitrite Scavenging Ability and SOD-like Activity of a Sterol Glucoside form Chrysanthemum coronarium L. var. spatiosum (쑥갓 스테롤배당체의 아질산염소거작용 및 SOD 유사활성)

  • From the total methanolic extract of Chrysanthemum coronarium L. var. spatiosum (Compositae), nitrite scavenging ability and superoxide dismutase (SOD)-like activity were analyzed as antioxidative characteristics. After successive partitioning with chloroform, n-butanol, and water, the chloroform fraction showed the most significant nitrite scavenging ability with an $IC_{50}$ value of 39 ppm compared with the values of vitamin C and chlorogenic acid, 15 ppm and 36 ppm, respectively. The active fraction was subjected to silica gel and Sephadex LH-20 column chromatography, and the compound was isolated and identified as ${\beta}-sitosterol-O-{\beta}-D-glucoside$ using $^{1}H-NMR$ and $^{13}C-NMR$ spectral data. The glucoside was further hydrolyzed and confirmed as a glycosylated ${\beta}-sitosterol$. The compound and its aglycone, ${\beta}-sitosterol$, showed different nitrite scavenging and SOD-like activity. The $IC_{50}$ value of nitrite scavenging ability of the compound was 335 ppm at pH 1.5, while that of its aglycone was 41 ppm. As for the SOD-like activity, the $EC_{50}$ values of the sterol and the glucoside were 1,291 ppm and >2,000 ppm, respectively, compared with those of vitamin C and chlorogenic acid, 38 ppm and 449 ppm, respectively.

Inhibitory Effect on Tyrosinase, ACE and Xanthine Oxidase, and Nitrite Scavenging Activities of Jubak (Alcohol filter cake) Extracts (주박 추출물의 아질산염 소거와 Tyrosinase, Xanthine Oxidase, ACE 저해 효과)

  • The purpose of this study was to investigate the tyrosinase, xanthine oxidase, angiotensin converting enzyme (ACE) inhibitory effects, and nitrite scavenging abilities of Jubak (alcohol filter cake, AFC) hot water and ethanol extracts. Tyrosinase inhibitory effects of Jubak hot water and ethanol extracts were above 85%. Nitrite scavenging ability was higher at pH 1.2 than at pH 4.2 and 6.0. Nitrite scavenging abilities of water and ethanol extracts were above 90% at pH 1.2. Xanthine oxidase inhibitory effect of Jubak hot water and ethanol extracts showed a lower, and the effect did not different by hot water and ethanol. ACE inhibitory effects of Jubak hot water and ethanol extracts were approximately 43~53% and 36~47%, respectively. Overall, our results indicate that Jubak hot water and ethanol extracts may have tyrosinase and ACE inhibitory effects and nitrite scavenging ability, but no xanthine oxidase inhibitory effect. Therefore, Jubak has potential as a cosmetic raw material. Additionally, Jubak could be used as a food additive.

Electron Donating Ability and Nitrite Scavenging Activity of Materials in a Traditional One-dish Meal (Bibimbab) (한국 전통 한 그릇 음식(비빔밥) 및 그 재료들의 항산화성과 아질산염 소거능)

  • The electron donating ability and nitrite scavenging activity were measured in ethanol extracts from the raw, simple cooked and seasoned materials of Bibimbab (mixed rice), a Korean traditional one-dish meal. The ethanol extracts of raw fragrant edible wild aster exhibited the highest electron donating ability $(80.28\%)$, as measured by the DPPH method. When Bibimbab was compared with Kimbab and hamburgers, which are popular food, it showed the highest antioxidative activity by the DPPH method. The nitrite scavenging ability was measured in an environment similar to in human digestive organs, i.e. pH $1.2\~6.0$. Generally, the antioxidative activity decreased in the sequence pH 1.2, 3.0 and then 6.0. The simple cooked materials, with the exception of fragrant edible wild aster, increased the antioxidative potential. The seasoned materials also had a tendency to increase the antioxidative potential. In conclusion, the ethanol extract of Bibimbab showed the highest nitrite scavenging activity. Therefore, the nutritional superiority of Bibimbab was confirmed through its electron donating ability, as measured by the DPPH method, and nitrite scavenging effect.

Antioxidant Activity and Nitrite Scavenging Ability of Olive Leaf (Olea europaea L.) Fractions (올리브 잎 분획물의 항산화기능과 아질산염 소거능력 평가)

  • In this study, the antioxidant activities and nitrite scavenging abilities of olive leaf fractions acquired from plants cultivated in Australia (Olea europaea L. var. Picual) and Spain (Olea europaea L. var. Hojiblanca) were evaluated. Oleuropein was found to be the major phenolic compound in the leaves, with the butanol fractions presenting the highest contents. Antioxidant activity was evaluated in terms of superoxide dismutase (SOD)-like activity, 1,1-diphenyl-2-picryl hydroxyl (DPPH) radical scavenging activity, and the inhibitory effect on the auto-oxidation rate of linoleic acid. The SOD-like activities of the olive leaf extracts ranged from 0 to 36.8%. DPPH radical scavenging activity was highest in the ethanol extract of the Australian cultivated olive leaves. Finally, the chloroform fractions of the extracts showed inhibitory effects on the auto-oxidation rate of linoleic acid as well as nitrite scavenging ability.

