Bioconversion of nitrogen oxides and reduction of ferric ions by probiotic lactic acid bacteria (프로바이오틱스 유산균에 의한 질소 산화물 전환 및 철 이온 환원활성)

  • Many lactic acid bacteria (LAB) have probiotic properties that exert various health benefits. In this study, the reduction potential of nitrogen oxide compounds and ferric ions by six LAB, including Lactobacillus kimchicus, L. lactis, L. casei, L. plantarum, L. rhamnosus GG, and Leuconostoc mesenteroides were evaluated. The L. kimchicus strain produced a substantial amount of nitrite reduced from nitrate added to the media, whereas the other five LAB strains did not. L. kimchicus also showed the most potent reducing activity of ferric to ferrous ions. However, the reduction potential of the autoclaved L. kimchicus was little pronounced. The scavenging activities of viable LAB or their cell lysates against different radicals were not consistent with the potency of the LAB's reducing ability. The present results indicate that L. kimchicus has a strong reduction potential for nitrogen oxides in viable status, and that this ability can be used as a probiotic property for various health benefits.

Application of Ganghwa Mugwort in Combination with Ascorbic Acid for the Reduction of Residual Nitrite in Pork Sausage during Refrigerated Storage

  • The application of ganghwa mugwort (GM), ascorbic acid (AC), and their combinations for reduction of residual nitrite contents was analyzed in pork sausages during storage of 28 d. Six treatments of pork sausages contained the following: Control (no antioxidant added), AC (0.05% AC), GM 0.1 (0.1% GM), GM 0.2 (0.2% GM), AC+GM 0.1 (0.05% AC + 0.1% GM) and AC+GM 0.2 (0.05% AC + 0.2% GM). Results showed that the mixture of 0.05% AC and 0.2% GM was most effective for reducing thiobarbituric acid reactive substances (TBARS) and residual nitrite contents than the control and GM added sausages alone (p<0.05). The color values of all treatments were significantly affected by adding GM (either alone or with AC). Additionally, the total color difference (${\Delta}E$) and hue angle ($H^{\circ}$) values of treatments added with GM were higher than those of the control as the amount of GM increased (p<0.05). However, there were no significant differences in the pH values between the control and all treatments during the storage period (p>0.05). Our results showed possible applications of antioxidant combination, for preventing the lipid oxidation and decreasing the residual nitrite levels of meat products.

Effect of $TO_3$ and $NO_2$ on Net Photosynthesis, Transpiration and Accumulation of Nitrite in Sunflower Leaves

  • Photosynthesis and transpiration rates were simultaneously measured in attached sunflower leaves(Helianthus annuusL. cv. Russian Mammoth) during exposure to $NO_2$ and $O_3$ to determine the effect of mixed gan on photosynthesis and the stomatal aperture. The application of $O_3$ alone reduced both the net photosynthetic and transpiration rates. An analysis of the $CO_2$ diffusive resistances indicated that the main cause affecting photosynthesis reduction during $O_3$ exposure was not the internal gas phase of the leaf $(rCO_2^{liq})$ but rather the liquid phase or mesophyll diffusive resistance $(rCO_2^{liq})$, suggesting that there is a very concomitant relation between photosynthetic reduction and $rCO_2^{liq}$. The application of NO2 alone caused a marked reduction of the net photosynthesis yet no significant reduction of transpiration, indicating that NO2 affects the $CO_2$ fixation processes with no inluence on the stomatal aperture. A greter reduction in the photosynthesis of sunflower plants was caused by the application of $NO_2$ alone as compared to a combination of $NO_2$ and $O_3$. $NO_2$ alone reduced the photosynthetic rate by 90%, whereas a mixture of NO2 and O3 reduced it by 50%.