Prediction of the Optimum Conditions for Microwave-Assisted Extraction of the Total Phenolic Content and Antioxidative and Nitrite-scavenging Abilities of Grape Seed (포도씨의 총페놀 성분, 항산화능 및 아질산염소거능에 대한 마이크로웨이브 추출조건 예측)

  • Response surface methodology (RSM) was used for the microwave-assisted extraction (MAE) of the effective components of grape seed, such as its antioxidative and nitrite-scavenging abilities. Microwave power (2,450 MHz, 0-160W), ethanol concentration (0-100%), and MAE time (1-5 min) were used as independent variables (Xi) for the central composite design to yield 16 different MAE conditions. The optimum MAE conditions were predicted for the dependent variables of the extracts, such as the total phenolic content ($Y_1$) antioxidative ability ($Y_2$), and nitrite-scavenging ability ($Y_3$), depending on different microwave powers, ethanol concentrations, and MAE times. The determination coefficients ($R^2$) of the regression equations for the dependent variables ranged from 0.8024 to 0.9498. The maximal values of each dependent variable were predicted at different MAE conditions, as follows: 3.19% total phenolic content at 142.32W, 44.30% ethanol, and 4.36 min, and 1.22 antioxidative ability at 84.44W, 56.60% ethanol, and 3.28 min. More than 99.5% nitrite-scavenging ability was predicted at pH 1.2-3.0, 30.80-106.58W, 49.32-55.18% ethanol, and 3.72-4.58min, respectively. The results indicated that the total phenolic content and anti oxidative ability showed a higher correlation with each other in that they were more influenced by microwave power than by the other variables, while the nitrite-scavenging ability was largely influenced by the ethanol concentration.

Total Phenolic Contents and Antioxidant Activities of Undaria pinnatifida and Capsosiphon fulvescens (미역과 매생이의 총 페놀함량 및 항산화성)

  • This study compared the total phenolic contents and antioxidant activities of two seaweed cultivars, Undaria pinnatifida (UP) and Capsosiphon fulvescens (CF), subjected to different drying methods. UP and CF were dried under two different conditions: vacuum drying (VD) at $20^{\circ}C$ and hot-air drying (HD) at $60^{\circ}C$. After drying, the total phenolic content, DPPH, and nitrite scavenging activities of the water extracts were determined. Total phenolic contents were 101.94 mg/100 g for UP and 171.35 mg/100 g for CF upon VD-20, and these values were significantly decreased to 67.59 mg/100 g for UP and 141.48 mg/100 g for CF upon HD-60. UP upon VD-20 and HD-60 had 46.17% and 35.20% DPPH radical scavenging activity, whereas CF upon VD-20 and HD-60 scavenged 57.73% and 35.22%, respectively. UP upon VD-20 and HD-60 had 40.36% and 40.01% nitrite scavenging activity at pH 1.2, whereas CF upon VD-20 and HD-60 scavenged 72.35% and 55.24%, respectively. Nitrite scavenging activities of UP and CF were reduced at pH 3.0.

Antioxidant and Xanthine Oxidase Inhibition Activities of Cynomorium songaricum Extracts

  • In this study, we evaluated the antioxidant activities and xanthine oxidase inhibition effects of water and ethanol extracts of Cynomorium songaricum. The ethanol extract of C. songaricum (EE) contained more phenolic and flavonoid compounds than the water extract (WE). The antioxidant activities of the extracts were increased as the concentration of the extract increased. The WE has better effectiveness than the EE for DPPH free radical scavenging activity and nitrite scavenging ability. The nitrite scavenging abilities of WE were 90.02% ($EC_{50}$ 653.15 ${\mu}g$/mL) at conditions of pH 1.2 and 2,000 ${\mu}g$/mL, and 84.34% ($EC_{50}$ 817.17 ${\mu}g$/mL) at pH 3.0. The EE has more effective SOD-like activity and XO inhibition than WE. The SOD-like activity of EE was 81.47% at a concentration of 2,000 ${\mu}g$/mL, $EC_{50}$ was 951.70 ${\mu}g$/mL. The xanthine oxidase inhibition of the EE, with an $EC_{50}$of 112.47 ${\mu}g$/mL, is greater than that of ascorbic acid, which was 192.50 ${\mu}g$/mL (p<0.05). These results suggest that the C. songaricum is a potentially useful antioxidant source for the development of nutraceuticals and medicines.

Effective Components and Nitrite Scavenging Ability of Root and Leaves a Angelica gigas Nakai (당귀와 승검초의 기능성 성분과 아질산염 소거능)

  • This research was conducted to examine the usability of Dangui (Angelica gigas Nakai, Root) and Seungumcho (Angelica gigas Nakai, Leaf) as functional food in aspects of their functional components and nitrite scavenging ability. Analysis of proximate composition showed that Dangui contains more moisture, crude lipid and crude protein than Seungumcho. On the other hand, Seungumcho contains more than twice mont of crude ash and crude fiber than Dangui. Dangui showed higher contents in phosphorus, iron, magnesium, and Seungumcho showed higher contents in copper, calcium, sodium, potassium compare to each other. Substances such as calcium, magnesium and iron that showed high contents in Dangui and Seungumcho are the most important inorganic substances. Total dietary fiber(TDF) of Dangui, sum of IDF and SDF, was 24.2%, and Seungumcho showed 28.18%. vitamin C contents of a 29.690.33 mg/100g appeared only in Seunggumcho. Total phenol contents of Dangui was 0.100${\pm}$002% and that of Seungumcho 0.0900${\pm}$008%. Nnitrite Scavenging ability of both water soluble and methanol soluble extracts were more than 90% at pH 1.2, and it decreased as pH level adjusted to pH 4.2, pH and 6.0.