  • During the 1970s, concern arose that cured meats contained high levels of residual nitrite and preformed nitrosamines. Therefore, the search for alternatives and alternative approaches to the use of nitrite have been still continued, however no complete alternative for nitrite has yet been identified. Recently, it was publicized in Korea that nitrite-containing meat products would be detrimental to health, about which consumers have been seriously concerned. Therefore, this study was carried out to inform the consumer of the safety status of nitrite and thereby to lead proper consumption of meat products. For assessing the safety of nitrite, data regarding the regulation for use, metabolism in human body, and dietary intake amounts of nitrite were collected and analyzed. The mean intake level of nitrite for Korean per capita was recently reported to be not more than 1% of ADI set by JECFA. On the contrary, a calculation indicated that the daily nitrite intake per capita from saliva by ingestion of vegetables in Korea would be about 300-fold higher than that from cured meats. In consideration of the low consumption amount of meat products per capita of Korean, that is, at least one fifth, compared to European and American, there is no particular reason to concern about the impairment of health by nitrite intake from meat products for Korean. However, any effort for the reduction of residual nitrite content in cured meats should be given with an idea to minimize the intake of nitrite even from the minor source.

Nitrate Reduction and Pigment Formation of Chinese-Style Sausage Mixes Caused by Micrococcaceae

  • This study investigated the nitrate reduction ability of Micrococcaceae on pigment formation in Chinese- style sausage. One hundred ppm sodium nitrite and 150 ppm sodium nitrate was added asepticly to ground pork which was then inoculated with $10^7CFU/g$ of either Micrococcus varians, Staphylococcus carnosus or Staphylococcus xylosus. All samples were cured at $20{^{\circ}C}$ or $30{^{\circ}C}$ for 3 days and then color, residue nitrite, nitrosyl pigment and residue nitrate were determined. The results indicated that samples inoculated with S. xylosus had higher a- and b- values due to nitrate reduction and pigment production after 3 days curing and these values were higher at the higher curing temperature. The nitrosyl pigment of the samples with S. xylosus had highest values after 3 days curing at both $20{^{\circ}C}$ and $30{^{\circ}C}$. However, sample inoculated with S. carnosus and S. xylosus had lower nitrate contents than the sample inoculated with M. varians. At $30{^{\circ}C}$ as well as S. carnosus and M. varians had a stronger decreasing in nitrate concentration during curing at $20{^{\circ}C}$. Moreover, samples inoculated with S. xylosus and S. carnosus had a higher residual nitrite content during curing at $20{^{\circ}C}$ or $30{^{\circ}C}$. In conclusion, two Staphylococci strains tested were most optimum starter cultures for improving pigment formation during Chinese-style sausage curing.

Effect of Electrochemical Redox Reaction on Growth and Metabolism of Saccharomyces cerevisiae as an Environmental Factor

  • The effect of an electrochemically generated oxidation-reduction potential and electric pulse on ethanol production and growth of Saccharomyces cerevisiae ATCC 26603 was experimented and compared with effects of electron mediators (neutral red, benzyl viologen, and thionine), chemical oxidants (hydrogen peroxide and hypochlorite), chemical reductants (sulfite and nitrite), oxygen, and hydrogen. The oxidation (anodic) and reduction (cathodic) potential and electric pulse activated ethanol production and growth, and changed the total soluble protein pattern of the test strain. Neutral red electrochemically reduced activated ethanol production and growth of the test strain, but benzyl viologen and thionine did not. Nitrite inhibited ethanol production but did not influence growth of the test strain. Hydrogen peroxide, hypochlorite, and sulfite did not influence ethanol production and growth of the test strain. Hydrogen and oxygen also did not influence the growth and ethanol production. It shows that the test strain may perceive electrochemically generated oxidation-reduction potential and electric pulse as an environmental factor.

Recycled packed-bed reactor for efficient denitrification (재순환 충전탑식 반응기를 이용한 효율적 탈질)

  • Recycled packed-bed reactor emploring immobilized microorganism was suggested in this paper for efficient denitrification. In the batch reactor, the effects of initial oxidation-reduction potential and nitrate concentration on denitrification were investigated. As the initial oxidation-reduction potential was decresed to -70 mV from +40 mV, the removal rate of nitrate was increased to 3.33 from 1.25 m9 NO$_3$$\^$-/-N/min under the experimental conditions. As the initial nitrate-N concentraion was increased to 200 mg/l, the removal rate of nitrate was proportional to the concentration of nitrate. When the concentration of nitrate-N was 400 mg/min, nitrite was detected, and when the initial nitrate-N concentration was reached at 1,000 mg/l, it took longer time for the complete nitrate removal. In order to decrease the initial oxidation-reduction potential and the nitrate-N concentration in the feed stream, the effluent was recycled to the influent stream in the packed-bed reactor. In the case of recycling, the initial oxidation-reduction potential was decreased to 30 mV from 150 mV, and the initial nitrate concentration could be decreased to 85 from 120 mg NO$_3$$\^$-/-N/l. As the result of recycling, the removal rate of nitrate was increased to 91.7% from 49.2%.

현장 Single Well Push-Pull 실험을 통한 탈질산화반응 각 단계의 반응속도 측정

  • Quantifying rates of microbial processes under subsurface conditions is difficult, and is most commonly approximated by laboratory studies using aquifer materials. In this study a single-well, 'push-pull' test method is adapted for the in situ determination of denitrification rates in groundwater aquifers. The rates of stepwise reduction of nitrate to nitrite, nitrous oxide, and molecular nitrogen were determined by performing a series of push-pull tests at an experimental well field of Korea University. A single Transport Test, one Biostimulation Test, and four Activity Tests were conducted for this study. Transport tests are conducted to evaluate the mobility of solutes used in subsequent tests. These included bromide (a conservative tracer), fumarate (a carbon and/or source), and nitrate (an electron acceptor). At this site, extraction phase breakthrough curves for all solutes were similar, indicating apparent conservative transport of the solutes prior to biostimulation. Biostimulation tests were conducted to stimulate the activity of indigenous heterotrophic denitrifyinc microorganisms. Biostimulation was detected by the simultaneous production of carbon dioxide and nitrite after each injection. Activity tests were conducted to quantify rates of nitrate, nitrite, and nitrous oxide reduction. Estimated zero-order degradation rates decreased in the order nitrate '||'&'||'gt; nitrite '||'&'||'gt; nitrous oxide. The series of push-pull tests developed and field tested in this study should prove useful for conducting rapid, low-cost feasibi1ity assessments for in situ denitrification in nitrate-contaminated aquifers.

Influence of Ammonia and Dissolved Oxygen Concentrations on Nitrite Accumulation in a MBR (MBR 반응조에서 아질산염 축적에 미치는 암모니아와 용존산소 농도의 영향 연구)

  • The complete oxidation of ammonia to nitrate is a distinctive two-step process divided into the oxidation of ammonia to nitrite(nitritation) by Nitrosomonas and the oxidation of nitrite to nitrate(nitratation) by Nitrobacter. The nitrogen removal via nitrite accumulation offers several advantages such as saving costs for aeration, saving carbon source and finally reduction of sludge discharge. In this work a suspended bioreactor coupled with membrane filtration(MBR) was used to find the process conditions of nitrite build-up. The MBR enables to reach sufficient nitrifying bacteria in the bioreactor, although the autotrophic bacteria can be easily washed out due to their lower growth rate. The dissolved oxygen concentration $c'_{O2}$ and ammonia concentration $c_{NH3}$ in the reactor were varied and investigated as parameters for nitrite accumulation. As a result the higher ammonia concentration in the reactor is very effective for starting nitrite build-up and the effect was strengthened in combination with lower dissolved oxygen concentration. With lower $c'_{O2}<0.3$ $mgL^{-1}$ $O_2$ and high $c_{NH3}=6.3\sim14.9$ $mgL^{-1}$ $NH_3N$ the 74% of the nitrite accumulation was achieved. Specially, it was found that the nitrite accumulation could occur not only in biofilm reactor as many references showed but also in the membrane bioreactor carried out in this study.
